Effects of Gamma Irradiation on Antioxidant, Antimicrobial Activities and Physical Characteristics of Sargassum thunbergii Extract (지충이 추출물의 항산화, 항균 활성 및 물리적 특성에 미치는 감마선 조사의 영향)

  • This study was carried out to determine the effect of gamma irradiation (3-20 kGy) on the antioxidant, antimicrobial activities and physical characteristics of Sargassum thunbergii (ST) extracts. When ST powder was treated by gamma irradiation, the yields and total phenolic compounds (TPC) of water extracts were increased, but radical scavenging activities were not changed. When ST extract was irradiated, the TPC and DPPH radical scavenging activities were increased. In addition, gamma irradiation of ST extract decreased viscosity and removed color. These results suggest that gamma irradiation would be a useful method for improving the physical characteristics of ST extract while maintaining native biological activities.

Quality assessment of Aster scaber cultured at different conditions for processed food production (재배환경에 따른 가공식품 소재로서 참취의 품질 비교)

  • Consumption of wild edible greens (WEG) has risen in Korea recently, and processing of WEG at greenhouses is increasing. In this study, we investigated chamchwi (Aster scaber Thunb), a major WEG, to assess the effect of growth conditions on nutritional quality and blanching process. Field-grown chamchwi had 1.5-2-fold more total phenolic compounds and anti-oxidative capacity than the house-grown one. Among functional compounds, quercetin was significantly higher (about 5-fold) in the field samples (approximately $430{\mu}g/g$ fresh weight) than in the house samples. Soluble sugars in field-grown chamchwi were 2-fold higher than in the house samples. For both the samples, blanching process decreased chamchwi quality significantly, but the degree of loss for each component was different. Consequently, field-grown chamchwi exhibited superior nutritional quality than greenhouse-grown one, and blanching process resulted in a similar decrease of nutritional quality in both the samples.

  • This study was conducted to evaluate the effects of microbial inoculation on sewage sludge composting. The number and species of microorganisms in sewage sludge sampled on February were higher than those sampled on August. The composting of sewage sludge is inhibited by the polyacrylamide cation, which was used as a coagulant and known to repress the growth of microorganisms. The growth of all microorganisms was inhibited by the addition of the polyacrylamide cation at a concentration of more than 0.8%. The species and viable counts of microorganisms were observed to increase during composting sewage sludge by inoculation of the effective microorganisms and addition of the pine tree sawdust as a bulking agent, compared with those without inoculation. A variety of organisms in compost(sewage sludge plus sawdust) were observed after composting for 30 days, such as Fragilaria sp., Proales sp., Vorticella sp., Schizothrix sp., Anabaena sp., Zoothaminium sp., Epstylis sp., Arcella sp., Balantidium sp., Actinophrys sp., Synedra sp., Euglypha sp., Ulothrix sp., Anacystis sp., and Clostium sp.

