체외배양조건이 체세포 핵이식란의 배발달 및 배반포의 동결-융해 후 생존성에 미치는 영향

  • 체세포 핵이식에 의한 산자가 양, 소, 염소, 쥐 및 최근에 돼지에서 보고되었지만, 생산된 배반포의 동결보존에 관한 연구결과는 보고되지 않고 있다. 따라서, 본 연구는 체세포 핵이식란의 활용과 산업화 촉진을 위해서 체세포 핵이식 유래 배반포의 동결성 향상에 배양조건이 미치는 영향을 조사하고자 실시하였다. 공여세포는 60일령의 태아세포를 10% FBS 가 첨가된 DMEM 에서 39℃, 5% CO₂의 incubator에서 배양하여 monolayer confluent 형성을 유도한 후 0.25%trypsin 처리하였다. 개 유방 선암종(Adenocarcinoma) 분비물의 세포학적 진단

  • 개 유방 종양은 개에서 가장 흔한 종양들이지만 유방 분비물을 통한 세포학 진단의 의의는 크다. 이들 종양의 진단상 병리조직학적 진단은 보편화되어 있으며 그 정확성도 매우 높다. 그러나 진단에 걸리는 시간과 비용 문제를 간과 할 수는 없다. 본 증례는 전남대학교 부속동물병원에 내원한 9살된 암컷 요크셔테리어 종으로서 유선에서 혈액과 고름이 섞인 분비물이 5개월 동안 관찰되었다. 이들 분비물을 슬라이드에 도말하여 diff-quik 염색후 세포학적 진단결과 다양한 형태의 종양세포, 적혈구 및 호중구가 관찰되었다. 종양세포는 대소부동한 다형태성의 핵을 가지고 있었고, 핵소체는 뚜렷하였으며 핵과 세포질 비율(N/C) 도 매우 높았다. 전형적인 유방 선암종(adenocarcinoma) 세포로 진단되었으며 유방절제후 병리조직학적 검사 소견도 세포학적 진단과 일치하였다. 유방 선암종 분비물의 세포학적 진단은 보다 간편하고 적은 비용으로 진단해 낼 수 있었다. 개 유방 선암종 분비물의 세포학적 진단은 국내보고를 쉽게 찾을 수 없었다.

The Ultrastructure of the Cutaneous Pigment Cells in the Amphibia (양서류 피부 색소세포의 미세구조)

  • The ultrastructures of the pigment cells in the Asiatic land salamander (Hynobius leechi) dorsal skin were obtained by means of electron microscope. The results were as follows; 1. The pigment cells of the epidermis consisted of the melanocytes in the germinal layer and of the melanophores distributing to the keratinocyte layer. The traits of these cells in the epidermis were as follows: A. The nuclei of the melanocytes were round or oval in shape and appeared as partly small or large infoldings of the nuclear envelope. B. Rough-surfaced endoplasmic reticulums and Golgi complexes were well developed in infranuclear cytoplasm. Many ribosomes were mainly distributed around the perinuclear portion. C. The melanosomes of the melanocytes were observed as a found or an oval shape and strong electron-dense or less electron-dense melanosomes were observed. D. The infoldings of the nuclear envelope in the melanophore were partly found deeper than those of the melanocyte. The cytoplasm of the melanophore filled with melanosomes caused organelles not to be observed in that. 2. The pigment cells in the dermis were composed of the xanthophores just beneath basement membrane and the melanophores in the connective tissue. The traits of these cells in the dermis were as follows: A. The xanthophores contained round or oval vesicles, and these vesicles were divided into 6 types (type I pterinosome, type II pterinosome, type III pterinosomes, type iv pterinosome, type V pterinosome, type VI pterinosome). B. Most of the nuclei of the melanophores in the dermis were elongate in shape, and a portion of the nuclear envelope was deep infolded. C. Becuase the cytoplasm was filled with the melanosomes of the same electron-density, organelles were not observed in the cytoplasm. D. Two processes of the melanophore in the dermis extended in parallel with a xanthophore and the cytoplasm in those processes were filled with the melanosomes.

