Optogenetics is the combination of optical and molecular strategies to control designated molecular and cellular activities in living tissues and cells using genetically encoded light-sensitive proteins. It involves the use of light to rapidly gate the membrane channels that allows for ion movement. Optogenetics began with the placing of light-sensitive proteins from green algae inside specific types of brain cells. The cells can then be turned on or off with pulses of blue and yellow light. Using the naturally occurring algal protein Channelrhodopsin-2 (ChR2), a rapidly gated light-sensitive cation channel, the number and frequency of action potentials can be controlled. The ChR2 provides a way to manipulate a single type of neuron while affecting no others, an unprecedented specificity. This technology allows the use of light to alter neural processing at the level of single spikes and synaptic events, yielding a widely applicable tool for neuroscientists and biomedical engineers. An improbable combination of green algae, lasers, gene therapy and fiber optics made it possible to map neural circuits deep inside the brain with a precision that has never been possible before. This will help identify the causes of disorders like depression, anxiety, schizophrenia, addiction, sleep disorder, and autism. Optogenetics could improve upon existing implanted devices that are used to treat Parkinson’s disease, obsessive-compulsive disorder and other ailments with pulses of electricity. An optogenetics device could hit more specific subsets of brain cells than those devices can. Applications of optogenetic tools in nonneuronal cells are on the rise.

Stress fiber (SF) alteration is mediated by cellular receptors, which, upon interaction with the extracellular counterpart, signal to the actin cytoskeleton for remodeling. This association is mediated by a variety of scaffold and signaling factors, which control the mechanical and signaling activities of the interaction site. The heterotrimeric transmembrane lymphotoxin α1β2 (LTα1β2), a member of the tumor necrosis factor (TNF) family of cytokines, including soluble homotrimeric lymphotoxin (LT α), plays an important role in lymphoid tissue architecture. Ligation between LTα1β2 and the lymphotoxin β receptor (LTβR) activates signal-cascade in fibroblastic reticular cells (FRCs). We found LTβR stimulation using an agonistic anti-LTβR antibody alone or combined with LTα or TNFα induced changes in the actin and plasticity of cells. To clarify the involvement of myosin underlying the alteration, we analyzed the effect of myosin light chain kinase (MLCK) with an MLCK inhibitor (ML7), the phosphorylation level of myosin light chains (MLC), and the level of phospho-myosin phosphatase target subunit 1 (MYPT1) after treatment with an agonistic anti-LTβR antibody for cytoskeleton reorganization in FRCs. The inhibition of MLCK activity induced changes in the actin cytoskeleton organization and cell morphology in FRC. In addition, we showed the phosphorylation of MLC and MYPT1 was reduced by LTβR stimulation in cells. A DNA chip revealed the LTβR stimulation of FRC down-regulated transcripts of myosin and actin components. Collectively, these results suggest LTβR stimulation is linked to myosin regarding SF alteration in FRC.

In this paper, the vestibular hair bundle feature model and integrated vestibular hair cell model were proposed. In conventional modeling studies of vestibular system, only partial mechanisms were modeled, such as the characteristics of the vestibular hair bundles without external forces or the action potential of synapse, and the study about action potential of vestibular afferent considering the characteristics of the vestibular hair bundle was not performed. The proposed integrated vestibular hair cell model reflects external forces considering negative stiffness features of each hair bundles with different regularities of hair cells and our model was compared with conventional model without external forces. As a result, irregular afferent and intermediate afferent with high ratio of firing frequency variations to the changes of external stimulation had small width of negative stiffness section, but the width of the negative stiffness of regular afferent with low ratio was similar to that of conventional negative stiffness features. And the proposed integrated vestibular hair cell model showed almost same results with conventional data with animal experiments in 11 chosen frequency bands. It is verified that our proposed hair bundle feature model is adequately modeled.