  • 최근 국내에서는 하천변 생태계를 대상으로 생태구조 및 서식처 평가 등 많은 연구가 진행되었음에도 불구하고 생태적 변화를 예측하고 예측된 결과에 따라 생태복원 대책을 수립할 수 있는 연구는 현재까지 미흡하다고 볼 수 있으며, 이는 생태복원 연구에 있어 주요과제라고 할 수 있다. 이에 반해 유럽과 미국에서는 수문학적 서식처 조건에 따른 식생 반응 예측 등 수문생태 모형에 대한 연구가 꾸준히 이루어져 왔으며, 최근에는 습지, 홍수터 등을 대상으로 생태보전 복원분야에 널리 응용되고 있다. 본 연구에서는 국외의 수문생태모형 적용사례를 분석하여 국내 적용가능성 및 문제점에 대한 타당성을 제시하고자 하였다. 그 대상으로는 유역 내에 홍수량 분담을 위한 천변저류지를 적용하였으며, 의미상으로 홍수터(범람원), 습지 역시 해당 범위에 포함할 수 있다. 천변저류지는 홍수기와 비홍수기에 따라 활용 측면을 구분할 수 있으며, 이런 맥락에서 천변저류지의 수위변화는 식생 변화의 중요한 요소(factor)가 될 수 있다. 그 외 범람시기, 범람일수, 건조기간 등을 포함한 수문요소 역시 수문생태 모델의 예측 변수인 식생 성장에 영향을 미치게 된다. 이러한 수문/생태시스템의 상호관계를 활용하고 모형의 형태를 국내 식생에 대한 생리학적 특성에 맞게 변화시킬 수 있다면 국내 천변저류지에도 충분히 적용 가능할 것으로 판단된다. 또한 잠재적으로 식생 성장에 영향을 줄 수 있는 다양한 환경조건 예를 들면 영양물질, 토양구조, 토사 퇴적 등을 적용대상지 환경에 맞게 포함하거나 고려할 수 있다면 모형의 재현성을 더욱 높일 수 있을 것이다.의 기대효과를 가져올 수 있으리라 생각되며 분석된 인자들은 수달 서식지를 위한 하천정비의 기초자료에 유용할 것으로 보인다.따른 유량측정망을 구축하는 것이다.의 의사결정 지원 도구가 될 것이다. 따라서, 본 연구에서는 도시유역의 물순환 해석을 위한 일련의 과정, 즉 자료의 조사 및 취득에서부터 물순환 해석 모형을 이용한 정량적 현황파악, 물순환 개선 기법 및 평가를 수행함에 있어 주요 착안점 및 실무에서의 기술적 가이드를 제공하고자 하였으며, 보다 세밀한 도시유역의 물순환 해석을 위하여 우리나라와 일본에서 적용이 활발한 물리적 기반의 분포형 모형(WEP, SHER, SWMM)의 적용사례를 통하여 국내 도시하천의 물순환 해석에 활용함에 있어서의 실질적인 적용절차 등을 제시하고자 하였다. 한다.호강유역의 급격한 수질개선을 알 수 있다.世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와 동년대에 동일한 목적으로 찬술되었음을 알 수 있다. $\ulcorner$경상도실록지리지$\lrcorner$(慶尙道實錄地理志)에는 $\ulcorner$세종실록$\lrcorner$(世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와의 비교를 해보면 상 중 하품의 통합 9개소가 삭제되어 있고, $\ulcorner$동국여지승람$\lrcorner$(東國與地勝覽) 에서는 자기소와 도기소의 위치가 완전히 삭제되어 있다.

A Mutant Arthrospira platensis M20CJK3 Showing Enhanced Growth Rate and Floatation Activity (생장 및 부상성이 향상된 남세균 돌연변이 균주 Arthrospira platensis M20CJK3)

  • A photosynthetic cyanobacterium Arthrospira platensis, well known for health food supplement, was studied as a target species for atmospheric $CO_2$ removal as well as biomass production. Although the biomass of A. platensis was massively produced in many countries, the recovery cost of its biomass is still high. The purpose of this study was to develop the A. platensis mutant strains which have enhanced growth rate and floatation activity to reduce the recovery cost. A. platensis KCTC AG20590 was treated with 0.24% ethyl methanesulfonate (EMS) for 20 min at room temperature. The mutant strain A. platensis M20CJK3 was finally selected by its morphological and physiological features. The morphology of the mutant A. platensis M20CJK3 was changed from loose-coiled form to tight-coiled form showing short pitch. The growth and $CO_2$ uptake rate of A. platensis M20CJK3 were improved about 15% and 17% compared with A. platensis KCTC AG20590, respectively. The floatation activity of A. platensis M20CJK3 was enhanced in 2-fold compared with that of A. platensis KCTC AG20590. Soluble proteins extracted from two strains were analyzed by two dimensional electrophoresis (2-DE) and MALDI-TOF MS/MS. Among 15 protein spots induced in 2-DE analysis, two spots were the proteins related to photosynthesis and electron transfer system of the other cyanobacteria. As a consequence, it seems that the tight-coiled mutant A. platensis M20CJK3 has an advantage of high growth rate and floatation activity which are beneficial for the mass cultivation and recovery.

