어류 초기 자어의 로티퍼 최초 섭이 일령과 포식수 분석을 위한 새로운 관찰 및 계수법

  • 로티퍼는 해산어와 담수어뿐만 아니라 식용어와 관상어에 이르기까지 모든 어류의 인공 종묘 생산 과정에서 초기 먹이생물로 이용되고 있는 중요한 동물 플랑크톤이다. 그리고 로티퍼를 초기 자어의 첫 먹이로 사용하는 것을 시작으로 알테미아, 코페포다 또는 초미립자 사료 등을 사용하는 어류 인공종묘 생산 과정에서의 먹이계열이 확립되어 있다. 그러나, 사육하고자 하는 어종에 따라 초기 자어기에서 로티퍼를 섭이하는 시기와 사육 일령별 자어의 로티퍼 포식수를 알 수 있는 정확한 방법이 없다는 이유로 지금까지는 자어의 로티퍼 최초 포식 일령 및 로티퍼 포식수를 개개의 연구자들 나름대로의 방법으로 관찰하여 왔다. 일반적으로 자어가 로티퍼를 포식하였는지의 여부 및 포식수는 자어 소화기내에 존재하는 소화가 진행되기 전의 로티퍼 몸체의 수를 계수하는 방법으로 이루어졌다. 로티퍼의 체내에는 섬모환으로 모은 수중의 먹이 현탁액을 소화기계로 이동시키기 전에 먹이를 잘게 다지고 부수는 기능을 하는 저작기 (trophi)가 존재한다. 그리고 이 저작기는 로티퍼의 종에 따라서 다른 형태를 하고 있는 것으로 알려져 있다. 이 연구에서는 자어가 섭이한 로티퍼는 어류의 소화기내에서 소화가 진행되는 과정에서 로티퍼의 체 구조 중 저작기가 쉽게 소화되기 어렵고 소화기관의 연동 운동에 의해서도 쉽게 그 모양이 손상되지 않는다는 것을 발견하고 이 저작기의 수를 계수하므로서 로티퍼의 최초 섭이 일령 및 일령별 로티퍼 포식수를 계수할 수 있는 방법을 검토하였다. 그 결과, 소화기내의 잔존 로티퍼 몸체만으로 계수 관찰하던 기존 방법에 비교하여 로티퍼의 저작기를 계수하는 방법을 이용할 경우 보다 신뢰도가 높은 결과를 도출할 수 있음을 확인하였다. 경제적 가치의 우수성 때문만은 아니다. 미국은 자국의 대두 생산자들이 생산해 낸 대두를 판매하면서 조성한 자조금을 모아 대두의 생산성을 향상시키기 위한 연구 개발과 국내외 시장을 확대하기 위한 마케팅 활동에 쓰고 있다. 이런 자조 노력은 비단 미국의 대두 농민들에게만 국한된 것이 아니다. 가깝게는 일본의 수산업과 해산어 양식산업의 발전은 중앙 및 지방 정부, 수협과 같은 어민 조합 및 연근해 수산업 종사자들이 출연한 기금을 통해 이루어지고 있음을 지난 8월 북해도의 양식산업을 시찰했을 때 확인할 수 있었던 사실이다. 이제 우리나라도 국민소득 1만불 시대를 넘어 2만불 시대를 열기 위한 청사진을 그릴 만큼 모든 경제의 규모가 확대되었을 뿐 아니라 질적으로 고도화되었다. 우리에게 필요한 것은 성숙된 산업사회에 걸맞는 각 산업 주체들의 의식이다. 한국의 양식산업도 자조 정신을 갖추어 생산성 향상에 초점을 맞춘 양식기술의 개발, 환경 보전을 고려한 양식장 관리 및 소비를 확대하기 위한 유통시스템의 선진화에 스스로 투자를 해야 할 시기에 와 있다.치가 완전히 익은 15일 후에는 무첨가 김치, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 젖산균 수는 5.4X107 cfu/g, 3.3X107 cfu/g, 8.6X106 cfu/g 및 2.6X106 cfu/g로 나타났다. 5. 발효가 진행 중에 대장균 군 수가 시간과 더불어 대장균군 수가 모든 구에 있어서 6일까지는 서서히 증가하다가 그 후부터는 대장균 군 수가 감소하였다. 즉, 발효초기에는 모든 구가 2.0X104∼4.0X105 cfu/g이었고, 6일 경에는 8.9X104∼4.5X105 cfu/g로 약간 증가하였으며 15일후에는 키토산 무첨가 김치가 2.0X

