Antioxidant Activity of Stevia Leaf Extracts Prepared by Various Extraction Methods (다양한 추출방법으로 조제된 스테비아 잎 추출물의 항산화 활성)

  • This study was carried out to evaluate the antioxidant activity of stevia extracts from Stevia rebaudiana Bertoni leaves. Stevia extracts were prepared by three different methods including hot water extraction (HWE) at $120^{\circ}C$ for 4 hr, vacuum extraction (VE) at $65^{\circ}C$ for 4 hr under 0.08 MPa, and fermentation of hot water extract (FHWE) using Lactobacillus buchneri. The antioxidant activities measured by radical scavenging activity, ferric-reducing antioxidant potential ability, and thiobarbituric acid reactive substance showed the highest values in vacuum extract. Also, the antioxidant activities of all extracts were higher than those of stevioside and rebaudioside at the same concentrations, known as the major active components in stevia. To define the antioxidative compound in stevia extracts, the total phenol content was measured, and it was shown that the highest contents of total phenolic compounds were in vacuum extract. These results suggest that the antioxidant activity of stevia extract was due to the phenolic compound components. In addition, vacuum extraction was the proper method to prepare stevia extract with higher antioxidant activity.

  • 최근 디스플레이, 태양전지 그리고 touch screen panels 등 optoelectronic 장치의 시장이 성장함에 따라 투명전극의 수요가 증가하고 있다. Indium tin oxide (ITO)의 좋은 특성 때문에 주로 투명전극에 많이 사용되고 있다. 그러나 화학적 안정성이 떨어지고, 휘어질 때 특성저하가 심하여 금속나노와이어, 탄소나노튜브, 전도성폴리머, 그리고 그래핀 등의 다른 투명전극의 연구가 활발히 진행되고 있다. 그 중에서 그래핀은 높은 전자 이동도(200000 cm2v-1s-1)와 휘어져도 전기적 크게 변하지 않는 특성 때문에 유망한 투명 전도성 전극 (Transparent Conductive Electrodes, TCEs)으로 연구되어왔다. 또한 다양한 속성 가운데, 높은 광 투과성은 그래핀의 가장 큰 장점이다 [1]. 최근, 화학 기상 증착 (Chemical Vapor Deposition, CVD) 등 다양한 제조 방법이 대량 생산을 위해 개발되었다. 그러나 이 방법은 비용이 많이 들며, 과정이 상당히 복잡하고 높은 온도 (${\sim}1000^{\circ}C$)를 필요로 한다. 따라서 용매 기반의 환원된 그래핀 산화물(Reduced Graphene Oxides, RGOs)이 최근 주목 받고 있다. 그러나 RGOs의 면저항이 높아 전극으로서 사용이 제한된다. 따라서 전기적 특성을 향상시키는 방법으로 단일 벽 탄소 나노튜브 (Single-Walled Carbon Nanotubes, SWNTs)를 혼합하거나 화학적 도핑을 통하여 면저항을 크게 향상시키는 연구가 활발히 진행되고 있다. 그러나 이런 화학적 도핑의 경우 박막이 공기 중에 직접 산소나 습기와 반응하여 전기적 특성이 저하되는 문제점을 가지고 있다 [2]. 이러한 문제를 해결하기 위해 AuCl3을 도핑한 박막에 내열성 및 내광성 등의 화학적 안정성이 뛰어난 PEDOT:PSS를 코팅하여 필름의 공기중의 노출을 막아 줌으로써 도핑의 안전성 및 전기적 특성을 최적화하였다. 본 연구에서는 간단한 dip-coating방법을 사용하여 4개의 RGO/SWNTs 박막을 흡착하였다. 다음으로 AuCl3를 도핑하여 면저항 $4.909K{\Omega}$, $4.381K{\Omega}$인 두 개의 샘플의 시간과 온도에 따른 면저항의 변화를 확인하였다. 그리고 필름의 도핑 안전성을 향상 시키기 위해 AuCl3를 도핑한 필름 위에 전도성 폴리머 PEDOT:PSS 코팅하여 면저항 $886.1{\Omega}$, $837.5{\Omega}$인 두 개의 샘플의 시간과 온도에 따른 면저항의 변화를 확인하였다. AuCl3 도핑된 필름의 경우 공기 중에 150시간 노출 시 72%의 면저항 증가가 발생하였지만 PEDOT:PSS가 코팅된 필름의 경우 5%의 면저항 증가가 나타나 확연한 차이를 보였다. 또한 AuCl3 도핑한 필름의 경우 $150^{\circ}C$에서 60시간동안 공기중에 노출되었을 때 525%의 면저항 증가가 발생하였지만 PEDOT:PSS가 코팅된 필름의 경우 58%의 면저항 증가를 나타내었다. 이것은 PEDOT:PSS가 passivation역할을 하여 필름이 공기에 노출된 부분을 막아주어 도핑된 필름의 면저항의 변화를 줄여 주었음을 알 수 있다.

