식물배양세포를 이용한 항산화연구

  • To understand the antioxidative mechanism in plant cell cultures, we investigated the levels of antioxidant enzymes and low molecular antioxidants in 100 cell lines derived from different plant species. SOD and POD activities in plant cell lines were significantly higher than intact plants. The cell lines from sweet potato (Ipomoea batatas) and cassava (Manihot esculeanta) showed the highest POD and SOD activities, respectively, suggesting that the cell cultures of sweet potato and cassava are good biomaterials for the mass production and molecular study of antioxidant enzymes. The average ascorbate content in plant cell lines was several hundred times lower than intact plants, whereas the glutathione content was 2-3 times higher than plants. Interestingly, the ratio of reduced and oxidized ascorbate and glutathione was different from plant species. In conclusion, the results strongly suggest that plant cell cultures are good biomaterials for the study of antioxidative mechanism and the production of useful components including antioxidants.

We developed the funtional lighting fixture which emit the lighting, negative(-)ion and Infraredray. The negative(-)ion and Infraredray refresh the air pollition and will arise nature healing power lot the human body and activate the body cell, and makes the good blood circulation and then we had the experiment to the Biological effect by the funtional lighting fixture.

Study on Construction of Soil Structure in Creation of Man-made Tidal Flat (인공 간석지 창출에 있어서 토양조성에 관한 연구)

  • The purpose of this study was to identify the controlling factors to construct tidal flat ecosystems having similar characteristics as natural ones. We transplanted the soil in a constructed tidal flat to a natural one and vice versa. Parameters monitored after these transplantations were silt content, organic matter, bacterial population and oxidation-reduction potential. Moreover, the relationship among silt content, organic matter and bacterial population was investigated by laboratory column experiment. The silt content, organic matter, bacterial population and vortical profile of oxidation-reduction potential in the soil transplanted from the constructed tidal flat to the natural one changed to similar values to those In the natural one. On the contrary, all the parameters for the soil transplanted from the natural tidal flat to the constructed one changed to similar values as those in the constructed one. The silt contents in these two transplanted soils were in proportion to the organic carbon contents and bacterial population. Similarly, the bacterial population in laboratory column experiment increased with the increase in silt and organic matter contents. It seemed to be important to select a place to enhance accumulative of silt and/or to maintain the silt content by hydrodynamic control of seawater in order to construct a tidal flat having similar characteristics as natural one.

Biological Activities of Rosaceae Plants Extracts (장미과 식물 추출물의 생물학적 활성)

  • DPPH radical-generating system was used to evaluate the antioxidant properties of the Rosaceae. The inhibitory effects of ethanolic extracts from Rosaceae plants were investigated on melanin biosynthesis which is closely related to hyperpigmentation. Of the Rosaceae extracts, Prunus sargentii, Rubus coreanus, Chaenomeles sinensis, Photinia glabra and Pyrus pyrifolia showed a potent inhibition of tyrosinase, the enzyme which converts 3-(3,4-dihydroxyphenyl) alanine (dopa) to dopachrome in the melanin biosynthetic process. Furthermore, MMT assay was used to check the cytotoxicity of extracts on the human foreskin fibroblast cell line, Hs68. Among the Rosaceae, bark of Prunus sargentii, bark wood of Photinia glabra and all parts or Chaenomeles sinensis showed more than $50\%$ inhibition of mushroom tyrosinase activity at 100 ug/mL and more than $80\%$ of strong DPPH radical-scavenging activity at 10 ug/mL. In audition to, they had no cytotoxic activity on Hs68. These results suggest that these extracts might be except a controler in pigmentation.

Combined Effect of 2,4,6-trinitrotoluene(TNT) and Cadmium on Uptake and Phytotransformation of TNT by Abutilion avicennae (TNT(2,4,6-trinitrotoluene)와 카드뮴의 복합오염이 어저귀의 TNT흡수 및 생물학적 전환에 미치는 영향)

  • Most of army depots contaminated with co-contaminants, 2,4,6-trinitrotoluene(TNT) and heavy metals. In phytoremediation for the TNT, heavy metals may inhibit mineralization, transformation and sequestration of TNT by the plant. We studied effect of cadmium on TNT degradation and transformation by Abutilion avicennae in hydroponic cultures. When cultured in 20 mgTNT/L and 1.3 mgCd/L, the plant displayed phytotoxicities; reduction of leaf fresh, leaf roil, chlorosis, leaf loss and fresh weight loss. Phytotoxicity was severer in the combined contaminants than in single contaminant. Because A. avicennae uptake just a little cadmium, 1.3 mgCd/L included in the TNT medium did not influece significantly TNT transformation, translocation and distribution by A. avicennae. Therefore, the soil solution containing cadmium would not affect TNT degradation by Abutilion avicennae in Army depots polluted with TNT.

