Selection and Identification of a Hyperparasite, Ampelomyces quisqualis 94013 for Biocontrol of Cucumber Powdery Mildew (오이 흰가루병 생물적 방제를 위한 중복기생균 Ampelomyces quisqualis 94013의 선발 및 동정)

  • 308 isolates of Ampelomyces sp. were isolated from powdery mildew fungi of 73 plant species in Korea for selection of biocontrol agents. An isolate 94013 isolated from powdery mildew fungus of red bean was selected as an effective biological control agent against cucumber powdery mildew in greenhouse. The morphological characteristics of the isolate is as follows. Pycnidia were sub-globose or elongated to pyriform, pale to dart brown, $52.5{\sim}82.5\;{\times}\;35.0{\sim}47.5\;(ave.\;62.5{\times}40.5){\mu}m$, and conidia were guttulate, straight cylindrical to fusiform, pale brown, $5.0{\sim}8.0\;{\times}\;2.5{\sim}4.3\;(ave.\;6.0{\times}3.0){\mu}m$. The isolate 94013 was identified as Ampelomyces quisqualis by morphological characteristics and rDNA ITS sequencing. The isolate A. quisqualis 94013 was different from that of the commercial product $AQ10^R$ in the rDNA ITS sequence.

Changes in Saponins Sugars and Amino Acids of White Ginseng during Storage Following Gamma Irradiation and Phosphine Fumigation (감마선과 Phosphine 처리된 백삼의 저장 중 사포닌, 당 및 아미노산 함량의 변화)

  • To investigate an alternative technique to phosphine fumigation, being used for controlling storage insects of dried ginseng, comparative effects of gamma irradiation (5 kGy) and fumigation on the content of moisture, saponins, free sugars, and amino acids of commercially-packaged white ginseng were evaluated immediately after and at the 6th month of storage at 20$^{\circ}C$ and 70% RH after both treatments. The initial moisture content (8.44%) of the sample was not changed with the treatments, and maintained the 10% levels of moisture until 6 months of storage. Major ginsenosides were stable to both treatments, but they showed some reduced content in the control and fumigated samples at the 6th month of storage. Gamma irradiation caused some decrease in the content of free sugars and the subsequent storage of the sample resulted in a further reduction in their content in the order of irradiated, fumigated and control samples. Although irradiation and fumigation brought about a decrease in the content of fee amino acids by about 5%, however the storage period for 6 months was shown more influential than the treatments used for the improvement of biological quality in stored white ginseng.

Physiological Characteristics of Actinomycetes Isolated from Turfgrass Rhizosphere (잔디 근권에서 분리된 Actinomycetes균주의 생리학적 특성)

  • Total 443 isolates of actinomycetes were isolated from turfgrass rhizosphere as potential biological control agents. The two isolates (S11 and S4) showed highest cellulase activity with compared to the other isolates that exhibited a clear zone of 1.2 mm around the colony on cellulose agar medium. S12 strain appeared the most active chitin degrading, which exhibited a 1.2 mm of clear zone. The highest proteolytic activity on skim milk agar was which exhibited a 7.5 mm of clear zone by S2 strain. S1 strain from the soli showed siderophore production ability, which exhibited a 0.6 mm of large clear zone on chrome azurol S agar. The antifungal activity of the volatile compound producing by 4 selected actinomycetes was investigated that inhibition rate against Rhizoctonia solani AG2-2 and Sclerotinia homoeocarpa. Growth inhibition effect of S8 isolate against S. homoeocarpa was appeared to 94.8%, S2 to 76.9%, S5 to 46.1% and S12 to 43.5%. The significant inhibition effects on mycelial growth of S. homoeocarpa were shown on media with four strains. The inhibition effect was the highest with S8 strain treatment at 94.8%.

Toxicity and Characteristics of Antifungal Substances Produced by Bacillus amyloliquefaciens IUB158-03 (Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 생화학적 특성 및 독성)

