  • GamnH-irradiated aqueous solutions of triglycine were studied using low voltage electrophoresis, paper chromatography, and determination of carbonyl compound. Four generalizations can be made as follows: 1. One component from irradiated triglycine formed a complex with copper ions and had higher electronegativity than did triglycine. 2. Five components from irradiated triglycine were separated by paper chromatography; of these, two were identified. 3. Carbonyl compound formation in irradiated triglycine increased up to $5{\times}10^{20}\;ev/ml$ and decreased at higher doses up to $1.9{\times}10^{21}\;ev/ml$ indicating its participation in secondary reaction in the complex system. 4. The possibility of hydrolytic brcabtgc of peptide bond was indicated by the separation of glycine from irradiated triglycine.lycine.

Secretory Overexpression of β-Agarase in Bacillus subtilis and Antibacterial Activity of Enzymatic Products (Bacillus subtilis에서 β-agarase의 분비형 과발현 및 효소분해산물의 항균활성)

  • The gene for ${\beta}-agarase$ of an Agarivorans sp. JA-1 was expressed in Bacillus subtilis DB104, 168 and ISW1214 strains for mass-production. Among 3 host strains, B. subtilis ISW1214 secreted the highest amount of recombinant ${\beta}-agarase$ with a specific activity of 201 U/mg and 360 mg of protein into culture broth. This was approximately 130-fold higher than the production in E. coli as an expression host. Recombinant enzyme produced neoagarooligosaccharides such as neoagarohexaose, neoagarotetraose, and neoagarobiose from agar. Produced neoagarooligosaccharides showed antibacterial activities against gram-negative E. coli and gram-positive B. subtilis at a concentration of 1.5%. These data suggest that neoagarooligosaccharides could be an useful preservative for food industry.

Biodegradation of Polychlorinated Biphenyls (PCBs) within Insulating Oil by Pseudomonas sp. P2 (Pseudomonas sp. P2에 의한 절연류 내의 Polychlorinated Biphenyls (PCBs)의 분해)

  • Polychlorinated Biphenyls(PCBs)의 생물학적 처리가 시도되고 있으며, PCBs를 분해할 수 있는 미생물을 이용할 수 있다. 따라서 본 연구에서는 폐기된 절연유의 생물학적 처리를 위하여 PCBs를 분해하는 균을 분해하였으며, 분해된 균을 이용하여 절연유 내의 Polychlorinated Biphenyls(PCBs) 분해를 회분식 실험에서 연구하였다. 대구의 신천으로부터 유일한 탄소원으로 Biphenyl을 포함하고 있는 고체배지에서 PCBs를 분해할 수 있는 Pseudomonas sp. P2 균주를 분해하였다. PCBs의 용해도를 높이기 위해 사용된 유화제 alkyl aryl ethoxylated phosphate가 200 mg/L에서는 Pseudomonas sp. P2 균주의 성장에 영향을 미치지 않았다. 1000 mg/L의 Biphenyl과 PCBs에 Pseudomonas sp. P2를 접종하여 160시간 배양후에 Biphenyl과 PCBs의 분해가 각각 97.5%, 58.0%였다. Biphenyl 1000 mg/L에서 최대성장율($\mu_{max}$)은 0.34 $day^{-1}$, 0.26 였다. 따라서 염소가 결합되지 않은 Biphenyl는 염소가 결합된 PCBs보다 분해가 빠르게 진행되었다. 또한 Pseudomonas sp. P2는 Biphenyl과 PCBs의 분해로 부터 유도된 황색의 분해대사산물을 확인하였다. 본 연구에서는 Pseudomonas sp. P2 균주가 절연유 내의 PCBs를 분해할 수 있다는 것을 확인하였다.

  • The structures of novel branched oligosaccharides synthesized by the acceptor reaction with dextransucrase from Leuconostoc mesenteriodes M-12 were proposed in accordance with the results obtained from enzymatic hydrolyses and a partial acid hydrolysis. The structure of branched oligosaccharide B4 was shown to be 62-O-$\alpha$-D-kojibiosylmaltose. Branched oligosaccharide B5 was shown to be 63-O-$\alpha$-D-kojibiosylpanose. By reacting the acceptor reaction products with endodextranase a novel branched oligosaccharide (D4) could be produced. D4 was derived from the result of endodextranase hydrolysis of oligosaccarides synthesized by the second acceptor reaction with dextransucrase and was resistant to endodextranase and glucoamylase. The proposed structure of D4 was 62-O-$\alpha$-D-kojibiosylisomaltose. Formation pattern of the acceptor reaction products smaller than d.p. 6 with linear or branched linkage was also shown.