Synthesis and Characterization of Octamethylenethiafulvalene Compounds with Osmium, Iridium, Platinium and Gold Chloride (Octamethylenethiafulvalene과 염화오스뮴, 이리듐, 백금 및 금 화합물의 합성과 특성에 관한 연구)

  • The charge-transfer compound (OMTTF)AuCl$_4$ was prepared from the direct reaction of octamethylenethiafulvalene (OMTTF) with HAuCl$_4{\cdot}xH_2$O in THF. (OMTTF)$_2PtCl_4$, (OMTTF)_2IrCl_6{\cdot}2H_2$O, and (OMTTF)Os$Cl_5{\cdot}THF$ were also formed using $H_2PtCl_6{\cdot}xH_2O$, $H_2IrCl_6{\cdot}xH_2O$ and $H_2OsCl_6$, respectively. The prepared compounds were characterized by magnetic (EPR, magnetic susceptibility), spectroscopic (IR, UV-Vis), electrochemical (CV) methods, and the powdered electrical conductivity measurement. The powdered electrical conductivities at room temperature were ~$10^{-7}S{\cdot}cm^{-1}$. The experimental results show that $OMTTF^+$ monocation radicals exist in all of the prepared compounds. The redox potential of OMTTF supports that $OMTTF^+$ is relatively stable. The magnetic properties indicate that there are significant magnetic interactions between the localized odd electrons on the central metal (Ir and Os) ions and the odd electrons resided on $OMTTF^+$ cation radicals in both (OMTTF) $_2IrCl_6{\cdot}2H_2O$ and (OMTTF)$OsCl_5{\cdot}THF$.

  • Mo isotope, one of highly redox-sensitive isotopes, has been shown to be useful tracers of geochemical processes. Many studies for Mo isotope have documented with the help of recently developed analysis tools, but it has not yet been documented in the Korea. In this study, we introduce two-stage column separation method of Mo using column tube (BioRad PolyPrep(R) column, 10 ml) and anion exchange resin (BioRad Resin AG(R) 1-X8, 200-400 mesh). Mo isotope ratios in the solid SRMs (BHVO-2, SDO-1, PACS-2) and liquid SRM (IAPSO) were measured on MC-ICP-MS (Multi-collector Inductively Coupled Plasma Mass Spectrometer) and then compared with reference Mo isotope ratios. Mo isotope ratios in our study overlap with reference Mo isotope ratios within analytical error.