  • Adsorption experiments for As(V) and As(III) onto the surfaces of aerobic Pseudomonas aeruginosa, which can be readily isolated from natural media, were conducted under nutrient-absent conditions. While a small amount of As(III) was adsorbed on the bacterial cell surfaces, As(V) was not effectively removed from the solution through adsorption. The result was likely due to the electrostatic repulsion between anionic compounds of aqueous As(V) and cell surfaces of f aeruginosa. However, the bacteria forming biofilm reduced a large amount of aqueous As(V) to As(III), which indicated that microorganisms in most oligotrophic, natural geologic settings can mediate the behavior of aqueous As. Biobarriers designed to remove the various heavy metals in contaminant plume may practically lead to the enhancement of toxicity and mobility of As.

Enantioconvergent Hydrolysis of Racemic Epoxides for Production of Enantiopure Epoxides and Vicinal Diols using Epoxide Hydrolases (에폭사이드 가수분해효소에 의한 광학수렴반응을 이용한 광학활성 에폭사이드 및 Vicinal Diol 제조)

  • One drawback of conventional kinetic resolution of racemic epoxides by epoxide hydrolase (EH) is that the theoretical yield can never exceed 50%. This 50% limitation can be overcome by using enantioconvergent process, in which both enantiomers of the racemic epoxide are transformed via stereochemically matching pathways into a single enantiopure diol as the sole product in 100% theoretical yield. In order to make a single enantiopure vicinal diol, the two enantiomers of the racemic epoxide must be hydrolyzed with retention and inversion of configuration each other. The EHs should be enantio- and regiospecific at the same time. The enantioconvergent hydrolysis with EHs and relevant biotransformation for preparing enantiopure epoxides and vicinal diols with a high yield are reviewed.

Effect of Oxygen on the Browning of Soy Sauce During Storage (간장의 저장 중 갈색화반응에 대한 산소의 영향)

  • Raw soy sauce and raw soy sauce added with several Maillard reactants (glucose, xylose, glycine), preheated or unheated, were stored under aerobic or anaerobic conditions to investigate the effect of oxygen on the browning of soy sauce. All experimental groups, except xylose-added groups, stored under aerobic condition browned about 2.5 times as much as those stored under anaerobic condition. Soy sauce added with xylose stored under aerobic condition browned about 1.5 times as much as that stored under anaerobic condition. Contrarily, there have been no consistent differences in the browning except the initial 10% difference between preheated and unheated samples, implying that biological (microbiological and enzymatic) browning was not important in the browning of commercially brewed soy sauce.

Effect of Saponin with Antioxidant Activity on Matrix Metalloproteinase in Human Dermal Fibroblasts (항산화 효능을 가진 사포닌이 사람섬유아세포에서 기질 금속 단백질 분해효소에 미치는 영향)

  • Saponin is a main component of ginseng widely known as an oriental traditional medicinal ingredient. A variety of biological effects of saponin has been reported, but its action related to skin regeneration has remained unclear so far. In this study, the effect of saponin on matrix metalloproteinase as well as its antioxidant effect in cell free system was examined in human dermal fibroblasts. First of all, as a result of investigating the effect of saponin on cell viability using MTT assay, it was shown to increase cell viability below 10 ${\mu}g$/ml, but it also showed cytotoxicity above 25 ${\mu}g$/ml. The antioxidant effect of saponin was exerted by inhibition of $H_2O_2$ in addition to reducing power above 1 ${\mu}g$/ml. In particular, saponin showed a protective effect on DNA oxidation. Furthermore, it was observed that saponin activates MMP-2 and increases MMP-1 activity in gelatin and casein zymography analyses, respectively, indicating that saponin could have potential a therapeutic agent for anti-aging and skin regeneration.