Preparation of Chitosan/Poly-${\gamma}$-glutamic Acid Nanoparticles and Their Application to Removal of Heavy Metals (키토산/폴리감마글루탐산 나노입자의 제조 및 중금속 제거에의 응용)

  • Chitosan is a natural polymer that has many physicochemical(polycationic, reactive OH and $NH_2$ groups) and biological(bioactive, biocompatible, and biodegradable) properties. In this study, chitosan nanoparticles were prepared using poly-${\gamma}$-glutamic acid(${\gamma}$-PGA) as gelling agent. Nanoparticles were formed by ionic interaction between carboxylic groups in ${\gamma}$-PGA and amino groups in chitosan. Chitosan(0.1~1 g) was dissolved in 100 ml of acetic acid (1% v/v) at room temperature and stirred overnight to ensure a complete solubility. An amount of 0.1 g of ${\gamma}$-PGA was dissolved in 90 ml of distilled water at room temperature. Chitosan solution was dropped through needle into beaker containing ${\gamma}$-PGA solution under gentle stirring at room temperature. The average particle sizes were in the range of 80~300 nm. The prepared chitosan/${\gamma}$-PGA nanoparticles were used to examine their removal of several heavy metal ions($Cd^{2+}$, $Pb^{2+}$, $Zn^{2+}$, $Cu^{2+}$, and $Ni^{2+}$) as adsorbents in aqueous solution. The heavy metal removal capacity of the nanoparticles was in the order of $Cu^{2+}$ > $Pb^{2+}$ > $Cd^{2+}$ > $Ni^{2+}$ > $Zn^{2+}$.

Modulatory Effect of Four Azulene Derivatives from the Fruiting Bodies of Lactarius hatsudake on Interferon-$\gamma$ Production (젖버섯아재비 자실체로부터 분리한 Azulene계 화합물이 Interferon-$\gamma$ 생성에 미치는 영향)

  • Investigation of secondary bioactive metabolites from the fruiting bodies of Lactarius hatsudake led to the isolation of four azulene derivatives by means of repeated column chromatography and preparative HPLC, and they were identified as 1-formyl-4-methyl-7-isopropyl azulene (1), lactaroviolin (2), 4-methyl-7-isopropyl-azulene-1-carboxylic acid (3), and 1-formyl-4-methyl-7-(1-hydroxy-1-methylethyl) azulene (4) by their physico-chemical properties and spectroscopic analysis. The isolated compounds were evaluated for the effects on modulation of cytokines in natural killer cell line (NK92 cells). Compounds 1 and 4 strongly inhibited IFN-$\gamma$ production in a dose-dependent manner, corresponding to 101.3 % and 92.7 % inhibition at 400 ${\mu}M$, and 11.9 % and 24.1 % at 100 ${\mu}M$, respectively, whereas compounds 2 and 3 showed weak inhibitory effect on INF-$\gamma$ production, corresponding to 45.9 % and 18.0 % inhibition at 400 ${\mu}M$.

