Germination Characteristics of PEG Priming Seed in Barley (Hordeum vulgare L.) (PEG로 프라이밍 된 보리(Hordeum vulgare L.)종자의 발아특성)

  • These experiments were conducted to evaluate the variability of seed germination, and seedling growth of PEG priming barley. The average germination percentage(AGP) of PEG priming seed was higher than control, but those were low with extend the treatment period. The AGP of washing seed after PEG priming was higher than unwashed seed, and that of redried seed after priming was lower than control. The germination time of priming seed was short compare to control seed, and that was prolonged with extend the priming period, and that of washing seed after priming was shortening, and that of redried seed after priming was prolonging. The emergence percentage(EP) of priming seed was higher than control, and the emergence time was shortest in 50% field moisture capacity soil. The seedling and root length of priming seed was shorter than control.

  • The seeds of three barley varieties of which initial seed vigor were different were used to measure seed vigor of accelerated aging techniques. A vigor index derived from the product of percent germination and plumule length was used to estimate seed vigor. The index was compared with the results of other tests. The results of warm germination test well suited to the measurements of seed vigor at the advanced stages of seed deterioration. Other vigor measurements except ATP and GADA values, showed close relationships with the vigor index. The measurements of plumule length in cold test and tetrazolium test were found to be appropriate for predicting seed quality.

Effects of PEG Treatment on Seed Viability and Seedling Emergence in Rice, Barley and Wheat (벼, 보리, 밀 종자의 PEG 처리가 종자활력과 포장출아에 미치는 영향)

  • The effects of priming with different polyethylene glycol (PEG 6, 000) solutions on the germination, emergence characteristics and early plant growth in rice, barley and wheat were investigated. Rice, barley and wheat seeds were subjected to various priming conditions of osmotic potentials (-0. 75, -1.00 and -1.50 MPa) of PEG, and incubation period were 5 days at 25, 20, 2$0^{\circ}C$, respectively. The percentage of water absorption was highest in barely, and lowest in rice, and all the treatments enhanced water absorption in barley, but showed no significant effects in rice and wheat. Respiration quotient was lower than none PEG treatment seeds, and those of barley and wheat were higher than rice regardless of PEG concentration, and that of rice increased with high PEG concentration. Total germination percentage of osmoconditioning seeds with PEG was higher than that of none treatment seed, and those of barley and wheat were significant. The artificially deteriorated seeds with PEG treatment seeds after ageing treatment could recover to nearly the same germination level as that of the control seeds. The effects of coating polymer were higher than osmoconditioning with PEG, and germination characteristics in rice showed varietal difference at PVP and waterlock at recoated seeds after PEG treatment. Osmoconditioning with PEG reduced mean germination and emergence time, but there was no difference among PEG concentrations. The plant height of PEG treaed seed in rice was taller and those of barely and wheat showed varietal difference, and those of polymer-coated seed after PEG treatment were different among the polymers. The dry weight of PEG treatment were different among the crops, and those were increased with the high PEG concentration. The emergence percentage of PEG-treated seed were higher than none-treated seed, and those were decreased with the increased PEG concentration, and the highest emergence percentage of rice, barley and wheat were 90, 50 and 50% soil moisture content, respectively. The time to emergence in rice was longer than barley and wheat, and those in rice was shortened in high soil moisture content, and barley and wheat were shortened in low soil moisture content.

A Study on the Efficient Germination of Barley Seed using Electrostatic Field (정전기장을 이용한 보리종자의 효율적 발아에 관한 연구)

  • This paper discusses methods for increasing the germination rate of barley seeds using low direct and alternating current power below 110V. The experimental apparatus used here is a parallel plate, with the bottom surface of the plate designed to be wider than the top surface to increase the size of the electrostatic field. As a result, three different magnitudes of electrostatic fields were created on the plates: the first ranging from 400V/cm to 600V/cm, the second from 600V/cm to 900V/cm, and the third from 2200V/cm to 2400V/cm. The finite difference method was applied to analyze the electrostatic field inside the parallel plate. The plant seeds used in the experiments were barley seeds produced domestically. The average germination rate of barley seeds using the presented electrostatic field in this paper was 57%, while it was 65% when using a microwave of 2.45GHz, compared to a control group with a result of 31%. An important difference between using the electrostatic field and the 2.45GHz microwave is the dry method and wet method. When applying these two methods to practical seed germination, it is necessary to consider the advantages and disadvantages of each experimental approach and choose the appropriate method accordingly.

