Evaluation of Antibacterial and Therapeutic Effects of a Sodium salts Mixture against Salmonella typhimurium in Murine Salmonellosis (나트륨 염 복합조성물의 마우스 살모넬라증에 대한 항균 및 치료효과)

  • Salmonellosis is a major bacterial zoonosis that causes self-limited enteritis to fatal infection in animals and food-borne infection and typhoid fever in humans. Multidrug-resistant strains of Salmonella spp. has increased over the last several decades and recently causes more serious problems in public health. The present study was investigated bacteriocidal effects of sodium chlorate, sodium azide, sodium cyanide, and sodium salts mixture containing sodium chlorate, sodium azide, and sodium cyanide on infection with S. typhimurium in macrophage RAW 264.7 cells, and antibacterial effects of sodium salts mixture for murine salmonellosis. In infection assay of S. typhimurium in RAW 264.7 cells, bacterial survival rates within macrophage in all treated groups was significantly reduced comparing to that of the control group with the passage of incubation time. Administration of sodium salts mixture showed a therapeutic effect for S. typhimurium infected ICR mice. The mortality of mice treated with sodium salts mixture was 70% until 12 days, while that of control mice was 100% until 9 days after S. typhimurium infection. The results of this study strongly indicate that sodium salts mixture has a potency treatment for murine salmonellosis.

  • The studies on the standard plate count, most probable number(MPN) and isolation of the Enterobacteriaceae, especially Salmonella species from stream waters of Kyong-Ju city were carried out from June to July, 1988. The relationship between water temperature, weather and standard plate count were observed. The locations where all of the MPN was higher than 1,600 were Mun-chon bridge, Wol-song bridge, Go-hun bridge and the area adjoining the Hyong-san river. The higher the water temperature, the more the viable counts without rain. Bacteriological survey of the Salmonella species was performed with the total 80 specimens isolated from 16 areas which are the main streams in Kyong-Ju city. The author identified S strains(2.36%) of Salmonella typhi, 5 strains(2.3%) of Salmonella paratyphi, 2 strains(1.42%) of Salmonella paratyphi B, 63 strains(29.72%) of other Salmonella groups by biochemical tests and serotyping tests with slide agglutination.

Antimicrobial Activity of Essential Oils from Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze on Escherichia coli and Salmonella typhimurium (대장균과 살모넬라균에 대한 박하와 배초향 정유성분의 항균활성)

  • For developing natural presevatives, essential oils of Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze were analyzed the composition of two oils and experimented on microorganism survival. Main components of Mentha arvensis oil were isomenthol (26.84%) and menthol (25.48%), and those of Agastache rugosa oil were estragole (79.83%) and limonene (4.13%) from GC-MSD analysis. Inhibition activities of Mentha arvensis oil against growth of Escherichia coli O157 : H7 ATCC 43895 and Salmonella typhimurium ATCC 7988 were observed from their clear zone $(9{\sim}14 mm\;&\;9{\sim}13\;mm)$, and that of Agastache rugosa oil were done from the clear zone $(13{\sim}20 mm\;&\;10{\sim}18\;mm)$ by concentration-dependent manner, respectively. In the inhibition test on CFU/ml of the microorganisms, both of the plant essential oils at concentration of 5 and 10 mg showed potent growth inhibition activities from 9 hour of incubation. Analysis using transmission electron microscope on E. coli also showed antimicrobial activities of the oils as deformation of the cell and loss of the intracellular materials.

Effects of Dietary Liquid Chitosan on the Performance of Broiler Chickens (액상 키토산의 급여가 육계의 생산성에 미치는 영향)

  • This study were conducted to evaluate effects of drinking concentrated liquid chitosan on performance of broiler. A total of 240 broiler chicks at 7 days of age were fed the commercial diet and water until 47 days of age, drinking water divided into three treatments, 0 ppm (control), 200 ppm (200) and 400 ppm (400) of concentrated liquid chitosan. The body weight gain and feed conversion ratio were not different by drinking concentrated liquid chitosan until 28 days of age, but body weight gain and feed conversion ratio were improve by drinking concentrated liquid chitosan over 28 days of age. Both body weight gain and feed conversion ratio were tend to improve and significantly improved (p<0.05) in 200 and 400 than control at 29 to 35 days of age, respectively. While body weight gain was tended to improve and significantly improved (p<0.05) in 400 and 200 than control at 36 to 47 days of age, respectively. There was a decrease in the microflora population of E coli. in the cecum contents and feaces of broilers by drinking concentrated liquid chitosan. This effect is higher in 400 than 200. The digestibility of dry matter and crude fat of feed were tend to increased in broiler by drinking concentrated liquid chitosan. Digestibility of crude protein of feed of broiler in 200 and 400 was significantly improved (p<0.05) as compared with those in control. These results indicated that when broiler chicks drinking concentrated liquid chitosan of 400 ppm, the improvement of the body weight gain, feed conversion ratio and feed nutrients digestibility, decreasing of microflora population of both E coli. and Salmonella in the cecum contents and feaces.

