Detection of Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp. and Staphylococcus aureus using duplex real-time PCR assay with melting curve analysis on fresh lettuce

  • In this study, two duplex real-time PCR approach with melting curve analysis is presented for the detection of Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp. and Staphylococcus aureus, which are important food-borne bacterial pathogens usually present in fresh and/or minimally processed vegetables. Reaction conditions were adjusted for the simultaneous amplification and detection of specific fragments in the ${\beta}$-glucuronidase (uidA, E. coli), thermonuclease (nuc, S. aureus), hemolycin (hly, L. monocytogenes) and tetrathionate reductase (ttr, Salmonella spp.) genes. Melting curve analysis using a SYBR Green I real-time PCR approach showed characteristic $T_m$ values demonstrating the specific and efficient amplification of the four pathogens; $80.6{\pm}0.9^{\circ}C$, $86.9{\pm}0.5^{\circ}C$, $80.4{\pm}0.6^{\circ}C$ and $88.1{\pm}0.11^{\circ}C$ for S. aureus, E. coli O157:H7, L. monocytogenes and Salmonella spp., respectively. For all the pathogens, the two duplex, real-time PCR was equally sensitive to uniplex real-time PCR, using same amounts of purified DNA, and allowed detection of 10 genome equivalents. When our established duplex real-time PCR assay was applied to artificially inoculated fresh lettuce, the detection limit was $10^3$ CFU/g for each of these pathogens without enrichment. The results from this study showed that the developed duplex real-time PCR with melting curve analysis is promising as a rapid and cost-effective test method for improving food safety.

  • The microhabitat of Iksookimia hugowolfeldi was surveyed in two sites, Geogeum Island and Jangheung. I. hugowolfeldi in Geogeum Island inhabits mainly pool. This water system consists of sand and cobble and has a shallow water (0.1~0.8 m depth) with slow currents (0.1 m/s). 0+ years old inhabits pool consist of sand and lives in a less than 0.3 m depth. I. hugowolfeldi in Jangheung population inhabits mainly pool consists of sand and cobble and lives in a shallow water (0.1~0.8 m depth) with slow currents (0.1 m/s) compared to the stream of Geogeum Island. With some different fish species, sympatric species collected at Geogeum Island and Jangheung population are 7 species in 4 family identically in the two sites.

Performance of UASB Reactor Treating Leachate from Acidogenic Fermentation of food Waste (음식물쓰레기의 산발효 침출액을 처리하는 UASB 반응조의 거동)

  • Leachate from acidogenic fermentation of food waste was effectively treated by the UASB reactor at $37^{\circ}C$. The efficiency of COD removal was consistently over 96% up to the loading rates of $15.8g\;COD/{\ell}{\cdot}d$. The methane production rate increased to $5.5{\ell}/{\ell}{\cdot}d$. Of all the COD removed, 92% was converted to methane and the rest presumably to biomass. At loading rates over $18.7g\;COD/{\ell}{\cdot}d$, the efficiency of COD removal decreased due to the sludge flotation and washout in the reactor, which resulted from short HRT of less than 10.6 hr. The SMA(specific methanogenic activity) analysis showed that the VFA-degrading activity of granule was the highest for butyrate, and the lowest for propionate. This result was consistent with the observation that the residual propionate concentration was the highest among the VFAs in the effluent. Typical granules were found to be mainly composed of microcolonies of Methanosaeta. Though the original seed sludge contained 64.3% of particles smaller than 1.4mm, the sludge particles had been growing during the fermentation, and at the final step of this study, 75.1% of the particles were found to be larger than 1.4 mm in the UASB reactor.

  • Thermophilic $H_2$ was produced for 1 year using a bench-scale continuous stirred tank reactor(CSTR). The CSTR was inoculated with anaerobically digested sludge after heat treatment and fed with a glucose-based medium. The reactor showed relatively short start-up period(30 days) and high maximal $H_2$ yield(2.4 mol $H_2/mol$ glucose). Keeping pH 5.0 or less suppressed methanogenic activity. Bacteria affiliated with Thermoanaerobacterium thermosaccharolyticum kept being dominant from approximately 40 days as determined by DGGE. Environmental perturbation(pH or temperature) caused the decrease of biomass concentration in the reactor and the instability of reactor performance, $H_2$ production rate and $H_2$ yield. The unstable performance was accompanied with high concentration of lactate in the effluent. Taken together, the poor recovery of CSTR after perturbations could be partly explained by low biomass concentration and/or metabolic shift of the major population in the CSTR.

