Studies on the Germination of Korean Ginseng(Panax ginseng C.A.Meyer) Seed I. Influences of Nonstratified Seed on Embryo Growth and Germination (인삼 종자의 발아에 관한 연구 I. 미개갑 종자 파종이 배생장 및 발아에 미치는 영향)

  • To define natural embryo growth and germination of the Korean ginseng (Panax ginseng C.A.Meyer) seed, freshly harvested and depulped seeds were seeded on nursery bed at Aug. 5 without stratification. At 100 days after seeding, embryo/endosperm length ratio of the seeds in nursery bed was higher than that of the seeds which were stratified with conventional method for 100 days. And also there was no significant difference between dehiscence ratio of the seeds in nursery bed and that from stratification with sand. The germination ratio of the ginseng seeds seeded without stratification at Aug. 5 was about 57% in average of 2 years and was not significantly lower than that of the seeds seeded after stratification at Nov. 15. And there were also no significant differencies of plant growth after germination between the ginseng seedlings from two seeding methods.

Development of Pollen-Derived Embryos and Ploidy Level of Their Regenerated Plants in Paeonia ladctiflora Pall. (작약(Paeonia lactiflora Pall.) 화분에서 유래된 배의 발달과 분화식물체의 배수성)

  • Pollen-derived embryos cultured on the hormone-free medium showed a low germination frequency (12.5%) and poor growth response after germination. The greatest frequency of germination (81.3%) was obtained from the embryos cultured on medium with 0.3mg/L GA$_3$.The greatest frequency of generation (81.3) was obtained from embryos cultured on medium with 0.3mg/L GA$_3$. The embryos precultured for 20 days on medium with 0.3mg/L GA$_3$were transferred to the medium with various combination of hormones such as IAA, kinetin, zeatin, 6-benzylaminopurin (BA) and Gh$_3$. The germination frequency of cotyledonary stage embryos showed above 72% on media with all of the hormonal combinations, but the embryos germinated on medium with 2mg/L BA or 0.1mg/L kinetin and 0.3mg/L GA$_3$ developed more vigorously into plantlets than those of other hormonal combinations. Torpedo-stage embryos cultured on medium with 0.3 mg/L Gh$_3$ were pretreated for 8 weeks at 2-week intervals at 4$^{\circ}C$, The germination frequency of the cold-preheated embryos increased with the increment of pretreatment period from 2 to 8 weeks. The greatest frequency of germination (73.3%) was obtained from the embryos pretreated for 8 weeks at 4$^{\circ}C$. The chromosomes of the root-tip cells of W plane grown for 40 days after germination were observed. Most of the regenerated plants were haploid (55.8%) or diploid (315%), but triploid (1.3%), tetraploid (5.2%), or aneuploid (6.5%) were also detected among them.

Amylolytic activity and Properties of Starch Granules from the Giant Embryonic Rices (발아 거대배아미의 당화력 및 전분입자의 이화학적 특성)

  • Rice seeds of 4 cultivars including Whachung-giant embryonic rice and Nampung-giant embryonic rice, as a group of the non-waxy rice cultivars, and Shinsunchal-giant embryonic rice and Whachungchal-giant embryonic rice, as that of the waxy rice cultivars, were germinated at $27^{\circ}C$ for 3 days to compare the changes in some physicochemical properties of the starch granules and the starch-hydrolysing enzyme activities during germination, respectively. ${\alpha}-Amylase$ activity of rices germinated for 3 days found to be higher than that of malt. Especially, Whachung-giant embryonic rice and Shinsunchal-giant embryonic rice were greater in activity than other rice cultivars and possessed the activities double that of malt. In contrast, ${\beta}-amylase$ of germinated rice found to be considerably less active than malt, although the giant embryonic rice group showed prevalent activity as compared o the normal rice group. With the starch granules, the amount of long glucose chains from amylose molecules were reduced in the non-waxy type giant embryonic rices, while the chain length increase was found in the waxy type giant embryonic rices. For the distribution profile of the glucose chain length from amylopectin molecules, we could observed that the chain length with DP (degree of polymerization) ranged 33 to 66 and 14 to 32 increased with the decreasing rate of that above 67 and below 13 regardless of starch waxiness. With non-waxy type of giant embryonic rices, susceptibility for glucoamylase were found to reduce along with germination, however, increase in susceptibility was observed with waxy rice types. In addition, we found the reduction in both initiation and termination temperature, and enthalpy for gelatinization.