Effects of Wild Plant's Fermented Extracts and Herbal Essential Oil on Barley Seed Germination (산야초 발효추출물과 허브 정유가 보리 종자 발아에 미치는 영향)

  • 본 연구는 4종의 산야초 추출물(개느삼, 엄나무, 참취, 60가지 산야초혼합 - 모두 원 식물재료를 설탕과 혼합하여 추출한 발효추출물을 15년 숙성시킨 원액의 3% 수용액으로 처리)과 8종의 허브 정유(베르가모트, 클라리세이지, 휀넬, 제라니움, 마조람, 타임, 로즈우드, 스페어민트 원액의 0.3% 농도로 처리) 및 2종의 호르몬(지베렐린, 카이네틴, 0.3%)을 2종의 보리종자(1998년산 새올보리, 2016년산 찰쌀보리) 발아율 향상에 영향을 미치는지를 알아보고자 수행하였다. 특히 새올보리 묵은 종자의 발아력 증진에 효과가 있는지 알아볼 목적으로 수행하였다. 묵은 종자 새올보리의 무처리구에서 8%와 43%의 낮은 발아세와 발아율을 보인 반면, 산야초 추출물 처리구에서 84~93%와 86~100%의 높은 발아세와 발아율을 보였다. 호르몬처리구는 새올보리에서 발아세와 발아율은 각각 43~87%와 55~95%였다. 또한, 찰쌀보리의 경우 무처리의 발아세와 발아율은 각각 54%와 91%였다. 반면, 산야초 추출물 처리구에서 92~100%의 발아세와 95~100%의 높은 발아율을 보였고, 호르몬 처리구에서는 각각 77~85%와 87%의 발아세와 발아율을 보였다. 무처리(증류수)와 2종의 호르몬 및 8종의 허브 정유에 비해 산야초 추출물의 발아 증진 효과가 현저하게 높았다. 특히 냉장고($4^{\circ}C$)에서 20년 보관한 묵은 보리종자에 대해서도 유의한 발아율 향상을 보였다. 정유는 대체로 발아를 억제하는 것으로 나타나 잡초 종자에 대한 발아억제(제초제 대용) 가능성을 시사한다.

Serological detection of barley stripe mosaic virus infection in the seeds of barley and wheat cultivars grown in Korea (혈청학적방법에 의한 보리와 밀종자의 보리${\cdot}$줄무늬 모자이크 바이러스 감염상조사)

  • An attempt was made to find out whether barley stripe mosaic virus (BSMV) occurs in barley and wheat cultivars in Korea. Modified Hamilton's technique for serological detection of barley stripe mosaic virus was used to assay for BSMV in mature seed embryos. Of the 51 barley and wheat seed lots assayed, BSMV was detected in 34 of them. BSMV was detected in covered, naked, and matting barley as well as in wheat. The level of BSMV infection in the infected cultivars varied from $2\~35\%$. This is the first reported occurrence of barley stripe mosaic virus in Korea.

Genetic Diversity of Korean Barley (Hordeum vulgare L.) Varieties Using Microsatellite Markers (Microsatellite 마커를 이용한 한국 보리 품종의 유전적 다양성)

  • Microsatellite markers were utilized to investigate genetic diversity among 70 Korean barley varieties (Hordeum vulgare). Ninety nine microsatellite primer pairs were screened for 9 varieties. Twenty primer pairs showed highly polymorphic. The relationship between markers genotypes and 70 varieties was analyzed. A total of 124 polymorphic amplified fragments were obtained by using 20 microsatellite markers. Two to nine SSR alleles were detected for each locus with an average of 6.2 alleles per locus. Average polymorphism information content (PIC) was 0.734, ranging from 0.498 to 0.882. A total of 124 marker loci were used to calculate Jaccard's distance coefficients for cluster analysis using UPGMA. Clustering group was divided 2 groups corresponding to 2-rowed and 6-rowed barley varieties. The phenogram was discriminated all varieties by markers genotypes. These markers may be used wide range of practical application in variety identification and genetic purity assessment of barley.

Effect of Nanobubbles on the Seed Germination (초미세버블이 종자발아에 미치는 영향)

  • For the last few decades, much research has been tried to improve crop’s characteristics such as crop yield, quality, seeding period and environmental adaptability. In this paper, the effect of nanobubbles on the germination rate of barley seed is experimentally investigated. The air nanobubble was generated in water by gas-liquid mixing method. The results were shown that the mean diameter and concentration of nanobubble fabricated in DI water are 133 ㎚ and 8.59✕108 particles/㎖, respectively. Also, the seed germination rate for the nanobubble water was approximately 40 % higher than that of pure water.