Characteristics and Toxicity Sensitivity of Korean Dominant Species Daphnia galeata for Ecotoxicity Testing: Comparative Study with Daphnia magna (생태독성실험을 위한 한국 우점종 유리물벼룩(Daphnia galeata)의 특성 및 독성민감도: 큰물벼룩(Daphnia magna)과의 비교연구)

  • Water flea is a widely used test species in the aquatic ecotoxicity test. In Korea, D. magna is currently used as a standard test species, however that do not inhabit in the Korean aquatic ecosystem. In this study, Daphnia galeata, which is a dominant species in the Korean aquatic ecosystem, was collected from domestic lake and investigated to suggest the D. galeata for ecotoxicity assessment in Korea. We investigated the characteristics, life span, and toxicity sensitivity compared with D. magna. The life span test of D. galeata was performed in this study, and then the results were compared to the sensitivity with D. magna to confirm the applicability for ecotoxicity assessment. The 48h-L(E)C50 values for seven heavy metals (As, Cd, Cr, Cu, Hg, Ni, and Zn) of D. galeata and D. magna were collected and analyzed. As a results, shorter lifetime, less reproduction, smaller body size of D. galeata were observed compared with D. magna. D. galeata was similar or more sensitive than D. magna for seven heavy metals. Therefore, we propose that D. galeata is a suitable test species for ecotoxicity testing in Korea.

Study on the Performances and Microbial Community in the Biofilm Process for Treating Nonpoint Source Pollutants (비점오염물질 처리를 위한 생물막 공정의 운전 및 미생물 군집의 특성)

  • In this study, biofilm process was introduced for treating nonpoint source pollutants. The ceramic media were provided for biofilm growth in the reactors. The packing ratio of ceramic media was 5% and 15(v/v)%, respectively. Thereafter, the reactors were operated intermittently with the different interevent periods such as 0, 5, 10 and 15 days, respectively. The removal efficiencies of COD and NH$_4{^+}$-N were investigated at the different operating conditions such as media packing ratio, temperature, and interevent period. Additionally, Polymerase chain reaction(PCR)-denaturing gel gradient electrophoresis(DGGE) and INT-dehydrogenase activity(DHA) test were conducted to observe the microbial community and activity in the biofilm. Consequently, the interevent period seemed to have no significant influence on the COD removal efficiency. COD was removed within 6$\sim$8 hours at 25$^{\circ}C$ and about 15 hours at 10$^{\circ}C$. DGGE profiles showed that the initial species of microorganisms were changed from seeded activated sludge into the microorganisms detected in sediments. INT-DHA test also showed that the activities of microorgnaisms were not decreased even in the 15 days of interevent period.

Degradation Behavior of Endocrine Disrupter Bisphenol-A in the Lake and Stream (호소 및 하천에서 환경호르몬 Bisphenol-A의 분해거동에 관한 연구)

  • Among the biodegradability tests, TOC-HANDAI and OECD method were utilized to examine the degree of biodegradation of endocrine disruptors, Bisphenol A (BPA) and Nonylphenol. Both methods used natural water microcosms and measured their biodegrada-bilities of BPA and Nonylphenol, in terms of TOC or DOC degradation with time for 28 days. Biodegradabilities for BPA, 73-78% with TOC-HANDAI method and 77-81% with OECD method were obtained respectively at the end of experiment. There was no difference in BPA degradation between two methods. BPA degradation was described by two distinct first order decay rates (k$_1$ and k$_2$) which could be separated by a simple visual fitting. Most of the initial decay reaction accelerated within 1-7 days with k$_1$of 0.24-0.34 $day^{-1}$. And the following another long term first order decay coefficient (k$_2$) showed 0.02-0.05 day$^{-1}$ with much flat slope. About 20-25% of initial BPA remained at the end of experiment. It suggests that the remaining TOC components in BPA biodegradation considered to be refractory metabolites of BPA. Nonylphenol at each sampling point was appeared to be mineralized 20-48% of initial TOC concentration. Consequently Nonylphenol seems more recalcitrant against biodegradation. BPA was not detected in the detection limit of ppb in the watershed of Daechung reservoir and Kum river. However 25 ppb concentration of BPA was detected at the influent of industrial wastewater treatment plant in Taejon.