Somatic Embryogenesis and Plant Regeneration with Embryogenic Tissue Lines in Pinus densiflora (소나무 배발생조직 라인 별 체세포배 유도 및 식물체 재분화)

  • This experiment was conducted to evaluate growth rate among 15 embryogenic tissue lines (ETLs), comparison of maturation on somatic embryos (SEs) with 13 ETLs and efficiency with various concentrations of gelrite on SEs germination in Japanese red pine (Pinus densiflora). In comparison of ETLs growth rate (folds) with 15 lines, the 05-4 line (5.3 folds) showed the highest rate, on the other hand, the lowest one was recorded in the line of 05-37 (1.4 folds). The 13 ETLs were tested for the extent of SEs production. The best production was recorded in the line of 05-4 (39.8/90 mg F.W.). However, most of ETLs, except 5 lines (05-4, 12, 21, 29 and 37), did not produce SEs at all, therefore, big differences in the ability of SEs production existed among the ETLs tested. Effects of various gelrite concentrations for SEs germination with 3 ETLs were also compared. The highest result was obtained from 0.2% gelrite concentration with 05-4 line (47.3%), there was a inclination that the rate of germination was gradually declined over 0.2% gelrite concentration with the 05-4 and 29 lines. respectively. In contrast, in the line of 05-37, no SEs germination occurred on medium with 0.1 or 0.2% gelrite. In conclusion, the growth rate, SEs production and germination frequency were appeared to deeply depended on the ETLs.

Effects of mechanical Injury, cotyledon elimination and nutrition for the germination of apple seeds (Malus pulmila var. domestica) (사과 종자 발아에 미치는 기계적 상처, 자엽제거 및 영양분의 영향)

  • When dehulled apple seeds (Malus pilmila var. domestica) were induced to germinate, the rate of germination, which was increased by piercing both the radicle and cotyledon, was related to the site of injury. Also, the germination rates of an embryo with ¾ cotyledon (¾ CE), embryo with ½ cotyledon (½ CE), embryo with ¼ cotyledon (¼ CE), and embryo without cotyledon (NCE) gradually decreased compared to an embryo with a normal cotyledon, but it significantly recovered on the agar media containing MS solution and/or 80 mg/L gibberellin (GA3) as nutrition. The results show that the germination of the embryo was inhibited by the partial removal of cotyledon and also demonstrated that the rate of germination could be significantly increased by adding GA3 (80 mg/L) and/or MS medium.

Studies on the Germination of Korean Ginseng (Panax ginseng C.A.Meyer) Seed II. Influences of Temperature and Seed Treatment on Embryo Growth and Germination (인삼 종자의 발아에 관한 연구 II. 온도 및 종자처리가 배생장 및 발아에 미치는 영향)

  • Freshly harvested and depulped Korean ginseng seeds were subjected to the seed treatment of removing endocarp plus surface sterilization with sodium hypocloride, surface sterilization only, and nontreated control. These seeds were stratified at temperatures of 5$^{\circ}$, 10$^{\circ}$, 15$^{\circ}$ and 20$^{\circ}C$ for 20, 40, 60, 80 and 100 days. Embryo growth of the ginseng seeds of which endocarp was removed was most rapid in each stratification temperature and that of sterilized seeds was slower than unsterilized seeds after 80 days stratification at 15$^{\circ}$ and 20$^{\circ}C$. About 15$^{\circ}C$ was an optimal stratification temperature for embry growth in ginseng seeds. Chilling treatment at 5$^{\circ}C$ for 100 days was needed for better germination of dehisced ginseng seeds. An optimal germination temperature for the ginseng seed following chilling treatment was about 15$^{\circ}C$.