Cooking condition for shelling process of raw oyster and contents of nitrogenous compounds in the cooked-released fluid of oyster (굴 가공 부산물의 이용에 관한 연구 (II) -통조림 원료 굴의 탈각 자숙조건과 자즙중의 질소화합물의 함량분포-)

  • Following the previous report(Kang et al, 1974), the conditions of cooking for shelling process of oyster were discussed, and the content of nitrogenous compounds in the cooked-released fluid of oyster by different cooking conditions has been investigated. The heating condition of $110^{\circ}C$ and 15 minutes was adequate for effective shelling and the condition of meat content in cooking of oyster. The contents of total nitrogen, protein nitrogen, nonprotein nitrogen and amino-nitrogen were seemed to be increased with heating time, and the tendency was to be more exact in the cooking condition of $110^{\circ}C$. The content of nitrogenous compounds at the cooking condition of $110^{\circ}C$ and 15 minutes was determined: total nitrogen, $225.6\;mg\%$ protein nitrogen, $54.4\;mg\%$; amino-nitrogen, $89.2mg\%$. The content of free amino acid was also analysed: arginine, $26mg\%$; histidine, $12.1mg\%$; tyrosine, $2.7mg\%$; leucine, $2.2mg\%$; lysine, $1.9mg\%$ Trace amounts of glutamic acid, isoleucine, serine, threonine, and glycine were detected.

  • The results of experiment to find out the optimum pasteurization condition by heating from $50^{\circ}C$ to $70^{\circ}C$ at intervals of $5^{\circ}C$ and to review chemical composition, microbiological aspects and keeping quality were summarized as follows: 1. The milk sample pasteurized at $50-70^{\circ}C$ showed the decreased pH value from 6.55 to 6.42 while the contents of protein, fat, lactose and ash did not have significant changes. 2. Non-casein nitrogen and filterable nitrogen decreased while casein nitrogen and non protein nitrogen increased as the heat treatment increased. 3. The content of calcium and vitamin C decreased and artificial digestibility for the pasteruized milk increased from 14.07% to 20.00% by the heat treatment. 4. Milk pasteurized at $65^{\circ}C$ had viable bacteria counts $1.3{\times}10^4/m{\ell}$, coliforms negative, psychrotrophic bacteria $9.5{\times}10^2/m{\ell}$ and 99.0% pasteurization effect. 5. Milk pasteurized at $65^{\circ}C$ did not show significant changes in chemical composition and microbiological aspects at $4^{\circ}C$ in refrigerator.

Studies on the Physico-chemical Properties and Sensory Evaluation of Pickled Quail Egg (메추리알 초란의 이화학적 특성과 기호성 평가)

  • This experiment was studied to investigate the physico-chemical properties and sensory evaluation in pickled quail egg during storage at 10 and $20^{\circ}C$ for storage periods 15 days. The weight change of the pickled quail egg were saw a big increase rate with the pickle during 10 days later, and the during storage at $20^{\circ}C$ were increased than the during storage at $10^{\circ}C$. Moisture, crude protein, crude fat and crude ash content of quail egg were 75.24, 12.13, 9.53 and $1.20\%$ respectively. The pH were showed increase pattern for pickle storage periods on 5 days at $10^{\circ}C$. Water soluble nitrogen of quail egg white saw decreased with during the pickle peroids at 10 and 15 days, and the non-protein nitrogen saw increased with according to pickle periods. The change of the viscosity by a rotation viscometer measurement saw increased with according to pickle period days. and the viscosity values saw increased on pickle periods at 5 days. The result of sensory evaluation of sample was S-3>S-2>S-1, in order.

