Draft genome sequence of Streptomyces sp. P3 isolated from potato scab diseased tubers (감자 더뎅이병 이병괴경으로부터 분리한 Streptomyces sp. P3 균주의 유전체 해독)

  • Streptomyces sp. P3 was isolated from potato scab diseased tubers in Pyeongchang, Gangwon-do, Republic of Korea in 2017. Here, we report the draft genome sequences of P3 with 9,851,971 bp size (71.2% GC content) of the chromosome. The genome comprises 8,548 CDS, 18 rRNA and 66 tRNA genes. Although strain P3 did not show pathogenicity both potato tuber assay and radish seedling assay, it possesses tomatinase (tomA) gene among conserved pathogenicity-related genes in well characterized pathogenic Streptomyces. Thus, the genome sequences determined in this study will be useful to understand for pathogenic evolution in Streptomyces species, which already adapted to potato scab pathogens.

Evaluation of Resistance of Phytopathogenic Bacteria to Agricultural Antibiotics (채소 재배에서 사용하는 농용 항생제에 대한 주요 식물병원세균의 저항성 평가)

  • Agricultural antibiotics are widely used to inhibit the growth of phytopathogenic bacteria involved in plant diseases. However, continuous antibiotic overuse in crop production may lead to the development of antibiotic resistance in phytopathogenic bacteria. This study was conducted to evaluate the resistance to three different agricultural antibiotics (oxytetracycline+streptomycin, streptomycin, and validamycin A) in 91 strains of phytopathogenic bacteria including Pectobacterium carotovorum, Pseudomonas syringae pv. actinidiae, Clavibacter michiganensis subsp. michiganensis, C. michiganensis subsp. capsici, and Xanthomonas arboricola pv. pruni. Bacterial growth in the presence of various concentrations of validamycin A was also assessed spectrophotometrically by analyzing the optical density. All strains did not grow when the cells were exposed to oxytetracycline+streptomycin or 100× of streptomycin. However, among the 91 strains, 4% and 2% strains showed bacterial growth at the concentrations of 1× and 10× of streptomycin, respectively. Furthermore, 97%, 93%, and 73% strains were resistant to the 1×, 10×, and 100× of validamycin A, respectively, and especially, P. carotovorum contained the highest resistance to the validamycin A. Minimum bactericidal concentration values of validamycin A did not correlate with the patterns of agricultural antibiotic resistance. Further studies are needed to understand the incidence and development of antibiotic resistance in phytopathogenic bacteria.

Monitoring of Pathogenic Bacteria in Organic Vegetables from Korean Market (국내 유통중인 유기농 채소류의 미생물 분포도 분석)

  • This study was undertaken to assess the microbiological quality and prevalence of pathogens in organic vegetables produced in Korea. A total of 189 organically grown vegetable samples (perilla leaf 50, lettuce 50, tomato 39, cucumber 50) were analyzed for the presence of aerobic plate count, Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, and Yersinia enterocolitica. The total aerobic plate counts were in the range of 4.2 to $7.7log\;CFU\;g^{-1}$ for perilla leaf, 5.0 to $8.0log\;CFU\;g^{-1}$ for lettuce, 4.0 to $7.5log\;CFU\;g^{-1}$ for tomato, and 6.6 to $8.6log\;CFU\;g^{-1}$ for cucumber. The highest counts were found in cucumber. E. coli O157:H7, Salmonella spp., S. aureus, L. monocytogenes, and Y. enterocolitica were not detected from any organically grown vegetable samples. This research suggests that continuous monitoring in organic vegetables is required to improve fresh produce safety.

