A Study on Verification for the Design of Bio Safety Level 3 Laboratory by using Multi-zone Simulation (멀티죤 시뮬레이션을 이용한 생물안전 3등급(BSL3)시설의 설계 검증에 관한 연구)

  • In Korea, since the implementation of the LMO Law, the interest of biosafety level 3(BL3) lab. is increasing. In this study, using CONTAM which is applying multizone modelling, the multizone simulation for design verification of BL3 lab. is performed. In BL3 lab., because required air change rate is greater than general estimated air-conditioning load and it is difficult to maintain room pressure difference efficiently, to maintain pressure difference between laboratory rooms is important through sealing condition of doors and proper airflow control of laboratory rooms. In this study, about BL3 lab,(M. tuberculosis research lab.), the multizone simulation for four kind of biohazard scenarios is carried out in the case of unexpected spread of contaminants in the laboratory room, anteroom, corridor and inside of BSC. Multizone simulation results show that these approach methods are used as a tool for the design and verification of BL3 lab.

  • 일반적으로 시설을 설계함에 있어서 고정하중(자중)은 부재단면이 결정되기 전이므로 추정에 의한 값을 사용한다. 그러나 생물생산시설에 관한 기준이 마련되어 있지 않기 때문에 국내 실정에 맞는 자료가 요청된다. 또한 구조설계 과정은 복잡하고 전문적 지식을 요하므로 전문가에 의해서 수행된다. 그러나 생물생산시설의 규모나 형태는 비슷한 경우가 많으므로 대표적인 몇가지 시설에 대하여 구조해석을 실시하고, 이것을 일반화함으로써 비전문가인 농민이나 생물생산분야 종사자가 간편하게 시설의 구조적 안전성을 진단할 수 있을 것으로 생각한다.(중략)

Effect of Wildlife Access on Microbial Safety of Irrigation Water Used in the Cultivation of Chinese Cabbage in Goesan (야생동물 출입이 괴산 지역 배추 재배 농업용수의 미생물 안전성에 미치는 영향)

  • Water is an important component in the production of fresh produce. It is mainly used for irrigation and application of pesticides and fertilizers. Several outbreaks cases related to fresh produce have been reported and water has been identified as the most likely source. On the other hand, wildlife has been identified as a possible source of the waterborne pathogens. The purpose of this study was to investigate the effect of wildlife access on irrigation water used in the cultivation of Chinese cabbage in Goesan. The frequency of wild animals access to upstream water source and the contamination level of bacteria such as Escherichia coli and Enterococci of irrigation water used in Chinese cabbage farm was examined. A total of 37 wildlife including the wild bear and water deer were observed in upstream of water source during the cultivation of Chinese cabbage. The result indicated the presence of hygienic indicator bacteria from the upstream where there is no human access. The contamination range of coliforms, E. coli, and Enterococcus spp. Detected in the irrigation water were 2.13~4.32 log MPN / 100 mL, 0.26~2.03 log MPN / 100 mL, and 1.43~3.49 log MPN / 100 mL, respectively. Due to low water temperatures, the contamination levels of coliform bacteria and E. coli in the irrigation water during harvesting time was lower compared to those recorded during transplanting of Chinese cabbage. However, no significant difference was detected in the number of Enterococci during the cultivation of Chinese cabbage. The results indicated the need to manage the microbial risk in irrigation water to enhance safety in cultivation of Chinese cabbage.

Efficacy of Commercial Sanitizers for the Inactivation of Listeria monocytogenes on the Processing Equipment for Enoki Mushrooms (팽이버섯 재배 현장에서 Listeria monocytogenes의 성장을 억제하기 위한 살균 처리 기술 개발)

  • The consumption of enoki mushrooms has been associated with cases of listeriosis produced by Listeria monocytogenes, highlighting the significance of sanitizing food-contact surface, such as the velcro used in welding processing of enoki mushrooms, to ensure microbial safety. We investigated the inhibitory activity of nine chemical disinfectants at regular concentrations against L. monocytogenes isolated from a mushroom farm environment. The bacterial suspension was prepared in phosphate buffered saline and mushroom extract broth and inoculated onto the velcro surface. After inoculation, most disinfectants reduced the initial 8 log CFU/coupon concentration by less than 2 log CFU/coupon during a 5-min treatment. Slightly acidic hypochlorous water showed a reduction of approximately 4 log CFU/coupon when tested for more than 30 min at the maximum allowable concentration of 200 mg/L. Sodium hypochlorite solution showed a reduction of approximately 5 log CFU/coupon when used at 100 mg/L for 60 min. Peracetic acid, at the maximum allowable concentration of 300 mg/L, showed the most effective reduction of 5 log CFU/coupon or more when the surface was treated with 37.5 mg/L for 30 min. These results indicate that peracetic acid can be used as the disinfectant strategy to control cross-contamination of L. monocytogenes on the velcro surface of plastic wrappers used in the welding processing of enoki mushrooms.