Thermal Acid Hydrolysis Pretreatment, Enzymatic Saccharification and Ethanol Fermentation from Red Seaweed, Gracilaria verrucosa (꼬시래기 홍조류로부터 열산가수분해, 효소당화 및 에탄올 발효)

  • The seaweed, Gracilaria verrucosa, was fermented to produce bioethanol. Optimal pretreatment conditions were determined to be 12% (w/v) seaweed slurry and 270 mM sulfuric acid at 121℃ for 60 min. After thermal acid hydrolysis, enzymatic saccharification was carried out with 16 U/ml of mixed enzymes using Viscozyme L and Celluclast 1.5 L to G. verrucosa hydrolysates. A total monosaccharide concentration of 50.4 g/l, representing 84.2% conversion of 60 g/l total carbohydrate from 120 g dw/l G. verrucosa slurry was obtained by thermal acid hydrolysis and enzymatic saccharification. G. verrucosa hydrolysate was used as the substrate for ethanol production by separate hydrolysis and fermentation (SHF). Ethanol production by Candida lusitaniae ATCC 42720 acclimated to high-galactose concentrations was 22.0 g/l with ethanol yield (YEtOH) of 0.43. Acclimated yeast to high concentrations of specific sugar could utilize mixed sugars, resulting in higher ethanol yields in the seaweed hydrolysates medium.

  • This study was conducted to establish a method to analyze biurea decomposed from azodicarbonamide in processed foods such as wheat flour and bread. New method was developed using high performance liquid chromatography mass/mass spectrometry to determine biurea in wheat flour and bakery products. The recovery rate was 94.3-112.5%. The limit of detection for biurea was 0.003 mg/kg, and the limit of quantification was 0.01 mg/kg. The monitoring results for biurea content using established methods showed that biurea was detected at 2.76 mg/kg in the azodicarbonamide-detected flour (detection rate, 2%). The detection rate in processed foods such as baked goods was 27% (16/59). The detection range was 0.19-18.01 mg/kg (average, 3.79 mg/kg). However, it was thought that the detection level was safe due to much lower values than the standard (45 mg/kg). As a result, the newly established biurea analytical method will contribute to the management of azodicarbonamide in processed foods such as wheat flour and bakery products.

  • Several Prosapogenins and sapogenins obtained by acid hydrolysis or alkaline cleavage of Korean red ginseng saponins were separated and identified by spectral and physical methods. Some of these degradation products showed the cytotoxic activities against various cancer cell lines, that is, A549, SK - OV - 3, L121O, P388 and K562. The significant difference of activity between stereoisooers was not approved and the activity was inversely proportional to the number of sugars binding to sapogenins. It was clear that diol type prosapogenins and sapogenins were more cytotoxic than triol type ones.

  • In order to study the degradation pattern of propanil in solution, the environmental factors such as temperature, pH and UV irradiation effect on propanil degradation were investigated. The degradation of propanil in solution was more rapid in high temperature than in low. The production amount of DCA was increased in high temperature, and then was decreased in a certain period of time because of conversion to TCAB, but the concentration of TCAB was maintained without more degradation. Propanil was rapidly hydrolyzed in alkaline solution as well as in strong acidic solution. Degradation product, DCA was rapidly produced and condensed to TCAB in strong acidic and alkaline condition. On exposur to ultraviolet light, 90% of original propanil was degraded within 20minutes, 0.3 ppm of DCA was produced in 10 minutes, and maintained the concentration throughout irradiation times.

  • 본 연구에서는 산발효산물과 하수슬러지의 혼합소화를 위한 적정 혼합비를 도출을 위하여 음식물찌꺼기 고온 산발효산물과 하수슬러지의 생분해 특성과 적정혼합비를 도출하고자 수행한 결과 음식물찌꺼기 고온 산발효액과 하수슬러지를 혼합비별로 회분식 혼합소화 실험을 실시 한 결과 혼합비 1:1에서 385ml CH$_4$/g VS$_{added}$로 가장 높은 메탄발생량을 나타내었다. 이러한 결과는 음식물찌꺼기와 하수슬러지를 1:1로 동일하게 혼합하여 비교 소화실험을 실시했을 때 의 293m1 CH$_4$/g VS$_{added}$와 비교하여 상대적으로 높은 메 탄발생량을 나타내었다.

와동 이장재의 임상적 응용

  • 치과보존학 영역에서 사용되는 각종 충전재는 다소의 차이는 있으나, 충전후 치질과 완전한 접착을 이루지 못하고, 충전물과 와동벽 사이에 미세공간을 형성하여 변연누출을 야기한다. 즉 타액, 세균, 음식물 분해산물등이 자유로이 이 미세공간을 통해 침투하여 치수와 건강 치질에 이차적인 위해작용을 하기 때문에, 충전의 결과를 더욱 효과적으로 개선시키기 위해서는 이 미세공간을 폐쇄하여 변연노출을 억제시키는 것이 필수적이다. 이에 사용되는 재료가 와동이장재이며, 이의 임상적 응용에 대해 기술하고저 한다.
