This study was conducted to investigate the effect of cryoprotectants on the storage stability of Lactobacillus fermentum SK152, which was isolated as a probiotic candidate. Solutions of 10% glucose, trehalose, dextrin, and skim milk powder were used as cryoprotectants. The survival rates of L. fermentum SK152 after freeze-drying were 5.6% (dextrin), 2.2% (skim milk powder), 1.7% (glucose), and 1.5% (trehalose), suggesting that dextrin was most effective at minimizing the cell death of L. fermentum SK152 by lyophilization. The survival rates of L. fermentum SK152 stored at 4℃ ranged from 37% (dextrin)-90% (skim milk powder) after 8 weeks, while those at 20℃ ranged from 4% (dextrin)-12% (skim milk powder) after 7 weeks, indicating that skim milk powder was the best at minimizing the cell death of L. fermentum SK152 during storage, irrespective of storage temperature, among the cryoprotectants used.
This study is focused on establishing the optimal conditions to enhance the production of antilisterial substances by Lactobacillus paracasei BK57 isolated from Baikkimchi. In addition, the growth and in situ lactic acid and bacteriocin production of this strain were investigated during the manufacture of fresh cheese. And then the efficacy of using Lactobacillus starter as a protective culture to improve the safety of fresh cheese against Listeria monocytogenes KCTC 3569 was estimated. Maximum growth rate and activity of antibacterial substances were obtained in Lactobacilli MRS broth at $37^{\circ}C$ with controlled pH 6.0 after 30 h of incubation under aerobic condition. However, the growth rate and antimicrobial activity of bacteriocin produced in whole milk supplemented with yeast extract (2.0%) as a substrate were lower than those obtained in MRS broth. Live cells and cell-free culture supernatant of BK57 strain were effective in the suppression of L. monocytogenes in milk, whereas the inhibitory of the bacteriocin obtained from BK57 strain was higher in BHI broth than in milk. During storage at $4^{\circ}C$ and $15^{\circ}C$ for 6 days, no significant difference was found in the cell viability and antimicrobial activity of BK 57 strain in fresh cheese. In samples held at two temperatures, there was at least a 15% reduction in the numbers of the pathogen in fresh cheese artificially contaminated with approximately $10^5CFU/ml$ of L. monocytogenes within 6 days. Our results demonstrated the usefulness of L. paracasei BK57 having antilisterial activity as a biopreservative in the cheese making process.
Lactic acid bacteria(LAB) fermented food was prepared with milk or egg white powder(EWP) and added with freeze drying protectant(FDP). 0.2% of Tween 80 or 1% of ascorbate was added to milk sample and 3% of raffinose or 1% of ascorbate was added to EWP sample. Effects of FDP on sensory property, volatile aroma compounds and physical property of LAB fermented food were investigated. In case of non-freeze dried samples, sensory properties of milk sample with ascorbate were slightly better than those of reference sample(milk), while sensory properties of EWP sample or EWP sample with FDP were slightly inferior to reference sample. Sensory properties of all of the freeze dried/reconstituted samples were not different. Sensory properties of milk sample with ascorbate were reduced by freeze drying/reconstitution, while those of sample with ascorbate were not changed. Although all of the volatile aroma compounds were reduced by freeze drying, the residual ratio was slightly different between milk samples and EWP samples. Difference in volatile aroma compounds between milk samples and EWP samples before freeze drying was relatively large, while difference between two sample groups after freeze drying/reconstitution was relatively small. Rheological properties of milk samples were markedly changed by freeze drying/reconstitution, while those of EWP samples were changed slightly.
In order to study the preservation effect of $-3^{\circ}C$ partial freezing method, tile growth and biochemical activity of microorganisms and the changes of K-value in mackerel press juice were investigated at $0^{\circ}C,\;-3^{\circ}C$ supercooling (liquid phase) and $-3^{\circ}C$ freezing (solid phase). The results obtained in this paper were follows : 1) The growth and biochemical activity of microorganisms were reduced at $-3^{\circ}C$ supercooling than $0^{\circ}C$ in spite of the small variation in temperature. 2) There were no growth and biochemical activity of microorganisms at $-3^{\circ}C$ freezing (solid phase). 3) The difference in the K-value between $-3^{\circ}C$ supercooling and $-3^{\circ}C$ freezing was remarkable in spite of the same temperature.
The processing conditions and quality of sardine surimi were examined: Raw sardine meat was separated, washed in 0.2% $NaHCO_3$ and 0.15% NaCl solution, and then dewatered by centrifuge. The dewatered sardine meat was chopped, mixed with 20% emulsion curd (soybean protein : water : refined sardine oil=1:5:2.6), 4% sorbitol, 4% sucrose, 0.2% polyphosphate and 0.1% sodium erythorbate by stone mortar. The mixed sardine meat was frozen with contact freezer, packed in carton box and then stored at $-25{\pm}2^{\circ}C$. The moisture, crude protein and lipid contents of the sardine surimi product was 73.3%, 15.0% and 6.9%, respectively. Fatty acid composition of product consisted of 28.8% of saturates, 24.3% of monoenes and 47.7% of polyenes and the major fatty acids were 16:0, 20:5, 18:1, 22:6 and 16:1. The results of changes in POV, TBA value, fatty acids, texture and sensory score of products during frozen storage showed that lipid oxidation and freeze denaturation of product could be retarded, and flavor enhanced by addition 20% emulsion curd and 0.1% sodium erythorbate. In an attempt to apply sardine surimi in producing surimi-based product, it was concluded that pollack surimi could be substituted with sardine surimi up to 40% without showing any significant changes in texture and taste of surimi-based product.