Studies on the chages in Free Amono Acids of Yellow Corvenia(Pseudosciaena manchurice) during Gulbi processing (굴비제조중 유리아미노산의 변화에 관한 연구)

  • Gulbi were processed by salting Yellow corvenia (Pseudosciaena manchurica) with in three ways: the dry salting method with bar salt, the dry salting method with purified salt or with the abdominal brine injection method with purified salt. The half of the sample was dried by the control system of temperature and humidity; the other part was dried by the natural condition. The moisture content of fresh Yellow corvenia muscle and eggs were 76.8%, and 68.2% while those of dried samples decreased to 57.7% and 45.3%, respectively. The total nitrogen content of fresh muscle and eggs were 11.0g% and 7.6g%, respectively (dry weight basis), which decreased slightly during salting and showed no significant changes during drying prosess. The protein nitrogen content of fresh muscle and eggs were 10.2g% and 7.5g%, which decreased during Gulbi processing. On the other hand, the nonprotein nitrogen content of both muscle and eggs increased. The content of free amino acids of fresh muscle and eggs were 508.8mg/100g and 1,110.6mg/100g, which increased to between 5.3 and 2.7 times, respectively after 25 days of drying. The composition patterns of free amino acids in muscle and eggs were similar to each other. The four amino acids - Ala, Glu, Lys and Leu - were most abundant in both fresh and dried samples. These amino acids are known as taste and flavour constituents.

  • Hanwoo has improved the quality of carcass traits. Especially, the higher the grade, the better the texture and taste of Hanwoo. One of the many factors that determine consumer preferences in food is the flavor. The flavor of food is expressed through heat treatment, and fat, carbohydrate and water-soluble compound are important ingredients for flavor. Therefore, The purpose of this study was to investigate the characteristics of fatty acids and carcass traits of 373 Hanwoos. The oleic acid (C18:1) and linolenic acid (C18:3) contents were 48.08% and 0.11%, respectively, while the carcass weight, Eye muscle area, back fat thickness and marbling score of the carcass trait were 383.73 kg, 83.88 ㎠, 10.91 mm, 3.89, respectively. The heritability of oleic acid was estimated to be 0.726, and the phenotypic correlation between eye muscle area and marbling score was estimated to be 0.105 and 0.141, respectively. The phenotype of the marbling score and eye muscle area were positively correlated with Oleic acid. Therefore, the improvement of these two traits seems to affect the increase of unsaturated fatty acid content.

  • The results of experiment to review the optimum pasteurization condition for the raw milk produced in Korea by using UHT procedure of $100-145^{\circ}C$, the changes in chemical composition, microbiological aspects and the keeping quality of the heat treated milk are summarized as follows: 1. In UHT milk sterilized at $100-145^{\circ}C$, the pH value decreased from 6.55 to 6.33 but protein, fat, lactose and ash did not show significance changes while casein nitrogen and non-protein nitrogen increased but non-casein nitrogen and filterable nitrogen decreased. 2. Calcium content decreased gradually from 119.8 mg/100 g of raw milk to 75.75 mg/100 g at $145^{\circ}C$ as the heat treatment increased and vitamin C decreased rapidly from 1.37 mg/100 ml to 0.82 mg/ 100 ml while artificial digestibility increased from 14.07% of raw milk to 26.0% as the heat treatment increased. 3. As the heat treatment increased, microorganism counts decreased to $0.5{\times}10^2/ml$ and were not found above $135^{\circ}C$ - coliforms and psychrotrophic bacteria from $100^{\circ}C$ thermoduric bacteria, thermophiles, mould and yeast from $125-130^{\circ}C$. Heat treatment above $135^{\circ}C$ showed 100% sterilization effect. 4. The result of preservation test for heat treated milk did not show any significant changes in titratable acidity and general composition at $4^{\circ}C$, $25^{\circ}C$ and $37^{\circ}C$ up to 15 days. Viable bacteria counts, coliforms and psychrotrophic bacteria were not found but loss of vitamin and increase in viable bacteria counts appeared after 20 days.

