인삼, 산양산삼 및 홍삼 추출물의 항비만 효과 (Anti-obesity effects of cultivated ginseng, -wild simulated ginseng and -red ginseng extracts)

  • This study was to investigate whether cultivated ginseng (CG), cultivated wild simulated ginseng (CWG) and cultivated red ginseng (CRG) extracts influences on the obesity. The saponin contents of 3 kinds of ginsengs were analysed by HPLC-ESI-TOF-MS. Total saponin contents were determined in CG on the most contents and since red ginseng has the highest PD (protopanaxadiol type) / PT (protopanaxatriol type) ratio, there may be differences between ginseng, wild ginseng, and red ginseng with respect to their pharmacological effects. Male C57BL/6J mice were fed a normal diet(N), HFD (60% Kcal fat, C), HFD with CG, CWG and CRG extracts (800 mg/kg) for 5 weeks. We observed change of total body weight, degree of hepatic lipid accumulation and immunohistochemical change of GLP-1 and insulin-secreting cells. Also this study attempts to use the physiological analysis method to analyze the changes of blood lipids, insulin and leptin concentration. The change of body weight and size of accumulated lipid droplets in liver lobules decreased in all of the experimental groups than the control(C) group. In the pancreas, the immunohistochemical density of insulin-secreting cells were significantly stronger in the CWG and CRG than C group. The levels of serum insulin and leptin significantly decreased 55.6%, 54.3% respectively in CWG and CRG. The changes of triglyceride, total cholesterol in serum decreased in CRG than the C group. Obesity related CG, CWG and CRG extracts might have contribute to improvement of obesity by regulating the levels of blood lipids and biochemical indicator of fat accumulation.

검정곰팡이의 분화(分化)에 따르는 균체내(菌體內)의 고인산(高燐酸)뉴크레오티드의 소장(消長)에 관한 연구(硏究) (Studies on the Highly-phosphorylated Nucleotides during the Differentiation of Aspergillus niger)

  • 경정곰팡이(Aspergillus niger)의 포자형성(胞子形成)을 액침배양법(液浸培養法)과 동조적방법(同調的方法)으로 여행(旅行)하면서 인산화합물(燐酸化合物), 고인산화(高燐酸化)뉴크레오티드, 및 RNA(핵산)(核酸)의 동태(動態)를 연구(硏究)하였다. 결과(結果)는 다음과 같이 요약(要約)된다. 1. 곰팡이의 포자형성시(胞子形成時)에 유기인산화합물(有機燐酸化合物)의 양(量)이 증가(增加)하였으며, 무기(無機) Polyphosphate의 양(量)은 반대(反對)로 감소(減少)하였다. 2. 무기인산양(無機燐酸量)은 변동(變動)하지 않았으며, 총인산화합물(總燐酸化合物)의 양(量)은 초기균사생장시(初期菌絲生長時)에는 감소(減少)하고 포자낭병시기(胞子囊柄時期)부터는 변동(變動)하지 않았다. 3. RNA핵산구획(核酸區劃)에 있어서 orcinol 반응양성구(反應陽性區)의 흡광도(吸光度)가 급격(急激)히 증가(增加)하였다. 그러나 UV흡광도(吸光度)는 완만하게 증가(增加)하였다. 4. 포자낭병(胞子囊柄)및 포자형성시기(胞子形成時期)에 guanosinetetraphosphate가 검출현(檢出現) 되었다. 5. 고분자양성(高分子量性) RNA (rRNA분획(分劃))의 polyacrylamide gel 전기영동(電氣泳動)을 2.6% gel로서 시행(施行)한 바 polyphosphate와 RNA의 결합물(結合物)이 상당양(相當量) 존재(存在)함을 알았다. 이상(以上)의 결과(結果)로 보아 검정곰팡이의 분화과정(分化過程)에 있어서 고인산화(高燐酸化) RNA 화합물(化合物)의 존재(存在)가 확인(確認)되었으며, 이 화합물(化合物) 가운데 guanosine tetraphosphate (G4P)의 존재(存在)가 진핵미생물(眞核微生物)인 곰팡이에서도 검출(檢出)되었음을 지적(指摘)한다.

