Effect of levamisole, selenium and tocopherol on the lymphocyte blastogenesis and production of antibody in korean native goat (Levamisole, selenium 및 tocopherol이 한국재래산양의 혈중 림프구기능 및 항체생성에 미치는 영향)

  • In this study, effect of levamisole, selenium and tocopherol on the lymphocyte blastogenesis and antibody production in Korean native goat were evaluated in vitro and in vivo. Lymphocyte blastogenesis of goat blood increased significantly (p<0.01 and p<0.05) when the cells were treated in vitro with levamisole at the concentration of $50{\sim}500{\mu}g/ml$, with selenium at the concentration of $0.062{\sim}1.0{\mu}g$ and with tocopherol nt the concentration on $12.5{\mu}g$. Increased lymphocyte blastogenesis was detected from 2 to 24 hours after oral administration of levamisole (2.5mg/kg of body weight). After 7 days, increased mitogenic response of lymphocytes was not detected. Meanwhile increased blastogenesis of lymphocyte from goats given the selenium-tocopherol mixture (selenium $100{\mu}g$-tocopherol 200IU/head/day) was detected from 10 days after feeding, and the tendency continued throughout the entire experimental period. When immune responses of goats against PPD were subjected to test by ELISA, the mean IgG titers of levamisolc group (1 : 1,800) and selenium-tocopherol group (1 : 960) were higher than that of control group (1 : 600) at 2 weeks after lst inoculation. At 3 weeks after lst inoculation and 1 week after 2nd inoculation, the significant (p<0.05) differences in IgG titers were detected among the three groups. The mean IgG titers of levamisole group, selenium-tocopherol group and control at that time were 1 : 20,480, 1 : 5,120 and 1 : 2,640, respectively. The IgG production of levamisole group was significantly (p<0.01) higher than that of control group.

Immunohistochemical studies on somatotropes and mammotropes in hypophysis of Korean native goat (한국재래산양 뇌하수체의 성장자극세포 및 젖샘자극세포에 관한 면역조직화학적 연구)

  • The localizations and morphological characteristics of immunoreactive cells for prolactin(PRL) and growth hormone(GH) antisera were studied with double immunohistochemistry in the adenohypophysis of the Korean native goat. PRL immunoreactive cells(mammotropes) and GH immunoreactive cells(somatotropes) were present in the pars distalis and pars tuberalis, but not in the pars intermedia. A few cells were stained with both PRL and GH antisera (somatomammotropes). The possessional percentages of mammotropes, somatotropes and somatomammotropes were 37.0%, 32.6%, 1.0% in females and 35.6%, 32.6%, 1.1% in males, respectively. Mammotropes were oval or round in shape, and $10{\sim}20{\mu}m{\times}17{\sim}25{\mu}m$ in size. These cells were distributed throughout the pars distalis, but were more abundant on the dorsal part adjacent to the hypophyseal cavity and along the lateral and ventral peripheral regions. Somatotropes were elliptical, triangular or polygonal in shape, and $8{\sim}17{\mu}m{\times}17{\sim}18{\mu}m$ in size. The distribution pattern of somatotropes was similar to that of mammotropes. Some somatomammotropes were intercalated between clusters of mammotropes while the others were dispersed independently among the secretory cells.

The karyotype of Korean native goat (capra hircus) (한국재래산양의 핵형분석)

  • We investigated the cytogenetic characteristics of Korean native goat(Capra hircus). Chromosome slides were prepared from peripheral blood cell cultures. GTG, GBG, RBG and CBG-banding techniques were employed on those slides. The high resolution karyotype of Korean native goat could be made with the incorporation of BrdU. Korean native goat has 60 chromosomes composed of 58 autosomes and XY or XX sex chromosomes. All of autosomes of Korean native goat were acrocentric chromosomes. X chromosome was submetacentric and Y chromosome was metacentric. The GTG, GBG and RBG-band patterns of Korean native goat were similar to those of other goats. CBG-band regions were distinct at the proximal portion of the long arms of all autosomes in Korean native goats. According to our investigation, there was no significant difference in chromosomal band patterns between Korean native goat and other goats. It might be necessary to use molecular genetic markers for clarifying the genetical characteristics of Korean native goat whose biological characteristics are not clearly defined.