Investigation on antimicrobial resistance genes of Salmonella spp. isolated from pigs and cattle (소와 돼지유래 살모넬라속균의 약계내성유전자의 특성에 관한 연구)

  • At the present study, it was aimed to detect virulence genes and antimicrobial resistance genes among 102 strains of 12 Salmonella serotypes isolated from pigs and cattle. In polymerase chain reaction (PCR), invA was detected from all strains of Salmonella spp., spvC was detected from Salmonella enterica serotype Enteritidis (S. Enteritidis) (100%), S. Bradenburg (75%), and S. Typhimurium (20.4%). Drug resistance related genes of 12 types were detected from all strains. TEM ($bla_{TEM}$) gene was detected from 51 (92.7%) of 55 $\beta$-lactams (54 ampicillin or 1 amoxicillin) resistance strains. 55 (100%) of 55 chloramphenicol resistance strains, 3 (100%) of 3 gentamicin resistance strains and 5 (100%) of 5 kanamycin resistance strains did contain cml, aadB, and aphA1-Iab, respectively. strB (89.9%), strA (88.4%), aadA2 (84.1%) and aadA1 (72.5%) were detected from 69 streptomycin resistance strains. sulII and dhfrXII were detected from 49 (100%) of 49 sulfamethoxazole/trimethoprim resistance strains, but sulI was not detected. tetA (97.9%) and tetB (21.6%) were detected from 97 tetracycline resistance strains. int gene was detected from 58 (56.9%) of 102 strains. 54 S. Typhimurium of 102 Salmonella spp. were attempted to detect drug resistance genes. TEM was detected from 44 (95.7%) of 46 $\beta$-lactams (45 ampicillin or 1 amoxicillin) resistance strains. cmlA was detected from 51 (100%) of 51 chloramphenicol resistance strains. aadA2 (100%), strA (100%), strB (100%), and aadA1 (79.6%) were detected from 54 streptomycin resistance strains. sulII (100%) and dhfrXII (100%) were detected from 49 sulfamethoxazole/trimethoprim resistance strains. tetA was detected from 54 (100%) of 54 tetracycline resistance strains. int gene was detected from 54 (100%) of 54 strains. The major drug resistance pattern and resistance gene profile were ampicillin, chloramphenicol, streptomycin, sulfamethoxazole/trimethoprim and tetracycline (ACSSuT) and TEM, cmlA, aadA1, aadA2, strA, strB, sulII, dhfrXII, tetA and int, respectively.

Molecular Detection and Epidemiology of Etiologic Agents among Children with Acute Gastroenteritis at a Secondary Hospital from 2015 to 2018 (2015년부터 2018년까지 일개 이차병원에서 동정된 소아 급성 위장염 원인 병원체의 분자진단과 역학의 임상적 연구)

  • Purpose: The multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) test developed recently can help detect enteric pathogens of acute gastroenteritis (AGE). This study aimed to investigate the epidemiology of pathogens in children with AGE using the multiplex RT-PCR. Methods: From May 2015 to June 2018, multiplex RT-PCR tests were performed to identify pathogens in the feces of pediatric patients diagnosed with AGE at a secondary hospital in Seoul, Korea. Results: Of the 1,366 stool samples examined for viral pathogens, 483 (35.3%) tested positive for ≥1 pathogen. Group A rotavirus (RV) was detected in 106 cases (7.8%). The positivity rate increased annually from 3.0% (8/263) to 16.7% (48/288) and surged in 2018 (P<0.001). Norovirus (NoV) GII was the most common viral pathogen (263/1,366, 19.3%), and the positivity rate did not increase during the 3 years. Of the 304 stool samples tested for bacterial pathogens, Campylobacter spp. was the most common bacterial pathogen (32/304, 10.5%), followed by Clostridium difficile (22/304, 7.2%) and Salmonella spp. (17/304, 5.6%). The positivity rate of these bacterial pathogens did not change significantly during the study period. Conclusions: NoV GII is the main pathogen in childhood AGE since the introduction of RV vaccine, yet the number of rotavirus-infected patients increased during our study, especially in 2018. Therefore, further research is needed including the possibility of emergence of novel RV strains. Campylobacter spp. is the predominant cause of bacterial AGE in children. For proper treatment, the clinical characteristics of the bacteria should be taken into consideration, and continuous monitoring is necessary.

