  • 난 발생 및 난 성숙을 위한 난소내의 제어기능의 이해는 흥미로운 연구대상이다. 그러나, 난포에 의하여 지엽적으로 분비된 어떤 스테로이드 물질도 난 발생 및 난 제어를 위해 관여하지 않는 것으로 알려져있다. 따라서, 난소내의 조절인자가 어떻게 난 발생 및 난 성숙을 위해 작용하는지를 구명하는 것은 난 발생 및 난 성숙을 위해 매우 중요한 연구과제라 할 수 있다. 본 총설에서는, 본 연구실의 최근의 업적을 중심으로 포유동물의 난 발생과 난 성숙을 위한 난소내의 조절인자에 대하여 정리하였다.

Increased mRNA Related Ovarian Maturation during Induction of Maturational Competence in Red Seabream, Pagrus major (참돔, Pagrus major의 성숙능력 유도시 증가된 난성숙 관련 mRNA)

  • This study has used differential display-PCR, to amplify genes transcribed during the ovarian maturation induced by human chorionic gonadotropin (hCG). The cDNA expressed at the times of acquisition of oocyte maturational competence in red seabream (Pagrus major) following treatment with hCG was amplified and cloned. A full-length of cDNA for p. major was isolated using differential display-PCR and 5'RACE. This cDNA clone contained 2,662 nucleotides including the open reading frame that encoded 434 amino acids. Homology analyses, using the GenBank and EMBL general database searches, indicated that the nucleotides sequence of the cDNA does not have high homology with any other genes. This cDNA was judged to be a gene, which induction of maturational competence coincides with increase of mRNA related ovarian maturation. Consensus sequences which were consistent with protein kinase C phosphorylation sites and casein kinase II phosphorylation sites were identified. in vitro, the transcription level of mRNA related ovarian maturation increased between 9hr and 24hr following treatment of ovarian follicles with hCG. It was also increased after GtH-II (300 ng/ml) stimulation. Furthermore, in vivo, mRNA related ovarian maturation was rarely expressed prior to the acquisition of oocyte maturational competence, but was strongly expressed after the acquisition of oocyte maturational competence, suggesting that the hCG induction of maturational competence is brought about by the de novo synthesis of the mRNA related ovarian maturation in p. major.

Protein Contents During Oocyte Development and Some Characteristics of Egg-Specific Protein in Lucilia illustris (연두금파리의 난세포성숙에 따른 단백질의 변화와 난특이성단백질의 특성)

  • Changes in protein content during oocyte development was measured and egg-specific protein was characterized from the eggs in Lucilia illustris. During normal development ovarian protein was rapidly increased at 72hr and reached maximum at 96hr after a protein meal, when the eggs were fully matured. Purified protein from the ovaries by gel filtration of DEAE-cellulose an Sephacryl S-200 was loaded on 7.5% native polyacrylamide gel electrophoresis and identified at ${R}_{f}$ 0.4 as egg-specific protein, which has a mol. wt of 110,000. A total of 13 amino acids in th egg-specific protein was identified and expecially asparagine, glutamic acid, and tyrosine were highly concentrated. Five fatty acids were also identified. It is suggested that there is a specific protein in the eggs of L. illustris except yolk protein synthesized and secreted by fat body.

  • To clarify characteristics of yolk protein of abalone, yolk protein was purified from the ovarian egg extracts of mature female Haliotis discus hannai by a gel chromatography of sepharose CL-4B. From the results of immuno-electrophoresis and Ouchterlony's diffusion test to male and female sera and ovarian egg extracts using antibodies raised against mature female and male sera and male sera and ovarian egg extrascts, it was identified that the mature female serum had female specific serum protein and its antigenecity shared with ovarian egg extracts. A single type of yolk protein was purified from ovarian egg extracts, and it was composed of two subunits. Their molecular weights were estimated to be approximately 166 KDa and 113 KDa by SDSPAGE. The antiserum against yolk proteins cross-reacted with a mature female specific serum protein and extracts of hepatopancreas of vitellogeing females, but did not reacted with extracts of hepatopancreas of mature male.

  • In vitro cultrue systems for the growth of sma-II oocytes and for meiotic maturation are expected to provide a new source of a large population of oocytes as well as assistance in basic physiological studies of oogenesis. Mouse oocytes mid-growth phase can complete grovvth and acquire full developmental capacity in vitro. On the other hand, growing pig oocytes need some other factors. FSH at a low concentration maintains the viability of both oocytes and granulosa cells, and hypoxanthine promotes the meiotic competence of the oocytes during culture period. Considerable improvement in the culture systems for growth of pig oocytes, suggested from mouse studies, and for oocyte maturation could help to develop this technology in larger species.

