Partial Purification and General Properties of Yeast Acetolactate Synthase (효모 Acetolactate Synthase의 부분 정제와 일반 특성 연구)

  • Acetolactate Synthase (ALS) was partially purified from the yeast and its basic biochemical studies were carried out. Yeast was grown in the minimum media containing 0.5% glucose, 51 mM $K_2HPO_4$, 22 mM $KH_2PO_4$, 8 mM $(NH_4)2SO_4,\;0.4\;m M\;MgSO_4$ for 18 hours at 37 $^{\circ}C$. The cell was ruptured in the buffer (20 mM phosphate buffer pH 7.0, 0.1 mM TPP, 0.5 mM DTT, 1 ${\mu}M$ FAD, and 1 mM MgCl_2$) following an overnight suspension. The supernatant fraction was collected from $10,000{\times}g$ and the enzyme was further purified by ammonium sulfate fractionation, DEAE-Sephacel chromatography and leucine-agarose chromatography. The enzyme activity was measured under the various conditions by the function of protein concentration, time, temperature, pH, and substrate. The optimum temperature was found to be 50$^{\circ}C$, optimum pH 8.0∼8.5. The kinetic parameters, $K_m\;and\;V_{max}$ were 8.4 mM and 17.9 nmol/mg/min respectively. Stability of the enzyme was studied with ethylene glycol and glycerol added to the enzyme solution. Both ethylene glycol and glycerol improved the enzyme stability up to 50%. The study of feedback inhibition showed that valine was a strong inhibitor while leucine was a weak inhibitor.

Numerical Implication of Concrete Material Damage at the Finite Element Levels (콘크리트 재료손상에 대한 유한요소상의 의미)

  • The principal objective of this study is to assess the hierarchical effects of defects on the elastic stiffness properties at different levels of observation. In particular, quantitative damage measures which characterize the fundamental mode of degradation in the form of elastic damage provide quite insightful meanings at the level of constitutive relations and at the level of structures. For illustration, a total of three model problems of increasing complexity, a 1-D bar structure, a 2-D stress concentration problem, and a heterogeneous composite material made of a matrix with particle inclusions. Considering a damage scenario for the particle inclusions the material system degrades from a composite with very stiff inclusions to a porous material with an intact matrix skeleton. In other damage scenario for the matrix, the material system degrades from a composite made of a very stiff skeleton to a disconnected assembly of particles because of progressive matrix erosion. The trace-back and forth of tight bounds in terms of the reduction of the lowest eigenvalues are extensively discussed at different levels of observation.

  • In the event of a building fire, it is important to control the smoke generated to ensure the safe evacuation of occupants. A natural smoke ventilator should be installed to exhaust the fire smoke in accordance with the Korean building Act and Code. On the other hand, the present law does not specify the contents regarding natural smoke ventilators sufficiently. The problems that occur in various parts, such as the installation target, installation location, free area, and the control and maintenance of natural smoke ventilators need to be solved. In this study, the problem of the current system was examined through domestic and foreign standards, preliminary research, and field investigations. In addition, suggestions for improvement are provided.

  • This paper proposes a new methodology for the real-time on-line quality monitoring of biofuel processes through the integration of infrared spectroscopy and chemometrics. A method of Partial Least Squares (PLS) in Chemometrics is employed for quantitative analysis of key components in bioethanol products. After a number of preprocessing methods and variable importance in projection (VIP) are used, Savitzky-Golay method showed the best performance in terms of spectrum correction, noise reduction, and model maintenance. The proposed method allows us to economically forecast the concentration of multiple impurities encountered with the production of bioethanol. The proposed system is also accurate enough ($R^2$ > 0.99) to replace the laboratory analysis.

  • In this study, a method of designing a neurointerface using neural network (NN) is proposed for controlling nonholonomic mobile robots. According to the concept of virtual master-slave robots, in particular, a partially stable inverse dynamic model of the master robot is acquired online through the NN by applying a feedback-error learning method, in which the feedback controller is assumed to be based on a PD compensator for such a nonholonomic robot. The NN for the online feedback-error learning can composed that the input layer consists of six units for the inputs $x_i$, i=1~6, the hidden layer consists of two hidden units for hidden outputs $o_j$, j=1~2, and the output layer consists of two units for the outputs ${\tau}_k$, k=1~2. A tracking control problem is demonstrated by some simulations for a nonholonomic mobile robot with two-independent driving wheels. The initial q value was set to [0, 5, ${\pi}$].

  • Cholesteryl pentyl carbonate $(C_{33}H_{56}O_3)$ is monoclinic, space group P21, with a = 12.484(3), b = 9.043(3), c = 14.053(3)$\AA$, ${\beta} = 94.12(2)^{\circ}$ and z = 2. The intensity data were measured for the 2969 reflections within sin $\theta/\lambda = 0.52 {\AA}^{-1}$, using an automatic four--circle diffractometer and graphite monochromated Mo-K$\alpha$ radiation. The atomic coordinates from cholesteryl octanoate were used in an initial trial structure and the structure was refined by full-matrix least squares methods. The final R-factor was 0.12 for 1164 observed reflections. The pentyl group has shortened bond lengths due to the high thermal vibrations in this region. Adjacent molecules are related by $2_1$ screw axis so that they are arranged in an antiparallel array, corresponding to the Monolayer Type II packing mode. There are close packings of cholesteryl groups within the monolayers. This packing type is similar to those of cholesteryl hexanoate, octanoate, hexyl carbonate and oleate.