General Composition of Mozzarella Cheese Made from Concentrated Milk Derived by Ultrafiltration (한외여과 농축유로 제조한 모짜렐라 치즈의 일반성분 조성에 관한 연구)

  • We compared the general composition of Mozzarella cheese manufactured by the traditional method and by an ultrafiltration technique. The comparison of the general components between raw milk and the retentate of ultrafiltration (CF 2 and CF 3) demonstrated the following. The fat, protein, and non-fat solid contents of the retentate were higher than those of raw milk, but the lactose content was lower. The effect of 3 factors-fat content (0.5% and 3%), bacterial populations (30,000 CFU/mL and 100,000 CFU/mL), and ratio of added starter and rennet (50, 65, or 80)-on Mozzarella cheese made by the traditional method and made using the retentate of ultrafiltration (CF 2 and CF 3) were compared. Total solids (TS), fat, lactose, total nitrogen (TN), water soluble nitrogen (WSN), non-protein nitrogen (NPN), and salt (NaCl) were affected by fat content, bacterial population, and the ratio of starter and rennet added, but the ash content was not affected. During storage at $4^{\circ}C$ over a period of 3 months, the TS, WSN, NPN, and NaCl contents increased significantly (p<0.05), but fat, lactose, and TN contents decreased significantly (p<0.05), while ash contents remained constant. In whey, the protein, lactose, and solids not fat contents were higher in cheese made from ultrafiltration retentate than in traditional Mozzarella cheese. These results indicate that ultrafiltration can be applied to producing several Mozzarella-type cheeses in order to meet consumer needs.

Studies on the Meat Production and Woolskin Processing of Sheep and Korean Native Goats for Increasing Farm Income as a Family Subsidiary Work (농가부업(農家副業)의 소득향상(所得向上)을 위한 양육생산(羊肉生産) 및 모피가공(毛皮加工)에 관(關)한 연구(硏究))

