Novel Purification and Characterization of Glucose oxidase from Aspergillus niger (Aspergillus niger Glucose oxidase의 새로운 정제 방법 및 특성)

  • Glucose oxidase(EC 1.1.3.4) was purified to electrophoretic homogeneity from Aspergillus niger by a combination of ammonium sulfate fractionation, ion exchange chromatography, and ultrafiltration. Two active fractions A and B, of glucose oxidase were obtained from the hydrophobic chromatography on phenyl sepharose CL-4B. The enzyme A and B were glycoproteins with the same denatured molecular weight of 78, 000 and had specific activities of 2, 191 and 1, 273-units/mg proteins, respectively. But the two enzymes showed differences in native molecular weight that was measured by HPLC gel filteration, maximum absorbtion wavelength and isoelectric point. The enzyme A oxidized $\beta$-D-glucose only and was resistant to sodium dodecyl sulfate. Activity optimum was found at $30^{\circ}C$ and pH 3.5. Also the enzyme A was inhibited greatly by $Hg^{2+}$(10mM). The results of chemical modification experiments suggested that cysteine and cystine residues might be involved in the active site of the enzyme A.

  • Hereafter, the housing construction industry in a domestic country has to give more relative importance to the redevelopment and the rebuilding of existing housing rather then the housing supply methods by the development of new housing sites, and those tendencies are rapidly spreading out. Also, because apartment occupies $50\%$ over of the existing housing, its maintenance, repair, remodeling and reconstruction got to be necessary area for the slumming prevention of a residential area and the disaster prevention occurred by durability lowering of building. But, as having focused on the housing supply methods by the development of new housing sites, the domestic construction industry has relatively carelessly dealt with deconstruction areas including maintenance, remodeling, redevelopment and reconstruction of old existing housing. Therefore the process of the deconstruction is not well suited to regulations and related technology. This paper suggests the present situation of C&D production and new demolition process through comparative analysis of the present demolition works.

The Basalts and Volcanic Process in the Seondol Cinder Cone, Seobjikoji Area, Jeju Island (제주도 섭지코지 선돌 분석구의 화산작용과 현무암)

  • The purpose of this study is to determine the petrology of basalt and the volcanic process in the Seondol cinder cone, Seobjikoji area, eastern Jeju Island. The Seondol cinder cone is mainly composed of spatters in the lower part, pyroclastic deposits including reddish brown blocks, ashes with volcanic bombs in the middle part, and dark black scoria deposits in the upper part. The volcanic sequences suggest volcanic processes that progress through Strombolian eruption and end with Hawaiian lava effusion which breached the cinder cone northwestward and extended over northwestward as lava delta and basalt emplaced as a volcanic neck in the central part of crater in the cinder cone. The age of basalt lava flows is about $95\;{\pm}\;3\;ka$. The basalts belong to transitional basalt and show products of fractional crystallization of clinopyroxene and olivine from a parental basalt magma on the basis of variation diagram of major, trace and rare earth elements. Basalts in the region of this study are plotted at the region of within plate basalt.

Gabor Descriptors Extraction in the SURF Feature Point for Improvement Accuracy in Face Recognition (얼굴 인식의 정확도 향상을 위한 SURF 특징점에서의 Gabor 기술어 추출)

  • Face recognition has been actively studied and developed in various fields. In recent years, interest point extraction algorithms mainly used for object recognition were being applied to face recognition. The SURF(Speeded Up Robust Features) algorithm was used in this paper which was one of typical interest point extraction algorithms. Generally, the interest points extracted from human faces are less distinctive than the interest points extracted from objects due to the similar shapes of human faces. Thus, the accuracy of the face recognition using SURF tends to be low. In order to improve it, we propose a face recognition algorithm which performs interest point extraction by SURF and the Gabor wavelet transform to extract descriptors from the interest points. In the result, the proposed method shows around 23% better recognition accuracy than SURF-based conventional methods.

  • This review paper was written to provide background information as well as future application for aquatic ecologists interested in using stable isotope. Stable isotope techniques has proved to be an extremely useful to elucidate a lot of environmental and ecological problems. Stable isotopes have been used as possible tracers to identify sources, to quantify relative inputs in a system. When utilized carefully, stable isotope tools provides apparent advantages for the scientists to find out the processes of material cycles in various environments and energy flows in natural ecosystems.