Studies on Electroanalytical Chemistry for the Oxygen Adducted Tetradentate Schiff base Cobalt(III) Complexes in Pyridine Solution (Pyridine 용액에서 산소 첨가된 네자리 Schiff base Cobalt(III) 착물들의 전기 분석화학적 연구)

  • Tetradentate Schiff base cobalt(II) complexes such as $Co(II)_2-N$, N-bis(salicylidene)-m-phenylendiimine; [$Co(II)_2(SMPD)_2(H_2O)_4$] and $Co(II)_2-N$, N-bis(salicylidene)-p-phenylendiimine: [$Co(II)_2(SPPD)_2(H_2O)_4$], and oxygen adducted cobalt (III) complexes such as [$Co(III)_2O_2(SMPD)_2(Py)_2$] and [$Co(III)_2O_2(SPPD)_2(Py)_2$] in pyridine solutions were synthesized. It was identified that the oxygen adducted cobalt(III) complexes have hexacoordinated octahedral configuration with pyridine and oxygen from the measurement of elemental analysis, AA, IR spectra, and TGA. The redox processes were investigated for the oxygen adducted complexes in 0.1M TEAP-pyridine solution, using cyclic voltammetry on the glassy carbon electrode. The redox processes of oxygen adducted Co(III) complexes result in $$[Co(III)_2-O_2-CO(III)]\rightarrow^{e^-}[Co(III)-O_2-Co(II)]\rightarrow^{e^-}[Co(II)-O_2-Co(II)]\rightleftarrows^{e^-}[Co(II)+Co(II)+O_2{\cdot}^-]\rightleftarrows^{e^-}[Co(II)+Co(I)+O_2{\cdot}^-]\rightleftarrows^{e^-}[Co(I)+Co(I)+O_2{\cdot}^-]$$.

Bacterial Productivity and Degradability of Organic Compounds in the Coastal Area of East Sea (동해 연안역의 박테리아 생산력과 유기물질의 분해능)

  • Bacterial population, signal compounds of pollution, bacterial productivity, heterotrophic activity and the activities of extracellular enzymes were analyzed four times from July 1994 to April 1995 at Daejin, Galnam, Pohang, and Ulsan in the coastal area of Bast Sea in order to investigate the state of eutrophication. The population of heavy metal resistant bacteria in Pohang was the highest among all stations. The bacterial populations which could utilize C -P compounds (phosphonate) showed the highest at Pohang that was expected for the industrial and agricultural effluents to inpour. It could be useful as an index of expectation about the influx of industrial sewages and agricultural effluents. The activities of extracellular enzymes for the degradation of organic matter were the highest in the Ulsan Chuyongam, but low in Daejin. These results showed positive relationship with the population of heterotrophic bacteria and bacterial productivity The ratio of poly-P and DNA in Pohang was the highest, and that in Ulsan, Galnam, and Daejin consecutively, corresponding with the distribution of heterotrophic bacteria. In case of the heterotrophic activities, each of the stations gives difference of seasonal variation in heterotrophic activities. Bacterial productivity seemed to be dependent on seasons; the highest in summer and the lowest in fall and winter. These seasonal variations of bacterial productivity seems to be similar with those of heterotrophic bacteria. It is suggested that the state of eutrophication could be evaluated by the criteria of the population sizes and the productivities of microorganisms, the amount of poly-P, heterotrophic activity, the activities of extracellular enzymes, etc.

Electrochemical Performances of Acid-Treated and Pyrolyzed Cokes According to Acid Treatment Time (산처리 시간별 산화 코크스와 열분해 코크스의 전기화학적 거동)

  • As an activation procedure, in this study, the oxidation treatment of needle cokes with a dilute nitric acid and sodium chlorate $(NaClO_3)$, combined with heat treatment, was attempted. The structures of acid-treated and pyrolyzed coke were examined with XRD, FESEM, elemental analyzer, BET, and Raman spectroscopy. The behavior of double layer capacitance was investigated with the analysis of charge and discharge. The structure of needle coke treated with acid was revealed to a single phase of (001) diffraction peak after 24 h. On the other hand, thecoke oxidized by heat treatment was reduced to a graphite structure of (002) at $300^{\circ}C$. The distorted graphene layer structure, derived from the process of oxidation and reduction of the inter-layer, makes the pores by the electric field activation at the first charge, and generates the double layer capacitance from the second charge. The cell using pyrolyzed coke with 24 h acid treatment and $300^{\circ}C$ heat treatment exhibited the maximum capacitance per weight and volume of 33 F/g and 30 F/mL at the two-electrode system in the potential range of 0~2.5 V.

