Characteristics and Virulence Assay of Entomopathogenic Fungus Nomuraea rileyi for the Microbial Control of Spodoptera exigua (Lepidoptera: Noctuidae) (파밤나방의 미생물적 방제를 위한 병원성 곰팡이 Nomuraea rileyi의 특성 및 병원성 검정)

  • To date, chemical control remains the most common way to reduce beet armyworm (Spodoptera exigua) populations. However, this insect has become more tolerant or resistant to many chemical insecticides and the insect larvae usually hide inside hollow, tube-like leaves of host plant so they were difficult to kill by spraying insecticides. The use of viral and bacterial insecticide to solve these problems has not been successful because of their novel feeding habit. To overcome these problems, in this study, the biological characteristics and virulence of an entomopathogenic fungus isolated from the cadaver of larvae beet armyworm were investigated. Isolated entomopathogenic fungus was identified as Nomeraea rileyi (Farlow) Samson by morphological examinations and genetic identification using sequences of the ITS, ${\beta}$-tubulin gene and EF1-${\alpha}$ regions. This fungus was named as N. rileyi SDSe. Virulence tests against 3rd larvae of beet armyworm were conducted with various conidial suspensions from $1{\times}10^4$ to $10^8$ conidia/ml of N. rileyi SDSe in laboratory conditions. Mortality rate of beet armyworm showed from 20 to 54% and the virulence increased with increasing conidial concentrations. Although N. rileyi SDSe showed low mortality rate against beet armyworm, it is expected that N. rileyi SDSe will be used effectively in the integrated pest management programs against the beet armyworm.

  • Flupyrazofos (O,O-diethyl-O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)thiophosphoric acid ester) is a new organo-phosphorous insecticide. Flupyrazofos has exhibited excellent activity against diamondback moth (Plutella xylostella, DBM), and it is highly activity against rice armyworm(Pseudaleta separata) and cotton caterpillar(Palpita indicae). Flupyrazofos has then revealed outstanding both rapidity and residual action for DBM, although no systemic actions were observed and no cross-resistances were found to the resistance strains (Op-R, Py-R, IGR-R). Also, susceptibilities of five local strains to flupyrazofos were similar to those of the susceptible strain. These results indicate that flupyrazofos has considerable potential for controlling diamondback moth, rice armyworm and cotton caterpillar in field.

Resistance of Diamondback Moth(Plutella xylostella L.) against the Pyrethroids (피레스로이드제에 대한 배추좀나방의 포장약제저항성의 변동)

  • This test has been carried out to clear the effectiveness of pyrethroid insecticides to the Diamond back moth, that was complained by a farmer. The results obtained are summarized as follows. Effectiveness of Pyrethroids to suceptable strain and the results of chemical anaysis on the ingredients of pyrethroikds were normal at recommended concentrations. The effect ualue of pyrethroid by which the blocks were only one time treated was 57-59%, whereas the blocks sprayed 4 consecutive times of pyrethroid didn't show any effectiveness as shown in larval increasing rate of 489-552%. Among the results of field test conducted from 1990 to 1991 at 7 area, Dae Gu area showed the highest Effectiveness as 95-98%. However, Pyong Taek area, showed the least effectiveness as 0% in effect value. The resistant population which was collected at Tae kwan Ryung area was resurrected susceptability as much as JMC, in case of Placing it in non-treatment environment, whereas the pupulation which was revealed to pyrethroids for 6 genetations showed as much as 341-544 times of resistance compared with JMC. The non-effectiveness of pyrethroids to Diamondback moth which was complainted by a certain farmer named Mr Ju, was clearly resulted by the development of insect resistance. In Korea, the resistance of Diamondback moth to pyrethroids has developed in some area, as well as the resistance of pyrethroids has developed according to the continuous use of Insecticides, and the susceptability resurrected by stopping the use of pyrethroids. The result from this test would suggest that the pyrethroid insecticides should be applied in turb with other insecticides.

