Effect of Dietary Vitamin A and E on Hepatic Lipid Metabolism in Adriamycin-Treated Rats (Adriamycin을 투여한 흰쥐의 간 지질대사에 미치는 비타민 A와 E의 급여 효과)

  • This study was conducted to evaluate the effect of dietary supplementation of vitamins A and E on hepatic lipid metabolism in adriamycin (ADR)-treated rats. Male Sprague-Dawley rats were assigned to 5 groups according to the dietary supplementation of vitamin A or E. Except control rats, a dose of 2mg ADR/kg of B.W. was injected to these animals intraperitoneally in the same day every week. Lipid peroxide values of liver were elevated by ADR treatment. But dietary supplementation of vitamin A or E significantly reduced these values. Catalase activity in rat liver was increased by ADR treatment, but glutathione peroxidase (GSH-Px) activity was decreased. This effect was somewhat modified by feeding dietary vitamin A or E. Superoxide dismutase (SOD) activity in rat liver was not changed by ADR treatment, but the combined use of ADR, vitamins A and E significantly induced this activity. Fatty acid composition of liver mitochondria was not affected by ADR treatment. The contents of hepatic triglyceride and cholesterol were elevated by ADR treatment. Dietary vitamin A reduced the increased hepatic triglyceride and cholesterol contents in ADR-treated rat.

Comparison of Extraction Methods for the Determination of Vitamin E in Some Grains (곡류 내 비타민 E 분석을 위한 추출방법의 비교)

  • For the determination of vitamin I using high performance liquid chromatography (HPLC), the most critical and time consuming step is the Quantitative extraction of all vitamin E homologs from sample matrix. Three different extraction methods were compared to determine vitamin E in grains. Saponification used alkaline hydrolysis followed by solvent extraction. Direct solvent extraction included dispersing the samples in hot water, addition of isopropanol and $MgSO_4$, and extraction of the analytes with extracting solvent with Polytron homogenization. Using Soxhlet extraction, the samples were extracted in a Soxhlet apparatus with hexane containing butylated hydroxytoluene (BHT). Vitamin E content in 14 grains were analyzed and compared by three different extraction methods. Generally, the highest values were observed from direct solvent extraction and Soxhlet extraction followed by saponification. a-Tocopherol equivalent (a-TE) levels in grains ranged from 0.32 a-TE/100 g in prosomillet to 5.12 a-TE/100 g in black rice.

Effect of Different Ethanol Treatments on the Oxidative Stress in Liver and Brain of SD Rats (알코올 투여 방법이 흰쥐의 간과 뇌에서 산화적 스트레스유발에 미치는 영향)

  • The effect of different ethanol feeding protocols on the oxidative stress in liver and blain of rats was studied. The rats were fed 5%-ethanol solution in drinking water (5%-EtOH group) or 2.5g ethanol/kg body wt. once daily intragastrically (2.5g-EtOH group). The levels of thiobarbituric acid reactive substances (TRARS) and vitamin EI in the liver, cerebrum and cerebellum were measured. In the liver of 5%-EtOH group, the level of TBARS was not changed, whereas vitamin I was significantly increased compared to control group. In the liver of 2.5g-EtOH group, the level of TBARS was significantly increased compared to control group and the vitamin E concentration was not affected. The levels of TBARS were increased and the vitamin E concentrations were decreased significantly both in the cerebrum and cerebellum in 5%-EtOH group as well as in 2.5g-EtOH group. These results show that lipid peroxidation and vitamin E concentration in liver were varied according to the conditions of ethanol treatment, however, the vitamin E contents in cerebrum and cerebellum were affected by both ethanol intoxications used in this study.

The Effects of Vitamin E and C on Serum Cholesterol and Antioxidative enzyme in ovariectomized rat (난소 절제 흰쥐의 혈청 콜레스테롤 및 항산화효소에 미치는 비타민 E와 비타민 C의 효과)

  • In order to observe the bioactivity of ovariectomized rats, ovariectomized group (Ovx), nonovariectomized group (Sham), ovariectomized Vitamin C-treat group (Ovx+Vit C), ovariectomized Vitamin E-treat group (Ovx+Vit E) and ovariectomized Vitamin C+Vitamin E-treat group(Ovx+Vit C+E) were made. Lipidperoxides of liver and kidney, serum total cholesterol and HDL-cholesterol were investigate as follows. Lipidperoxides of liver and kidney in Ovx group were 1.78 times and 1.61 times increased compared to Sham group respectively. But, they were significantly decreased in Ovx+Vit C group, Ovx+Vit E group, Ovx+Vit C+E group compared to Ovx group. Serum total cholesterol in Ovx group was increased 2.57 times compared to Sham group. Injections of each substance such as ascorbate, tocopherol, mixture (C+E) make data of Cholesterol become low. When especially Vit C is injected, the data of cholesterol lowed by about 94%. Serum HDL-cholesterol in Ovx group decreased 36.7% compared to Sham group. And as the result of the measurement of SOD, Catalase, and GPx which are antioxidant enzyme, SOD and Catalase activities in Ovx group much higher than in Sham group. Based on the results, it is supposed that more produced free radicals increased antioxidant enzyme. And it is also thought that vitamin can inhibit aging by reducing antioxidant enzyme.

  • Tocopherol, Se and garlic powder were dieted to hatched chick breeding. The Se content of certain organs is influenced by garlic powder supply. The high Se content for the testis was a function of the vitamin E uptake. Effect of low dose of Se on the growth and survival of rat were examined under the diet of $2\;{\mu}g/ml$ of Se in drinking water either in the form of $Na_2SeO_3$ or $Na_2SeO_4$. The females were dead in early ages while the males were not influenced by dieting the selenite did not make males dying rapidly at early ages and males were less growth depressed. The previously known fact that garlic act as a tonics may be attributable to its high contents of Se and sulfur-containing amino acids which are closely related to vitamin E. Further details on the dietary mechanisms of the Se, vitamin E, and garlic powder are described in this paper.

Effect of Ascorbic Acid Supplementation on Hepatic Microsomal and Mitochondrial Cytochrome P450 System in Diabetic Rats (비타민 C의 보강이 당뇨쥐의 간 소포체와 미토콘드리아의 Cytochrome P450계에 미치는 영향)

  • This study was performed to investigate whether ascorbic acid can modulate the induction of CYP2E1 and prevent the lipid peroxidation which may cause diabetic chronic complication. Diabetes was induced by intraperitoneal injection of streptozotocin to 5-week-old male Sprague-Dawley rats(150~170g). Normal and diabetic group was randomly divided into three groups each; Control(CON, no supplementation), SUP1 (50mg/d ascorbate supplementation) and SUP2(250mg/d ascorbate supplementation). Ascobic acid was prepared daily by dissolving in drinking water and supplied for 4 weeks. There was no difference in hepatic microsomal and mitochondrial P450 contents between normal and diabetes. Hepatic microsomal N-nitrosodimethylamine(NDMA) demethylase activity, which repre-sents contents of CYP2E1, was elevated in diabetes, but not significantly. The NDMA demethylase activity of diabetic SUP2 group was significantly lower activity than that of the diabetic CON group. However, no difference in hepatic mitochondrial NDMA demethylase activity was observed between the diabetes and the normal group. The result suggests that the induction of CYP2E1 in diabetes can be alleviated by ascorbic acid supplementation at the dose of 50mg1d. In addition, ascorbic acid supplementation showed dose-dependent reduction of hepatic microsomal TBARS contents in diabetic rats.