Interactions among Carbon Isotope Discrimination, Water Use Efficiency and Nitrogen Nutrition in Wheat and Barley (밀과 보리에 있어서 탄소동위원소차별, 수분이용효율, 질소영양간의 상호작용)

  • Large and small seeds (44 and 22 mg per caryopsis) of a spring wheat (cv. Kulin) and a spring barley (cv. Skiff) were sown at two nitrogen rates (equivalent to 10 and 32 g m$^{-2}$ ) in well-watered pots under outdoor conditions to determine the effects of seed size and nitrogen (N) nutrition on water use efficiency (WUE) and carbon isotope discrimination ($\Delta$) and to evaluate interaction among $\Delta$, WUE and N nutrition in wheat and barley. Barley produced, on average, 105% more biomass (root+shoot dry weight) than wheat at stem elongation because of early vigor. By anthesis this difference had disappeared as wheat had 16% more biomass than barley which headed 3 days earlier. Compared to plants grown from small seed, plants grown from large seed had much greater biomass in wheat than in barley at stem elongation and anthesis. Higher N nutrition increased average biomass of wheat and barley by 40 and 31%, respectively, at anthesis. Barley had 35 and 20% greater WUE (biomass gained/transpiration) than wheat at stem elongation and anthesis, respectively, and 2.0 to 3.6% lower $\Delta$ in aboveground shoots depending on growth stages and plant parts than wheat which had a greater stomatal conductance than barley. Seed size had a variable effect on WUE and did not affected $\Delta$ values. Water use efficiency was not affected by N rate at stem elongation in wheat and barley whereas WUE was increased 2 and 7%, respectively, in wheat and barley at anthesis with increasing N from 10 to 32 g m$^{-2}$ . High N plants had about 2.5% lower $\Delta$ values regardless of growth stages than low N plants across species and seed sizes. Carbon isotope discrimination was negatively correlated with WUE at anthesis but not at stem elongation.

Evaluation of Cultivation Limit Area for Different Types of Barley owing to Climate Change based on Cultivation Status and Area of Certified Seed Request (기후변화에 따른 맥종별 재배실태와 보급종 보급지역에 의한 재배한계지 평가)

  • This study was conducted to determine the extent to which climate change is expanding areas in which barley can be successfully cultivated. In 2019 and 2020, we collected data on areas that had requested certified seeds from the Korea Seed and Variety Service to determine potential cultivation areas. In addition, we surveyed the growth and yield of different types of barley in fields. Certified seeds of hulled and dehulled barley were requested by farmers across Korea from the Korea Seed and Variety Service in both years. Areas that were provided with certified seeds were considered potential barley cultivation areas. The varieties and use rates of certified seeds varied based on the barley type and region. For example, certified seeds of dehulled barley in 2019 and 2020 were not used in some areas, whereas in others, these seeds constituted 100% of the seeds sown for barley crops. In 2019 and 2020, the average sowing days in Korea were from October 17 to November 9 for dehulled barley, October 26 to November 13 for hulled barley, October 19 to November 5 for malting barley, and October 3 to November 1 for naked oats. Thus, the sowing days of the barley types varied depending on the area and year they were used. For example, in the case of hulled barley in Jeonnam, some farmers sowed until December 12. The yield per 10 a of barley cultivation was typically higher in the main production areas than in the cultivation limit areas. In extreme cases, harvest was impossible in some cultivation limited areas, such as Gangwon-do. Based on the current 20-year January minimum average temperature (JMAT) in Korea (2002-2021), climate change scenarios suggest that barley cultivation is feasible, provided that the minimum temperature in January is no lower than -10℃, -8℃, and -4℃ for hulled barley, dehulled barley, and for malting barley and naked oats, respectively. Additionally, cultivation of barley across South Korea seems feasible based on data on certified barley seeds by area. Although both JMAT and certified seed data suggest that barley cultivation across Korea is feasible, our survey results of barley growth and yield showed that harvest was impossible in certain cultivation areas, such as Gangwon-do. Therefore, climate change scenarios related to the cultivation limits of different barley types need to be re-estimated by factoring in survey data on the growth and yield of crops within those cultivation areas.
