A Study on the Pollution of Polycyclic Aromatic Hydrocarbons(PAHs) in the Surface Sediments Around Gwangyang Bay (광양만 주변해역 표층퇴적물에서의 다환방향족탄화수소류(PAHs)의 오염에 관한 연구)

  • PAHs(Polycyclic Aromatic Hydrocarbons) are widespread contaminants in the marine environment. They are of mainly anthropogenic origin from urban runoff, oil spill and combustion of fossil fuels. Some PAHs are potentially carcinogenic and mutagenic to aquatic organism The contamination of PAHs in the coastal environments has not been well known yet in Korea. This study was carried out to survey the contamination of PAHs in sediments around Gwangyang bay. The Yeosu petrochemical industrial complex, POSCO(Pohang steel company) and Gwangyang container harbor are located around the bay. PAHs in sediment samples were extracted in soxhlet extractor and were identified and quantified by GC-MS(Gas Chromatography-Mass Spectrometry) TOC(Total Organic carbon) and textural parameters in sediment samples were also analyzed 13 species of PAHs were detected at all of the surface sediments. Total PAHs concentrations in the surface sediments ranged from 171.40 to $1013.54{\mu}g/kg$ dry wt.. In most of the surface sediments, Naphthalene was the highest in the range of 14.08 to $691.39{\mu}g/kg$ dry wt. and Anthracene was the lowest in the range of 0.49 to $22.66{\mu}g/kg$ dry wt.. The correlation coefficients between individual PAHs and Total PAHs in the surface sediments were relatively higher in the low molecular compounds such as Naphthalene and Phenanthrene. In the relationship of the P/A(Phenanthrene/Anthracene) ratio and F/P(Fluoranthene/Pyrene) ratio, P/A ratio was generally above 10 and F/P ratio was shown to be above 1 in all sediment samples. These data indicate that PAHs in sediments around Gwangyang bay seem to be of both pyrolytic and petrogenic origin. Total PAHs in the surface sediments were correlated with TOC and textural parameters. The values of PAHs in the surface and core sediments were lower than the biological effect guidelines.

Analysis of Genetic Relationship of Cordyceps militaris in Korea by Random Amplified Polymorphic DNA (한국산 번데기동충하초의 RAPD 분석에 의한 종내 그룹의 유전적 유연관계 분석)

  • Seventy two isolates of Cordyceps militaris collected from 11 sites in Korea, including two isolates from ATCC, were used to assess genetic variation within Cordyceps militaris. The anamorph stage and cultural characteristics of C. militaris were observed through microscope and investigated on PDA respectively. The anamorphs of C. militaris were identified to be Verticillium. Isolates of C. militaris showed different growth rates, morphology and color. Fifty six isolates of single ascospore and seventy two isolates of mass ascospore from C. militaris were analysed using by Random Amplified Polymorphic DNA (RAPD) for genetic relationship analysis. Fifty six single ascospore isolates fell into two groups by phenogram constructed from distance values using the UPGMA method in NTSYS-pc software: group A from artificial fruit body of C18 except for isolate 51; group B from artificial fruit body of C738. The average genetic distance value within group A is 0.150 and group B is 0.163. The average genetic distance value between the two groups is 0.221. The average genetic distance value within 56 single ascospores is 0.207 and 72 mass ascospores is 0.330. Genetic relationships were not found among 72 mass ascospore isolates obtained from eleven geographically distant populations.

Current Status of Cattle Genome Sequencing and Analysis using Next Generation Sequencing (차세대유전체해독 기법을 이용한 소 유전체 해독 연구현황)

  • Thanks to recent advances in next-generation sequencing (NGS) technology, diverse livestock species have been dissected at the genome-wide sequence level. As for cattle, there are currently four Korean indigenous breeds registered with the Domestic Animal Diversity Information System of the Food and Agricultural Organization of the United Nations: Hanwoo, Chikso, Heugu, and Jeju Heugu. These native genetic resources were recently whole-genome resequenced using various NGS technologies, providing enormous single nucleotide polymorphism information across the genomes. The NGS application further provided biological such that Korean native cattle are genetically distant from some cattle breeds of European origins. In addition, the NGS technology was successfully applied to detect structural variations, particularly copy number variations that were usually difficult to identify at the genome-wide level with reasonable accuracy. Despite the success, those recent studies also showed an inherent limitation in sequencing only a representative individual of each breed. To elucidate the biological implications of the sequenced data, further confirmatory studies should be followed by sequencing or validating the population of each breed. Because NGS sequencing prices have consistently dropped, various population genomic theories can now be applied to the sequencing data obtained from the population of each breed of interest. There are still few such population studies available for the Korean native cattle breeds, but this situation will soon be improved with the recent initiative for NGS sequencing of diverse native livestock resources, including the Korean native cattle breeds.