  • 2차원 영상 기반의 세포 분석은 2차원 평면 상에서의 관찰만 가능하므로 정확하고 객관적인 분석에 한계가 있다. 따라서 이러한 한계를 극복하고자 컨포컬 현미경을 통해 획득한 연속적인 2차원 단면 영상들로 구성된 볼륨 데이터를 볼륨 및 표면 렌더링을 통해 가시화 하여, 세포핵에 대한 다양한 각도에서의 형태 관찰이 가능하도록 하였다. 또한 세포핵 볼륨에서 ROI 추출을 통해 국부 영역 분석의 효율성을 높였다. 그리고 이 결과를 바탕으로 정확한 암 세포의 계측 및 정량적 분석을 통해 이후 자궁암 환자에게 최적의 진단 및 지료를 제공할 수 있는 연구 기반을 마련하고자 한다.

Cytogenetic Properties of Bovine Reconstituted Embryos by Cell Cycle-Controlled Nuclear Transfer (소 수정란의 세포주기조절 핵이식에 의한 재구축배의 세포학적 특성)

  • This study was conducted to investigate the cytogenetic properties, in vitro development, and their relationship in the bovine reconstituted embryos following cell cycle-controlled nuclear transfer. Sixteen-cell stage embryos were treated by nocodazole, and after release from nocodazole treatment, their blastomeres were separated and allowed to subsequent cleavage. Blastomeres within 1.5 h post cleavage(hpc) and at 3hpc were transferred to enucleated oocytes at MII-phase or S-phase. Donor nuclei transferred into M II-phase recipients underwent various nuclear remodeling, such as extrusion of a polar body(PB)-like structure, premature chromosome condensation(PCC) and chromatin modifications. These nuclear remodeling patterns varied by the time post cleavage of donor blastomeres. Developmental rate to the blastocyst stage differed with time post cleavage of donor blastomeres and existence of a PB-like structure. Whereas do-nor nuclei transferred into S-phase oocytes did not undergo PCC and other major modifications, and their developmental potentials less depended on the nuclei types. This result confirms that the nuclear remodeling type differs with donor and recipient cell cycle stage, which affect the development of reconstituted bovine embryos.

  • It is important to obtain conn cytodiagnosis to classify background, cytoplasm, and nucleus from the diagnostic image. This study mose an algorithm that detects and classifies carcinoma cells of the uterine cervix in Pap smear using features of cervical cancer. It applies Median filter and Gaussian filter to get noise-removed nucleus area and also applies Kapur method in binarization of the resultant image. We apply 8-directional contour tracking algorithm and stretching technique to identify and revise clustered cells that often hinder to obtain correct analysis. The resulted nucleus area has distinguishable features such as cell size, integration rate, and directional coefficient from normal cells so that we can detect and classify carcinoma cells successfully. The experiment results show that the performance of the algorithm is competitive with human expert.

Nucleus Segmentation and Recognition of Uterine Cervical Pop-Smears using Region Growing Technique and Backpropagation Algorithm (영역 확장 기법과 오류 역전파 알고리즘을 이용한 자궁경부 세포진 영역 분할 및 인식)

