The Taxonomic Consideration of Leaf Epidermal Microsturcture in Glechoma L. (Nepetinae, Lamiaceae) (긴병꽃풀속(Glechoma L., 꿀풀과)의 잎표피 미세구조에 대한 분류학적 검토)

  • A comparative micromorphological study was examined on the leaves of the genus Glechoma and related genera (Nepetinae, Lamiaceae) by scanning electron microscopy (SEM) in order to evaluate their significance in the taxonomy. The leaves of taxa Marmoritis, Nepeta sect. Glechomanthe, G. hederacea var. longituba (Korea) are revealed amphistomatic type, while the remnants of taxa had hypostomatic type. The size range of the guard cells is $12.50-28.75{\times}9.17-21.25{\mu}m$: the smallest one was found in M. pharicus ($12.50-15.83{\times}9.17-11.25{\mu}m$), while the largest one was measured to G. hederacea var. longituba (Korea: $28.75-28.88{\times}21.25-21.38{\mu}m$). The stomatal type of genera Agastache, Dracocephalum was mostly diacytic, however for the rest rarely together with anisocytic and anomocytic, except G. hederaca var. longituba (Korea), Meehania urticifolia by having combined with diacytic and anomocytic. The shapes of epidermal cells are differ from in abaxial and adaxial side, and dived with two types (e.g., platelet, stripe pattern). Five types (three glandular, two non-glandular hairs) of trichomes are distributed in leaves. Among trichomes, long and stalk capitates glandular trichome, subsessile glands are different from studied taxa so that leaf micromorphological characters are significance features in the taxonomy.

  • During the course of the field trip to Mt. Baekdu and Manchurian region especially along the border of North Korea, young flower buds were collected and fixed to analyze the meiotic configuration of chromosomes and to discuss the cytotaxonomic characteristics of these areas. A total of 29 taxa with 31 populations of vascular plants were analyzed chromosomally. Among them, 26 taxa with 28 populations were collected from Mt. Baekdu which include Papaver radicata var. pseudoradicata, Bupleurum euphorbioides, Oplopanax elatus, Silene jenisseensis, and Gentiana jamesii and three taxa collected from Manchurian region which include Scrophularia buergeriana, Ostericum grosseserratum and Callistephus chinensis. The first report of chromosome numbers were provided here for those species of Artemisia lagocephala as 2n = 9II, Cacalia komaroviana as 2n = 30II, Chrysanthemum zawadskii var. alpinum as 2n = 27II and Rodiola rosea as 2n = 11II,

Nature of Phytoliths and their Applications in Botany (식물규소체의 특성과 식물학적 응용)

  • Phytoliths (plant stones) mean all types of mineral deposition formed in or between plant cells. However, most of the researches on the phytoliths so far have been focused on the opaline silica bodies (silicified phytoliths) formed by grasses. Therefore, the phytolith is usually refered to opaline silica body in a narrow sense. Phytoliths are very useful in grass classification, and their usefulness as microfossils is also proved in several other fields, including soil science, paleoecology, and archaeology. Recently attention has been paid to the possibility that they might function as carcinogen. The purpose of this review is to present an overview of the nature and analysis of phytoliths and their applications in botany as well as other fields of sciencess.encess.

  • The taxonomical properties of strain J-275L isolated from soil as a microorganism which produces extracellular adenine deaminase and cultural conditions for the enxyme production were studied. The hyphae of strain J-275L is fragmented into rod-or coccus-like elements. The elements of fragmented aerkal hyphae has smooth surfaces. The cell wall of the organism contains LL-diaminopimelic acid. Mycolic acid are not produced. As a result of taxonomical studies, strain J-275L is designated as Nocardioides sp. J-275L. The optimum medium for the enzyme production from Nocardioides sp.J-275L wascomposed of 0.5% peptone, 0.5% dextrin, 1% yeast extract, and 0.2% $K_{2}HPO_{4}$. The optimum initial pH of the medium was pH 7.5.

  • Morphological characters of three taxa belonging to three genera (Fuirena, Kobresia, and Scleria) in Korean Cyperaceae were reexamined. The anatomical patterns of stems and leaves, and epidermal patterns of achenes and leaves were investigated by a scanning electron microscope (SEM) and a light microscope (LM). Based upon the results, the scientific and Korean names, which have not been clearly established, were reviewed. Morphological characters such as length and width of stem, leaf, bract, scale, achene, and shape of achene, spikelet, style base, scale, perianth, and anatomical characters such as shape of stem and leaf, shape of achene epidermal cell and silica body of leaf, size and frequency of stomatal complex, presence of bulliform cell were useful for an identification of the three taxa.

