Morphological and Molecular Characterization of Alternaria Isolates from Solanaceous Crops (가지과 작물에서 분리한 Alternaria 속 균의 형태적, 분자생물학적 특징)

  • Twenty five isolates of Alternaria were obtained from various solanaceous crops in Korea. For all isolates, morphological characteristics of the conidia were determined and compared with those of representative isolates of A. solani and A. tomatophila. A selection of the isolates and the representative Alternaria isolates were evaluated for Pathogenicity to potato, tomato, egg plant and red pepper. Molecular characteristics of 17 isolates of Alternaria inculding the representative isolates were determined using sequence analysis of IRS rDNA and histone H3 gene, and URP-PCR analysis. Based on morphological characteristics, the isolates from the solanaceous crops were grouped as identical or very similar to either A. tomatophila (ATO), A. solani (ASO), and unidentified Alternaria sp. (ASP). Isolates of ASO were moderately pathogenic to all the solanaceous crops tested, but ATO isolates were highly pathogenic to tomato and the ASP isolate was pathogenic only to potato. Among the molecular markers used in this study, the URP-PCR analysis was found to be appropriate for taxonomic resolution of these species. Based on the conidial morphology, pathogenicity test and molecular characteristics, A. tomatophila (early blight of tomato) could be distinguished from A. solani (early blight of potato), and the Alternaria sp. (ASP) from potato, which was closely related to ASO in conidial morphology, was considered as a new species.

Screening of effective control agents against bacterial soft rot on Chinese cabbage in alpine area (고랭지 배추 무름병 방제를 위한 우수약제 선발)

  • Bacterial soft rot by Erwinia carotovora subsp. carotovora is one of the diseases causing the biggest problem in Chinese cabbage. Chemical screening was conducted to select effective agents for controlling bacterial soft rot. Control effect of antibiotics, plant activator, and Biokeeper (avirulent Erwinia) to soft rot were tested by in vitro assay, nursery test, and field experiment. The in vitro assay was done by paper disc method and potato slice method. The nursery test was performed by using mineral oil inoculation method with consistent disease induction. The in vitro assay showed that streptomycin, oxolinic acid, bronopol, and copper hydroxide significantly suppressed the growth of pathogenic bacterium and the decomposition of potato slice. However, plant activators including acibenzolar-S-methyl did not show the suppressive effect on the growth of pathogenic bacterium and the decomposition of potato slice. When applied by the nursery test condition using mineral oil inoculation method with Chinese cabbage 'Kangruckyeurum', Biokeeper, oxolinic acid, antibiotics streptomycin, validamycin, and copper compound provided 83.5%, 95.2%, 91.2%, 57.5% and 79.9% in control efficacy, respectively. However, the control effect of acibenzolar-S-methyl showed to be low to cause phytotoxicity. Also acibenzolar-S-methyl showed a significant control effect in the field experiment with Chinese cabbage 'Sanchon' in 2000, but the field experiment with Chinese cabbage 'Kangruckyeurum' in 2001 revealed it had phytotoxicity to Chinese cabbage. Such a difference was considered to be caused by differences in phytotoxic reaction of Chinese cabbage cultivars to the chemical. Streptomycin+copper, copper hydroxide and Biokeeper showed 79.7%, 71.9% and 60.9% in control efficacy, respectively, in the field experiment with Chinese cabbage 'Sanchon' in 2002.

The Effect of Psychological Factors on Postoperative Pain in Gastric Tumor Patients after Endoscopic Submucosal Dissection (내시경 점막하 박리법을 시술 받은 위종양 환자에서 심리적 요인이 시술 후 통증에 미치는 영향)

  • Objectives : Since pain in cancer patients is an important factor that greatly affects the quality of life and prognosis, many attempts have been actively made to reduce the pain. Despite continuous effort on reducing pain after Endoscopic submucosal dissection (ESD), research has not been done on psychological factors as much as on biological factors affecting pain. The objective of this study is to investigate the psychological factors affecting postoperative pain in gastric tumor patients who underwent gastric ESD. Methods : 91 gastric tumor patients who visited National Health Service Ilsan Hospital in Korean between May 2015 and June 2016, and received ESD were evaluated. Baseline characteristics including sociodemographic factors, anxiety, depression, and resilience were evaluated before the procedure. Multivariate logistic regression was done to analyze factors affecting postoperative pain. Results : The group with high postoperative pain showed lower alcohol consumption and higher depressive symptom scores than the group with low postoperative pain. Also, the group with high postoperative pain showed lower total resilience score with lower subtotal scores in self-control and positive item. Multivariate logistic regression analysis of the postoperative pain showed that patients with lower score in self-control of resilience [odd ratio (OR), 0.911 ; 95% CI, 0.854-0.971, p=0.004) reported more pain after ESD. Conclusions : This study showed that patients with lower self-control ability of resilience felt more pain after ESD. Among the psychological factors evaluated in this study, resilience of an individual seems to have effect on pain.

