Determination of Nitrite-scavenging Activity of Seaweed (해조류에 의한 아질산염 소거활성 분석-연구노트-)

  • Nitrite plays an important role in the formation of carcinogenic nitrosamine. In order to effectively inhibit the formation of carcinogenic nitrosamine in food and biological systems, scavenging of residual nitrite is necessary. In screening test of nitrite-scavenging effect of seaweed, The methanol extract from Phaeophyta was more effective than those from Rhodophyta and Chlorophyta. Among the brown algae family, Laminariaceae, A. cribrosum (90.4$\%$) and E. cava (80.1$\%$), E. stolonifera (79.0$\%$) that belong to genus Ecklonia showed marked nitrite-scavenging effect at pH 1.2. Furthermore, these algae showed potent reducing power and radical scavenging activity. The nitrite-scavenging effect was correlated with level of reducing power.

  • 목적 : Acetylcholine은 콜린과 아세트산의 에스테르로 인체에서 중요한 신경전달물질로 Acetylcholine-sterase(AChE)라는 효소에 의해 분해된다. Alzheimer's disease, ataxia, myasthenia gravis, Parkinson' disease 등의 질환에 AChE 억제제가 사용되어 왔으며 최근 한약재의 AChE 억제 효능에 관한 연구들도 진행되고 있다. 봉독은 관절염, 통풍 등의 질환에 응용되어 왔으며 진통효과 및 항염증작용에 대한 임상적, 실험적 연구가 많이 보고되어 왔으나 AChE 억제효과에 대한 연구는 아직까지 보고된 바 없다, 본 연구에서는 봉독약침액과 봉독의 과민반응 유발항원 중 하나인 Phospholipase A2 억제효능이 있는 것으로 알려진 상백피를 혼합한 상백피봉독약침액의 AChE 억제효과를 알아보았다. 방법 : PC12 세포주에서 추출한 AChE와 0.1, 0.01 and $0.001mg/m{\ell}$ 농도의 봉독약침액 및 상백피봉독약침액을 60분간 반응시켰다. 효소면역측정법(ELISA)을 이용하여 흡광도를 10분, 30분, 60분 경과시 각각 측정한 후 효소활성저해도(%)를 계산하였다. 효소활성저해도(%) = [(Cc - Ce)/Cc] ${\times}$ 100 Cc : 대조군 흡광도, Ce : 실험군 흡광도 결과 : 1. 봉독약침액은 0.1, 0.01, $0.001m{\ell}/mg$의 농도에서 30분 경과 후부터 유의성 있는 억제효과를 나타내었다. 2. 상백피봉독약침액은 $0.1m{\ell}/mg$ 농도에서 10분 경과 후부터 유의성 있는 억제효과를 나타내었고, $0.01m{\ell}/mg$ 농도에서 30분경과 시 유의성 있는 억제효과를 나타내었다. 3. 봉독약침액과 상백피봉독약침액의 AChE 억제효과 비교에서 봉독약침액의 억제효과가 상백피봉독약침액 보다 뛰어났다. 요약 : 봉독약침액과 상백피봉독약침액의 AChE 억제효과를 확인하여 두 군 모두 유의성 있는 결과를 얻을 수 있었다. 앞으로 알츠하이머병이나 치매와 같은 신경퇴행성 질환에 대한 봉독의 임상적 활용 및 보다 넓은 범위에 대한 연구가 필요할 것이라 사료된다.

  • PCBs was degraded by bacterium, which was classified as a strain of Alcaligenes aquamarinus. Its PCB-42 degradation was maximized when grown on mineral salts medium containing 0.1% of PCB-42 as a sole carbon source at $25^{\circ}C$ and initial pH $7.0{\sim}8.0$, and also shaking culture was effective for it. The addition of glucose and peptone were effective for the degradation of PCB-42, but metal ions were not effective.

Comparison of General Composition of Cooked Krill and Alcalase Optimization for Maximum Antioxidative Activity by Using Response Surface Methodology (자숙크릴의 일반성분 분석과 항산화 활성을 위한 반응표면법에 의한 알카라제 가수분해 최적화)

  • The objective of this study is to optimize enzymatic hydrolysis of cooked krill by using Alcalase. To optimize krill hydrolysis on such dependent variables as TCA, DPPH-scavenging, and Fe-chelating activities by using Alcalase, independent variables of hydrolysis pH and temperature were investigated Their formulas and three dimensional graphs were obtained by using SAS and Maple softwares, respectively. For comparison of general composition of raw krill, its contents of moisture, crude protein, crude fat, and ash were 17.48%, 53.74%, 15.66%, and 10.21%, respectively, and for cooked krill, its contents were 4.80%, 71.84%, 5.26%, and 15.09%, respectively. The composition of fatty acids for cooked krill was similar to that of raw krill. The most abundant fatty acid was palmitic acid(16:0) and the following order was oleic acid(18:1), eicosapentaenoic acid (20:5), palmitoleic acid(16:1), and docosahexaenoic acid(22:6). For DH optimization of hydrolysates from cooked krill, its result was pH 8.5 and $66.6^{\circ}C$ hydrolysis temperature for the maximum DH of 29.4% For DPPH-antioxidative optimization of hydrolysates from raw krill, its maximum result of 27.1% was obtained in the hydrolysis condition of pH 7.4 and $67.5^{\circ}C$. For Fe-chelating optimization of hydrolysates from cooked krill, its maximum result of 24.9% was in the condition of pH 8.7 and $65.5^{\circ}C$. These results can be used for basic data for using krill products and other fish products as bioactive ingredients.

