Effect of Nutrients and pH on the Growth and Sporulation of Four Entomogenous Hypomycetes Fungi (Deuteromycotina) (배지의 영양원 및 pH가 수종 곤충기생균의 균사생장 및 포자생산에 미치는 영향)

  • Growth of Metarrhizium flavoviride var. minus and Hirsut lle strigosa showed good yield in the carbon source media adding dextrose, starch and saccharose, but Hirsutella sp. from korea greq well in the other media except in the dextrose media. Yeast extract was necessary for the mecelial growth of the fungi, but the fungi tested in this experiment showed a difference in the amount of required yeast extract. Growth of Nomurea rileyi was fastidious in the carbon and nitrorgen sourced media and the optimum pH of the media for growth was at 6.7. Sporulation of M. Flavoviride var. minus was high on media, containing 1%~2% of yeast extract as nitrogen and carbon source media, but N. rileyi sporulated abundantly on the media with nitrogen and dextrose.

  • A photoautotrophic micropropagation methodology in liquid culture medium has a number of advantages for large-scale propagation of plants. This paper describes an improved system for the mass propagation via somatic embryogenesis of the medicinal plant Kalopanax septemlobus Nakai. Somatic embryo-derived young plantlets of K. septemlobus were cultured either under heterotrophic conditions with sucrose on half-strength MS medium with $30gL^{-1}$ sucrose, under heterotrophic conditions without sucrose, or under photoautotrophic conditions (MS liquid medium without sucrose under forced aeration) for four weeks before transferring the plantlets for acclimatization. Plantlets grown under photoautotrophic conditions had more leaves, higher chlorophyll content, a higher net photosynthetic rate (NPR), and a higher survival rate. The results indicate that the photoautotrophic conditions with a forced ventilation system are effective in enhancing the growth of plantlets and the rate of net photosynthesis. The plantlets grown under photoautotrophic conditions had a high survival rate (92%) upon ex vitro transplantation. Our study shows that autotrophically produced plantlets acclimatize better and sooner upon ex vitro transplantation than conventionally cultured plants.

Screening of optimum nutrient supplement of corncob as a main substrate for bottle culture of Oyster mushrooms (느타리버섯 병재배시 주재료 콘코브배지에 적합한 영양원 탐색)

  • In this study we carried out to find suitable nutrient supplement of corncob as a main carbon source for bottle culture of oyster mushroom. Data from chemical analysis of Kapok seed cake(KP) treatment showed 20.2 of C/N ratio, $0.28g/cm^3$ of bulk density and 74.2% of porosity. Mushroom yield of KP treatment is 158g/bottle that is similar to control(150g/bottle). However, the price of KP is 400 won/kg that is cheaper than 550 won/kg of cotton seed cake(CS). The highest REV(relative feed value) also was observed KP treatment. A further study is required determine practical animal performance by feeding the corncob-based KP.

Phylogenetic Analysis of Oligotrophic Bacteria Found in Potable Groundwater (음용 지하수중에 분포하는 저영양세균의 계통학적 해석)

  • In order to investigate the ecological aspect of bacteria on groundwater, water samples were collected from various regions. Total of 318 strains were isolated from diluted nutrient broth (DNB) agar medium, and investigated their growth pattern on nutrient broth (NB) medium. As a result, all the isolated strains were divided into two groups, NB and DNB organisms. Growth of DNB organisms were suppressed in full strength NB medium but not in DNB medium, which were called oligotrophic bacteria in this study. Proportion of DNB organisms occurred in the frequency of 50-98% in potable groundwaters (CW, CJ, DPG, CJG1), however, it was 23,46% in polluted site (TJ, NPG1). One hundred and two strains were identified as oligotrophic bacteria and their phylogenetic characteristics were determined by using 16S rDNA sequencing. Based on the phylogenetic analysis, they were found to fall into three major phylogenetic groups: belonging to the Proteobacteria $\alpha$-(49 strains), $\beta$-(50 strains), $\gamma$ -(3 strains) subdivisions. The phylogenetic analysis suggested that microbial diversity of potable groundwater is more complex than that obtained in the past investigation.

Optimization of Media Composition and Cultivation for the Mycelial Growth of Agrocybe Cylindracea (Agrocybe Cylindracea의 영양배지 조성 및 배양조건의 최적화)

  • Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.

Comparison of Four Different Isolation Media for Staphylococcus aureus (황색포도상구균 분리배지 비교)

  • Performance test was carried out between selective media which are generally used in Staphylococcus aureus isolation from food. Sensitivity, determined according to the appearance of characteristic colonies when 30 different S. aureus strains were tested, resulted as Baird-Parker agar (RPF)> $Petrifilm^{TM}$ Staph Express plate> Baird-Parker agar> Mannitol salt agar. Also, the four different media showed the same selectivity because all tested media did not produce the false positive colonies. Recovery efficiency from the artificially inoculated foodstuff was almost the same for the tested media. Presumptive colonies were collected from the dried fishery product using Mannitol salt agar and collected strains were tested on 4 different selective agar. Almost presumptive strains did not show the false positive colonies except for S. carnosus ssp carnosus. This strain was identified as false positive colonies on Mannitol salt agar, Baird-Parker agar and $Petrifilm^{TM}$ Staph Express plate. But Baird-Parker agar (RPF) did not show the false positive colonies with the same strains. So, it was concluded that the Baird-Parker agar (RPF) has more higher selectivity than other tested media in this experiment.

Production of Biosurfactant Using Bacillus spp (Bacillus spp.를 이용한 Biosurfactant 생산공정)

  • Among the bacterial strains isolated from chungkook-jang, Bacillus subtilis CH-1, Bacills circulans K-1 and Bacillus subtitis (natto) N-1, Bacillus subtitis CH-1 showed the highest productivity of biosufactant. A-medium was selected for the basal medium in the large scale production of biosurfactant, and modified to synthetic medium which containing 2% glucose, 0.3% soy peptone, and mineral salts. The surface tension was reduced to maximal value after 96 hr after fermentation, about the 43% of initial tension. Temperature and initial pH of medium was not critical factor for the biosurfactant production. The yield of crude biosurfactant was 6 g/L under the optimum condition.

  • This study aimed to replace the imported Chinese complete medium for Lentinula edodes with a newly-developed complete medium that is suitable for export as well. Three media compositions that supported fast mycelium growth and higher density, compared to that in the control, were chosen. Culture in the T13 medium reduced the incubation period for 'Nongjingo' by 12 days and for 'Cham-aram' by 10 days, compared to that required for the control; in addition, the number of days required for browning was greatly reduced for both varieties. The quantity of each mixed medium was increased according to the composition from the 1st to the 5th cycle by 5.9% for 'Nongjingo' and 12.6% for 'Cham-aram' in T13, compared to that in the control. A mixed medium comprising oak sawdust + Douglas fir sawdust + corn flour (40:40:20, v/v) was selected as the most suitable complete medium.