천연기념물 한국 산양의 혈액 및 혈액 화학적 정상표준범위 (Hematologic and biochemical reference intervals for natural monument Korean goral (Naemorhedus caudatus))

  • Korean goral (Neamorhedus caudatus) is registered as a natural monument number 217 by South Korea Cultural Heritage Administration. It is also recognized as the endangered species I by Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). In this study, blood samples of Korean goral were collected to make reference intervals. Blood sampling was conducted on 19 numbers of Korean gorals (ten females, nine males), which were reared in Korean Goral Restoration Center. For total samples, the reference intervals of hematological parameters were: white blood cell $7.69{\sim}10.87K/{\mu}{\Lambda}$; hematocrit 36.73~46.18%; red blood cell $10.72{\sim}12.86K/{\mu}{\Lambda}$; hemoglobin 12.79~15.14 g/dL; mean corpuscular volume 33.15~36.75 fL; mean corpuscular hemoglobin 11.53~12.23 pg; mean corpuscular hemoglobin concentration 32.64-35.91 g/dL and red blood cell distribution width 39.2~43.46%. For total samples of each parameters, the following results were obtained for serum biochemistry: glucose 111.81~153.77 mg/dL; blood urea nitrogen 22.35~28.91 mg/dL; creatine 1.22~1.84 mg/dL; phosphate 4.57~6 mg/dL; calcium 8.7~9.1 mg/dL; total protein 6.53~6.92 g/dL; albumin 3.1~3.48 g/dL; globulin 3.26~3.62 g/dL; alanine aminotransferase 56.7~158.56 U/L; aspartate aminotransferase 230.35~473.06 U/L; alkaline phosphatase 178.06~332.47 U/L; gamma-glutamyl transpeptidase 131.6-~181.24 U/L; total bilirubin 1.47~2.12 mg/dL; cholesterol 46.48~71.52 mg/dL; amylase 16.3~26.03 U/L; sodium 150.43~153.88 mmol/L; potassium 3.98~4.6 mmol/L and chlorine 109.48~113.26 mmol/L. The ranges of values were similar campared to previous studies except in the case of RDW value, which showed higher range than the RDW value of a previous study. The reference intervals from this study will be useful data for treatment and management of gorals.

한국재래산양(韓國在來山羊)의 정자발생(精子發生)에 관한 연구(硏究) (Studies on Spermatogenesis in Korean Native Goat)

  • This study was conducted in order to observe the changes in cellular associations of seminiferous tubules from 8 to 20 weeks of age and to obtain the cycle and relative duration of the seminiferous epithelia from 24 to 32 weeks of age. Twenty-eight Korean native male goats were used in the experiment and divided into 7 groups, consisting of 4 goats each, with four weeks intervals from 8 to 32 weeks of age. The results were summarized as follows; 1. Gonocytes were seen at 8 weeks of age, however they were not observed as from 12 weeks. Both type A-spermatogonia and type B-spermatogonia occurred from 8 weeks, while primary spermatocytes were found from 12 weeks. Secondary spermatocytes and spermatids appeared from 16 weeks, and increased in numbers sequentially until 32 weeks of age. Spermatozoa were observed at first at 20 weeks of age. 2. Type A-spermatogonia appeared approximately twice as many at stage 2 as compared to stage 1, while the same numbers of cells were seen in both stages 1 and 8, showing the least number among 8 stages of the cycle of the seminiferous epithelia. The type B-spermatogonia were found during the stage 5 to 8, not to be detactable during stage 1 to 4. The number of primary spermatocytes of the leptotene phase increased markedly during stage 1 to 4, and decreased afterwards. The primary spermatocytes of the pachytene phase were shown the least in number at stage 4. The secondary spermatocytes could be seen only at stage 4 and the largest number of spermatids was seen at the stage 4 among 8 stages. 3. The relative frequencies of each stage among stages 1 to 8 of the cycle of the seminiferous epithelia were 27.5, 17.5, 12.8, 5.8, 8.9, 8.3, 12.0 and 7.2% respectively. 4. Some of the nuclei of Sertoli cells transformed from the "parallel" type to the "perpendicular" type. This evolution took place from stage 1 to 6, when the number of "perpendicular" type nuclei reached a peak and the number was decreased in the rest of the stages. Thus, establishment of spermatogenesis in Korean native goats was completed at the age of 20 weeks.