A Study on the Investigation and Application of Microbial Pathogens of Major Insect Pests of Forest in Korea (중요산림해충의 병원미생물 개발에 관한 연구)

  • The study has been carried to investigate a possibility to control several major insect pest of forest by microbial pathogens existing in nature as one of the biological control measure. Microorganisms including polyhedral virus isolated from diseased fall webworm were total of 4 kinds pathogenic microbes among these 4 kinds were polyhedral virus and Bacillus .species. Control effect of these two pathogens appeared to be $70.6\%$ and $49.5\%$, respectively, when they were compared with those of control plot that was $27.8\%$. Each one of bacterium species and fungus species were isolated from diseased Japanese alder leaf beetle. Pathogenisity to the healthy beetle was recognized by the fungus species, while the bacterium showed none of pathogenisity. The fungus was identified as Beauveria sp. and its effect on the beetle control was $96.2\%$ while untreated plot showed $49.2\%$ of dead beetles in the same period. Fifteen species of microbes were isolated from diseased larvae of pine gall midge. Six species out of 15 showed certain level of insecticidal effect to the larvae of the insects. The highest efficiency was showed by a fungus species, Fusarium sp. and was followed by Bacillus SP. I, Spicaria sp. pathogens isolated from larvae of pine gall midge did not affected to both of Japanese alder leaf beetles and fall webworms in any means.

Detection of Pseudomonas tolaasii causing brown blotch disease in water from oyster mushroom cultivation farms by PCR (PCR을 이용한 느타리버섯 재배사 물로부터 세균성갈색무늬병 병원균 Pseudomonas tolaasii 검출)

  • Pseudomonas tolaasii causing brown blotch disease was detected by PCR from water samples collected from the oyster mushroom cultivation farms to find the contamination level of the pathogen in water. Sixteen water samples (28.1%) contain less than 1,000 cfu, 31 samples (54.4%) contain 1,001-10,000 cfu, 6 samples (10.5%) contain 10,001-100,000 cfu, and 4 samples (7%) contain of bacteria per milliliter. P. tolaasii-specific DNA band was amplified in 3 samples (5.3%) by nested-PCR and in 20 samples (35.1%) by immunocapture (IC)-nested PCR respectively. These results suggest that IC-nested-PCR was much more sensitive than nested-PCR in detection of P. tolaasii and a quite few waters using for oyster mushroom cultivation were contaminated with P. tolaasii.

Bacteriological Monitoring of Radiology Room Apparatus in the Department of Radiological Technology and Contamination on Hands of Radiological Technologists (영상의학과 촬영실 장비와 방사선사의 손 오염의 세균학적 모니터링)

  • Distribution of microorganisms were examined for the bucky tables in the radiology rooms of the department of radiological technology, the aprons, handles of various apparatus, handles of mobile radiological apparatus, and hands of the radiological technologists. As a result, relatively larger amounts of bacteria were found on the handles of the mobile radiological apparatus and the aprons. Among the isolated bacteria, Acinetobacter baumanni (7.3%), Klebsiella pneumoniae (6.7%), Staphylococcus aureus (3.9%), Serratia liquefaciens (1.7%), Enterobacter cloaceae (0.6%), Providenica rettgeri (0.6%) are known as the cause of nosocomial infection (hospital acquired infection). In addition, similar colonies were also found on the hands of the radiological technologists such as microorganisms of Klebsiella pneumoniae (8.4%), Staphylococcus aureus (6.6%), Yersinia enterocolotica (5.4%), Acinetobacter baumanni (4.2%), Enterobacter cloaceae (2.4%), Serratia liquefaciens (1.8%), Yersinia pseuotuberculosis (18%), Enterobacter sakazakii (1.2%), and Escherichia coli (0.6%). In particular, this result indicates clinical significance since Staphylococcus aureus and Escherichia coli show strong pathogenicity. Therefore, a continuous education is essential for the radiological technologists to prevent the nosocomial infection.