Changes in Physicochemical Properties and Antioxidant Activities according to Different Harvest Times in Black Rice (Oryza sativa L.) (흑미 수확시기에 따른 이화학적 특성 및 항산화 활성 변화)

  • This study compared physicochemical properties and antioxidant activities according to different harvest times in five varieties of black rice (BR). Five cultivars of BR, Nunkeunheukchal (NKHC), Sinnongheugchal (SNHC), Sintoheugmi (STHM), Josaengheugchal (JSHC), and Heugjinju (HJJ), were selected for the study. Cultivars were transplanted on June 20th and cultivated by the Department of Southern Area Crop Science, NICS in Miryang, Korea. Heading dates of NKHC, SNHC, STHM, JSHC, and HJJ were Aug. 17th, Aug. 30th, Sep. 1st, Aug. 20th, and Aug. 12th, respectively. The five different harvest times were as follows; 35, 40, 45, 50, and 55 days after heading date. The highest level of anthocyanin was 35 days after heading date in all varieties. Levels of anthocyanin in NKHC, SNHC, STHM, JSHC, and HJJ were 164, 203, 251, 67, and 210 mg/100 g, respectively. Content of anthocyanin significantly decreased 35 days after heading in four varieties, excluding JSHC. Highest DPPH $IC_{50}$ values were 57, 29, 10, and $27{\mu}g/mL$ in NKHC, SN HC, STHM, and JSHC extracts, respectively. Highest ABTS $IC_{50}$ values were 24, 13, 9, and $11{\mu}g/mL$ in NKHC, SNHC, STHM, and JSHC extracts, respectively. The antioxidant activities were highest 35 days after the heading date and significantly decreased in four varieties, excluding HJJ. The antioxidant activity of HJJ showed no significant difference, according to harvest time. This study suggests that the content of anthocyanin and antioxidant activity could be considered as factors affecting optimal harvest period to produce highly qualified black rice.

Development of Near-Infrared Reflectance Spectroscopy (NIRS) Model for Amylose and Crude Protein Contents Analysis in Rice Germplasm (근적외선 분광광도계를 이용한 벼 유전자원 아밀로스 및 단백질 함량분석을 위한 모델개발)

  • The objective of this research was to develop Near-Infrared Reflectance Spectroscopy (NIRS) model for amylose and protein contents analysis of large accessions of rice germplasm. A total of 511 accessions of rice germplasm were obtained from National Agrobiodiversity Center to make calibration equation. The accessions were measured by NIRS for both brown and milled brown rice which was additionally assayed by iodine and Kjeldahl method for amylose and crude protein contents. The range of amylose and protein content in milled brown rice were 6.15-32.25% and 4.72-14.81%, respectively. The correlation coefficient ($R^2$), standard error of calibration (SEC) and slope of brown rice were 0.906, 1.741, 0.995 in amylose and 0.941, 0.276, 1.011 in protein, respectively, whereas $R^2$, SEC and slope of milled brown rice values were 0.956, 1.159, 1.001 in amylose and 0.982, 0.164, 1.003 in protein, respectively. Validation results of this NIRS equation showed a high coefficient determination in prediction for amylose (0.962) and protein (0.986), and also low standard error in prediction (SEP) for amylose (2.349) and protein (0.415). These results suggest that NIRS equation model should be practically applied for determination of amylose and crude protein contents in large accessions of rice germplasm.

Diagnostic Value of MRI in Schwannoma (신경초종 진단에 있어 자기공명영상촬영(MRI)의 진단적 가치)

  • Purpose: To diagnose soft tissue tumor, such as lipoma and Schwannoma, magnetic resonance imaging (MRI) is sufficient in most cases. However, various characteristics are found in MRI images of Schwannoma, thus other type of tumors are often misdiagnosed as Schwannoma with MRI images. In this study, we evaluate the diagnostic value of specific MRI findings of Schwannoma. Materials and Methods: From January 2002 to May 2013, 104 patients who are suspected as Schwannoma rith MRI images are included in data, and the final diagnosis is confirmed with biopsy. Patients are divided into group 1 and group 2 who are confirmed as Schwannoma and other disease with biopsy, respectively. Results: 92 patients were diagnosed as Schwannoma (group 1) and 12 patients were diagnosed as other disease (group 2). We investigate the diagnostic value of specific MRI findings of Schwannoma. 41 patients of group 1 (45%) and 0 patients of group 2 (0%) showed target sign, 47 patients of group 1 (51%) and 2 patients of group 2 (17%) showed fascicular sign, 44 patients of group 1 (48%) and 5 patients of group 2 (42%) showed fat split sign, 28 patients of group 1 (30%) and 1 patients of group 2(9%) showed nerve entering and exiting sign, and 8 patients of group 1 (9%) and 6 patients of group 2 (50%) showed none of four specific findings on their MRI images. 52 patients of group 1 (57%) and 5 patients of group 2 (42%) have tumors on the pathway of nerve. Target sign could be considered as the best diagnostic value of the sign we investigate (p<0.05). Conclusion: Although specific MRI findings have powerful diagnostic value, patients are often misdiagnosed as Schwannoma with MRI findings. Therefore, if patients who are suspected as Schwannoma based on MRI findings have no target sign on their MRI images, we should consider the possibility of other disease.