  • Hemal nodes and hemolymph nodes are lymphoid organs which share morphologic and functional characteristics of lymph nodes and spleens. Hemal nodes and hemolymph nodes are normally present in Korean native goats. Hemal nodes bad extensive subcapsular and deep sinuses distended by a great number of erythrocytes, and no typical cortex and medulla were observed. Blood vessels commonly occurred, but lymph vessel was not observed in the hemal node. Hemolymph nodes had distinct cortex and medulla, and also had afferent and efferent lymph vessels. The aim of the present study was to obtain new information on the distinct morphological structures of hemal nodes and hemolymph nodes according to ages, and have the basic data for their functions. Goats are divided into 5 groups, consisting of 3 animals aged 1, 3, 6, 10, and 12 months. The morphological studies of the organs were carried out by gross anatomy, light microscopy and immunohistochemistry. During aging, there was an increase in the size of the organs, while there were no significant changes of their numbers, locations and colors. As the goat got older, the lymphatic nodules of hemal nodes were more developed, and the number of macrophage containing phagocytosed erythrocytes was more increased. As the goat was younger, the lymphatic tissues of hemolymph nodes were less developed. There was no difference in distribution of T- and B- lymphocytes according to ages.

  • There have been a number of studies of gonadotropes secreting LH and FSH in the adenohypophysis, but the pattern of hormone storage and secretion of these cells still remains a controversial matter. In this study, we examined whether gonadotropes contained both of LH and FSH, and if so, how these hormones were distributed within the secretory granules. Hypophyseal sections of Korean native goat were simultaneously immunostained for LH and FSH antisera by protein A-gold technique. It was found that most gonadotropes contained both FSH and LH, but hormone storages in the secretory granules were some different among cells. Three types of gonadotropes were identified by the shape and size of the secretory granules and their hormone storage patterns. One type(I) of gonadotropes contained oval secretory granules, which immunoreactivity for FSH and LH were very weak. The size of secretory granules ranged from 160 to 310nm in diameter. Most granules contained both FSH and LH, but some contained only one of them. In another type(II) of gonadotropes, the immunreactivity and hormone storage patterns of the secretory granules were similar to those of type I cells. However, the secretory granules were round in shape and larger in size than those of type I. The other gonadotropes(type III) were distinctly distinguished by plenty of hormones in their secretory granules which were densely packed with numerous immunolabelled gold particles. These data are some inconsistent with other results that have been obtained in other ruminants like as cattle and sheep.

Ultrastructure of hemal node and hemolymph node in Korean native goat (한국재래산양 혈절과 혈림프절의 미세구조)

  • Hemal nodes and hemolymph nodes are lymphoid organs that share morphologic and functional characteristics of lymph nodes and spleens. The aim of the present study was to obtain new informations on the distinct morphological structures of hemal nodes and hemolymph nodes according to ages, and to get the basic data for their functions in Korean native goats. Goats were divided into 5 groups, consisting of 3 animals aged 1, 3, 6, 10 and 12 months, respectively. Ultrastructural features of the organs were observed by transmission and scanning electron microscopes. The sinuses of hemal nodes and hemolymph nodes were lined by endothelial-like reticular cells which had euchromatin-rich nuclei and many cytoplasmic processes, surrounding collagen fibrils. Macrophages containing phagocytosed erythrocytes were often noted in the diffuse lymphatic tissues of hemal nodes and hemolymph nodes. Some mast cells were in contact with the plasma cells near the blood vessel. Hemal nodes and hemolymph nodes had venous sinusal-like vessels which were different from the deep sinus. The lymph vessels with valves were observed in the capsule of the hemolymph node. There were no ultrastructural differences of the organs in the age different groups of the animals. These results suggest that hemal nodes and hemolymph nodes may take part in hemopoiesis, blood filtration and immune reaction in Korean native goats.