Levamisole, selenium 및 tocopherol이 한국재래산양의 혈중 호중구 및 복강 대식구의 기능에 미치는 영향 (Effects of levamisole, selenium and tocopherol on the functions of blood neutrophil and peritoneal macrophage of Korean native goats)

  • In this study, the immunomodulating effects of levamisole, selenium and tocopherol on blood neutrophils and peritoneal macrophages of goat were evaluated in vitro and in vivo. The functions of blood neutrophils and peritoneal macrophages were assayed by random and direct migration, phagocytosis of S aureus, production of superoxide and hydrogen peroxide. The results obtained were summarized as follows: In vitro trials 1. Levamisole treatment enhanced the random and direct migration of goat blood neutrophils when compared with untreated cell, and a significant (p<0.01) enhancement was noticed at the concentration of $100{\mu}g$ for direct migration and $50{\mu}g{\sim}1,000{\mu}g\;per\;ml$ of culture medium for random migration. There was no influence of selenium and tocopherol on random and direct migration of neutrophil at all of treatment concentration. 2. Neutrophils produced higher levels of superoxide by lcvamisole treatment at the concentration of $100{\mu}g$ and by selenium treatment at the concentration of $1.0{\mu}g$, but the production of hydrogen peroxide was not increased. Tocopherol had no effect on the production of antimicrobicidal oxygen metabolites of neutrophils at various concentrations. 3. No differences of phagocytic activity were observed when neutrophils were treated with three substances. In vivo trials 1. Blood neutrophils of goats orally administered levaraisole showed significantly (p<0.05) higher random migration from 2 to 24 hours after feeding (2.5mg/kg of body weight). Augmentation of random migration of neutrophil from goats orally administered selenium-tocopherol mixture (selenium $100{\mu}g$-tocopherol 200IU/head/day) was observed at 10 days and the significant (p<0.05) increase was shown from 30 days after feeding and continued throughout the feeding periods. 2. There was no effect on phagocytic activity and production of antimicrobicidal oxygen metabolites of neutrophils from goats administered levamisole or selenium-tocopherol mixture. 3. Random migration, production of superoxide and hydrogen peroxide and S aureus phagocytic activity of peritoneal macrophages of goats administered 300ml of levamisole-thioglycollatc medium mixture $(2.5{\mu}g/ml)$ into peritoneal cavity increased significantly (p<0.01 or p<0.05) when compared with those of goats administered thioglycollate medium alone.

한국재래산양 송과체와 앞쪽목신경절의 관계규명을 위한 면역조직화학적 연구 (Immunohistochemical studies on the relationship between pineal body and superior cervical ganglia of the Korean native goat)