  • The purified antifungal substances produced by Bacillus amyloliquefaciens IUB158-03 was positive to ninhydrin but negative to aniline, suggesting that the antifungal substance could be a peptide. FAB-MS, UV adsorption spectrum, and amino acid composition analysis revealed that the molecular weight of the antifungal substance was 1042 and that maximal adsorption was at 220 nm and 277 nm. The antifungal substance was composed of $Asn_3$, $Gln_2$, $Ser_1$, $Gly_1$, and $Tyr_1$. The composition and structural characteristics of antifungal substance were analysed by $^1H$-NMR spectrum, $^1H$-COSY, HMQC, which revealed that the compound belongs to the iturin A family. Temperature and pH had little effect on the stability of the antifungal substance in the ranges of $-70{\sim}121^{\circ}C$ and pH 6.0~10.0, respectively. It showed strong antibiotic activity against fungi. An in vitro cytotoxicity test using NIH3T3 cell showed that the antifungal substance does not have cytotoxicity. The number of circulating leukocytes and the hematobiological analysis of the mice administered with the antifungal substances was similar to those of the control group, indicating no cytotoxicity in vivo. Therefore, the antifungal substances extracted from culture broth of Bacillus amyloliquefaciens IUB158-03 have future potential as biocontrol agents against plant diseases caused by fungi.

  • In this study, strains isolated from soil samples collected from Busan, Changwon, and Jeju Island were examined to verify their abilities of phosphate solubilization and nitrogen fixation, production of indole-3-acetic acid (IAA), siderophore, and 1-aminocylopropane-1-carboxylyic acid (ACC) deaminase in order to select strains that promote plant growth and play a role in biocontrol of pests or pathogens. According to the results of this study, most of the isolated strains were found to have ability of phosphate solubilization, nitrogen fixation, IAA production, siderophore production, and production of ACC deaminase. These isolated strains might help plant growth by directly improving absorption of nutrients essential for phosphate solubilization and nitrogen fixation. In addition, they can promote plant growth and control resistance to plant diseases through extracellular enzyme activity and antifungal activity. In addition, most of the selected strains were found to survive in various environmental conditions such as temperature, salinity, and pH. Therefore, Pseudomonas plecoglossicida ANG14, Pseudarthrobacter equi ANG28, Beijerinckia fluminensis ANG34, and Acinetobacter calcoaceticus ANG35 were finally selected through a comparative advantage analysis to suggest their potential as novel biological agents. Further studies are necessary in order to prove their efficacy as novel biological agents through formulation and optimization of effective microorganisms, their preservation period, and crop cultivation tests.

Status of Fusarium Wilt Incidence on Summer Radish and Etiological Characteristics of the Causal Fungus in Korea (고랭지 여름 무에서 시들음병 발생 현황과 병원균의 병원학적 특성 연구)

  • Incidence of Fusarium wilt was surveyed in fields of summer radish in Gangwon province in Korea in 2018 and 2019. The disease started in early July and spread rapidly in hot summer of late July and August and in severe case, reached up to 80% in a field in Gangneung area. Symptoms in the seedling stage include poor growth and browning of internal tissue of root. During mid-growth, the leaves of diseased plant turned yellow over time, the surface of the roots changed from white to blackish, and the vascular tissues turned brown. A total of 23 isolates was obtained from the diseased plants and identified as Fusarium oxysporum f. sp. raphani by elongation factor-1α and intergenic spacer sequence analysis. Pathogenicity of the isolates was tested by artificial inoculation to the radish and other plants. All the isolates tested were pathogenic to radish plant, although there were differences in virulence on radish 11 cultivars. However, the isolates were not virulent to other plants except some cruciferous vegetables including Brussels sprouts, rocket, stock, and turnip. The results of pathogenicity test showed that it is necessary to rotate with crops other than cruciferous vegetables in order to prevent Fusarium wilt from radish fields.

Complete Genome Analysis of Hyphantria cunea Nucleopolyhedrovirus Isolated in Korea (한국에서 분리한 미국흰불나방 핵다각체병 바이러스의 전장 유전체 분석)

  • The morphology and whole genome sequence of Hyphantria cunea nucleopolyhedrovirus W1 (HycuNPV-W1) isolated in Korea were analyzed for the use as an eco-friendly control agent against H. cunea. The HycuNPV-W1 had irregular tetrahedral polyhedra with a size of 1.5-2.2 ㎛ which is similar to that of previously reported HycuNPV isolated in Korea. As a result of whole viral genome analysis, HycuNPV-W1 was composed of 131,353 bp, which is 1,606 bp shorter than that of the previously reported HycuNPV. The G+C content was 45% and six of the homologous repeated regions were found, so there was no significant difference from the previous report. As a result of ORF analysis, HycuNPV-W1 contains total of 145 ORFs which is three ORFs less than the previous report, while two ORFs were exclusively found in HycuNPV-W1. The functions of these ORFs remains unclear and are not considered to have a significant influence on the characteristics of the HycuNPV. The genome vista analysis showed that the overall sequence identity between HycuNPV-W1 and the previously reported HycuNPV was very high. The whole genome of HycuNPV-W1 analyzed was found to be similar to those of the previously reported HycuNPV, however, it is supposed to be a novel resource in Korea with different isolate.

