Effects of Solarization and Green Manual Crops for Promotion of Fall Patato Grawth (가을감자 生産性向上을 위한 綠肥作物栽培와 太陽熱 土壤消毒의 效果)

  • This study was conducted to effect promotion of fall potato growth in the place which green manual crops had been previously cultivated, crushed and tilled and polyethylene transparent vinyl had been mulched from Aug. 5 to Aug. 29 in 1997. The maximum temperature of below-ground at the depth of 10cm was $42^{\circ}C$ during the mulching period, and the pH and the organic compound content of soil was very increased by the cultivation of green manual crops and the mulching of polyethylene transparent vinyl. The total yields of green manual crops which had been previously cultivated were as the following orders; the hybrid of sudan grass, soybean, alfalfa, otherwise, fresh weight of stems and the tuber yield of fall potato were significantly affected in the plot of soybean + fall potato after the soil solarization with the mulching. As these results, it was possible to improve crops growth and to sterilize soil by the mulching treatment of polyethylene vinyl after cultivation of green manual crops during the high temperature of the summer season.

Growth and Yields of Korean Soybean Cultivars in Drained-Paddy Field (국내 육성 콩 품종의 논 재배에 따른 생육반응과 수량성)

  • With various Korean domestic soybeans, growth and yields analysis were conducted to select the suitable soybean cultivars for cultivation in paddy field. Distinctive aspects of the soybean growth were observed in paddy field such as retarded growth of top plants and roots, relatively higher T/R ratio followed by overgrowth of top plant. However, growth and yields were significantly different among the cultivars showing 134kg/10a in Paldokong and 385 kg/10a in Doremikong. At V5. and R2 stage, highly positive correlations $(r=0.76^{**}\~0.91^{**})$ were observed between leaf area and dry weight of top plant and/or root. T/R ratio was negatively correlated with dry weight of root $(r=-0.37^*)$ at V5 stage, while significantly correlated with leaf area $(r=-0.37^{**})$ and dry weight of top plant $(r=0.65^{**})$at R2 stage. Among the characters, only 100-seed weight was significantly correlated with yield. Considering the growth characters, 37 cultivars could be included in 3 different groups and genotypic properties such as maturity and growth habit were similar in each group. Nine cultivars in group 1 showed retarded growth from V5 to R2 stage, relatively lower T/R ratio, and good seed ripening. Average yields of the cultivers was 257kg/10a. In group 2, 12 cultivars showed higher T/R ratio due to overgrowth of top plant and lowest average yields (230 kg/l0a) due to poor seed ripening. Sixteen cultivars in group 3 grew fast from V5 to R2 stage representing late maturity traits, low T/R ratio, and good seed ripening. Average yields of the cultivars was highest among groups showing 270kg/l0a. In results, stable self-sufficiency of soybean yields could be expected by selective cultivation with high yielding cultivars ranging from 301 to 385kg/10a, such as Shinpaldalkong 2, Sohokong, Doremikong, Keumkangkong, Bukangkong, Dajangkong, and Geomjeongkong 2, or with cultivars included in group 3.

The characterization of transgenic Chrysanthemum under low temperature condition (저온저항성 유전자가 도입된 국화 형질전환체 특성)

  • Previous studies on genetic transformation of chrysanthemum using cold regulated gene (BN115) have been conducted and the PCR and Real-Time PCR based method to determine the presence of the transferred cold regulated gene in the chrysanthemum was established. To check whether over-expression of BN115 gene in transgenic chrysanthemum will enhance their tolerance to cold stress, the transgenic chrysanthemum were grown under low temperature condition and several cold signalling including growth characteristics, stoma size and shape, SPAD value and ion leakage test were investigated. The transgenic chrysanthemum in the low temperature growth chamber grow much faster in term of the height, number and size of the leaves than those of wild-type plants and damage of transgenic plant caused by the low temperature was much less than that of wild-type plants. The stoma type and size of transgenic plant leaves grown at $5^{\circ}C$ were much similar to of wild-type plant cultured on $25^{\circ}C$ It has been found that SPAD value of transgenic plants was much higher than those of wild-type, but the EC density being lower under low temperature condition.

Current status and outlook on genetic transformation of fruit trees in Korea (과수 형질전환 국내 연구 동향 및 전망)

  • The paper reviewed research status and outlook of genetic transformation in fruit trees in Korea. Genetic transformation has been long considered as an alternative technique overcoming limitation of conventional breeding and conducted since early 1990's. An efficient genetic transformation was established with major cultivars of apple, even in Fuji and Gamhong, and some transgenic apple plants have been transferred into the greenhouse for further analysis of gene expression. A few successes of genetic transformation have been reported and application to a variety of cultivars tried in citrus and kiwifruit. Successful genetic transformation has not been reported in the other fruit trees including grapevine, yet and it is considered being at the level of research. Those factors including replacement of antibiotics as a selective agent, use of transgenic rootstocks and manipulation of gene expression at proper parts and developmental stages have been prerequisites for the rapid commercialization of transgenic fruit plants.