  • 자궁 경부 세포진 영상의 핵 영역 분할은 자궁 경부암 자동화 검색 시스템의 가장 어렵고도 중요한 분야로 알려져 있다. 자궁 경부 세포진 영상은 배경과 세포의 영역이 확실히 구분되지 않는 경우가 많기 때문에 이들을 확실히 구분하는 것이 매우 중요하다. 본 논문에서는 이러한 문제점을 해결하기 위해 자궁 경부 세포진 영상에서 Region growing 기법을 적용하여 세포 영상을 분할한다. Region growing 기법은 화소간의 유사도를 측정하여 영역을 확장하여 분할하는 방법이다. 세포와 배경이 분할된 영상을 일정 임계값을 이용하여 영상을 이진화 한 후, 8방향 윤곽선 추적 알고리즘을 이용해 세포 영역을 추출한다. 추출된 세포 영역을 원 영상인 RGB 컬러로 변환한 후에 K-means 알고리즘을 적용하여 각 세포 영역의 RGB 화소를 R, G, B 채널로 각각 분리하여 클러스터링한다. 클러스터링된 각각의 R, G, B 채널의 클러스터 값을 이용하여 HSI 모델로 변환시킨 후에 세포핵 영역의 Hue 정보를 추출한다. 추출된 세포핵의 특징을 오류 역전파 알고리즘을 적용하여 정상 세포와 비정상 세포를 분류하고 인식한다.

Morphological Study on the Post-hatching Development of the Pineal Gland in Korean Pheasant (Phasianus colchicus Karpowi) (한국산 꿩 송과샘의 부화후 발달에 관한 형태학적 연구)

  • The pineal gland of the bird occupies a key position in the phylogenetic evolution of this organ. Therefore, the purpose of this study was to investigate the developmental changes of the pineal gland during post-hatching period in Korean pheasant. The pheasants were sacrificed at 1-day-, 1-month-, 2-month-, and 6-month-old after hatching. The morphological characteristics of a pineal glands were determined in all pheasants using light microscope, and transmission electron microscope. Connective tissue originated from the capsule divided the pineal parenchyma into incomplete lobules. The parenchyma was consisted of pinealocytes and supportive cells. These parenchymal cells were arranged in the forms of solid lobules as well as incomplete follicles. At the follicular lumen, membraneous lamellar complexes and blob -like structures were present. Pinealocyte, a predominent cell type, had euchromatic nucleus, and showed the segmental organization. The bulbous apical portion had scanty free ribosomes and occasional cilia associated with basal bodies. The constricted neck, transitional portion from apical to pericarya had junctional complexes with adjacent supportive cells, and had microtubules. Cell body contained abundant mitochondria, well-developed Golgi complex, rough endoplasmic reticulum (RER) and free ribosomes. Basal processes extended from the base of the cell soma toward the basal lamina and contained 60∼90 nm dense cored vesicles. Supportive cells, another major type of the parenchyma, were characterized by the dense and elongated nucleus, and contained moderate number of mitochondria, RER, developed Golgi complex, free ribosomes and a few dense bodies in the perinuclear cytoplasm. Slender processes of supportive cells interposed between the pinealocytes and often bordered the basal region of the parenchyma. These results indicate that the pinealocytes of the pheasant are not rudimentary photoreceptor cells, and appear to have secretory function. Further studies will be required to confirm the morphological characteristics of pineal gland in adult pheasant during breeding and nonbreeding season.

Study on evaluating the significance of 3D nuclear texture features for diagnosis of cervical cancer (자궁경부암 진단을 위한 3차원 세포핵 질감 특성값 유의성 평가에 관한 연구)

  • The aim of this study is to evaluate whether 3D nuclear chromatin texture features are significant in recognizing the progression of cervical cancer. In particular, we assessed that our method could detect subtle differences in the chromatin pattern of seemingly normal cells on specimens with malignancy. We extracted nuclear texture features based on 3D GLCM(Gray Level Co occurrence Matrix) and 3D Wavelet transform from 100 cell volume data for each group (Normal, LSIL and HSIL). To evaluate the feasibility of 3D chromatin texture analysis, we compared the correct classification rate for each of the classifiers using them. In addition to this, we compared the correct classification rates for the classifiers using the proposed 3D nuclear texture features and the 2D nuclear texture features which were extracted in the same way. The results showed that the classifier using the 3D nuclear texture features provided better results. This means our method could improve the accuracy and reproducibility of quantification of cervical cell.