Analysis on the Reliability and Influence Factors of Refraction Traveltime Tomography Depending on Source-receiver Configuration (송수신기 배열에 따른 굴절 주시 역산의 영향 인자 및 신뢰성 분석)

  • In land seismic exploration, irregular surface topography and weathering layer in near surface distorts the reflected signals of data. Therefore, typical land seismic data should be compensated for this distortion by static correction. To perform the static correction, near-surface velocity is required, which can be obtained by seismic refraction survey. However, land seismic data is often acquired in a limited form of geometry depending on the equipment availability, accessibility condition, and permission for the survey site. In this situation, refraction analysis should be performed using reflection data because it is impossible to acquire refraction-oriented data due to limited source and receiver geometry. In this study, we aimed to analyze the reliability of the results obtained by refraction traveltime tomography when using reflection data with a limited number of sources and receivers from irregular surface topography. By comparing the inversion result from irregular topography with that from flat surface, we found that the surface topography affects the reliability of the inversion results to some degree. We also found that the number of sources has little effect on the inversion results unless the number of sources are very small. On the other hand, we observed that velocity distortion occurred in the overlapped part of receiver arrays when using a limited number of receivers, and therefore suggested the size of the least overlapping ratio to avoid the velocity distortion. Finally, we performed numerical tests for the model which simulates the surface topography and acquisition geometry of the survey region and verified the reliability analysis of inversion results. We identified reliable areas and suspicious area of the inverted velocity model by applying the analysis results to field data.

Proposal of a New Design Method of the Pile-Bent Structure Considering Plastic Hinge (단일 현장타설말뚝의 소성힌지를 고려한 최적설계법 제안)

  • In this study, a new design method of Pile-Bent structure considering plastic hinge was proposed on the basis of the beam-column model. To obtain the detailed informations, the optimized cross-section ratio between column and pile was analyzed to induce the plastic hinge at the joint section between the pile and column. Base on this study, the optimized diameter ratio of pile and column can be obtained below the inflection point of the bi-linear curve depending on the relations between column-pile diameter ratio ($D_c/D_p$) and normalized lateral cracking load ratio ($F/F_{Dc=Dp}$). Moreover, through comparisons with field cases to find out in-depth limit in which minimum concrete-steel ratio could be applied, in-depth limits ($L_{As=0.4%}$) normalized by the pile length ($L_p$) proportionally decrease as the pile length ($L_p/D_p$)increases up to $L_p/D_p=17.5$, and beyond that in-depth limit converges to a constant value (${\simeq}0.3$).

Intra- and Inter-Variation of Protein Content in Soybean Cultivar Seonnogkong (선녹콩 개체간 및 개체내 단백질 함량 변이)

  • Soybean [Glycine max (L.)] is a major source of protein for human and animal feed. Inter- and intra-genotype variation of soybean protein has been investigated by soybean researchers. However, limited sample amount of soybean single seed there is no report that investigated intra-plant variation of soybean protein within soybean plant. Recently a non-destructive NIR (near-infrared reflectance) spectroscopy using single seed grain to analyze seed protein was developed. The objectives of this study were to understand variation of seed protein content within plant and to determine the amount of minimum sample size which can represent protein content for a soybean plant. Frequency distribution of protein content within plant showed normal distribution. There was an intra-cultivar variation for protein content in soybean cultivar Seonnogkong. Difference of protein content among single plants of Seonnokong was recognized at 5% level. Seeds in lower position on plant stem tended to accumulate more protein than in higher position. There was significant difference for protein content between sample size 5 seeds and sample size of more than 5 seeds (10, 20, 30, 40, and 50 seeds) at a soybean plant with 57 seeds however no difference was recognized among sample size (5, 10, 20, and 30 seeds) at a soybean plant with 33 seeds. Around 20% seeds of soybean from single plant needed to determine the protein content to represent protein content of single soybean plant. This study is the first one to report evidence of intra-plant variation for proteincontent which detected by non-destructive NIR spectroscopy using single seed grain in soybean.

Clinical Parameters Predicting Responsiveness to Treatment in Enuresis Patients (야뇨증 치료반응 예측에 관계하는 평가지표)

  • Purpose : We tried to find out the clinical parameters which predict the outcome of treatment in children with enuresis. Methods : Enuresis patients who visited our hospital during 2003-2007 were included. Parameters such as age, gender, height, weight, minimal voided volume, maximal voided volume, maximum functional bladder capacity, frequency of voiding, urine S,G. before and after sleep were measured and an enuresis diary was also recorded. The reduction in wetting frequencies were classified into three groups; none(<50%), partial(50-90%) and complete(90%) response groups. We also compared the 'initial responders' who showed improvement(${\ge}50%$) during the 2 weeks of evaluation and behavioral therapy to the 'initial non-responders'. Results : Parameters mentioned above showed no significant relation to the treatment out-come. The response rate during the 2 weeks of the evaluation period was 32%(49/151) [complete in 1.3% (2/151), partial in 29.6% (47/151)]. Two-months' treatment responses were complete in 14(40%), partial in 19(54.3%) and none in 2(5.9%) responders(n=35), while they were 10(13.5%), 46(62.2%) and 18(24.3%), respectively in the non-responders(n=73) (P<0.05). Conclusion : We suggest that initial 'responsiveness' can be used as a predictor for good treatment outcome in patients with enuresis.
