Identification of host plant species of Balanophora fungosa var. indica from Phnom Bokor National Park of Cambodia using DNA barcoding technique (캄보디아 프놈보콜국립공원의 Balanophora fungosa var. indica의 숙주식물에 대한 DNA barcoding 기법을 통한 동정)

  • During the floristic survey on Phnom Bokor National Park, Kampot, Cambodia, we encountered Balanophora fungosa var. indica, which is a tropical holoparasitic plant. To identify its host species, we collected host roots and trees nearby and tried to identify them using DNA barcoding approach. We applied plastid rbcL and matK gene regions as DNA barcode markers, and successfully amplified and sequenced the markers from 15 host roots and seven tree samples. Obtained host root sequences were identified as Primulaceae, Celastraceae, Myrtaceae, and Oleaceae, while trees nearby are Oleaceae, Myrtaceae, Sapindaceae, Rosaceae, Clusiaceae, Ericaceae, and Lauraceae. At genus level, host species are identified as Myrsine, Euonymus, Syzygium, and Olea, but failed in species discrimination. Myrsine (Primulaceae) and Olea (Oleaceae) are reported here as host species of B. fungosa var. indica for the first time. Further sampling and comparative work, and DNA barcoding will help recognize the biodiversity of the area and host species of Balanophora, together with their evolution.

Geographical Patterns and Biodiversity of Endophytic Fungi Isolated from Scirpus L. s.l. in Korea (고랭이류에서 분리한 내생균의 생물다양성 및 지리적 양상)

  • We investigated the biodiversity of endophytic fungi of Bolboschoenus planiculmis and Schoenoplectus triqueter in Korea. Twenty five of these host plants were collected from six sampling sites. Internal transcribed spacer analysis identified a total of eighty five isolates from the collected plants. The isolates comprised nineteen taxa; most of the isolates (92%) belonged to class Dothideomycetes, followed by Sordariomyetes (4.8%), Agaricomycetes (1.6%), and Eurotiomycetes (0.8%). The most prevalent fungus was Cladosporium perangustum in B. planiculmis and Macrospora scirpicola in S. triqueter. There are very few studies of endophytic fungi in halophytes and these have reported biodiversity patterns different from those of previous studies in Korea. Our results should provide useful information for future research in Korea.

Purification and Amplification of Garlic Latent Virus (마늘 잠복 바이러스의 순수분리 및 증식)

  • 한국 마늘에 감염되어 있는 주된 바이러스 중의 하나로 알려진 마늘 잠복 바이러스 (GMV)의 분자 구조와 병 발생 메카니즘을 이해하기 위하여, 국부 감염 숙주 식물인 Vicia faba에 연속적으로 감염시킴으로써 마늘 잠복 바이러스를 정제하였고, GMV에 대한 전신 감염성 숙주 식물로 생각되는 leek에서 GLV를 대량으로 증식시켰다. 투과전자현미경을 이용하여 바이러스 입자를 관찰한 결과, 마늘 바이러스들의 입자의 길이는 200-2000 nm의 분포를 보였으나 입자의 대부분은 600-900 nm의 범위 안에 존재하였다. 반면 순수 분리된 GLV 입자는 평균 690 nm의 길이를 보여주었고, 유연한 실 모양이었다. SDS-PAGE 분석으로 혼합 감염된 마늘 잎으로부터 분리된 마늘 바이러스의 구조 단백질은 분자량 24,500-38,000 Da의 분포를 나타내었으나, GLV 껍질 단백질의 분자량은 34,000 Da으로 나타났다.

Isolation of N2-fixing Microorganism from the Root of Non-leguminous Crops (수종(數種)의 비두과(非豆科) 식물(植物)로 부터의 질소고정균(窒素固定菌)의 분리(分離)에 관(關)한 연구(硏究))

  • A number of nitrogen-fixing microorganisms were isolated from roots of non-legumious crops and evaluated for their nitrogen-fixing ability on their host crops. The results are summarized as follows: 1. Among nitrogen-fixing isolates obtained from 9 different crops, several isolates showed relatively higher level of acetylene reduction activity on Spinach, Chinese cabbage, Cucumber, mustard and Egg plant. 2. When the isolates were inoculated into 4 host crops, a number of isolates were found to grow well on Spinach, Chinese cabbage and Cucumber, but particularly well on the former. 3. Results of this study suggest that nitrogen-fixing a symbiotic microorganisms are also present and able to grow well on roots of non-legumious crops, and stimulate plant growth by promoting growth and differentiation of roots.

