The A, C, F colonies of Apis mellifera ligustica Spin. and D, V colony of Apis mellifera caucasia Gorb. bees were collected from 2005-2007. Consequently, inbred lines were derived from the bees of original colonies by matting in the isolated island with due regard for pure breeding. This project helps in the selection of colonies with higher production capacity, aiming to improve honey and royal jelly production and breeding programs. Twenty-three standard morphological traits of honeybee were evaluated, and samples were compared with the data of the two original subspecies. The result suggested that 8 traits partly preserved in bees of inbred lines, and the bees from A. m. ligustica preserved more traits than bees from A. m. caucasia. Among the studied inbred lines, the F line is distinguished by an increase in leg parameters, considered as a favorable phenotypic trait of inbred lines. Importantly, bred of beelines in the same area can be classified as remote and isolated areas. Therefore, we observed differences of inbred lines with the origin subspecies in description acquired with morphometric characteristics as a result of adaptation, breeding, purebred individual lines used as an important resource for breeding novel cross-breeding colonies.
농촌진흥청 국립축산과학원(원장 라승용)은 지난 15년간의 재래닭 수집과 순계 복원, 연구 및 개량을 통해 '우리맛닭'을 탄생시켰다. 재래닭 복원 프로젝트는 1994년부터 1997년까지 4년간 대한양계협회에서 '재래닭육용화 사업'의 일환으로 서울대학교 오봉국 박사(본회 고문)의 진두지휘 하에 대학교 수 16명, 연구관 8명 등 연인원 84명과 9억2천만원의 예산이 투입되어 진행됐으며, 11개의 과제를 수행해 마침내 재래닭 순계 복원에 성공하였다. 연구진은 이후 연구 결과물을 축산과학원으로 이관하여 후속연구를 지속토록 하였으며, 축산과학원은 이로부터 우수한 실용계를 생산하는 연구에 착수하게 됐다. 이러한 과정을 거쳐, 경제적으로 불리한 기존 재래닭의 문제점을 보완하여 산란성과 산육성이 우수한 닭으로 개발한 것이 바로 '우리맛닭'이다. '우리맛닭'은 축산과학원이 보유한 9개 순계 모군으로부터 성장이 빠른 부계통과 맛이 좋고 알 생산이 많은 모계통의 교잡을 통해 맛이 좋고 성장이 빠른 실용계로 생산된다. '우리맛닭'은 콜라겐 함량이 높아 탄력감과 쫄깃한 느낌의 육질을 지니고 있으며, 이를 접하는 소비자들의 반응도 좋게 나타나 그 가능성이 긍정적으로 점쳐지고 있다.
This study was conducted to develop herbicide-resistance domestic maize plants by introducing the CP4 5-enol-pyruvylshikimate-3-phosphate synthase (CP4 EPSPS) gene using Agrobacterium tumefaciens-mediated immature embryo transformation. Immature embryos of five genotypes (HW1, KL103, HW3, HW4, HW7) were co-cultivated with strains Agrobacterium tumefaciens (strain C58C1) containing the binary vector (pCAMBIA2300) carrying Ubiquitin promoter-CP4 EPSPS gene and Cauliflower mosaic virus 35S (CaMV35S) promoter-nptll gene conferring resistance to paromomycin as a selective agent. The presence and expression of CP4 EPSPS transgene were confirmed by PCR, RT-PCR and Northern blot analysis, respectively. Also, the resistance to glyphosate in the transgenic maize ($T_1$) was analyzed by shikimate accumulation assay. The frequency (%) of paromomycin-resistance callus was 0.37, 0.03, 2.20, 2.37, and 0.81% in pure lines HW1, KL103, HW3, HW4 and HW7, respectively. EPSP transgene sequences were amplified in putative transgenic plants that regenerated from paromomycin-resistance calli of two inbred lines (HW3, HW4). Of them, RT-PCR and Northern blot analyses revealed that the transgene was only expressed in two transgenic events (M266, M104) of HW4 inbred line, and a mild glyphosate resistance of transgenic event (M266) was confirmed by the lower shikimate accumulation in leaf segments. These results demonstrate that transgenic maize with herbicide-resistance traits in Korean genotype can be genetically obtained.
This study evaluated disease resistance responses of pure lines of tomato plants at various temperature conditions against Ralstonia solanacearum strains isolated from Korea. Evaluation of six tomato lines with various strains of R. solanacearum showed that many strains can infect the resistant lines of tomato plants previously known as highly tolerant to bacterial wilt. One of the most virulent strains, SL341 (race 1 and biovar 4) caused severe infection on all six tomato lines, irrespective of temperature. In contrast, a moderately virulent strain SL1944 (race 1, biovar 4) showed the remarkable difference in disease progress on some resistant lines dependent on temperature. Moneymaker and Bonny Best were susceptible to SL1944 at all tested conditions with different temperature. However, tomato lines, such as Hawaii 7998, Hawaii 7996, Bblocking which were previously known as highly tolerant lines, were severely infected by SL1944 at relatively higher temperature ($35^{\circ}C$ for 14 hr light and $28^{\circ}C$ for 10 hr dark cycle). The disease progress at high temperature was much faster than those at low temprature on the same tomato line and those on Moneymaker and Bonny Best at the same high temprature. This result suggested that R. solanacearum strains isolated in Korea were highly virulent to bacterial wilt resistant tomato lines and some strains may cause severe infection on those plants at higher temperature.
To estimate the genetic characteristics and cumulative power of discrimination (CPD) within Korean native commercial chicken, we used a total of 395 genomic DNAs from six breeds population (Korean Native Red chicken: R, Korean Native Yellow chicken: Y, Korean native Commercial Chicken: C, Ogal chicken: S, Hy-Line Brown: H, White Leghorn: W). Genetic diversity indices including mean allele number among loci, unbiased heterozygosity ($h_i$) within locus, effective number of alleles ($N_e$) and polymorphism information content (PIC) as well as the unbiased average heterozygosity (H) among loci in the populations were calculated using the generated allele frequencies by each marker. Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The nearest distance (0.119) was observed between the C and Y strains. The generated unbiased average heterozygosity among loci in each population was integrated to the global formula of CPD and the result demonstrated that the CPD within the six chicken populations was 99.461%.