C-fos는 원종양유전자(proto-oncogene)인 v-fos의 세포 동족체로써, 성장인자나 신경전달 물질에 의해 수분 내에 다양한 형태의 세포에서 활성화된다. Fos단백질은 스트레스와 통증 과정의 신호전달기전에서 세포활동을 조절하는 3차전령으로 활동한다. 열충격 단백질(Heat shock protein : 이하 HSP)은 계통발생학적으로 초기 척추 동물에서부터 발현되며 생체방어체계의 중요한 인자로 세포가 고열, 외상, 허혈 등의 스트레스에 직면했을 때 발현이 증가하는 단백질로 알려져 있다. 본 연구에서는 캡사이신(capsaicin)으로 말초 신경병변을 유발시킨 후 통각신경활성의 지표로 이용되는 원종양 유전자인 c-fos의 발현과 열 또는 스트레스로 야기되는 손상에 대한 조직의 방어작용으로 발현되는 HSP 70의 발현을 동시에 관찰함으로서, 급성으로 유발된 말초 신경병변의 확인과 동시에 실험동물 체내에서 방어적인 역할을 밝히는 일환으로 이 실험을 실시하였다. 본 실험의 결과는 다음과 같다; 1. 척수 등쪽뿔 천층(Laminae I and II)에서 각각 c-fos와 HSP70을 항원으로 하는 면역조직화학적 방법으로 염색한 표본에서 0.9% NaCI 투여 2시간 후 c-fos와 HSP70의 양성을 나타내는 세포는 전혀 없음을 알 수 있었다. 2. 척수 등쪽뿔 천층에서 c-fos 단백질을 항원으로 하는 면역조직화학적 방법으로 염색한 표본에서 Capsaicin 투여 2시간 후 c-fos 단백질에 양성을 나타내는 세포가 많이 발현됨을 육안적 관찰로서 알 수 있었다. 3. 척수 등쪽뿔 천층에서 HSP70을 항원으로 하는 면역조직화확적 방법으로 염색한 표본에서 Capsaicin 투여 2시간 후 HSP7O의 양성을 나타내는 세포가 보통수준으로 발현됨을 육안적 관찰로서 알 수 있었다. 이 실험의 결과로 볼 때, 화학적인 신경병변 유발물질에 의한 손상을 방어하기 위해서 체내에는 내인성 물질이 형성될 것이라는 추측과 c-fos 가 다른 유전자의 발현을 유도한다는 점을 함께 고려 하였을때, Capsaicin에 의한 말초 신경병변에서 c-fos 발현이 많이 나타나는 것은 손상을 방어하는 물질의 생성에 관여하기 때문이며, 방어물질 중 이 실험에서 본 HSP70도 증가한 내인성 방어물질의 하나라고 할 수 있을 것이다.
최근 우리나라는 활동량은 적어지고 식생활의 서구화로 열량섭취가 증가하여 성인뿐만 아니라 소아에서도 비만증이 급격하게 증가하고 있다. 소아 및 청소년의 비만은 대개 증식형 비만 형태로 지방세포수가 일단 증가하면 체중조절이 어렵고, 특히 복부비만은 성인 비만으로 이행하기 쉬우며 실제로 어렸을 때 비만에 걸리면 그 중 60~80%가 성인 비만으로 이행된다고 보고되는 바 사회적 관심이 요구된다. 또한 성인과 마찬가지로 소아 비만 자체로도 고혈압, 고지혈증, 심혈관질환, 당뇨 등 다양한 합병증뿐만 아니라 우울증 및 다른 정신적 문제를 동반할 수 있어 보다 적극적인 치료가 필요하다.
초음파 시스템은 진단 영상뿐만 아니라 초음파 수술과 같이 초음파를 사용하여 특정 세포를 제거하거나, 또는 일반적인 수술을 위한 가이드 영상을 제공하는 등 그 활용이 증가되고 있다. 반면, 기존 초음파 시스템은 크기가 커서 클리닉 센터나 종합병원 등에서 이동성 및 공간 효율성이 낮고 환자에 관한 주치의들의 진단 활동 반경이 매우 제한적이라는 불편함을 초래한다. 본 논문은 서버 클라이언트 기반의 무선 초음파 진단 시스템에서 프루브가 수신한 초음파 반사 신호를 무선으로 실시간 전송하기 위해서 압축센싱과 산술 부호의 연접을 통한 손실 압축 알고리즘을 제안한다.

유해 남조 대발생(Harmful Algal blooms, HABs)이 담수호에 발생하면 마이크로시스틴과 같은 독성물질과 맛·냄새 물질을 생성하여 상수원이용과 친수활동을 방해한다. 그래서 유해 남조 대발생 전 유해남조 세포수를 예측하여 선제적 대응하는 것은 중요하다. 따라서 본 연구는 머신러닝기반 Random Forest(RF)를 활용하여 팔당댐 앞의 유해남조 세포수를 예측하는 모델을 개발하고 성능을 평가하고자 한다. 모델 구축을 위해 2012년 4월부터 2021년 12월까지의 팔당호(삼봉리, 경안천) 및 남북한강(의암댐~이포보)권역의 조류, 수질, 수리/수문, 기상 자료를 수집하여 입력 및 출력 자료로 이용하였다. 수집된 데이터에는 다양한 입력변수들이 있어 남조 세포수 예측 성능 비교를 위한 전체 26개 변수 적용과 통계학적으로 상관관계가 높은 12개 변수 적용을 통해 모델을 구축하였다. 입력, 출력 자료로 이용한 유해남조 세포수는 로그변환된 값으로 사용하였으며 일반적인 조류 시료 채취기간이 7일이므로 7일 후를 예측하기 위한 모델을 구축하였다. 구축한 모델의 성능은 실측데이터와 예측데이터의 R2로 산출하여 평가하였다. 전체 26개 입력변수로 모델 구축 후 학습 및 검증 수행 결과 R2의 학습 0.803, 검증 0.729로 나타났고, 유해남조 세포수와 유의미한 상관관계를 보이는 12개 입력변수로 모델 구축 후 학습 및 검증 수행 R2은 학습 0.784, 검증 0.731로 나타났다. 두 모델의 성능을 살펴본 결과 입력변수 개수의 변화에 따른 성능차이는 크지 않은 것으로 나타났으며, 남조세포수 예측을 위한 모델로서 활용가능함을 알 수 있었다. 향후 연구에서는 Random Forest 외 다른 기계학습 모델들과 딥러닝 모델을 통해 남조세포수 예측 성능이 높은 모델을 구축해볼 필요성이 있다.