  • A series of experiments was planned for practical application of rhizobia in grass lands in Korea. This is the report for the studies mainly on the isolation and characterization of alfalfa nodule bacteria, and on the assessment of their nodulation abilities and nitrogen fixation capacities. 1. Total number of 47 strains was isolated from nodules which were taken from alfalfa grown in Daekwanyong, Cheju and other places. 2. Morphological and cultural characteristics of the strains were studied, and attempts. were also made to investigate their antigenic properties and to demonstrate lysogenic strains. The results were; i) the isolates varied in their cultural characteristics on yeast mannitol broth and agar, and in degree of congo red absorption; ii) similarities in their antigenic prorerties were found between the strains: SU 47/M-11, M-13/M-15, and M-3/M-5; iii) no lysogeny was found in the strains. 3. Plant infection test by test tube method in light room were carried out to elucidate the ability of the strains to nodulate Luna alfalfa and of the capacity of such nodules to fix atmospheric nitrogen. The isolates were grouped info non-invasive, ineffective, or effective to the legumes. Those strains which produced effective nodules, supporting similar/higher level of growth as nitrate control were: M-8, 9, 14, 20, 21, 25 and 34.

Washing Effect of Marketing Mungbean Sprouts on Morphological Characters and their Color (출하용 숙주나물의 세척 유무에 따른 형태 및 색상 변화)

  • Traditionally mungbean(Vigna radiata L.) sprouts has been eaten soon after washing. The study was carried out to measure the effect of washing before packaging on morphological characters and color of mungbean sprouts. The seeds of cv. Zhong Lu 1 were soaked in 50 ppm BA solution immediately before 4 hour aeration and then cultured for 6 days. The sprouts were washed immediately before packaged with PE envelops or not, and then stored 5 days at $8^{\circ}C$. Their morphological characters, fresh and dry weights, and colors were measured everyday. Compared to washed, nonwashed sprouts had more lateral roots although the two sprouts did nearly same in hypocotyl and root lengths, hypocotyl diameter, fresh and dry weights. Non-washed sprouts, moreover, showed higher brightness in hypocotyl and root and cutting resistance in hypocotyl although there were not significant differences in color a and b, meaning that the latter ones were more rapidly changed during their storage. In non-washed sprouts, number of lateral roots, hypocotyl length and diameter, total fresh and dry weights were nearly same up to 3 days and afterward were declined. Brightness and color b of hypocotyl were decreased with increased storage period although cutting resistance of hypocotyl was since 3 days after storage.

Diversity of Arbuscular Mycorrhizal Fungi of Woody Plants in Mt. Munan (문안산의 목본식물과 공생하는 수지상균근균의 다양성)

  • This study was conducted to reveal the diversity of arbuscular mycorrhizal fungi (AMF) in Mt. Munan, based on the morphological and molecular characters. Soil and root samples were collected from the rhizosphere of host plants including Lindera obtusiloba, Stephanandra incisa, Styrax obassis and Symplocos sawafutagi and AMF were trap-cultured with Sorghum bicolor as a host plant in a greenhouse. The spores were extracted from the cultured soils and five species were identified using morphological and molecular characteristics; Acaulospora longula, A. mellea, Ambispora leptoticha, Gigaspora margarita and Paraglomus occultum. The distribution of AMF showed different trends according to host plants. The dominant AMF species were A. longula in L. obtusiloba, A. leptoticha in S. incisa, S. obassis and S. sawafutagi.

Population Changes of Arbuscular Mycorrhizal Spores in the Different Soil Environments (환경변화에 따른 내생균근 포자증식의 변화)

  • The association of soil environments and sporulation of arbuscular mycorrhizae was investigated using pot cultures. Increased arbuscular mycorrhizal spores as well as the better growth of the host plants were observed when sorghum (Sorghum bicolor) was treated with different soil conditions using several fertilizers with different concentrations. For to five fold increase of sporulation of arbuscular mycorrhizae was noticed depending on the mycorrhizal species. Although there were some differences between the four arbuscular mycorrhizae species and the conditions of soil environments, maximum populations of spores were reached at about 30-40 days after cultivation. The populations of four arbuscular mycorrhizal species was individually fluctuated. Also, the growth rate of host plants were different from the fertilizers over 2-3 times, but the increase of spores were not influenced by it.

Isolation and Structure Determination of Phytotoxins from a Phytopathogenic Fungus Bipolaris sorokiniana (식물병원균 Bipolaris sorokiniana로부터 제초활성 물질의 분리 및 구조결정)

  • Two phytotoxic compounds, 3-methoxybenzoic acid (MBA) and 3-hydroxy benzoic acid methyl ester (HBAME), were purified and structurally characterized by instrumental analyses from a culture of a phytopathogenic fungus, Bipolaris sorokiniana. During the isolation procedure, the toxic components were monitored by the assay using rice (Echinochloa crusgalli) and Italian ryegrass (Lolium multiflorum). These compounds had a molecular formula of $C_8H_8O_3$ and inhibited the root growth of the plant seedlings over 90% at a level of 1000 ppm. This is the first report of herbicidal activity of MBA and HBAME purified from B. sorokiniana.