Adenoviral-Mediated Ref-1 Overexpression Potentiates NO Production in Bradykinin-Stimulated Endothelial Cells (Bradykinin으로 자극한 혈관내피세포에서 Ref-1의 세포내 과발현에 의한 NO 생성 증진 효과에 대한 연구)

  • The dual-function protein redox factor-1 (Ref-1) is essential for base excision repair of oxidatively damaged DNA and also governs the activation of many redox-sensitive transcription factors. We examined the role of Ref-1 in regulation of nitric oxide (NO) synthesis employing adenoviral-mediatedoverexpression of Ref-1 in bradykinin-stimulated endothelial cells. Intracellular NO was detected with the NO-sensitive fluorophore DAF-2. Overexpression of Ref-1 potentiates bradykinin-stimulated NO production in endothelial cells. And, cells ifected with AdRef-1 showed higher fluorescence intensity compared with uninfected or AdD1312-infected cells. In parallel with this, over expression of Ref-1 also stimulated endothelial NO synthase (eNOS) enzyme activity, compared with unifected or AdD1312-infected cells, in bradykinin-stimulated cells as well as in unstimulated cells. These results suggest that Ref-1 implicates in endothelium-dependent vasorelaxation resulting from NO production in vascular system.

Column Bioleaching of Arsenic from Mine Tailings Using a Mixed Acidophilic Culture: A Technical Feasibility Assessment (혼합 호산성 박테리아를 이용한 광미로부터 비소의 Column Bioleaching : 기술적 평가)

  • Heap bioleaching for detoxification of mine tailings is a promising technology; however, long-term studies that aim to understand the potential of this process are scarce. Therefore, this study assesses the feasibility of column bioleaching as an alternative technology for treatment of mine tailings with high concentrations of arsenic during a long-term experiment (436 days). To accomplish this objective, we designed a 350-mm plastic column that was packed with 750 g of mine tailings and inoculated with an acidophilic bacterial culture composed of A. thiooxidans and A. ferrooxidans. Redox potential, pH, ferric ion generation, and arsenic concentration of the off-solution were continuously monitored to determine the efficiency of the technology. After 436 days, we obtained up to 70% arsenic removal. However, several drops in removal rates were observed during the process; this was attributed to the harmful effect of arsenic on the bacteria consortium. We expect that this article will serve as a technical note for further studies on heap bioleaching of mine tailings.

Quality Stability and Antioxidant Activity of Red Ginseng Stored for Long Periods (장기저장 홍삼의 품질안정성과 항산화효과)

  • Samples of red ginseng. which had been manufactured and packaged by the Korea Monopoly Corporation. were stored at ambient temperatures $(12-28^{\circ}C)$ and humidities $(55-68\%)$ for one to nine years to examine their overall quality stability and. in particular. antioxidant activity. The approximate compositions. contents of various solvent extracts. and TLC and HPLC patterns of ginsenosides in the samples which are otherwise susceptible to oxidation. were stable as judged by the changes of the TLC and GLC patterns of the lipids and fatty acids. It was also found that polyunsaturated fatty acids such as linoleic (C18: 2) and linolenic acid (C18: 3) present in the samples had been very stable during the long storage periods. The intensity of the brown color of the red ginseng samples increased significantly with storage time. The pH of the aqueous extracts of the samples also increased slightly during storage. The coloration changes seem to indicate that extensive browning reactions had occurred during storage. The reducing powers of aqueous and ethanol extracts and antioxidant activity of ethyl acetate extracts also increased with storage time. The increase in reducing powers and antioxidant activity appeared to be directly attributable to the increased amounts of non-enzymatic browning reaction products formed progressively during the long storage periods. Therefore. it seems that those antioxidative compounds. which will be progressively formed in red ginseng through non-enzymatic browning reactions during the manufacturing process and long-term storage. will not only contribute to their overall quality stability but also have some significant relationship with their antiaging pharmacological effects.
