Physicochemical and Sensory Properties of Red Pepper Extract treated with Enzyme Complex (복합효소를 이용한 고추 추출액의 이화학적 및 관능적 특성)

  • The ground red pepper puree was treated with cellulase (C treatment), pectinase (P treatment), amylase (A treatment) and/or enzyme complex (CP, CA, PA and CPA treatment) for 2~8 hours to improve the yield and bioactivity of extracts. And physicochemical and sensory properties of red pepper extracts were evaluated. The extraction yield of the control was 38.84%, which was lower than those of the enzyme treatments. And extraction yields of enzyme complex treatments were higher than that of single enzyme treatments. Especially, extraction yield was increased to 74.37% by cellulase + pectinase + amylase complex treatment (CPA treatment). The soluble solid and reducing sugar contents were higher in the extracts treated with enzymes compared with the control. CA and CPA treatment showed the highest soluble solid and reducing sugar contents. No significant changes in lightness, redness and yellowness of the control and the samples by enzyme treatments were observed during 2~8 hours experiments. The sensory evaluation results revealed that panelists preferred the extracts with enzyme treatments to the control. Therefore, enzyme treatment for red pepper extracts is a good method to improve the yield and sensory properties.

Extraction and Purification of Agar from Gelidium amansii (우뭇가사리로부터 한천의 추출 및 정제)

  • The effect of different treatments on the quality and yield of purified agar produced from Gelidium amansii has been studied, and the extraction condition ol agar produced from G. amansii has been examined. The contents of ash, sulfate in agar produced from g. amansii collected from different places were $2.63\~2.92\%\;and\;1.38\~1.78\%$, respectively. Yields and gel strength of agar produced from G. amansii collected from different places were $31.6\~46.8\%\;and\;496\~887g/cm^2$, respectively. It was effective to extract agar at $120^{\circ}C\;for\;2\~3hrs$. Agar was purified by D.W. washing, EDTA washing, chitosan treatment, CPC treatment, PEG treatment, ethanol precipitation, acetone precipitation and propanol precipitation. The mineral contents of agar produced from Gelidium amansii were Na (2934ppm), Ca (2472ppm), Mg (2259ppm), K (2527ppm), P1(81.1ppm), Fe (66.4ppm), Al (71.7ppm), Zn (29.7ppm) and Pb (ND: not detected), respectively.

Characteristics of Heat Recovery Rate and Fouling according to Structures and Materials in Heat Exchangers (열교환장치의 구조 및 재질에 따른 열회수율과 파울링의 발생 특성)

  • We researched characteristics of heat recovery rate and fouling according to structures and materials in heat exchangers like water preheater and air preheater. Economizer and air preheater have used in thermal electric power plant. we made small incinerator and heat exchangers to carry out simulated experiment. We observed fouling formation and change of heat recovery rate, combusting powdered coal for 24 hr. In economizer, fin tube type had the largest amount of fouling formation, followed by tube line type > pipe type > auto washing type according to structures. As heat recovery rate, fin tube showed highest recovery rate, followed by auto washing type > pipe type > tube line type. In air preheater, fin tube type had the largest amount of fouling formation, followed by fin plate type > pipe type > pipe type coated by teflon > pipe type coated by ceramic according to structures. And then, heat recovery rate showed the same oder.

Development of Osmotolerant Mutant, Candida magnoliae M26 and the Determination of the Optimum Concentrations of Carbon and Nitrogen Sources to Improve Erythritol Yield (에리스리톨의 수율 향상을 위한 Candida magnoliae의 삼투압 내성 변이균주의 개발과 탄소원 및 질소원의 최적 농도 결정)

  • Experimental studies were carried out to develop an osmotolerant mutant of Candida magnoliae JH and to determine the optimum concentrations of carbon and nitrogen sources to improve erythritol yield and productivity. Mutants of C. magnoliae JH were prepared by treatment with ethylmethane sulfonate, and osmotolerant mutants were isolated from the agar plate colonies containing 2.5 M KCI. Among the mutants isolated, one mutant M26 was finally selected based on erythritol yield and productivity. In shake flask culture, glucose proved to be the best carbon source and the optimum yeast extract concentration was 5 g/L based on 100 g/L glucose. The erythritol yield and productivity of mutant M26 were greater than wild type in 100 g/L glucose medium. in the fermentation experiments, erythritol production increased with increased glucose concentration, up to a limit of 250 g/L. The maximum concentration of erythritol achieved 127.5 g/L, and the yield and productivity of erythritol production were 51.0% and 0.63 g/L-h, respectively.

Effect of Extrusion Process on the Change of Components in Ginseng (압출성형이 인삼의 성분변화에 미치는 영향)

  • This study was carried out to investigate the effects of the extrusion process on the change of components in ginseng. The extraction yields from ginseng by distilled water extraction were highest in the extruded ginsengs, whereas it was lowest in the white ginseng. The contents of crude saponin were highest in the extruded ginseng, and they increased as the extrusion temperature was raised. The total contents of 11 kinds of ginsenosides increased in the order of red, white and extruded ginsengs. In particular, red ginseng showed higher contents of Rg1, Rg3 and Rb2, whereas Re was highest in white ginseng. In addition, the contents of Rg2, Rh1, Rh2 and Rg3 in the extruded white ginseng became higher. Free sugar contents were greatest in red ginseng. However, they were lowest in the extruded ginseng. White ginseng had a greater L value, whereas extruded ginseng demonstrated higher a and b values. In conclusion, the extraction yields, the contents of saponin, and ginsenoside-Rg2, Rh1, Rh2 and Rg3 were increased through the extrusion process.