Studies on the Pathogenic Pseudomonas Causing Bacterial Diseases of Cultivated Mushrooms in Korea 2.   • Tests were performed on 232 bacterial strains (71 strains of Pseudomonas tolaasii and 161 white line reacting organisms, WLRO) isolated from cultivated mushrooms. As results, P. tolaasii was divided into 5 groups on the basis of the phenotypical characteristics for the strains, and group 3 was the major one including 48 (62%) out of the total 71 strains. WLRO were classified into 23 groups, and group 10 was the major group (65 strains, 30% of the total WLRO tested). A white line was well formed at 22$^{\circ}C$ and at 4 mm distance between P. tolaasii and WLRO colonies in their dual culture on Pseudomonas agar F medium within 36-hr incubation, but not formed at $25^{\circ}C$ even for 72-hr incubation. The morphological, cultural and biological properties of P. tolaasii group 3, and the main group of WLRO, group 10, were different only in the light of pathogenicity. Also group 2 of P. tolaasii had the characteristics similar to group 24 of WLRO which was pathogenic to cultivated mushrooms, suggesting the P. tollaasiii and WLRO strains may be the same species although their white line forming ability and pathogenicity were more or less different from one another. Subculture of the strains in P. tolaasii group 1 induced the variation of their pathogenicity, white line forming ability and utilization of sodium benzoate and sodium tartrate, and thus it can be inferred that they were converted to strains having the characteristics of group 3 or WLRO groups.

Microbial Hazard Analysis of Manufacturing Processes for Starch Noodle (당면의 제조공정별 미생물학적 위해요소 분석)

  • The purpose of this study was to identify control points through microbiological hazard analysis in the manufacturing processes of starch noodles. Samples were collected from the ingredients, manufacturing processes, equipment and environment. Microbiological hazard assessments were performed using aerobic plate counts (APC), Enterobacteriaceae (EB), E. coli and five pathogens including B. cereus, E. coli O157:H7, L. monocytogenes, Salmonella spp., and S. aureus. The APC levels in raw materials were from 2.12 to 3.83 log CFU/g. The contamination levels after kneading were 4.31 log CFU/g for APCs and 2.88 log CFU/g for EB counts. APCs decreased to 1.63 log CFU/g and EB were not detected after gelatinization, but their levels slightly increased upon cooling, cutting, ripening, freezing, thawing, and separating. The reuse of cooling and coating water would be a critical source of microbial increase after cooling. After drying, APCs and EB counts decreased to 5.05 log CFU/g and 2.74 log CFU/g, respectively, and the levels were maintained to final products. These results suggest that the cooling process is a critical control point for microbiological safety, and the cooling water should be treated and controlled to prevent cross contamination by pre-requisite program.

  • The bacterial soft-rot disease is one of the most critical diseases in vegetables such as Chinese cabbage. The researchers isolated two bacteria (Pseudomonas kribbensis and Pantoea vagans) from diseased tissue samples of Chinese cabbages and confirmed them as being the strains that cause soft-rot disease. Lactic-acid bacteria (LAB), were screened and used to control soft-rot disease bacteria. The researchers tested the treatments with hypochlorous acid water (HAW) and LAB supernatant to control soft-rot disease bacteria. The tests confirmed that treatments with the HAW (over 120 ppm) or LAB (Lactobacillus plantarum PL203) culture supernatants (0.5 mL) completely controlled both P. kribbensis and P. vagans.

Effects of Bisroot in the Diet on Growth, Body Composition, Immume responses of Nile Tilapia, Oreochromis niloticus (사료 중 비스루트가 나일틸라피아, Oreochromis niloticus의 성장, 체조성 및 면역 방응에 미치는 영향)

  • The present study investigated the effects of Bisroot, that contains live bacteria (Bacillus polyfermenticus, Bacillus mesentericus, Streptococcus faecalis, & Bifidobacterium breve) and digestive enzymes (protease, lipase), on the growth, body composition and immune response of Nile tilapia fingerlings. One percent of the Bisroot was added to the experimenta feed. All exprimental fish were fed for 60 days. The weigh gains among the experimental fish were not significntly different (P>0.05). Hematocrit value, hemoglobin, total protein, glucose, GOT, and GPT were unaffected by Bisroot treatment. However, it was observed that glucose, GOT, and GPT value in the fish that were fed Bisroot, were lower than the control. The complement activity ($CH_50$) tended to be significantly increased by Bisroot treatment, but not lysozyme activity. Phagocytosis and respiratory burst activities of macrophages in the head kidney were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities of non-specific immune responses. When fish were challenged with a virulent strain of Edwardsiella tarda, the Bisroot treated fish were more resistant than the control. The present results suggest that the introduction of Bisroot into the diet of Nile tilapia could increase their resistance against bacterial infection, reduce fish mortality, and offers economic benefits.
