Cysteine antagonism of captafol induced toxicities in rats 1. Effects on hematological and serum biochemical values (랫트에 있어서 captafol의 독성에 대한 cysteine의 방어 작용 1. 혈액학 및 혈청 생화학적 성상에 미치는 영향)

  • This experiment was carried out to study the preventive effect of cysteine on the toxicities of captafol to the hematological and serum biochemical values. A single dose of captafol(5mg/kg BW, ip) was given to male Sprague-Dawley rats and its toxicities were evalutated by body weitht changes, autopsy findings, absolute organ weight, and hematological and serum biochemical parameters. The single dose of captafol caused significant decreases in body weitht, and absolute liver weight, as-cites, fibrin clot in abdominal cavity, adhesion of liver lobes significant elevation of number of RBC, hemoglobin concentration and serum AST activity, and decreased of serum phospholipid level. Where as cysteine(over 58mg/kg BW) given immediately after captafol appeared to prevent the ascites, fibrin clot in abdominal cavity and liver lobe adhesion. It also prevented the liver and blood, especially RBC toxicites. The results suggest that cysteine and other compounds containing sulfhydryl groups may protect the subjects from captafol-induced toxicity.

Prevention of Abdominal Re-adhesions by Sodium Carboxymethylcellulose in Dogs with Abdominal Adhesions (개에서 복강유착시 Sodium Carboxymethylcellulose를 이용한 재유착의 예방)

  • This study was carried out to prevent the abdominal re-adhesions in dogs. Two randomized groups of dogs were subjected to scratch the ileal serosa. Routine abdominal closure was performed on two groups. Second laparotomy was performed one month later for the scoring of the abhesion formation. The adhesions were separated physically after the scoring of the adhesions. In control goup saline was instilled into abdomen and in treated group 1% sodium carboxymethylcellulose(SCMC) solution was instilled into abdomen before the abdominal closure. Third laparotomy was performed another month later to evaulate the adhesions. Reduced adhesion formation following the instillation of 1% SCMC was reported. 1% SCMC is recommended to prevent the re-adhesions as well as adhesions.

  • Vaginal tumors are the second most common types of tumor in canine female reproductive tract, and most of these tumor are diagnosed as leiomyoma. These tumors are categorized by intraluminal or extraluminal form, and tend to develop as hormone dependent manner. In this report, we describe two cases of extra-luminal form of vaginal tumor. Both tumors were located in abdominal cavity, and adhered to entire surrounding internal organs, including urinary bladder, rectum, and particularly a part of ureter. Surgical excision was performed via laparotomy without damage to the surrounding internal organs and ovariohysterectomy, as well. A diagnosis of vaginal leiomyoma was established based on histopathology.

A Comparison of Three Suture Techniques on Adhesion in End-to-end Intestinal Anastomosis of Dogs (개에서 세가지 단단장문합 봉합법에 따른 유착비교)

  • In this study, we evaluated effects of three anastomotic techniques of small intestine on adhesions in the dog. Twenty six healthy mixed dogs were randomly assigned to three groups. Group I(n = 8) was sutured with a simple continuous suture, group II(n = 7) was sutured with a simple interrupted approximating suture and group III(n = 11) was sutured with a single layer continuous Connell suture. On completion of any intestinal anastomosis, a pedicle of greater omentum was wrapped around the suture line in all experimental dogs. One percent sodium carboxymeth-ylcellulose (5ml/kg) was administrated into the abdomen by feeding tube prior to closing the last part of peritoneum in all dogs. Postoperative adhesions were evaluated at 14th day after operation. The adhesions consisted primarily in two dogs in group I, three dogs in group II and group III. There were adhesions between intestinal serosal surfaces in eight dogs in all groups, but there were no intestinal serosa-visceral peritoneum adhesion and intestinal serosa-mesentery adhesion. Mean adhesion scores were less than score 2 in all groups. Between anastomotic site and omental graft, there were 13.13$\pm$4.97 mm (mean$\pm$S.D.) adhesion formation in group I and 17.29$\pm$4.68 mm in group II and 14.64$\pm$3.80mm in group III. A simple continuous suture resulted in the least adhesion formation and a simple interrupted approximating suture resulted in the greatest adhesion formation among the groups. However, there were no significant differences among three suture techniques in the severity of adhesions. Intestinal intussusception only encountered in one dog during the 14 days, the dog operated and survived. Daily monitoring of temperature, activity, appetite, defecation and micturition were done. All of those vital signs were within normal values and there were no obvious differences among the groups. In conclusion, even though there were no significant differences among three groups, a simple continous suture pattern is recommended to prevent adhesions when operating intestinal anastomosis in dogs.