Hand Motion Design for Performance Enhancement of Vision Based Hand Signal Recognizer (영상기반의 안정적 수신호 인식기를 위한 손동작 패턴 설계 방법)

  • This paper proposes a language set of hand motions for enhancing the performance of vision-based hand signal recognizer. Based on the statistical analysis of the angular tendency of hand movements in sign language and the hand motions in practical use, we construct four motion primitives as building blocks for basic hand motions. By combining these motion primitives, we design a discernable 'fundamental hand motion set' toward increasing the hand signal recognition. To demonstrate the validity of proposed designing method, we develop a 'fundamental hand motion set' recognizer based on hidden Markov model (HMM). The recognition system showed 99.01% recognition rate on the proposed language set. This result validates that the proposed language set enhances discernaility among the hand motions such that the performance of hand signal recognizer is improved.

토양내 총 NAPL과 공기접촉 NAPL의 측정을 위한 분별 NAPL 분배 추적자 기술의 개발

  • Gaseous partitioning tracer test has been used for determining the volume and spatial distribution of residual non-aqueous phase liquid (NAPL) in the unsaturated soils. In this study, an experimental method for measuring the content of gas-exposed NAPL as well as that of total NAPL in a sand during air sparging was developed. Two different gaseous phase NAPL-partitioning tracers were used; n-pentane, with very low water solubility, was used as the tracer that partitions into NAPL that is only in contact with the mobile gas, and chloroform, with fairly good water solubility, was selected for detecting total NAPL content in the sand. Helium and difluromethanewere used as the non- reactive tracer and water-partitioning tracers, respectively. Using n-decane as a model NAPL (NAPL saturation of 0.018), 25.6% of total NAPL was detected by n-pentane at the water saturation of 0.68. Oniy 9.1% of total NAPL was detected by n-pentane at the water saturation of 0.84. This result implies that the quantity of gas-exposed NAPL increased about three times when the water saturation decreased from 0.84 to 0.68. At the water saturation of 0.68, more than 90% of total NAPL was detected by chloroform while 65.8% of total NAPL was detected by chloroform at the water saturation of 0.84. Considering that the removal rate of NAPL during air sparging for NAPL-contaminated aquifer is expected to be greatly dependent upon the spatial arrangement of NAPL phase with respect to the mobile gas, this new approach may provide useful information for investigating the mass transfer process during air-driven remedial processes fer NAPL-contaminated subsurface environment.

Hierarchical Hidden Markov Model for Finger Language Recognition (지화 인식을 위한 계층적 은닉 마코프 모델)

  • The finger language is the part of the sign language, which is a language system that expresses vowels and consonants with hand gestures. Korean finger language has 31 gestures and each of them needs a lot of learning models for accurate recognition. If there exist mass learning models, it spends a lot of time to search. So a real-time awareness system concentrates on how to reduce search spaces. For solving these problems, this paper suggest a hierarchy HMM structure that reduces the exploration space effectively without decreasing recognition rate. The Korean finger language is divided into 3 categories according to the direction of a wrist, and a model can be searched within these categories. Pre-classification can discern a similar finger Korean language. And it makes a search space to be managed effectively. Therefore the proposed method can be applied on the real-time recognition system. Experimental results demonstrate that the proposed method can reduce the time about three times than general HMM recognition method.

  • Recently, Web applications, such as Stock Image and Image Library, are developed to provide the integrated management for user's images. Image hash techniques are used for the image registration, management and retrieval as the identifier and many researches have been performed to raise the hash performance. This paper proposes GLOCAL image hashing method utilizing the hierarchical histogram which based on histogram bin population method. So far, many researches have proven that image hashing techniques based on histogram are robust image processing and geometrical attack. We modified existing image hashing method developed by our research team. The main idea is that it makes more fluent hash string if we have histogram bin of specific length as shown in the body of paper. Finally, we can raise the magnitude of hash string within same context or feature and strengthen the robustness of hash.

Minimum Classification Error Training to Improve Discriminability of PCMM-Based Feature Compensation (PCMM 기반 특징 보상 기법에서 변별력 향상을 위한 Minimum Classification Error 훈련의 적용)

  • In this paper, we propose a scheme to improve discriminative property in the feature compensation method for robust speech recognition under noisy environments. The estimation of noisy speech model used in existing feature compensation methods do not guarantee the computation of posterior probabilities which discriminate reliably among the Gaussian components. Estimation of Posterior probabilities is a crucial step in determining the discriminative factor of the Gaussian models, which in turn determines the intelligibility of the restored speech signals. The proposed scheme employs minimum classification error (MCE) training for estimating the parameters of the noisy speech model. For applying the MCE training, we propose to identify and determine the 'competing components' that are expected to affect the discriminative ability. The proposed method is applied to feature compensation based on parallel combined mixture model (PCMM). The performance is examined over Aurora 2.0 database and over the speech recorded inside a car during real driving conditions. The experimental results show improved recognition performance in both simulated environments and real-life conditions. The result verifies the effectiveness of the proposed scheme for increasing the performance of robust speech recognition systems.