염소 난포란의 체성숙시 streptomycin에 의한 성숙능력 억제 효과

  • 항생제는 일반적으로 포유동물 난자의 체외 성숙배지나 배양배지에 첨가된다. 그러나 이들 항생제들이 포유동물의 체외 난자성숙에 미치는 영향들은 아직 완전히 검토되지 않고 있다. 본 연구에서는 염소 난포란에서 체외성숙 능력과 그 후의 단위생식 활성화에 penicillin, streptomycin 또는 gentamycin이 영향을 미치는지에 대하여 조사하였다. 도축장으로부터 수집된 난소로부터 난자-난구세포 복합체들을 회수하여 5개의 처리구 [1) Control: TCM-199 medium with no antibiotics, 2) TCM-199 with 100 IU/ml penicillin, 3) TCM-199 with 50 ug/ml streptomycin, 4) TCM-199 with 50 ug/ml gentamycin, 5) TCM-199 with both 100 IU/ml penicillin and 50 ug/ml streptomycin]에서 24시간 동안 성숙시켰다. 그리고 성숙된 난자들은 ionomycin 처리에 의한 단위생식 활성화를 시킨 후 6-diethlaminopurine(6-DMAP)에서 노출시킨 다음 5개의 항생제 처리구에서 48시간 동안 배양하였다. 24시간 동안 체외성숙이 유기된 난자에서 제 1 극체가 뚜렷이 방출된 것을 관찰하여 성숙율은 얻었고, 단위생식 활성화는 48시간 후 4-cell 단계의 난분할 관찰에 의해 평가하였다. 24시간 동안 체외성숙이 유기된 미성숙 염소 난자의 성숙비율은 69.1~73.8%로 나타났으나 Streptomycin 처리구에서는 42.5~45.7%로 나타나 유의적인 차이가 인정되었다. (p<0.01) 그러나 48시간 후의 성숙난자의 난할비율에서 5개의 처리구들 사이에는 유의적인 차이가 없었다. penicillin과 gentamicin 처리 집단들은 성숙비율과 48 시간 후 4-cell 단계로의 난분할 비율에 크게 영향을 미치지는 않았다. 그러므로 streptomycin은 미성숙 염소 난자의 체외성숙을 억제하지만 성숙된 난자에서 다음 단계로의 배발달에는 영향을 주지 않는 것이 나타났다.

Effectof of Hormonal Treatment of Induced Maturation and Ovulation in the Spotted Halibut, Verapser variegatus (호르몬 처리에 의한 범가자미, Verasper variegatus 의 성숙과 배란유도)

  • Hormone induction in maturation and ovulation was carried out with spotted halibut (Verasper variegatus) which normally does not spawn spontaneously in captivity. Prior to spawning females were treated with human chorionic gonadotropin(HCG 265-678 IU/kg BM) 17 ${\alpha}$-hydroxy 20-${\beta}$-dihydroprogesterone (17 ${\alpha}$ 20${\beta}$ OHP 0.5-1.0${\mu}$g/kg BW) and luteinizing hormone-releasing hormone analogue pellet (LHRHa pellet 63-81 ${\mu}$g/kg BW) In all the trials HCG injection succeeded in inducing ovulation at 265-300 IU/kg BW. HCG at 620 or 678 IU/kg BW was ineffective in inducing a good rate of ovulation. Final maturation and ovulation were not stimulated by treatment of 17 ${\alpha}$ 20${\beta}$OHP and LHRHa pellet. These results suggest that HCG injection (around 300 IU/kg BW) is more effective compared with LHRHa pellet implantation.

The Oogenesis of Chinese minnow, Leuciscinae, Teleostei (경골어류 황어아과 버들치의 난자형성과정)

  • Chinese minnow, Rhynchocypris oxycephalus is a teleost belonging to Leuciscinae, Cyprinidae. The oogenesis and ultrastructure of egg envelope in Chinese minnow were investigated by light and electron microscopes. The ovary was of white yellowish and ellipsoidal shape with the major axis 30 mm and the minor axis 7mm. Cytoplasm of oogonia was basophilic and many nucleoli were located at inside of nuclear membrane. In primary oocytes, yolk vesicles were distributed only in the marginal area and egg envelope was not formed on the outside of an egg. In secondary oocytes, the egg envelope was formed and yolk vesicles in the cytoplasm were increased than the earlier stage. The basophilic substance of cytoplasm was changed to acidic. In case of matured egg, thickness of egg envelope and size of egg were increased. The yolk vesicles were changed to yolk mass in accordance with development. The outer surface of egg envelope was covered by microvilli-structures, and had a micropyle on the area of animal pole. Egg envelope consisted with 2 layers, an adhesive outer layer with microvilli-structures and fibrillar inner layer. In conclusion, the oogenesis of Chinese minnow was characterized by the increase in cell size, the formation and accumulation of yolk, and the decrease of basophilic substance in the cytoplasm. The oogenesis of Chinese minnow seems to share common patterns in Cyprinidae, but these ultrastructural unique characters of egg envelope can be utilized in taxonomy of teleost.

Ecdysteroid Titer during Metamorphosis and the Effect of Ecdysteroid on Oocyte Develoment on Phormia regina (검정금파리의 변태기에 따른 엑디스테로이드와 난세포성숙에 미치는 엑디스테로이드의 효과)

  • The ecdysteroid titers of representive developmental stages of the blackblow fly, Phormia regina, were determined by radioimmunoassay and the effect of ecdysteroid on the oocyte maturation was investigated. Prior to every molts ecdysteroid levels began to increase sharply, suggesting ecdysteroid was the major component for egg-larval, larval-larval, and larval-pupal transformation. A difference in the levels of ecdysteroid between male and female was ob¬served during adult life span. Following the protein meal, ecdysteroid in the females increased rapidly to a maximum at 96 hr of age when terminal oocyte fully matured. Effect of ecdysteroid on oocyte development was determined for control and ecdysone-treated female flies after the liver-feeding. The growth of oocyte in the flies treated by $\mu$g of ecdysone, along with the control flies, was not facilitated. When the flies treated by 5 $\mu$g of ecdysone, however, duration of oocyte maturation was shorter than those of other two groups. This can be suggested that oocyte development in P. regina is due to the critical level of ecdysone.