Careful Approach for Ureteronephrectomy in a Dog with Hydronephrosis causing Dense Adhesion (치밀한 유착조직을 형성한 수신증을 가진 개에서 신요관절제술을 위한 세심한 접근)

  • Unilateral ureteronephrectomy was performed in a dog presented with unilateral hydroureter and hydronephrosis. On laparotomy, severe adhesion of the dorsomedial border of the affected kidney to the abdominal aorta longitudinally and caudal pole of the kidney firmly to the ipsilateral ovary were observed. Since it was not possible to ligate the renal artery and vein separately due to severe adhesion tissue, two mass ligations were applied in the medial border of kidney, and the adhesion tissue was transected distal to each ligature. Then, blunt and sharp dissections were performed to isolate dorsomedial border of the kidney from the abdominal aorta, but they brought out unexpected hemorrhage caused by incision of renal artery and it has been suspected that two mass ligations could not include renal artery, which was adhered to dorsomedial border of the kidney. The hemorrhage was controlled by double ligations and electrocautery. In this case report, we recommend that if isolation of renal vessels is impossible due to strict adhesion of kidney to adjacent tissue, the mass ligation could be chosen to isolate and exteriorize the kidney and placed carefully not only at medial border of the kidney, where the renal vessels come into the kidney anatomically but also additional border of the kidney where adhesion tissues are formed.

Using of Polypropylene Mesh for Peritoneal Defect induced Gossypiboma in a Shih-Tzu Dog (시츄견에서 Polypropylene mesh를 이용한 gossypiboma에 의한 복막 결손 수복 증례)

  • A 10-year-old spayed female Shih-tzu dog with a left-sided caudal abdominal swelling of 2-month clinical duration was referred to the Konkuk University Teaching Hospital (KUVTH). We confirmed the presence of an abdominal mass by radiography and ultrasongraphy. A gossypiboma was identified between bladder and small intestines at surgery and removed. The resulting peritoneal adhesion was severe. But, the peritoneal defect did not allow easy repair of the abdominal wall. Therefore, polypropylene mesh (PPM) was used in repair. The patient was good condition after surgery with no caudal abdominal swelling. We followed the patient for the next 6 months with no complication.

  • A massive mycetoma was observed in the abdominal cavity of a 3-year-old spayed female Labrador Retriever dog originated from France. The mycetoma lesion involved adhesion with one-half of the spleen, several portions of the small and large intestine, and the diaphragem. Histologically the distinct hypertrophy of the intestinal muscle layer, wide-spread deposition of the hemosiderin through the affected abdominal organs and thrombosis of splenic arterioles with atheromatous change were recognized as persistant, debilitating changes of the affected organs. From the histological appearance of the fungal colonies it was presumed that the causative fungus to be identified as Allescheria boydii.