  • The purpose of the study was to find out possible ways for increasing farm income through the sheep and Korean native goats farming, and to investigate meat productivity, wool productivity; woolskin utility, physiological characteristics and correlation between economical college animal farm of the Chungnam National University and sample farms in the suburbs of Dae jeon City were selected for feeding 20 heads of Corriedale wethers and another 20 heads Korean native kids as research materials for the periods of 5th May-26th November, 1977. The data such as growth rate, carcass, viscera weight, blood picture and plamsa components, hebage intake and economic traits were obtained and analysed. The result of the study are summarized as follows: 1. Meat production and quality 1) After 196days of feeding, the body weight of sheep and Korean native goats was increased by two times of those at the beginning of the trial, i.e. 20kg and 8kg respectively. 2) There was no significance of growth rates of sheep in housing and grazing. 3) The growth rate of Korean native goats were excellent at the mountainous areas of Gong ju-Gun where infectious diseases were not found 4) Accroding to the body measurements of 18-month-old sheep, percentages of hip height, body length, rump length, chest depth, chest width, hip width, chest girth and forearm circumference to the withers height were 103,%, 104%, 33%, 44%, 31%, 23%, 135% and 15% respectively, and those of hip height, body length, chest depth and chest girth of 8-month-old native goats to the withers height were 106%, 109%, 46% and 122,% respecitively. As a result, it was found that the percentage of hip height, body length and chest depth of Korean native goats were higher than those of sheep while that of the chest girth of goats was lower. 5) In the carcass data, 47, $52{\pm}2.27%$ of carcass percentage, $34.61{\pm}1.62%$ of lean meat, $26.07{\pm}2.51%$ of viscera, $9.75{\pm}1.4%$ of bone, and $20.95%{\pm}2.14%$ of woolskin for sheep, and $45.58{\pm}5.63%$ of carcass percentage, $27.62{\p}3.81%$ of meat, $34.86{\pm}4.16%$ of viscera, $11.66{\pm}1.83%$ of bone, $3.63{\pm}1.61%$ of skull and $9.26{\pm}2.41%$ of woolskin for native goats were obtained. 6) The contents of moisture, crude protein, crude fat and crude ash in native goat meat were much similar in both plots of housing and grazing. It was, however, known that the contents of moisture and protein were higher in grazinrg than in housing, while fat content was lower in grazing plots. 7) The weights of visceral organs shown similar tendency for both of sheep and native goats. For the weights of liver, heart, kidney and spleen, significance was not reconized among the treatments. Those of rumen, reticulum, small and large intestine were heavier in grazing than in housing, while the amount of visceral fat was heavier in housing. 2. Wool productivity and woolskin 1) The wool production of sheep for 7 months was $3.88{\pm}1.02kg$, and wool percentage, staple length, straighten length, wool growth per day and number of crimps were $9.27{\pm}1.48%$, 8. $47{\pm}1.00cm$, $10.63{\pm}0.99cm$, $0.40{\pm}0.04cm$ and $2.78{\pm}0.40$ respecitively. 2) The tensile strength and tear strength of woolskin treated by alum tanning were highest on the skin obtained from rump, i.e. $1,351kg/mm^2$ and $2,252kg/mm^2$ respectively, and they are in order of loin and shoulder. 3. Utilization and improvement of pasture. 1) The difference of herbage intake of native goats was not recognized between grazing and tethering, but the intake in the afternoon was s lightly higher than that in the morning. However the hervage intake of sheep was superior in grazing and in the afternoon. 2) The cultivation effect was lower in the native goat plots due to their cultivation abilities, in other words, the establishment rates of pasture by hoof cultivation were 60.25% in the goat plots and 77.35% in the sheep plots. 4. Correlation among economical traits. 1) The correlation between live weight of sheep and daily gain was higher. On the other hand, the correlation between other traits was not significant except that live weight, daily gain and lean meat percentage to the length of thoracic vertebrae. The live weight of native goats and meat production were highly correlated, and high correlation was also found between weights of carcass and meat. However, negative correlation was shown between viscera weight and live weight as well as daily gain. 2) The correlatoin between fleece weight of sheep and other traits such as live weight, daily gain and fleece percentage is very high at the 1% siginficant level, and this means that rapid-growth individuals can produce much fleece. 3) The correlation between the factors such as weights of live body, lean meat and viscera of sheep and body measurements, i. e. chest girth and body length was highest, and weights, of carcass and lean meat was highly correlated to chest width and depth. It will be therefore reasonable that the meat productivity estimates will have to be made on the basis of chest girth and body length. The meat production traits of native goats were highly correlated to the most of body measurement data, and the correlation coefficient between chest girth and weights of live body, carcass, lean meat and bone percentage was very high, i. e. 0.992-0.974 in particular. The correlations of meat production traits to chest depth, forearm circumference, body length were 0.759-0.911, 0.759-0.909 and 0.708-0.872 respectively. Therefore, the meat production of native goats will have to be estimated on the basis of chest data. 5. Blood picture and plasma components. 1) The number of erythrocyte and MCHC of native goats were $12.93{\times}10^6/mm^3$ and 36.14%, and those of sheep were $10.68{\times}10^6/mm^3$ and 36.26 respectively. The values of native goats were significantly higher than those of sheep. 2) The hemoglobin concentration, PVC, MCV and MCR of native goats were 10.92 g/100ml, $23.40{\mu}^3$ and 10.94 pg, and those of sheep were 11.73 g/100ml, 36.25 ml/100ml, $33.97{\mu}^3$ and 30.2 ml/100ml 8.43 pg respectively. The values of native goats were significantly lower those of sheep. 3) The number of leukocytes of native goats was significantly higher than that of sheep, that is, $11.64{\times}10^3/mm^3$ in native goats and $9.32{\times}10^3/mm^3$ in sheep. 4) In differential count of leukocyte, neutrophil was significantly high in native goats while lympocyte in sheep. On the other hand, the basophil, eosinophil and monocyte were not significant between native goats and sheep. 5) The amounts of total protein and glucose in the plasma of native goats were 6.2g/100ml and 53.6mg/100ml, and those of sheep were 5.6g/100ml and 45.7mg/100ml, which means that the values of native goats were significantly higher that those of sheep. The amount of total-lipid of native goats(127.6mg/100ml) was significantly than that of sheep(149.6mg/100ml). 6) The amount of non-protein nitrogen, cholesterol, Ca, P, K, Na and Cl were not different between native goats and sheep. 6. Economic analysis. 1) The gross revenue of a farm which fed native goats and sheep was 4,000won per head and the optimum size for feeding them in a farm as a subsidiary work is 5-10 heads. 2) Since there was no difference between housing and grazing, they can be fed in group for farm's subsidiary work. 3) They can be also fed by youths and house wives in the suburbs of cities, because labour requirement is estimated as only two hours per days for feeding 5 heads of native goats and sheep.