The Value of ICAM-1 Expression and the Soluble ICAM-1(sICAM-1) Level as a Marker of Activity in Sarcoidosis: The Relationship Between the ICAM-1 Level and the Clinical Course of the Disease (유육종증의 활동성 지표로서의 ICAM-1)

  • Background: The natural course of sarcoidosis is variable from spontaneous remission to significant morbidity or death. So the assessment of disease activity is important but no single parameter was generally accepted as a good marker. Recently several studies suggested that adhesion molecules, especially ICAM-1 can be a marker, but there are some controversies. And only few data are available about the relationship of ICAM-1 with clinical follow-up course. Methods: We measured the expression of adhesion molecules on BAL cells by flow cytometry and the level of soluble ICAM-1(sICAM-1) in serum and BALF at the time of diagnosis in 12 patients with active disease and 7 inactive sarcoidosis(5 male, 14 female, mean age: $39.4{\pm}10.7$ years, mean follow-up : $20{\pm}15$ months). Follow-up clinical course were compared with the changes in serum sICAMA-1 level and the adhesion molecule on BAL cells. Results: In the patients with active disease, the ICAM-1 on AM(RMFI: $3.68{\pm}1.71$) and sICAM-1 level in serum($582{\pm}193$ng/ml) and BAL fluid($47.8{\pm}16.5$ng/ml) were all higher than those of 7 inactive disease(RMFI: $1.89{\pm}0.75$, p=0.0298, serum: $294{\pm}117$ ng/ml, p=0.0049, BALF: $20.9{\pm}8.3$ ng/ml). In the active sarcoidosis, ICAM-1 on AM(RMFI : $1.51{\pm}0.84$) and serum sICAM-1 were decreased after the therapy($250{\pm}147$ ng/ml) but no significant change was noted in inactive disease. Also we found the initial ICAM-1 on AM and serum sICAM-1 had a significant correlation with the degree of improvement in PFT after the therapy. During the follow-up, the disease relapsed in 4 patients after the discontinuation of steroid and the serum sICAM-1 level went-up again at the time of relapse. Conclusion: Our data suggest that the serum sICAM-1 level and the ICAM-1 expression on AM can be a good marker of disease activity and also a predictor of outcome in sarcoidosis.

Genetic and Environmental Effects on the Lactation Yield and Milk Compositions in Holstein Cow (Holstein종 유우(乳牛)의 비유량(泌乳量) 및 유조성분(乳組成分)에 미치는 유전(遺傳) 및 환경(環境)의 효과(效果))

  • This study was conducted to estimate the genetic and environmental effects on lactation yield and milk compositions in Holstein cows. The data analysis were the records of 159 cows rearing at Nation Animal Breeding Institute from 1990 to 1991. The least square means were estimated on milk and fat yield in lactation yield, and the percent of fat, protein, solids-not-fat and total solid in milk composition. The results obtained are summarized as follows: 1. The average yield of milk and fat in 305 days were $7,26.56{\pm}3,57.24$ and $254.65{\pm}44.94Kg$ and the percent of fat, protein, solids-not-fat and total solid were $3.69{\pm}0.43$, $3.32{\pm}0.41$, $9.15{\pm}0.49$ and $12.75{\pm}0.96$ and the coefficients of variation were 18.68, 17.64, 11.88 and 12.34% for milk yield, fat yield, fat percent and protein percent, respectively. 2. The effect of sires was highly significant at 1% level in milk and fat yield and fat percent, and significant at 5% level in protein and total solid percent. Among the sires, B, L and O sire were superior in milk yield with 7,571.22, 7,499.11 and 7,420.58 Kg, and A, F and K sire were superior in protein percent with 3.75, 3.64 and 3.65, respectively. 3. The effect of parity was highly significant at 1% level in milk yield, and significant at 5% level in fat yield. Among the parities, the 3rd parity was superior in milk and fat yield with 7,634.54 and 274.98 Kg, and the 4th and over was superior in fat and protein percent with 3.90 and 3.50, respectively. 4. The effect of calving seasons was highly significant at 1% level in milk yield, and significant at 5% level in fat yield, and the percent of fat, protein and total solid. Among the calving seasons, spring and winter were superior in milk yield with 7,310.31 and 7,364.57 Kg, also spring and winter were superior in protein percent with 3.68 and 3.52, respectively.