Karyotype and Insecticide Resistance of the Green Peach Aphids (Myzus persicae Sulzer) collected from Tobacco fields (연초포장에서 발생하는 복숭아혹진딧물의 핵형과 살충제 저항성)

  • Karyotype of apterous green peach aphid (Myzus persicae SuIBer) populations occurring in tobacco fields and their resistance to insecticides were investigated. All of the 16 clones collected from tobacco fields had same chromosome number of 2n=12. Among them, 5 clones had the normal karyotype, regardless of color morph, but chromosomal translnations between #1 and #3 autosomal chromosomes were noted in the other 11 clones. All of the aphid clones were highly resistant tolambda cyhalothrin, a pyrethroid pesticide, having over 20 times higher concentrations of LD90 than recommended concentration (10ppm) for the aphid. However, their resistance to acephate, demeton 5-methyl and pirimicarh varied depending on the clones, among which green-colored clones were generally more resistant to the pesticides than red-colored ones. No association was found between the insecticide resistance and the karyotype of the aphid in this experiment.

Effect of Temperatures on the Growth of Susceptible and Malathion Resistant Green Peach Aphid Strains (Malathion 저항성 및 감수성 복숭아혹진딧물의 온도에 따른 발육특성)

  • This test has been carried out to evaluate the effect of temperature on the growth of the insecticide susceptible strain, URY-O nomal genotype and insecticide resistant strain, O-RT abnormal genotype, and ABURABI nomal genotype. The nymphal periods were not significantly different between URY-O and O-RY strains at $25^{\circ}C$. At $30^{\circ}C$, susceptible strain URY-O could give birth to offsprings almost nomally, while resistant strain O-RY could not produce any offspring for 20 days which results in nymphal death. The numbers of offsprings of strain URY-O and strain ABURABI were not different between $25^{\circ}C\;and\;28^{\circ}C$, but strain O-RY, when it was reared at $28^{\circ}C$, could produce offsprings only 10% of those at $25^{\circ}C$. Body weight of strain URY-O and strain ABURABI were 0.22mg/female and 0.27mg/female, respectively at $28^{\circ}C$, however that of O-RY was only 0.16mg/female, showing considerable difference between normal and abnormal genotype. Substrain O-RY(+) which has high esterase activity showed poor reproduction ability(0.8 progenies per $G_{1}$ individual than substrain O-RY(-) (3.4 progenies per $G_{1}$ individual) which has low esterase activity at $28^{\circ}C$

Insecticide Susceptibility of Western Flower Thrip, Frankliniella occidentalis (Thysanoptera: Thripidae) on Horticultural Crops in Gyeonggi Area (경기지역 원예작물 꽃노랑총채벌레 약제 감수성)

  • This study was conducted to monitor the insecticidal susceptibility of western flower thrip, Frankliniella occidentalis, which damage horticultural crops in the Gyeonggi area. Bioassays were conducted under laboratory and greenhouse conditions by using the recommended concentrations of commercial insecticides being used for the control of thrips. Neonicotinoid insecticides, especially acetamiprid (8%) soluble powder (SP), clothianidin (8%) SC, imidacloprid (10%) wettable powder (WP) and thiacloprid (10%) water dispersible granule (WG) were not toxic to F. occidentalis collected from horticultural crops. However, F. occidentalis collected from vegetable greenhouses was extremely susceptible to acetamiprid + spinetoram (6 + 4%) suspension concentration (SC), clothianidin + spinetoram (6 + 4%) SC and methoxyfenozide + spinetoram (6 + 4%) SC, which resulted in over 90% control of thrips. In the greenhouse test, spinetoram (5%) WG, which caused 100% F. occidentalis mortality in the laboratory test, showed 87.4, 88.0, and 98.3% control at 3, 6, and 9 days after treatment, respectively, while imidacloprid (10%) WP showed below 44% control. From the results of this study, spinosin insecticides, such as more than over 4 and 10% of spinetoram and spinosad, and pyrrole insecticide, such as 5% chlorfenapyr, are recommended for the effective control of F. occidentalis.

A Report on the Control Effects of Automatic Dry Fog Sprayer on Aphids in Greenhouse Crops (시설 온실에서 진딧물에 대한 무인 연무방제기의 방제 효과 보고)

  • The area of greenhouse horticulture has increased dramatically since the early 1990s in Korea. However, area per farmer still very small and diverse pest species occurs in greenhouse abundantly because of its protected condition. To manage pests, farmers use many pesticides frequently. Incessant and improper use of chemicals provoked pesticide resistance in pest and poisoning of farmers. In this study, we investigated the efficiencies of automatic dry fog sprayer for controlling two aphid species, Aphis gossypii and Myzus persicae, in greenhouse horticulture. When doors and side windows were completely closed in automatic dry fog sprayer treatment greenhouse, aphids were controlled similar level with conventional spraying method (percent control: 61.0~94.1%). However, when half of the doors and side windows of the greenhouse were opened, aphids were controlled poorly (percent control: 36.0~54.4%).