Comparisons of Protein-bound Polysaccharide Contents Obtained from Mycelial Cultured Broth and Fruit body of Coriolus versieolor (구름버섯의 균사배양액 및 자실체로부터 얻어진 단백다당체의 수율비교)

  • This study was performed to obtain higher yield rate of protein-bound polysac-charide extracted from cultured broth and fruit body of Coriolus versieolor, basidiomycetes. 16002 strain produced 0.58%, the highest yield rate among four strains, and there was no significantly different yield in other three strains. The optimal liquid media were CVT-III to obtain higher yield of protein-bound polysaccharide comparing with the various culture media. Compared with several materials for substitution of medium source, the residue extract media of Coriolus versieolor fruit-body were yield rate similar to control, and addition of the extracts of ginseng residues increased 11.4-48%. And also compared with fruit bodies that were artificially cultured on the oak longs, 16002 strain produced the highest yield similar to the result of cultured broth. Fruit body of the prematures stage was more effective to obtain high yield rate.

Estimating Impervious Surface Fraction of Tanchon Watershed Using Spectral Analysis (분광혼합분석 기법을 이용한 탄천유역 불투수율 평가)

  • Increasing of impervious surface resulting from urban development has negative impacts on urban environment. Therefore, it is absolutely necessary to estimate and quantify the temporal and spatial aspects of impervious area for study of urban environment. In many cases, conventional image classification methods have been used for analysis of impervious surface fraction. However, the conventional classification methods have shortcoming in estimating impervious surface. The DN value of the each pixel in imagery is mixed result of spectral character of various objects which exist in surface. But conventional image classification methods force each pixel to be allocated only one class. And also after land cover classification, it is requisite to additional work of calculating impervious percentage value in each class item. This study used the spectral mixture analysis to overcome this weakness of the conventional classification methods. Four endmembers, vegetation, soil, low albedo and high albedo were selected to compose pure land cover objects. Impervious surface fraction was estimated by adding low albedo and high albedo. The study area is the Tanchon watershed which has been rapidly changed by the intensive development of housing. Landsat imagery from 1988, 1994 to 2001 was used to estimate impervious surface fraction. The results of this study show that impervious surface fraction increased from $15.6\%$ in 1988, $20.1\%$ in 1994 to $24\%$ in 2001. Results indicate that impervious surface fraction can be estimated by spectral mixture analysis with promising accuracy.

Visual Log Grading and Evaluation of Lamina Yield for Manufacturing Structural Glued Laminated Timber of Pitch Pine (리기다소나무 원목형질 조사 및 구조용집성재 제조 수율 평가)

  • Pitch pine (Pinus rigida) has been planted in Korean forests for several decades, primarily for erosion control and use as a fuel supply. To enhance its value, and especially potential use as lamina for structural glued laminated timber (glulam), log quality and lumber yield of pitch pine were evaluated in this study. Trees from pure pitch pine stands with an average diameter at breast height of 32 cm were felled and bucked into 3.6m long 15 cm minimum butt-end diameter logs. Over 80% of the logs were classified to No.2 or No.3 visual grade group. Upon sawing total lumber yield was 55.2%, 39.9% for structural glulam lamina, 7.2% for louver, and 8.1% for miscellaneous use. The final lumber yield for manufacturing structural glulam, after cross-cutting to eliminate knots and finger jointing, was only 15.3%. To enhance this manufacturing yield requires that the rate of knot-included lumber used as lamina be raised. However arrangement of the knot-included lamina, whose mechanical properties need to be accurately evaluated, must be optimized to minimize any reduction to the structural glulam strength. The log quality and lumber yield of pitch pine evaluated in this study are expected to facilitate proper planning for wood product manufacture in the Korean lumbering and glulam industrial field, which has not previously dealt with this species.

Bacterial Cellulose 생산균 KJ-1으로 부터 uv와 NTG mutant들의 cellulose 생산수율의 증가

  • This study was performed to improve the yield of bacterial cellulose(BC) by UV and NTG mutagenesis of strain KJ-1 which produced largely BC. some mutants showed high BC productivity with twice elevation compared to that the wild strain KJ-1. A difference was found in production and bioconversion phase of synthesized organic acid, such as gluconic acid, 2-keto gluconic acid, and 5-keto gluconic acid between mutants and strain KJ-1 in the static culture. The organic acid produced in secondary metabolism phase, were more rapidly consumed in the culture with the mutants than that the parent strain after glucose in the broth was conversed to a limiting substrate. Therefore, we suggested the reason for increasing of BC production that the mutant strain consumed more efficiently synthesized acids as substrates than that of the parent strain.

  • Maize starches with different amylose content were repeated autoclaving-cooling cycles up to 4 times, and the yield of resistant starch (RS) from autoclaved maize starches was investigated by enzymatic-gravimetric method and ${\alpha}-amylase$ treatment. With increasing amylose content in starch and the number of autoclaving-cooling cycles, RS yield was also increased, regardless of isolation method. Enzymatic-gravimetric method severely hydrolyzed amorphous region of autoclaved maize starches. Crystalline region was obtained more effectively by enzymatic-gravimetric method than by ${\alpha}-amylase$ treatment.