Phylogenetic implication of seed coat sculpturing in subtribe Agrimoniinae (Rosaceae) (장미과 짚신나물아족 종피형태의 계통분류학적 고찰)

  • Seed morphology in the subtribe Agrimoniinae (Rosaceae) was examined using scanning electron microscopy to identify distinct characters and evaluate their evolution in a phylogenetic framework for five genera in the subtribe: Agrimonia L., Aremonia Neck. ex Nestl., Hagenia J.F. Gmel., Leucosidea Eckl. & Zeyh., and Spenceria Trimen. All genera have one or two mature achenes in a fruiting hypanthium. In the seed coats, the cell shape, size, wall features, and sculpturing vary across genera. Of most significance is the presence of papillae structures in both Agrimonia and Aremonia. Through the mapping of papillae features onto phylogenetic trees, either one or two changes in seed coats are hypothesized. The phylogenetic tree inferred from four nuclear and six chloroplast regions of sequence data suggests that at least two steps of papillae sculpturing on seed coats are required. On the other hand, in the phylogenetic tree of a low-copy nuclear gene, one independent evolutionary step is postulated to explain the current character states. In the latter hypothesis, the seed coat sculpturing also supports a monophyletic relationship for cosmopolitan Agrimonia and European endemic Aremonia. The seed coat sculpturing provides valuable information for inferring phylogenetic relationships at the generic level in the subtribe Agrimoniinae.

  • Microscopic structures of epidermis and palynology and isozyme analysis were examined to find out the intergeneric and interspecific relationships and consequently to confirm the position of Hanabusaya aiatica as an endemic genus among 4 genera and 9 species belonging to the Hanabusaya asiatica, Korean endemic, and its allied groups. In the examination of microscopic structures of epidermis, cell patterns of perianth and ultrastructure of seed coat were found to be useful characters for the identification of the 4 genera and 9 species. Palynological characters such as microscopic structure and overall morphology of pollen grains were, however, not enough to distinguish them because of the great variabilities in these traits. Isozyme analysis showed that H. asiatica was very closly related to Campanula punctata and C. takesimana, though there were variations among populations and collected areas in some classified groups, depending on classified groups. Based on these results, the position of H. asiatica as an endemic genus was well confirmed.

Comparative Study of Melanin Content in Corneocyte with Skin Color (각질세포 내 멜라닌 정량과 피부색의 비교 연구)

  • Melanin is synthesized by the melanocytes in basal layer of epidermis and distributed over all the layers of skin including corneocytes in stratum corneum, outermost layer of the epidermis. Melanin of corneocytes was stained using Fontana-Masson method and quantified by image analysis. The correlation between melanin contents and skin color value was estimated in the skin of 71 Korean women. Melanin covering area (MCA) showed good correlation with $L^*$ value and $ITA^{\circ}$ (Individual Typology Angle) (r = 0.6049, 0.6651, respectively). MCA can be used as new parameter for skin color study and has potential application for evaluating the efficacy of the skin whitening cosmetics.

  • To examine the leaf epidermal microstructure, nine species in five genera (Daphne L. - 4 spp., Diarthron Turcz. - 1 sp., Edgewarthia Meisn. - 1 sp., Stellera L. - 1 sp., Wikstroemia Endl. - 2 spp.) of the Korean Thymelaeaceae were investigated by light microscopy (LM) and scanning electron microscopy (SEM). The stomata of stuo야ed taxa were 'hypostomatic type' and the size range of guard cell was $13.8-34.4{\times}8.7-22.9{\mu}m$: the smallest size of stomata was found in Diathran linifolium ($15.9{\pm}2.6{\times}10.0{\pm}1.3{\mu}m$), while the largest one was measured to Daphne adara ($32.8{\pm}1.6{\times}20.7{\pm}1.3{\mu}m$). The stomatal complex was anomocytic in the most studied taxa, except Daphne kiusiana by having combined with anisocytic together. The shapes of epidermal cells are undulate anticlinal wall. The size range of epidermal cell was $20.7-61.0{\mu}m$; the smallest size of epidermal cell was found in Stellera charnaejasme ($26.0{\pm}1.9{\mu}m$), on the other hand the largest one was found in Edgeworthia chrysantha ($53.6{\pm}3.1{\mu}m$). The well-developed flaky epicuticular waxes can be divided three kinds of pattern - (1) smooth in comparison, not entire platelets and scattered, (2) isolated flake-like platelets, mostly paralleled, sparsely, (3) flake-like platelets, flat, membraneous, protruding from the surfaces at varying angles and densely. Two types of trichome are recognized; (1) Type I: uniseriate trichome of striate surface (D. genkwa, Diarthron linifalium, E. chrysantha, W. ganpi and W. trichotama), (2) Type II: multicellular trichome of papillose surface, uncinated 3-4 nodes (Diathron linifolium). Finally, the systematics significance of the leaf micromorphological features in identification and elucidation of Korean Thymelaeaceae, especially between or within the genera including among the species is also briefly discussed.

  • Morphological characters of 14 taxa in 6 sections Ischnostachya(one taxon), Anomalae(one taxon), Capitellatae (five taxa), Debiles (one taxon), Capillares (one taxon) and Molliculae (five taxa) of genus Carex (Cyperaceae) were reexamined. The epidermal patterns of perigynium, achene and leaf were investigated using by using a scanning eletron microscope (SEM) and a light microscope (LM). Morphological characters such as rhizome, spike, width of leaf, leaf sheath, involucre, involucre sheath, beak of perigynium, perigynium shape, and leaf epidermal patterns were useful for the identification of observed fourteen taxa. The number of spike, arrangement of psitillate or staminate spike and presence of bract were very useful characters to divide the treated 14 taxa into two groups. A key based on data was presented here.