Monitoring on Insecticide Resistance of Major Insect Pests in Plastic House (시설 재배 작물 주요 해충에 대한 약제저항성 모니터링)

  • This study was carried out for looking into the status of susceptibility of vinyl house insect pests to insecticides. The Thrips (Thrips palmi and Frankliniela occidentalis), Mites (Tetranychus urticae), Aphids (Aphis gossypii) and Whitefly (Trialeurodes vaporariorum) were captured at various areas where the host crop was being cultivated and the susceptibility level of each pest insect was investigated. The susceptibility of each pest insect varied by insect species and areas where they were caught. The tested insecticides showed good control effect to palm thrips in 2000, but in 2003 showed decrease of effect to them. Western flower thrips showed low susceptibility to neonicotinoids, imidacloprid and thiamethoxam, but high to chlorfenapyr, spinosad, emamectinbenzoate and fipronil. Antibiotic insecticides, abamectin and milbemectin, and chlorfenapyr were very effective on mite control and dicofol still had good effectiveness to it despite of long year use. No aphid species showed resistance to pyrethroid and carbamate insecticides. Relatively new insecticides such as imidacloprid, spinosad, pymetrozine were effective to whitefly, but not were organophosphates, carbamates and pyrethroids.

A Study on the Rate of Occurrence according to Watering Methods, Susceptibility and Chemical Control of Chrysanthemum Nematode (국화잎 선충에 대한 저항성 검정, 관수방법에 따른 국화잎선충의 피해율 및 화학적 방제에 관한 연구)

  • The susceptibility of 8 chrysanthemum varieties to Apelenchoides ritzemabosi, Shuhonothikara and Sagakiku were resistant by 11.7% and 14.5% each infested leaf, Sinrokusiogio, Dangonanako and Sintoa were susceptible as above 50%, Gold wave and Tenju were modertae by 24 to 26%. Percentage of infested leaf was positively correlated with the number of epidermal hair but not with leaf area, leaf thickness, stomatal size, number of stomata and length of epidermal hair. In effect of watering method, Polyethylene film cover plot was lowest by 9.4%, next conventional watering plot by 50%, Conventional watering and spray plot was the highest by 62%. The infested leaf rate was closely related with a number of rainy days than the amount of rainfall. Effect of chemical soil treatment, Temik showed the lowest infested leaf rate by 5%. There was no significant difference between the chemicals except Temik, but these are markedly effective in compare to control.

Susceptibility of Field Populations of Plutella xylostella and Spodoptera exigua to Four Diamide Insecticides (4종의 Diamide계열 살충제에 대한 배추좀나방과 파밤나방의 지역계통별 감수성 평가)

  • The present study evaluated the susceptibility of field populations of Plutella xylostella and Spodoptera exigua larvae to four diamide insecticides-chlorantraniliprole, cyantraniliprole, cyclaniliprole, and flubendiamide. All the four diamide insecticides induced 100% mortality in the populations from Seongju (SJ) and Geochang (GC) when treated at a concentration recommended for P. xylostella. However, a very low insecticidal activity was observed in the population from Pyeongchang (PC) with 42.3% 3 d after treatment with chlorantraniliprole. The relative resistance ratio of P. xylostella to cyclaniliprole was similar or low (0.1-6.3 folds) in all the four areas. However, the population from PC exhibited a high resistance ratio to chlorantraniliprole (1,196.3 folds), cyantraniliprole (105.6 folds), and flubendiamide (191.6 folds) compared with that of the susceptible strain P. xylostella. Further, the populations of S. exigua from Cheongju (CJ), Jindo (JD), and Yeonggwang (YG) were not completely controlled by the 4 diamide insecticides. These populations also showed high relative resistance ratio when compared with that of the susceptible strain S. exigua. In particular, the populations from CJ and JD showed 100,000-fold higher resistance ratio to flubendiamide. A comparison of susceptibility of S. exigua larvae to chlorantraniliprole between 2014 and 2017 showed that chlorantraniliprole induced 100% mortality in all populations in 2014, whereas a very low insecticidal activity was observed among the populations in 2017. The results of the present study indicate that the insect pests rapidly develop resistance to diamide insecticides. Therefore, alternative insecticides with a different mechanism of action and comprehensive control strategy are needed. This study can serve as a basis to control pests effectively using diamide insecticides.