Distribution and Trend Analysis of the Significant Wave Heights Using KMA and ECMWF Data Sets in the Coastal Seas, Korea (KMA와 ECMWF 자료를 이용한 연안 유의파고의 분포 및 추세분석)

  • The coastal wave environment is a very important factor that directly affects the change of coastal topography, the habitat of marine life, and the design of offshore structures. In recent years, changes in the wave environment due to climate change are expected, and a trend analysis of the wave environment using available data sets is required. In this paper, significant wave heights which are measured at six ocean buoys (Deokjeokdo, Oeyeondo, Chibaldo, Marado, Pohang, Ullengdo) have been used to analyze long-term trend of normal waves. In advance, the outlier of measured data by Korea Meteorological Administration have been removed using Rosner test. And Pearson correlation analysis between the measured data and ECMWF reanalysis data has been conducted. As a results, correlation coefficient between two data were 0.849~0.938. Meanwhile, Mann-Kendall test has been used to analyze the long-term trend of normal waves. As a results, it was found that there were no trend at Deokjeokdo, Oeyeondo and Chibaldo. However, Marado, Pohang and Ullengdo showed an increasing tendency.

Development of an Efficient Method of Screening for Watermelon Plants Resistant to Fusarium oxysporum f. sp. niveum (수박 덩굴쪼김병에 대한 효율적인 저항성 검정법 개발)

  • This study was conducted to establish an efficient screening method for watermelon plants resistant to Fusarium wilt (FW), which is caused by Fusarium oxysporum f. sp. niveum (Fon). An HA isolate was prepared from a wilted watermelon plant in Haman-gun and identified as F. oxysporum f. sp. niveum based on morphological characteristics, molecular analyses of ITS (internal transcribed spacer) and TEF (translation elongation factor $1{\alpha}$) sequences, and host specificity on cucurbits including watermelon, melon, oriental melon, and cucumber. The assay for disease response of watermelon differentials indicated that the HA isolate was race 0. Among seven liquid media tested, the highest amount of Fon spores was produced from V8-juice broth, which was selected as a medium for mass production of Fon. The disease assay for 21 watermelon and 11 watermelon-rootstock cultivars demonstrated that 20 watermelon cultivars except for 'Soknoranggul' were susceptible; 'Soknoranggul' was moderately resistant. All the tested rootstock cultivars were highly resistant to the HA isolate. The evaluation of disease development depending on various conditions suggested that an efficient screening method for FW resistance in watermelon plants is to dip the roots of 10-day-old seedlings in spore suspension of $1.0{\times}10^5-1.0{\times}10^6conidia{\cdot}mL^{-1}$ for 30 min., to transplant the seedlings to plastic pots with a fertilized soil, and then to cultivate the plants at $25^{\circ}C$ for 3 weeks.

A New Screening Method for the Selection of Calla Lily Zantedeschia aethiopica Cultivars Resistant to Calla Lily Soft Rot (칼라의 무름병 저항성 품종 선발을 위한 검정 방법 개발)

  • This experiment was carried out to establish the screening methods for the selection of Zantedeschia aethiopica cultivars resistant to calla lily soft rot caused by Pectobacterium carotovorum subsp. carotovorum. A center of leaf and petiole tissues of four Z. aethiopica cultivars were wounded and inoculated with three different inoculum concentrations, $1{\times}10^7cfu/mL$, $1{\times}10^8cfu/mL$, and $1{\times}10^9cfu/mL$, of EccNHRI-21 isolate. And they were evaluated at 4, 18, and 26 hours after inoculation. The lesion of maceration was developed well in the leaf tissues inoculated with $1{\times}10^9cfu/mL$ concentration. And evaluation of resistance in 18 hours after inoculation was correlated with field resistance positively. Using this method, ten Z. aethiopica commercial cultivars and four wild types were screened. 'Crowbrough' and 'White Cutie' were selected as highly resistant genotypes and 'Mont Blanc' and 'Silky White' as resistant genotypes. 'Wedding March' and 'Kiwi Blush' were evaluated as moderate resistant and the others including 'Childsiana' were susceptible. And all of four wild types were evaluated as more than moderate resistant. In spite of control through cultural, biological and chemical methods, the use of resistant cultivars is most efficient to overcome calla lily soft rot. This newly developed screening method was helpful for breeding new varieties resistant to calla lily soft rot.

Development of Efficient Screening Methods for Melon Plants Resistant to Fusarium oxysporum f. sp. melonis (멜론 덩굴쪼김병에 대한 효율적인 저항성 검정법 개발)