  • The ultrastructural investigations of the parotid gland of Korean native goat were carried out by transmission electron microscopy. The results were as follows; 1. The acini of parotid gland were composed of light and dark acinar cells. 2. In the light acinar cells, the secretory granules were classified into three types according to their electron densities and dense bodies. One type of granules was low electron density and had no dense bodies. Another type was low electron density and had dense bodies, and the other type was low electron density and had granular dense bodies. 3. The secretory granules of dark acinar cells showed high electron density and were also calssified into three types by dense bodies as the same way as in the light acinar cells. 4. The intercalated ducts consisted of simple cuboidal epithelium. The nuclei of epithelial cells were oval or round form, located at the central part, and had infolding nuclear membranes and one or two nucleoli. 5. The cells of both of the striated and excretory ducts were composed of light cells, dark cells, specific light cells and basal cells. 6. The nerve terminals were distinguished into two types. One had large granular synaptic vesicles, and another had small agranular synaptic vesicles.

  • The development of reticulum in fetuses between 60, 90, 120 days of gestation and neonates of Korean native goats was investigated by light, scanning electron microscopy. The results were summarized as follows; 1. In the 60-day-old fetuses, the stomach was developed and differentiated into four compartments of rumen, reticulum, omasum, and abomasum. The reticular epithelial layers were differentiated into two zones; a small dark basal and a large light luminar zones. The wall of reticulum resembled that of the rumen except that the mucosa was in the cranio-dorsal region of the reticulum. 2. In the 90-day-old fetuses, the light luminar zone of the reticulum was about 10-16 times thicker than the dark zone. The outlines of the reticular ribs were visible. 3. In the 120-day-old fetuses, the wall of the reticlum had also increased in thickness. The reticular mucosa exhibited an irregular luminar surface and the invaginations had differentiated into large regularly arranged ones separated by 3-5 and small irregularly arranged ones. 4. In the neonate, the luminar surface of the reticular mucosa demonstrated clear furrows, at which the superficial cells of the light zone had undergone degenerative changes. 5. Scanning electron microscopic studies; In the 60-day-old fetuse, numerous microvilli were observed on the superficial epithelial layer of shape or dome like at 120 days. In the neonate, the reticular papillae liked the little finger.

Several undulations of various depths involving the basal zone, basement membrane and lamina propria were observed at irregular within the light zone. 4. In the neonate, the wall of the rumen had increased in thickness and the luminar surface had become wavy due to the appearance of several shallow furrows in the luminar surface of the epithelium especially between the tips of the relatively more developed undulations. 5. Scanning electron microscopic studies: In the 90-day-old fetuses, numerous microvilli and microridges were observed on the superficial epithelial layer of the rumen. The ruminal papillae were already formed at 120 days of gestation. In the 120-day-old fetuses, the ruminal papillae formed short dome-shape. In the neonate, the ruminal papillae formed pebble-shape or short finger like.

  • Six research areas - Hongcheon, Sangju, Jinan, Punggi, Hamyang and aricultural ginseng field near Sangju- were selected to compare microenvironments and photosynthetic capacity. Monthly mean temperature, precipitation and number of hours of clean days have to be identified because these factors are strongly related to the growth and photosynthesis of forest ginseng. Generally, monthly mean temperature in Sangju was highest at June. Monthly mean precipitation in Hongcheon was higher than other areas. In addition, numbers of clean days in Punggi were highest on May and June. Obviously, photosynthetic capacity of younger age class of forest ginseng grown in every 5 places are getting decreasing when forest ginseng becoming olds.