  • The pineal body have been known to be affected by superior cervical ganglia, and most of its nerve fibers containing peptidergic neurotransmitters have been considered to be originated from this ganglia. To confirm this relationships, some peptidergic neurotransmitters were identified in both of pineal body and superior cervical ganglia of the Korean native goat, which were divided into two group; breeding season and non-breeding season. The localizations of two catecholamine-synthesizing enzymes; tyrosine hydroxylase (TH) and dopamine beta-hydroxylase (DBH), were investigated by immunohistochemistry in the superior cervical ganglia and the pineal body of adult Korean native goats. Substance P (SP), calcitonin gene-related peptide (CGRP), vasoactive intestinal polypeptide (VIP), neuropeptide Y (NPY) and galanin (GAL) were also identified in these organs by immunohistochemical and double immunofluorescent methods. In superior cervical ganglia, immunoreactivities for TH and DBH were confirmed in the same ganglion cells. The immunoreactivites for SP, VIP(only in male), NPY and GAL were identified in both of ganglion cell bodies and nerve fibers in the ganglia. CGRP immunoreactivity, however, was observed only in nerve fibers. Most NPY- and VIP-immunoreactive(IR) ganglion cells also contained TH. SP and TH were colocalized in the cell bodies, but not in the nerve fibers. TH immunoreactivity was shown in almost all of ganglion cells in the superior cervical ganglia. The immunoreactivity for NPY had some seasonal variation and was stronger in breeding season than in non-breeding season. In pineal body, lots of TH-IR fibers were observed throughout the parenchyma including the pineal stalk and most of them also contained DBH. SP- and NPY-IR fibers were also immunostained with TH or DBH. But a few SP- and NPY-IR fibers were not colocalized with TH or DBH. Exceptionally, a bipolar neuron-like cell was observed to be immunostained with NPY in the pineal body. A few CGRP and GAL-IR fibers were observed, while VIP-IR fibers were not present. It is concluded that most TH- and DBH-IR fibers as well as the peptidergic immunoreactive fibers of the pineal body might be originated from the superior cervical ganglia. Some peptidergic immunoreactive fibers, however, might be come from other regions of brain. We also suggest that NPY in pineal body plays a important role for pineal function. The seasonal variation of NPY immunoreactivity indicates that the synthesis and use of NPY may be different between in breeding and non-breeding seasons.

한국재래산양의 태아 및 신생아 기관의 조직발달에 관한 조직화학적 및 전자현미경적 연구 1. 광학현미경적 주사 및 투과전자현미경적 연구 (Histochemical and electron microscopical study on the tracheal development in fetuses and neonates of Korean native goats 1. Light microscopic, scanning and transmission electron microscopical studies)

  • The development of trachea in fetuses on 60, 90 and 120 days of gestation and neonates of Korean native goats was investigated by microscopy and scanning and transmission electron microscopy. The results were summarized as follows; Light microscopic studies: 1. In the 60-day-old fetuses, the tracheal walls were differentiated and divided into four layers of the mucosa, submucosa, muscle and cartilage, and adventitia. The tracheal epithelium is composed of stratified ciliated columnar epithelium at 60- and 90-day-old fetuses while the epithelium observed at 120-day-old fetuses was pseudostraified ciliated colummar epithelium. 2. In the 90-day-old fetuses, tracheal glands extended into the submucosa and peripheral area of the tracheal cartilage. The blood vessels were observed in the submucosa and adventitia. The elastic and collagenous fibers were observed in the tracheal walls. 3. In the neonates, the tracheal walls consisted of mucosa with well-developed folds, submucosa, tracheal glands, muscle and cartilage, collagenous and elastic fibers, and adventitia, which were more developed than those of 120-day-old fetuses. The tracheal epithelium was developed as that in adults. Scanning electron microscopic studies: 4. In the 60-day-old fetuses, most of tracheal epithelial cells were nonciliated but short microvilli were sporadically observed on the luminal surface. On rare occasions, a few cells have solitary cilium. 5. In the 90-day-old fetuses, the ciliated cells appeared increasingly and cilia elongated longer than those of 60-day-old fetuses. 6. In the 120-day-old fetuses, the nonciliated cells covered with microvilli in dome-shape were barriered by thick carpet of cilia. The nonciliated cells also have many papillary projectons on the apical surface. 7. In the neonates, the nonciliated cells in tracheal epithelium were covered compactly with numerous cilia, and many secretory droplets were found on the cilia. Transmission electron microscopic studies: 8. In the 60-day-old fetuses, nonciliated cells of the tracheal epithelium contain large amounts of glycogen granules in the supernuclear and subnuclear areas meanwhile a few cell organelles were formed. Cilia were well formed along the apical cell membranes of the ciliated cells. Also found in the ciliated cells were basal corpuscles, mitochondria and short chains in granular endoplasmic reticulum(GER). Between the epithelial cells presented were well-defined junctional complex with zonula occludens and desmosomes. The nuclei were variable in size and shape. The more developed nucleoli were observed conspicuosly. 9. In the 90-day-old fetuses, nonciliated cells contained large glycogen granules. Accumulated glycogen granules were observed in the subnuclear and supranuclear portion of the cytoplasm. A few short microvilli were covered with glycocalyx. Ciliated cells contained numerous mitochondria and short chains of GER. 10. In the 120-day-old fetuses, the ciliated cells contained numerous mitochondria, abundant short chains of GER and nucleoli. Nonciliated cells contained some Golgi complex and mitochondria. The cell borders were well-defined and distinct junctional complex with zonula occludens, desmosomes, and interdigitorum. 11. In the neonates, well-developed goblet cells were observed in the tracheal epithelium. Ultrastructures of ciliated and nonciliated cells on the tracheal epithelia were similar in pattern as those in adults.