Diffusion-Weighted MR Imaging of Various Intracranial Diseases : Clinical Utility (다양한 두개강내 질환의 확산강조 자기공명영상 : 임상적 유용성)

  • Purpose : To evaluate the clinical utility of diffusion-weighted imaging by analyzing the signal intersity of lesions in patients with various intracranial diseases. Materials and Methods : difusion-weighted MR imaging was prospectively perormed in randomly selected 70 patients with various intracranial idseases. They consisted of 20 patients with acute infarct, 21 patients with chronic infarct of small vessel disease, 14 patients with primary intracranial tumor, three patients with brain metastasis, five patient with brain abscess, five patients with brain abscess, five patients with cerebral hemorrhage, one patient with neurocysticercosis, and one patient with epidermoid cyst. the diffusion-weighted images were obtained immediately after routine T2-weighted imaging on a 1.5T MR unit using single shot spin echo EPI technique with 6500 ms TR, 107ms TE, $128{\times}128$ matrix, 1 number of excitation, $24{\times}24$ field of view, 5-7 mm slice thickness, 2-3 mm inter-slice gap. The diffusion-gradients (b value of ($1000s{\;}/{\;}textrm{mm}^2$)) were applied along three directions(x, y, z). On visual inspection of diffusion-weighted images, the signal intersity of lesions was arbitrarily graded as one of 5 grades. In quantitative assessment, we measured the signal intensity of all the lesions and the contralateral corresponding normal area using round region of interest(ROI), and then calculated the signal intensity ratio of the lesion to the normal brain parenchyma. Results : On visual inspection, markedly hyperintense signals were seen in all cases of acute infarct, brain abscess, epidermoid cyst, and neurocysticercosis in degenerating stage. In all cases of cerebral hematoma, the very high signal internsity was intermingled with low signal intensity. focal very high signal intersity was also seen in a solid portion of the tumor in a patient. the mean signal intensity ratios of all those lesions to the normal brain parenchyma were above 2.5. Gliosis, solid component of brain tumor, brain metastasis, and vasogenic dedma appeared isointense to the normal brain parenchyma in 71%, 64%, 100%, and 67%, respectively ; the mean signal intensity ratios of those lesions to the normal brain parenchyma ranged 1.15 to 1.28 and there was no significant difference among these(p>0.1). Cystic cerebromalacia and necrotic or cystic portions in tumor were markedly or slightly hypointense, and the mean signal intensity ratios were 0.45 and 0.42, respectively. Conclusion : Very high signal intensity of acute infarct, brain abscess, epidermoid cyst, and cystic neurocysticercosis in degenerating stage on diffusion-weighted images may be helpful in differentiating from other diseases that are hypointense or isointense to the normal brain parenchyma. It may be especially useful differentiation of brain abscess from brain tumor with necrotic or cystic portion.

The Analysis of Productivity of Oak Stand following to Site Quality and Crown Class (I) (입지(立地)와 수형급(樹型級)에 따른 참나무임분(林分)의 생산력(生産力) 분석(分析) (I))

  • The structures of oak stands were studied in relation to their productivity. Three plots classified by the site quality following to altitude, were selected on the oak stand located at Naryongri Eunsanmyun Buyeogun. The site qualities of the stand studied were generally estimated to be moderate in respect to some soil physical and chemical properties. Quercus variabilis was dominant species in the vegetation composition of the stand studied. The crown densities of the stand were varied from 65.4% in plot I to 78.2% in plot III and the parts, occupied with oak trees, ranged from 44.4% in plot I to 65.9% in plot III in the density. In the contrast to crown density, the growing stocks of oak trees ranged from $3.937m^3$ in plot I (73.4% of plot total) to $2.075m^3$ in plot III (84.3% of plot total). The occupied ratios, measured by crown class, exhibited dissimilarity between crown projection area and volume, and also the ratios brought into different patterns by plot. Highly significant correlations were proved between crown projection areas and volumes of individual oak trees by plot, but not proved in the relations by crown class. The cumulative growth, current annual increment and mean annual increment displayed various patterns by plot and by crown class in the heights, D. B. H. s and volumes of oak trees. The maximum values of current annual increment of height were generally recorded in 10 to 20 years, earlier than those of D. B. H. The mean annual increment of height, D. B. H. and volume maintained similar levels after about 10 years and fluctuated irregularly. The crown projection area and volume following to thinning decreased in the order of the thinning methods of grade A < grade B < crown thinning$90m^3/ha$ in about 40 years in the productivity of volume of the oak stand studied.