Transgastric Endoscopic Cholecystectomy in a Dog : Natural Orifice Transluminal Endoscopic Surgery (개에서 내시경을 이용한 경위장관 담낭절제술 1예 : 자연개구부 내시경수술)

  • Transgastric endoscopic cholecystectomy was successfully accomplished in a 1-year-old, 15 kg, female, mongrel dog. Single-working channel flexible gastric endoscope was used with the aid of one abdominal laparoscopic port. Gastrotomy was performed using endoscopic needle knife at the ventral antral region. Through the gastric incision endoscope was advanced and retroflexed for the visualization of gallbladder. For the better exposure of surgical field, gentle traction was applied at the fundus of the gallbladder using laparoscopic grasping forceps. Cystic duct and artery was ligated using endoclips. After transecting the duct and artery, gallbladder was dissected using endoscopic coagulating grasping forceps and needle knife. Resected gallbladder was retrieved through the mouth and gastric incision site was sutured using endoclips. There was no evidence of bile leakage or stomach leakage on postoperative day (POD) 3. On POD 16, gastric endoscopy and laparoscopy was performed. Gastric endoscopy revealed complete adhesion of incision site. The content of the peritoneum appeared healthy, with no sign of infection, bile staining, or organ injury. The omentum was adhered over resected gallbladder fossa and the serosal surface of gastrotomy site. This is the first report of NOTES cholecystectomy in the dog and provides new concept of cholecystectomy of the dog.

Comparative Effect of Carboxymethylcellulose, Chondroitin, and Carboxymethylchitosan on Preventing Intraperitoneal Adhesion Formation in Rats (Rat에서 carboxymethylcellulose, chondroitin 및 carboxymethylchitosan의 복강유착방지 효과의 비교)

  • This study was performed to compare the effects of carboxymethylcellulose (CMCE), chondroitin sulfate (Chondron), and carboxymethylchitosan (CMCH) on preventing intraperitoneal adhesion. As a result, the tensile strength of adhesions formed between the parietal peritoneum and the ileal serosa was significantly decreased in the groups of three different kinds of anti-adhesive agents. The distance of adhesion site was slightly increased in the treatment groups comparing control group. In the CMCH group, the inflammatory cell infiltration, collagen hyperplasia, and neovascularization were significantly lower than those of control group. It was observed that the damage at intestinal serosa was significantly decreased in the chondron and CMCH groups comparing control group. Therefore CMCH may be useful as a anti-adhesive agent in the prevention of intraperitoneal adhesion in rats.

Protective Effect of the Inhibition of PAF Remodeling and Adhesion Molecule on the Oxidative Stress of the Lungs of Rats Given Endotoxin Intratracheally (내독소에 의해 유도된 급성 폐 손상에서 PAF Remodelling 및 Adhesion Molecule의 억제가 폐장내 Oxidative Stress에 미치는 영향)

  • Backgraound : There have been many reports on the pathogenesis of sepsis-induced acute respiratory distress syndrome(ARDS) but, the precise mechanism has not been elucidated. This study examined the protective effect of an inhibition of platelet activating factor(PAF) remodeling and the adhesion molecule on the oxidative stress of the lungs in rats with an endotoxin induced acute lung injury(ALI). Methods : ALI was induced in Sprague-Dawley rats by instilling an E-coli endotoxin into the trachea. Ketotifen and fucoidan were used respectively to inhibit PAF remodeling and adhesion molecule. The lung leak index, lung myeloperoxidase(MPO) activity, bronchoalveolar lavage(BAL) fluid neutrophil count and lyso PAF acetyltransferase activity(AT), were measured and an ultrastructural study and cytochemical electron microscopy were performed. Results : The lung leak index, lung MPO activity, BAL fluid neutrophil count and lyso PAF AT activity was higher in the endotoxin-treated rats. In addition, severe destruction of the pulmonary architecture and increased hydrogen peroxide production were identified. These changes were reversed by ketotifen. However, fucoidan did not appear to have any protective effects. Conclusion : The inhibition of PAF remodeling appeared to be effective in decreasing the endotoxin-induced ALI. In addition, this effect might be derived from the inhibition of neutrophilic oxidative stress. However, the inhibition of the adhesion molecules by fucoidan appeared to be ineffective in decreasing the endotoxin-induced ALI.