  • Several species of the genus Aphidius are used in biological control programs against aphid pests throughout the world and their behavior and physiology are well studied. While the importance of sensory organs in their behavior is understood, their antennal structure remains largely unknown. In this study, the external morphology and distribution of the antennal sensilla on the antennal of both female and male adults of A. colemani were described using scanning electron microscopy (SEM). Generally, the filiform antennae of males ($l,515.2{\pm}116.5\;{\mu}m$) are longer than females ($l,275.1{\pm}103.4\;{\mu}m$). Antennae of this species are made up of radicle, scape, pedicel and flagellomeres. Male and female antennae differed in the total number of flagellomeres as 15 in males and 13 in females. The result of SEM observation was characterization and grouping of seven types of receptors into morphological classcs: Bohm bristles, Sensilla placodea, 2 types of S. coeloconica and S. trichodea, and S. basiconica as with a tip pore and with wall pores. In addition, the possible functions of the above sensilla types arc discussed in light of previously published literature; mechanoreception (Bohm bristles, S, coeloconica I & II and S. trichodea) and chemoreception (S. placodea, S. basiconica type I & II).

Antifungal Activity of Bacillus sp. AM-651 Against Phytophthora capsici (고추역병 유발병원균 Phytophthora capsici에 대한 Bacillus sp. AM-651의 항진균활성)

  • Biological antagonists of Phytophthora capsici were isolated from soil in Gyeongbuk, Korea. Among the isolated bacteria, a Bacillus sp. was identified from l6S rDNA sequence analysis and named Bacillus sp. AM-651. Bacillus sp. AM-65l strain which can strongly a antifungal activity against Phytophthora capsici. Culture conditions for the maximum production of the antagonistic substance were optimized. The production of antibiotic were high on modified Davis mineral medium pH 7 at $30^{\circ}C$. The medium for highest production of the agonistic substance optimized. It is composed the best activity on glucose, $(NH_4)_2SO_4$ and $K_2HPO_4$ at 0.5%, 0.1%, and 0.7%, respectively. By time course of culture solution selected Bacillus sp. AM-65l, the culture solution after 48hrs had strongly growth inhibition rate against P. capsici. And culture solution of Bacillus sp. AM-651 was stable within a pH range $5{\sim}11$ and temperature range $4{\sim}70^{\circ}C$. Bacillus sp. AM-651 cultured broth shown fungal growth inhibitory activity against B. sorokiniana, B. cinerea, R. solani avove and beyond P. capsici and comparatively showed a high activity against C. gloeosporioides, B. dothidea, B. cinerea and F. graminearum by agar diffusion method.

In vivo Antifungal Activity of Pyrrolnitrin Isolated from Burkholderia capacia EB215 with Antagonistic Activity Towards Colletotrichum Species (탄저병균에 대하여 길항작용을 보이는 Burkholderia cepacia EB215로부터 분리한 Pyrrolnitrin의 항균활성)

  • An endophytic bacterial strain EB215 that was isolated from cucumber (Cucumis sativus) roots displayed a potent in vivo antifungal activity against Colletotrichum species. The strain was identified as Burkholderia cepacia based on its physiological and biochemical characteristics, and 16S rDNA gene sequence. Optimal medium and incubation period for the production of antifungal substances by B. cepacia EB215 were nutrient broth (NB) and 3 days, respectively. An antifungal substance was isolated from the NB cultures of B. cepacia EB215 strain by centrifugation, n-hexane partitioning, silica gel column chromatography, preparative TLC, and in vitro bioassay. Its chemical structure was determined to be pyrrolnitrin by mass and NMR spectral analyses. Pyrrolnitrin showed potent disease control efficacy of more than 90% against pepper anthracnose (Colletotrichum coccodes), cucumber anthracnose (Colletotrichum orbiculare), rice blast (Magnaporthe grisea) and rice sheath blight (Corticium sasaki) even at a low concentration of $11.1\;{\mu}g/ml$. In addition, it effectively controlled the development of tomato gray mold (Botrytis cinerea) and wheat leaf rust (Puccinia recondita) at concentrations over $33.3\;{\mu}g/ml$. However, it had no antifungal activity against Phytophthora infestans on tomato plants. Further studies on the development of microbial fungicide using B. cepacia EB215 are in progress.