Isolation and Identification of Lipolytic Enzyme Producing Pseudomonas sp. OME and Optimization of Cultural Conditions (지방분해효소 생산균 Pseudomonas sp. OME 의 분리 동정 및 배양조건 최적화)

  • Lipolytic enzyme-producing bacteria were isolated from edible oil mill effluents on tributyrin agar medium. The shake-flask-scale studies yielded a promising isolate and it was identified as Pseudomonas sp. An OME using various microbiological observations such as cultural, microscopic, and biochemical tests was undertaken and confirmed using PIBWIN bacterial identification software. Lipolytic enzyme production was screened with oils such as sunflower, caster, coconut, tributyrin, and olive. Amongst these, olive oil showed an increased lipase production 6.1 U/ml. In view of the highest lipolytic enzyme production with olive oil, further optimizations were carried out using olive oil as a carbon source. Lipolytic enzyme production was optimized by a conventional 'one variable at a time' approach and the significant factors were further analyzed statistically using response surface methodology (RSM). The effect of physical factors such as incubation time, temperature, initial medium pH, and nutritional factors such as concentration of olive oil and yeast extract were examined for lipase production. Lipolytic enzyme secretion was strongly affected by three variables (incubation time, concentration of yeast extract and olive oil). Therefore, the interaction of these three factors was further optimized using response surface methodology. The optimized conditions of lipase production using response surface methodology yielded a maximum of 9.62 U/ml with optimum conditions for incubation, yeast extract and olive oil concentrations were found to be 48 hr, 0.3 g. and 0.9 ml. respectively.

Protein Quality Evaluation and Effect of Plasma Lipid Contents of Acid Hydrolysates of Cocoon inn Rats Fed by High Cholesterol, High Triglyceride and High Sucrose Diet (누에고치 산 가수분해물의 단백질의 질적 평가와 고콜레스테롤, 고지질, 고당질식이 흰쥐의 혈장지질에 미치는 영향)

  • Acid hydrolysates of cocoon was gained by acid hydrolysis of 2 N HCl, 11$0^{\circ}C$, 48 hours, neutralization and desalting from the cocoon. The amino acid compositions of acid hydrolysates of cocoon were glycine 43.25%, alanine 34.39%, serine 10.05% and valine 2.44%. The contents of essential amino acid was 10.05%. Food efficiency ratio of acid hydrolysates of cocoon group was equal to the reference protein, casein. Liver weight, GOT, GPT activity, serum albumin and serum total protein level of rats were not significantly different among the experimental groups. Therefore, the protein acid hydrolysates of cocoon is not of high quality. When the rat fed with high cholesterol, high lipid, and high sucrose diet was administered with 5% acid hydrolysates of cocoon, its plasma lipids concentration of acid hydrolysates of cocoon was favorably affected: its triglyceride was decreased, and the level of phospholipid and HDL cholesterol were increased. There was also an unfavorable effect: the levels of LDL cholesterol and total cholesterol went up. Therefore, the acid hydrolysates of cocoon is not a good protein food source, but is can be used a cosmetic, medical, or packing material. Further research will reveal how it will affect or improve plasma lipid.

Effect of Diflubenzuron on Silkgland Development of Silkworm, Bombyx mori (Lepidoptera: Bombycidae) (Diflubenzuron이 누에의 실샘 발달에 미치는 영향)

  • This study was investigated to know the effects of diflubenzuron(DFB) on the larval silkgland development of the silk-worm, Bombyx mori (L.). It has been known as a prohibitor of chitin synthesis mostly on the species of the Order Lepidoptera. In this work, the effects of the DFB concentrations (2.5$\times$10$\^$-1/,2.5$\times$10$\^$-2/, and 2.5$\times$10$\^$-3/$\mu\textrm{g}$/$\mu$l on the various larval stages, were investigated in terms on the silkgland. The macro- and microstructure of cell membrane of silkgland, and the differences of silkgland weight and water contents treated by DFB are also surveyed. As the tesults, the silkgland weight depended sensitively on not the DFB concentration but the DFB treatment period. The longer DFB treatment period, the lighter dried silkgland weight and the heavier water content of the silkgland. White opaque(WO) emerged in the middle silkgland of DFB treated larva. From the scanning election microscope observation, the cell membrane of silkgland of DFB treated larva was distinctly more compact compared to that of control. The WO was evidently resulted from the obstacles of normal transformation of silk protein through the cell membrane of middle silkgland.