pH 및 전해질 조절 산양산삼(山養山蔘) 증류약침(蒸溜藥鍼)의 Apoptosis에 관(關)한 실험적(實驗的) 연구(硏究) (An Experimental Study on Apoptosis of Cultivated Wild Ginseng Distilled Herbal Acupuncture by controlled pH and Electrolyte)

  • Objective : To compare and examine how adjustment of pH and electrolyte can affect the efficacy of cultivated wild ginseng distilled herbal acupuncture, we've administered pure cultivated wild ginseng distilled herbal acupuncture and pH and electrolyte adjusted cultivated wild ginseng distilled herbal acupuncture on A549 human lung cancer lines. Then mRNA and proteins which take parts in apoptosis were examined. Methods : Pure cultivated wild ginseng distilled herbal acupuncture treated group was set as the control group and pH and electrolyte adjusted cultivated wild ginseng distilled herbal acupuncture groups were administered on A549 human lung cancer lines. Cell toxicity was carefully examined and from the analysis of DNA fragmentation, RT-PCR, and Western blot, manifestation of mRNA and proteins which are associated with apoptosis were inspected. Results : The following results were obtained on apoptosis of A549 human lung cancer lines after administering pH and electrolyte adjusted cultivated wild ginseng distilled herbal acupuncture. 1. Measuring cell toxicity of lung cancer cells, higher cell toxicity was detected at pH and electrolyte adjusted groups and the results were concentration dependent. 2. Through DNA fragmentation, we were able to confirm cell destruction in all groups. 3. Experiment groups treated with cultivated wild ginseng distilled herbal acupuncture showed inhibition of Bcl-2 and COX-2 at mRNA and Protein level, whileas increase of Bax was shown. 4. Manifestation of p21, p53, Cyclin E, and Cyclin D1 were confirmed in all groups. 5. Extrication of Cytochrome C was detected at all groups, as well as increased activity of the enzyme caspase-3 and caspase-9, and PARP fragmentation were confirmed. Conclusions : From the above results, we can carefully deduce cell destruction of A549 human lung cancer lines were induced by Apoptosis. At the same concentration level, cell destruction efficacy was better with adjusted pH and electrolyte. Cultivated wild ginseng distilled herbal acupuncture also showed decrease of Bcl-2 and COX-2, as well as increase of Bax. Since cultivated wild ginseng distilled herbal acupuncture increases manifestation of p21, p53, Cyclin E, and Cyclin D1, it affects cellular cycle and through these phenomena, we can consider extrication of Cytochrome C, increase of caspase, and PARP fragmentation are the results.

