Development and Validation of the Analytical Method for Oxytetracycline in Agricultural Products using QuEChERS and LC-MS/MS (QuEChERS법 및 LC-MS/MS를 이용한 농산물 중 Oxytetracycline의 잔류시험법 개발 및 검증)

  • An analytical method was developed for the determination of oxytetracycline in agricultural products using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method by liquid chromatography-tandem mass spectrometry (LC-MS/MS). After the samples were extracted with methanol, the extracts were adjusted to pH 4 by formic acid and sodium chloride was added to remove water. Dispersive solid phase extraction (d-SPE) cleanup was carried out using $MgSO_4$ (anhydrous magnesium sulfate), PSA (primary secondary amine), $C_{18}$ (octadecyl) and GCB (graphitized carbon black). The analytes were quantified and confirmed with LC-MS/MS using ESI (electrospray ionization) in positive ion MRM (multiple reaction monitoring) mode. The matrix-matched calibration curves were constructed using six levels ($0.001{\sim}0.25{\mu}g/mL$) and coefficient of determination ($r^2$) was above 0.99. Recovery results at three concentrations (LOQ, $10{\times}LOQ$, and $50{\times}LOQ$, n=5) were from 80.0 to 108.2% with relative standard deviations (RSDs) less than of 11.4%. For inter-laboratory validation, the average recovery was in the range of 83.5~103.2% and the coefficient of variation (CV) was below 14.1%. All results satisfied the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and the Food Safety Evaluation Department guidelines (2016). The proposed analytical method was accurate, effective and sensitive for oxytetracycline determination in agricultural commodities. This study could be useful for safety management of oxytetracycline residues in agricultural products.

Degradation Ability of Fungicide Myclobutanil by Several Soil Bacteria (수종(數種) 토양세균(土壤細菌)에 의한 살균제(殺菌劑) Myclobutanil의 분해력(分解力))

  • This study was carried out to isolate some bacterial strains which had potentiality of good degrader of fungicides from herbicide free soil and to clarify degradation of a fungicide mycrobutanyl[2-p-chlorophenyl-2-(1H-1,2,4-triazole-1-ylmethyl)-hexanenitrile]. Ten strains of the gram-positive and the gram-negative bacteria were isolated and identified. Most of them vigorously proliferated at 55ppm of mycrobutanil, but the stains were not grown when more than 70ppm of this fungicide were treated Staphylococcus spp. I, Actinobacillus spp. III, and another I of the isolated bacteria degraded more than 35% of the treated mycrobutanil. These three strains could utilize mycrobutanil as nitrogen and carbon sources. Mycrobutanil was rapidly decomposed by these strains when applied once or three times. Tested bacteria gradually increased in growth when mycrobutanil was applied repeatedly. Degradation of mycrobutanil and growth of these bacteria were greater in pH 5.5, and they were high in the order of $28^{\circ}C$ > $18^{\circ}C$ > $38^{\circ}C$.

  • Spiroxamine, one of fungicides, is used to control powdery mildew in various crops and black yellow sigatoka in bananas. The major strength of spiroxamine is to control powdery mildew in various crops and bananas yellow sigatoka in bananas. The compound has shown a high level of activity, good persistence and crop tolerance. Besides powdery mildew, good control of rust, net blotch and Rhynchosporium diseases been indicated in cereals, together with a complementary activity against Septoria diseases. In 2017, the maximum residue limit (MRL) of spiroxamine established in Korea. According to Ministry of ood and rug afety) regulations, spiroxamine residues defined only parent compound. Thus, a analytical method is needed to estimate the residue level of the parent compound. The objective of this study was to develop and validate analytical method for spiroxamine in representative agricultural commodities. Samples were extracted with acetonitrile and partitioned with dichloromethane to remove the interfering substances. The analyte were quantified and confirmed liquid chromatograph-tandem mass spectrometer (LC-MS/MS) in positive-ion mode using multiple reaction monitoring (MRM). Matrix matched calibration curves were linear over the calibration ranges ($0.0005{\sim}0.1{\mu}g/mL$) for the analyte in blank extract with coefficient of determination ($r^2$) > 0.99. For validation purposes, recovery studies will be carried out at three different concentration levels (LOQ, 10LOQ, and 50LOQ) performing five replicates at each level. The recoveries 70.6~104.6% with relative standard deviations (RSDs) less than 10%. All values were consistent with the criteria ranges in the Codex guidelines (CAC/GL40, 2003) and MFDS guidelines. proposed analytical method be used as an official analytical method in the Republic of Korea.

  • These experiments were performed to investigate the effects of sublethal doses of fungicides (validamycin A 3 Sp and neoasozin 6.5 Lq) and insecticides (diazinon 34Ec and decamethrin 1Ec) on the possible ways to the resurgence of brown planthoppers (BPH) (Nilaparvata lugens $St{\aa}l$). The possible factors tested in this experiment were fecundity of BPH, longevity, egg-hatchability, amount of honeydew excreted, population build-up following the pesticide treatments in the next generation, and hopperburn rate of potted rice plants. Among the insecticides tested, the applications of diazinon and decamethrin in rice fields may induce the BPH resurgence and accelerate the hopperburn of rice plants through stimulating the fecundity, increasing the adult longevity, promoting the feeding activity and causing the positve population build-up of the BPH

Risk Assessment of Operator Exposure During Treatment of Fungicide Dithianon on Apple Orchard (사과 과수원에서 농약살포시 살균제 Dithianon의 농작업자 위해성 평가)

  • BACKGROUND: Dithianon (75%) formulation were mixed and sprayed as closely as possible by normal practice on the ten farms located in the Mungeong of South Korea. Patches, cotton gloves, socks, masks, and XAD-2 resin were used for measurement of the potential exposure of dithianon on the applicators wearing standardized whole-body outer and inner dosimeter (WBD). This study has been carried out to determine the dermal and inhalation exposure to dithianon during preparation of spray suspension and application with a power sprayer on a apple orchard. METHODS AND RESULTS: A personal air monitor equipped with an air pump, IOM sampler and cassette, and glass fiber filter was used for inhalation exposure. The field studies were carried out in a apple orchard. The temperature and relative humidity were monitored with a thermometer and a hygrometer. Wind speed was measured using a pocket weather meter. All mean field fortification recoveries were between 85.1% and 99.1% in the level of 100 LOQ (limit of quantification), while the LOQ for dithianon was $0.05{\mu}g/mL$ using HPLC-DAD. The exposure to dithianon on arms of the mixer/loader (0.0794 mg) was higher than other body parts (head, hands, upper body, or legs). The exposure to dithianon on the applicator's legs (3.78 mg) was highest in the body parts. The dermal exposures for mixer/loader and applicator were 10 and 8.10 mg, respectively, from a grape orchard. The inhalation exposure during application was estimated as 0.151 mg, and the ratio of inhalation exposure was 11.2% of the dermal exposure (inner clothes). CONCLUSION: The dermal and inhalation exposure on the applicator appeared to be 4.203 mg - 25.064 mg and $0.529{\mu}g-116.241{\mu}g$, respectively. The total exposures on the agricultural applicators were at the level of 2.596 mg - 25.069 mg to dithianon during treatment for apple orchard. The TER showed 3.421 (>1) when AOEL of dithianon was used as a reference dose for the purpose of risk assessment of the mixing/loading and application.