Palmitoyl tripeptide (M330) showed higher antimicrobial activities than methyl paraben or phenoxy ethanol through minimum inhibitory concentration (MIC) test. However, when the M330 was added into cosmetic formulation, white precipitates formed due to the electrostatic interaction between M330 and carbopol (carboxy vinyl polymer) as a thickener in cosmetics, and the viscosity of cosmetics decreased sharply. Also, the antimicrobial activities of M330 in cosmetics became lower than those of methyl paraben or phenoxy ethanol. Thus, the encapsulation of M330 in ethosome vesicle was attempted in order to recover the declined antimicrobial activities of M330 in cosmetics and prevent the precipitates from forming. When ethosome-encapsulated M330 was added into cosmetics, the precipitates did not form, and the decrease in the viscosity of cosmetics was not large compared to the addition of unencapsulated M330. Challenge tests showed that antimicrobial activities against gram negative bacteria were improved by the encapsulation of M330, but the encapsulation was not effective against gram positive bacteria and fungus. A combination of M330 with EDTA showed synergistic inhibitory potential against C. albicans. After coencapsulation of M330 and EDTA in ethosome, antimicrobial activities proved to be higher than those of unencapsulated M330 and EDTA.
The effect of irradiation on the shelf-life of commercial ham products and its quality were investigated during eight weeks storage at $30^{\circ}C$ following 3-8kGy irradiation. Total bacterial count of unirradiated group was proliferated from. $5.4{\times}10^{3}$ per gram of sample to $10^{6}$ after two weeks storage but the bacterial count in irradiated group was propagated to the same level after three to five weeks storage and the growth of coliform group was shown negative. The freshness of stored ham was better in irradiated group than in unirradiated one and the changes in components related to the quality of ham were almost the same between unirradiated and irradiated group. Discoloration and irradiation odor were positive in 8kGy irradiated group according to the sensory test, but 5 kGy irradiated group was not only good in texture but also in the extension of shelf-life to more than two times as compared with that of unirradiated one.
Processing conditions and food components of frozen seasoned anchovy meat products were investigated. The separated anchovy meat was chopped, mixed with 12.8% emulsion curd, 0.5% table salt, 2.0% sugar, 0.4% sodium bicarbonate, 0.2% polyphosphate, 0.2% monosodium glutamate, 0.3% onion powder, 0.1% garlic powder, 0.1% ginger powder, 3.0% soybean protein, and 0.2% sodium erythorbate by remodeled stone mortar. This seasoned anchovy meat was frozen with contact freezer, Packed in a carton box and then stored at $-25{\pm}2^{\circ}C$. The major fat acids of product were linoleic, oleic, palmitic, docosahexaenoic, linolenic, palmitoleic, eicosapentaenoic acid. Amino acid composition of product were mainly consisted of Glu, Asp, Leu, Lys and Ala. The taste compounds of product were IMP 160.0 mg/100g ; free amino acids such as Glu, His, Ala, Leu 503.7 mg/100g ; total creatinine 158.3 mg/100g and small amounts of betaine, TMAO.

The physicochemical properties of dextran culture fermented by Leuconostoc citreum S5 were evaluated by the addition of various polysaccharides and heat-treatment. The consistency of dextran culture increased with the addition of carrot juice residue (CJR) in submerged culture, resulting in the highest consistency of 150 $Pa{\cdot}s^n$ and viable cell counts of $2.36{\times}10^9$ CFU/mL at 20% level of CJR. The dextran culture showed the pseudoplastic behavior and its consistency was greatly increased with the addition of various polysaccharides at 2% level. Addition of glucomannan indicated the highest consistency of 1275 $Pa{\cdot}s^n$ and their heat-treatment resulted in the increase of consistency except for glucomannan. After heat-treatment, the fermented dextran culture containing CJR fortified with gellan gum and carrageenan showed great change in rheology, indicating the highest consistency and hardness value resulted in the great increase of elastic and viscous moduli. The dynamic viscoelastic properties of dextran culture were greatly increased by the heat-treatment after fortification of various polysaccharides. Thus, the consistency and viable cell counts of dextran culture were increased with the addition of CJR. The rheological properties of dextran culture were manipulated by the fortification of various polysaccharides and heat-treatment.

This study examined the immune response of Philippines eel (Anguilla bicolor) to the oral administration of fermented Sparassis crispa stipe extract for 6 weeks. The S. crispa extract fermented with Lactobacillus plantarum showed a higher total phenol content (301.68 ppm) and DPPH radical scavenging activity (63.9%) than those fermented with other strains. Therefore, L. plantarum was selected as a suitable starter culture for the fermentation of S. crispa stipe. The eels were fed a commercial diet supplemented with 1% of fermented S. crispa stipe extract for 6 weeks. The mortality rate of the eels fed the supplemented diet was significantly lower than those of the control after 6 weeks. The lysozyme activity of the serum was increased significantly (12.33 ${\rightarrow}$ 54.66 units) after 6 weeks in the eel fed supplemented diets of fermented S. crispa stipe. The serum of the eel fed the supplemented diet of the S. crispa stipe extract showed higher bactericidal activity. These results suggest that both the S. crispa stipe extract and fermented S. crispa stipe have strong potential to activate the innate immune response of the Philippines eel.