주정박 사일리지 제조시 인삼박의 혼합이 사일리지의 품질에 미치는 영향 (Effects of Adding Ginseng Meal on the Quality of Distillers Feed Silage)

  • 본 연구에서는 타피오카 주정박에 인삼박을 혼합하여 사일리지를 제조하였을 때, 사일리지의 품질 (시험 I)과 건물 섭취량 및 소화율 (시험 II)에 미치는 영향을 조사하였다. 사일리지 조제시 수분함량을 낮추기 위하여 소맥피를 10% 수준으로 첨가하였으며, 인삼박의 혼합비율은 원물기준으로 0, 15 및 30% 수준 (3 처리)으로 하였다. 처리당 5반복으로 20kg 씩 비닐백에 넣고 공기를 배제하여 포장한 후 $20{\sim}25^{\circ}C$에서 30일간 숙성시켜 공시하였다. 시험 II의 소화 시험에서는 10개월령의 재래산양 수컷 12두 (평균체중 14 kg)를 공시하여서 처리당 4반복이 되게 하였다. 7일 동안의 적응기간을 거친 후, 5일간 소화율을 조사하였던 바, 얻어진 결과를 요약하면 다음과 같다. 1. 인삼박의 혼합 비율이 증가함에 따라 조단백질, NDF 및 ADF의 양이 증가하는 경향을 나타내었으며, 특히 인삼박 30% 첨가구는 인상박 무첨가구와 인삼박 15% 침가구에 비하여 유의하게 높은 결과를 나타내었다 (P<0.05). 그리고 조지방 함량과 hemicellulose 함량은 처리구간에 유의한 차이가 인정되지 않았다. 2. pH는 인삼박을 혼합하므로서 유의하게 감소하였으며, 인삼박 처리구간에 는 유의한 차이가 인정되지 않았다. 젖산 함량은 인삼박 혼합비율이 증가함에 따라 유의하게 많아졌으며(P<0.05), 총산도 인삼박 첨가구가 무첨가구에 비하여 유의하게 많은 결과를 나타냈다. 초산은 인삼박 30% 첨가구가 유의하게 적었다. 한편 낙산은 인삼박 첨가구가 무첨가구에 비하여 유의하게 적었다(P<0.05). 3. 사일리지의 섭취량은 인삼박 첨가구가 무첨가구 보다 유의하게 많았다. 건물 소화율은 인삼박을 많이 혼합할수록 증가하였다. 위의 결과로 볼 때 주정박에 인삼박을 혼합하여 사일리지를 조제하는 것은 주정박 만으로 조제할 때 보다 더 좋은 품질의 사일리지를 만들 수 있으며, 기타 유사 재료에 대해서도 광범위한 적용이 가능할 것으로 사료된다.

  • 본 연구에서는 산림유역의 최상류지역에 위치한 일시하천(ephemeral stream)과 간헐천(intermittent stream)이 상존하는 수변지역을 대상으로 지하수위의 계절적인 변화 양상을 파악하였다. 3곳의 산림유역(총 12개 소유역)에서 일시하천 및 간헐천의 수변구역 보호를 위한 최적관리기법을 포함하는 4개의 처리구(BMP1, BMP2, clearcut, reference)를 적용하였다. 지하수위는 각 소유역별 25개소(총 300개소)에서 2주 간격으로 측정되었으며, 벌채 전 1년과 벌채 후 2년, 총 3년 동안 관측되었다. 전체적으로 지하수위의 반응은 강우량의 영향이 큰 것으로 관찰되었다. 또한, 산림벌채전과 비교하여, 벌채 후 2년 동안 지하수위는 유의적으로 증가하였다. 특히, 벌채 1년 후(2008) 개벌처리구에서 28.2cm, 벌채 2년 후(2009) BMP2에서 54.2cm로 가장 크게 증가하였다. 그러나, 개벌처리구와 BMP2의 지하수위 변화 특성을 비교할 때, 벌채된 목재의 재적과 지하수위의 변화는 직접적인 연관성이 없는 것으로 나타났다. 산림벌채로 인한 지하수위의 증가는 �㎱굇璲� 보다 건조기간 동안 더 크게 나타났으며, 이러한 계절적인 추이는 산림벌채 후 식생구조의 차이에서 야기되는 증발산양의 변화에 기인하는 것으로 사료된다.