Autotrophic Perchlorate-Removal Using Zero-Valent Iron and Activated Sludge: Batch Test (영가철과 활성슬러지를 이용한 독립영양방식의 퍼클로레이트 제거: 회분배양연구)

  • Perchlorate ($ClO_4^-$) is a contaminant found in surface water and soil/ground water. Autotrophic perchlorate-reducing bacteria (PRB) use hydrogen gas ($H_2$) as an electron donor to remove perchlorate. Since iron corrosion can produce $H_2$, feasibility of autotrophic perchlorate-removal using zero-valent iron (ZVI) was examined in this study using activated sludge that is easily available from a wastewater treatment plant. Batch test showed that activated sludge microorganisms could successfully degrade perchlorate in the presence of ZVI. The perchlorate biodegradation was confirmed by molar yield of $Cl^-$ as perchlorate was degraded. Scanning electron microscope revealed that rod-shaped microorganisms on the surface of iron particles used for the autotrophic perchlorate-removal, suggesting that iron particles could serve as supporting media for the formation of biofilm as well. DGGE analyses revealed that microbial profile of the inoculum (activated sludge) was different from that of biofilm sample obtained from the ZVI-added enrichment culture used for $ClO_4^-$-degradation. A major band of the biofilm sample was most closely related to the class Clostridia.

Seed Production and Cultivation of Ecklonia stolonifera Okamura, Phaeophyta (갈조 곰피(Ecklonia stolonifera Okamura)의 종묘생산과 양성)

  • The seed production of Ecklonia stolonifera Okamura was studied under laboratory conditions through the embryonic sporophyte stage and the field cultivation was conducted in eastern coast of Korea. The germination of zoospores occurred within 3 days and the growth of gametophytes was most rapid at $25^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$. Sporophyte growth was highest at $20^{\circ}C$ and $20\;{\mu}mol{\cdot}m^{-2}s^{-1}$ and lowest at $25^{\circ}C$ and $80\;{\mu}mol{\cdot}m^{-2}s^{-1}$. In the nursery culture of E. stolonifera lasting for 2 weeks in January, the initial blade length of E. stolonifera (about $500\;{\mu}m$) grew to $526.3{\pm}176.0\;{\mu}m$ at water temperature of $12.05^{\circ}C$. The blade length and width reached their maxima in July, after which the ends of blade and stem began to degrade with the increase in water temperature. The degraded end of the blade started to regenerate in October, when water temperature began to decline. This species can be considered a potential candidate for aquaculture, increasing in the availability of raw material and aiding in recovery of seaweed bed.

Analysis of Archaeal Community in Autotrophic Perchlorate-degrading Enrichment Culture (독립영양 방식으로 퍼클로레이트를 분해하는 농화배양 내 고세균 군집 분석)

  • Perchlorate ($ClO_4^-$) is an emerging contaminant detected in soil, groundwater, and surface water. Previous study revealed bacterial community in the enrichment culture tdegraded perchlorate using elemental sulfur as an electron donor. Quantitative and qualitative molecular methods were employed in this study to investigate archaeal community in the enrichment culture. Real-time qPCR showed that archaeal 16S rRNA gene copy number in the culture was about 1.5% of bacterial 16S rRNA gene copy number. This suggested that less archaea were adapted to the environment of the enrichment culture and bacteria were dominant. DGGE banding pattern revealed that archaeal community profile of the enrichment culture was different from that of the activated sludge used as an inoculum for the enrichment culture. The most dominant DGGE band of the enrichment culture was affiliated with Methanococci. Further research is necessary to investigate metabolic role of the dominant archaeal population to better understand microbial community in the perchlorate-reducing enrichment culture.