Sterilization Efficacy of Washing Method Using Based on Microbubbles and Electrolyzed Water on Various Vegetables (다양한 채소류에서 마이크로버블 및 전기분해수의 세척 살균 효과)

  • The sterilization efficacies of various washing solutions on the surfaces of vegetables such as sesame leaves, lettuce, and mini-cabbage were investigated. The washing solutions were tap water (TW), microbubble water (MB), electrolyzed water (EW), and microbubble electrolyzed water (MB+EW). After Escherichia coli and Bacillus cereus were artificially inoculated onto the surfaces of vegetables, each vegetable was washed for 1, 3, and 5 min with TW, MB, EW 100 (100 mg/L of available chlorine), EW 200 (200 mg/L of available chlorine), MB+EW 100, and MB+EW 200. The washing efficacy of MB was slightly higher than that of TW, and EW was more effective than MB (p<0.05). In all instances, the sterilization efficacies of MB+EW 100 and MB+EW 200 were higher than those of EW 100 and EW 200 (p<0.05). Thus, MB+EW offers an effective means of reducing the studied microorganisms in a short time period. The MB+EW washing method provides microbial reduction on the surfaces of various vegetables and enhances the microbiological safety of the vegetables.

Uncertainty decomposition in water resources projection considering interaction effects (교호작용 효과를 고려한 수자원 전망의 불확실성 분해)

  • Water resources projection typically consists of several stages including emission scenarios, global circulation models (GCMs), downscaling techniques, and hydrological models, and each stage is a source of total uncertainty in water resources projection. Several studies proposed methods to quantify the relative contribution of each stage to total uncertainty, and we call such analysis uncertainty decomposition. Uncertainty decomposition enables us to investigate the stages yielding large uncertainties and to establish the uncertainty reduction plan that reflects them. Interactions between stages is one of the important issues to be considered in uncertainty decomposition. This study suggests a new uncertainty decomposition method considering interaction effect. The proposed method has an advantage of decomposing the total uncertainty to the uncertainty from each stage considering both the main and interactions effects. We apply the proposed method to streamflow projection for Chungju Dam basin. The results show that the uncertainties from the main effects are larger than the uncertainties from interaction effects in both summer and winter. Using the proposed uncertainty decomposition method, we show that the GCM stage is the largest source of the total uncertainty in summer and the downscaling technique stage is the one in winter among the following four stages: emission scenarios, GCMs, downscaling techniques, and hydrological models.

Effects of Higher-Order Laue Zone Reflections on HRTEM Images for illumination along an off-Bone Axis of a Crystal (비 결정 축(off-zone axis)으로 입사된 빔에 대한 고 분해 투과전자현미경 이미지에서 HOLZ 반사 빔의 효과)

  • In this paper we explored a possibility of observation for effects of higher-order Laue zone(HOLZ) reflections on high resolution transmission electron microscope(HRTEM) images for illumination along an off-zone axis of a crystal. The analysis of the observation could give useful three dimensional crystal structure information. For the image simulation the Howie-Whelan equation was used with modification of including HOLZ reflections. This study clearly indicates that HRTEM images for a very thin crystal tilted by a few degrees from a zone axis show the effects of HOLZ reflections and contain some information of atomic arrangements along the zone axis.

  • The memory effect of silver and aqua regia in the vessel by their permeation was studied during the microwave digestion of samples under the operating conditions of high temperature and pressure. The extent of permeation in vessels with the solution containing silver 5 mg has been shown to be $0.97{\mu}g/mL$ for the digestion of $HNO_3$ solution and $0.47{\mu}g/mL$ for the digestion of oil sample under condition of MDS 180T program ($180^{\circ}C$, 220 psi).

  • In this paper, we propose a method to improve the background sound separation performance by adding a Wiener filter to the end of the non - negative matrix factorization method. In the case of a mixed voice signal with background sound, a part that has not yet been completely separated may remain in the signal that separated first by the non-negative matrix factorization method. In this case, it can be reduced in proportion to the size of the residual signal due to the Wiener filter, so that the background sound separation or reduction effect can be expected. Experimental results show that the addition of the Wiener filter is more effective than the case of applying the non-negative matrix factorization method.

The Development of Whitening Cosmetic Ingredient Having Activity of Melanin Degradation (멜라닌 분해능을 지닌 미백용 기능성 화장품원료의 개발)

  • Extensive research was carried out for inhibition of melanin formation as development of whitening cosmetic ingredients. But degradation of melanin itself was not intensively pursued as development of cosmetics. In this study, novel melanin degradation enzyme was developed and characterized. Also this enzyme production process was optimized and formulation was tried using micro encapsulation technique.