Effect of Fucoidan Extracted from Hizikia fusiforme on Intestinal Villi and Salmenolla spp. in Broiler Chicks (톳추출 Fucoidan이 병아리 장내 Villi 및 Salmonella 균주에 미치는 영향)

  • This study was conducted to investigate the in vivo and in vivo antibiotic effect of crude fucoidan extracted from Hizikia fusiforme, and to investigate any possible structural changes of broiler chick's intestinal villi by the supplementation of fucoidan. Total 84 broiler chicks were randomly assigned to 7 treatments, control and Salmonella typhimurium infection groups. The broiler chicks was infected with Salmonella typhimurium at third days, and antibiotics, fucoidan, dried Hizikia fusiforme, dried Undaria pinnatifida and yeast cell debris was respectively supplemented for each group. Each treatment had 4 chicks with three replications. Extraction yield of crude fucoidan from Hizikia fusiforme was 5.453%. Antibiotic effect of fucoidan was not detected in vitro, inhibition zone and micoorganism growth test. Weight gains of broiler chicks were tend to higher in fucoidan treatment group and yeast cell significance was not found. In in vivo test, the number of viable Salmonella typhimurium was low in the antibiotics and fucoidan treatment groups. The intestinal villi were short in the fucoidan and marine algae treatment groups. The intestinal villi were densely distributed on the large intestinal wall, but the morphology was not different among treatments.

Effects of Dietary Korean Mistletoe on Performance and Blood Characteristics in Broilers (겨우살이의 급여가 육계의 생산성 및 혈액 특성에 미치는 영향)

  • The objectives of this study were to examine the effects of Korean mistletoe (KM) on performance and blood characteristics in broiler chickens. A total of four hundred eighty, 1-d-old male broiler chicks (Ross) were divided into 4 groups with 4 replicates of 30 birds each. The treatments were NC (antibiotics-free diet), PC (NC diet added antibiotics), 0.5% KM (NC diet added 0.5% of KM) and 1.0% KM (NC diet added 1.0% of KM), and the birds were raised for 49 days. There were no significant differences in feed intake and body weight gain among treatments. On d 49, the relative length of jejunum in 1.0% KM tended to be increased or significantly (P<0.05) increased compare to those of other groups. On d 35, the colony forming unit of Salmonella spp. of ceca significantly (P<0.05) decreased by feeding KM. The counts of lymphocyte of KM significantly (P<0.05) increased compare to those of the PC.

Evaluation of Functional Properties of Onion, Rosemary, and Thyme Extracts in Onion Kimchi (양파, 로즈마리, 타임의 기능성에 관한 연구)

  • This study was conducted to evaluate the functional properties of onion, rosemary and thyme extracts which are the ingredients of onion Kimchi. Onion extract showed a significant difference in antioxidative effect based on peroxide value and thiocyanate method. Antioxidative activity of rosemary extract was similar to those of BHA and BHT. but thyme extract did not show any antioxidative effect.. Onion, rosemary, and thyme extracts showed antimicrobial activities against gram positive bacteria but onion and thyme extracts did not against E. coli, Candida, and molds. Rosemary extract demonstrated a strong activity against L. plantarum which is a major lactobacillus in Kimchi fermentation, and Micrococcus luteus. Onion, rosemary, and thyme extracts showed an effect or hangover relief effect by lowering the alcohol concentration of blood in rat. Blood pressure of the male spontaneous hypertensive rat was suppressed by onion extract after 3 days of feeding, but rosemary and thyme extracts were not effective for lowering blood pressure.

Inhibition Effect of Sanitizers against E. coli and a Hygienic Condition on the Surface of Utensils and Equipments Used to Food Service (급식기구 표면의 위생상태 및 대장균 소독효과)

  • This study was conducted to investigate inhibition effect of sanitizers against E. coli and a hygienic condition on the surface of utensils and equipments which were used to food service. Samples swabbed using cotton were collected from the surface of stainless steel, wood and plastic utensils and equipments which were used to food service at four elementary schools in Busan and analyzed by measuring the total, coliform and Salmonella spp. count. Total plate counts were 10$^4$~10$^{5}$ CFU/100 cm$^2$ in most of utensils and equipments except plastic cutting board. There were lots of coliforms in slicer (2.8$\times$10$^1$CFU/100 cm$^2$) and peeler (1.1$\times$10$^1$CFU/100 cm$^2$). It was indicated that the sanitary condition of some utensils and equipments such as slicer and peeler should be improved promptly. To investigate inhibition effect of sanitizers against E. coli, the surface of utensils and equipments used in food service was treated at different concentration of sodium hypochlorite for 1 minute and 3 minutes, respectively. The plastic utensils and equipments were most effective aganist E. coli at 100 ppm sodium hypochlorite for 3 minutes. But the stainless steel and wood were most effective at 200 ppm sodium hypochlorite for 3 minutes. It was also treated with 70% ethyl alcohol for 10 seconds and 30 seconds, respectively. The stainless steel utensils and equipments were most effective aganist E. coli at 10 seconds, but plastic and wood were most effective at 30 seconds. Therefore, the results of this study indicated that standardization of disinfection method of utensils and equipments used in food service should be given and sanitation training for dietition should be conducted continuously.