  • This study was conducted to establish an efficient screening system to identify melon resistant to Fusarium oxysporum f. sp. melonis. F. oyxsporum f. sp. melonis GR was isolated from infected melon plants collected at Goryeong and identified as F. oxysporum f. sp. melonis based on morphological characteristics, molecular analyses, and host-specificity tests on cucurbits including melon, oriental melon, cucumber, and watermelon. In addition, the GR isolate was determined as race 1 based on resistance responses of melon differentials to the fungus. To select optimized medium for mass production of inoculum of F. oxysporum f. sp. melonis GR, six media were tested. The fungus produced the most spores (microconidia) in V8-juice broth. Resistance degrees to the GR isolate of 22 commercial melon cultivars and 6 rootstocks for melon plants were investigated. All tested rootstocks showed no symptoms of Fusarium wilt. Among the tested melon cultivars, only three cultivars were susceptible and the other cultivars displayed moderate to high resistance to the GR isolate. For further study, six melon cultivars (Redqueen, Summercool, Superseji, Asiapapaya, Eolukpapaya, and Asiahwanggeum) showing different degrees of resistance to the fungus were selected. The development of Fusarium wilt on the cultivars was tested according to several conditions such as plant growth stage, root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease. On the basis of the test results, we suggest that an efficient screening method for melon plants resistant to F. oxysporum f. sp. melonis is to remove soil from roots of seven-day-old melon seedlings, to dip the seedlings without cutting in s pore s uspension of $3{\times}10^5conidia/mL$ for 30 min, to transplant the inoculated seedlings to plastic pots with horticulture nursery media, and then to cultivate the plants in a growth room at 25 to $28^{\circ}C$ for about 3 weeks with 12-hour light per day.

Gnawing and Escaping Behaviors of Monochamus alternatus (Coleoptera: Cerambycidae) in a Confined Environment: Suggesting a Bioassay Method of Netting for Adult Escape Prevention (인위적 구속환경에서 솔수염하늘소의 쏠기와 탈출행동: 성충탈출 방지용 그물망의 생물검정법 제안)

  • The Japanese pine sawyer, Monochamus alternatus Hope, is a representative vector of the pine wood nematode, Bursaphelenchus xylophilus, which causes wilting symptoms in pine trees. A control method using a net has been introduced, which is an alternative method to the fumigation for the control of dead pine trees by pine wilt disease. This study was carried out to investigate the factors that induce gnawing and escaping behaviors of M. alternatus. The behaviors were examined after M. alternatus adult was placed in a confined space at different temperatures. M. alternatus adults could escape through mesh net torn by gnawing when they were confined in a space of 30 mm or less in diameter. The success rate of escape was high at 20 to $30^{\circ}C$, and no adults escaped at $15^{\circ}C$. The enticement of M. alternatus adults by food didn't affect the success rate of escape. In the case of not being confined in a narrow space, the escaping hole could not be formed because the gnawing was not concentrated on one part. M. alternatus moved its body in a narrow space using the tarsus of middle and hind legs, and made an escape hole by concentrically gnawing the obstacle on the front side with mandible, and showed a behavior of getting out while supporting the body by supporting the front legs. The present results will be able to use as an important basic information for evaluating the performance of mesh net which confines M. alternatus adults and suggested by alternative method to fumigation technology.

Determination of an Effective Method to Evaluate Resistance of Bottle Gourd Plant to Fusarium oxysporum f. sp. lagenaria (박 덩굴쪼김병 저항성 검정조건 구명)

  • Fusarium wilt caused by Fusarium oxysporum is a devastating disease limiting production of watermelon in Korea. The best way to control diseases is to use resistant gourd rootstock on watermelon. This study was conducted to establish an efficient screening method for resistant bottle gourd to Fusarium oxysporum f. sp. lagenaria. To develop an efficient inoculation method, incubation temperature after inoculation (15, 20, 25, and 30℃), inoculum concentration (1 × 105, 5 × 105, 1 × 106, and 5 × 106 conidia·mL-1), and growth stages of seedlings (7, 10, 13, and 16 days) was investigated. Disease development of Fusarium wilt of bottle gourd was little affected by differences in incubation temperature and growth stages of seedlings. But resistant lines were more susceptible and appeared more severe symptoms at the higher inoculation level. Taken together, we suggest that an effective screening method for resistant gourd plant to Fusarium wilt is to dip the roots of 10-day old seedlings in spore suspension of 1 × 105 - 1 × 106 conidia·mL-1, for 30 min, to transplant the seedlings into a non-infected soil, and then to incubate the inoculated plants in a growth room at 25℃ for 3 weeks to develop Fusarium wilt.

Applied Analysis for Metabolic Profiling of Trace-level Amino Acid in Biological Fluid (생체시료 중 미량 아미노산 대사 프로필을 위한 분석법 응용)

  • The universality of low molecular weight metabolites (i.e. amino acids, steroid hormones) allows rapid and straightforward investigation of biochemistry of genetically un-characterized species. Thus in vivo metabolic profiling of amino acid in combination with multivariate data analysis (metabolomics) offers great potential in comparative biology. In this paper, amino acid profiles in biological fluid (media) were studied by using HPLC/FLD. HPLC procedure for amino acids require the formation of derivatives due to the low absorption of the free compounds. o-Phthalaldehyde (OPA) used in association with a thiol, such as 3-mercaptopropionic acid (3-MPA), is one of the most popular and sensitive reagents, which yield quickly fluorescent iso-indoles at room temperature. To improve unstability of OPA/3-MPA derivatization, we optimized injector programs for fixed injection times. Linear regressions for the standard curves were linear in the range 0.5 - 100.0 ppb, giving correlation coefficents above 0.99. The detection limit were 1.70 pmol(GLU) - 23.81 pmol(SER). It is practically useful when the amount of sample is very low on single cells.