흰쥐에 사료 섭취량의 3%(G3군)와 5% (G3군)인 과량의 생마늘(즙)을 1개월간 경구 투여하여 체중 증가율과 사료 섭취량은 큰 영향을 받지 않았다. 흰쥐 외관과 활동도는 별다른 변화가 없었으나 해부 검사에서 G3군 흰쥐일부와 G3군 대부분의 흰쥐의 위벽점막에 홍반(erythema)를 관찰할 수 있었다. 간조직의 현미경 검사에서 G3군의 3마리 흰쥐에서 주로 간 실질세포으 퇘행성 변화와 염증세포의 침윤 등을 관찰했고 G5군의 7마리 흰쥐는 주로 간실질세포의 염증성세포 침윤과 충혈 및 경미한 간세포 괴사를 보였다. GOT와 GPTalc alkaline phosphatase 활성과 BUN수준에서 G3군과 G5군은 dose dependent fashion으로 모두 대조군보다 유의성있게 증가하였다. Total cholesteroltnwns과 hemoglobin함량은 G3군과 G5군에서 유의적인 변화를 보이지 않았다. 이상의 본 실험 결과를 보면 흰쥐에 과량의 마늘을 장기간 투여할 때 명백하게 독성이 나타남을 관찰할 수 있었다.
Buprenorphine, one of the mixed agonist-antagonist opioid drugs was used to inverstigate the opioid receptor on frog sciatic nerve A fibers. Action potentials were recorded for 4 hrs by a sucrose gap apparatus which were separated by four rubber membranes. To examine the one of the mechanism of action of buprenorphine, meperidine or naloxone was added after or before the treatment of buprenorphine. The results of this experiment were as follows: 1. Buprenorphine suppressed significantly the compound action potentials of frog sciatic nerve, and the maximal effects were shown both at $10^{-4}\;M$ and at $10^{-8}\;M$. 2. The dose-response relationship of buprenorphine on the depressant effect in frog sciatic nerve was biphasic and inverted U-shaped. 3. Buprenorphine blocked the effect of Meperidine $(10^{-3}\;M)$ on this preparation. 4. The depressant effcct of Buprenorphine on frog sciatic nerve was blocked by $10^{-8}\;M$ naloxone. From the above results, buprenorphine acts as one of agoinist-antagonistic effect on frog sciatic nerve, and the opioid receptor on this preparation is located on or near the intracellular opening of the sodium channels, which are sensitive to naloxone.
Iks 칼륨 전류에 관여하는 KCNQ1유전자의 S140G 돌연변이는 심방세동에 영향을 미치는 대표적인 돌연변이 유전자로, 심방세동과 S140G 돌연변이의 상관관계를 밝히기 위한 연구들이 많이 진행되어 왔다. 하지만 S140G 돌연변이 유전자가 심방 세동 환자의 심실 반응에 영향을 미칠 수 있다는 선행연구를 비롯하여 심방과 심실의 활동전위에 영향을 미칠 수 있는 가능성이 있음에도 불구하고, KCNQ1 S140G 돌연변이 유전자의 심실세동에 대한 영향과 그 메커니즘에 대한 연구는 부족하다. 따라서 본 연구는 KCNQ1 S140G 돌연변이 유전자가 심실세동에 미치는 영향에 대한 컴퓨터 시뮬레이션 연구를 통해 그 상관관계를 밝히고자 하였다. 이를 위해 1차원 세포 모델을 비롯하여 2차원 심실세동 반응과 3차원 전기 생리학 및 기계적 수축 시뮬레이션을 진행하였다. 3차원의 전기생리학 및 기계적 수축 시뮬레이션에서는 심실의 박출 활동을 확인하기 위한 정상 박동 시뮬레이션과 심실 세동 발생시의 심실의 변화를 확인하기 위한 세동 시뮬레이션을 각각 진행하였다. 그 결과 KCNQ1 S140G 돌연변이로 인해 심실의 Iks가 증가되었으며, 그로 인해 심실의 활동 전위기간(APD)과 불응기(ERP)가 단축되는 것을 확인할 수 있었다. 또한 활동전위 지속 곡선(APDr)과 불응기 지속 곡선(ERPr)이 완만하게 나타났으며, 심근세포의 전도파장이 감소하였다. 3차원 정상 박동 시뮬레이션의 결과 표준형에서 보다 KCNQ1 S140G 돌연변이형에서 심실이 소모하는 ATP의 양과 박출계수가 감소하였다. 3차원 세동 시뮬레이션 결과 표준형에서는 심실세동이 종결되었으나, S140G 돌연변이 형에서는 심실세동이 종결되지않고 유지되었으며, 심실세동이 빠르게 발생하였다. 결론적으로, KCNQ1 S140G 돌연변이로 인해 증가된 심실의 Iks는 심실의 박출 효율을 감소시키고 심실세동을 발생시키고 유지시키며, 부정맥 발생의 위험성을 높일 수 있다.