The Effects of Germination Conditions on GABA and the Nutritional Components of Barley (발아조건에 따른 보리의 GABA 함량 및 영양성분 변화)

  • Steeping and germination conditions were investigated in order to produce barley containing a high ${\gamma}$-aminobutyric acid (GABA) content, and the changes in GABA and nutritional components before and after germination were also evaluated in this study. Water absorption rates of three barleys increased alongside both steeping time and steeping temperature. The highest GABA contents of the three barleys, 10.4-14.1 mg/100 g, were obtained from the steeping condition of $25^{\circ}C$ for 24 hr. The GABA contents of germinated barleys ranged from 14.3 to 20.9 mg/100 g, increasing by 3.9 to 14.6 times compared with raw barley. The crude lipid, crude ash and total mineral contents were slightly decreased after germination. The major fatty acids of the three barleys before and after germination were linoleic and palmitic acids. ${\beta}$-glucan contents of three barleys were slightly decreased after germination.

Factors Influencing Somatic Embryo Induction and Plant Regeneration in Aralia elata Seem. (두릅(Aralia elata)의 체세포배 유도, 발아 및 식물체 재분화에 미치는 요인)

  • In order to find optimum conditions for somatic embryogenesis from different individual (2-year-old) in Aralia elata were cultured on MS medium supplemented with 1.0 mg/L 2,4-D, 3% sucrose, and 0.3% gelrite. We also investigated the effect of MS medium salt concentration, BA and ABA on the embryo germination and plant regeneration. While noticeable difference was observed on somatic embryo induction among different individual tree, no apparent difference was seen in both germination and regeneration frequencies. Compared with nonembryogenic calli, embryogenic calli tended to look yellow and/or pale brown in color, slowly growing and soft in their texture. Regardless of BA or ABA treatment, half-strength MS salt medium proved to be better than full strength MS medium in both embryo germination and plant regeneration. Both hypocotyl and cotyledon developments were slightly promoted by adding 0.1 mg/L BA. However, its effect on germination and regeneration seemed inferior to control. ABA treatment on somatic embryos at their torpedo and early cotyledonary stages resulted in poor response in germination and regeneration. Although most regenerated plantlets varied greatly in cotyledon number and shape, they could be developed into normal plants after 4 weeks in culture. More than 95% plantlets were acclimatized in an artificial soil mixture, successfully transplanted to nursery bed and grew normally without any phenotypic abnormalty.

Effect of Density Separation after Pretreatment on Embryo Growth and Radicle Emergence of Carrot(Daucus carota L.) Seeds (당근 종자의 전 처리후 비중선이 배생장과 발아에 미치는 영향)

  • Carrot(Daucus carota L.) seeds (CV Danver 126) were primed and then separated by density differences to improve both the percentage and time of radicle emergence. Seeds for priming were soaked in aerated distilled water for 2 days (water imbibed), 25% solution of polyethylene glycol(PEG) 6000 for 6 or 10 days, salt solution of 0.2M KNO$_3$+0.1M $K_2$HPO$_4$ for 6 or 10 days, or mixed with Agro-Lig with 90% moisture content for 6 days and 70% moisture content for 6 or 10 days (SMP) at 2$0^{\circ}C$, respectively. The greatest embryo growth and the highest radicle emergence were observed from the seeds treated SMP with 90% moisture content for 6 days among the primed treatments. After the SMP treatment, the seeds were separated into density classes with a float-sink procedure using aqueous solution of Maltrin 600 with 0.02/cm$^3$ density increments. The lower density classes of the carrot seeds, the more embryo growth, the higher and the faster rates of radicle emergence were exhibited in order from 1.06 to 1.14 density classes of the carrot seeds treated SMP.

Germination Arrest of Carrot Somatic Embryos Cultured in Liquid Medium (액체배지배양에서 당근 체세포배의 발아 억제 현상)

  • Cotyledonary somatic embryos after being cultured in a liquid MS medium for 1 week were subcultured on a solid MS medium and then the embryos germinated at a rate of 92%, but the rate was lowered by extending the culture period of the embryos on a liquid medium: 26% germination on a liquid medium culture for 4 weeks. Somatic embryos subcultured on the liquid medium showed the normal elongation of hypocotyl and radicle but in part showed secondary embryogenesis on hypocotyl and callus formation on and around the root-hypocotyl juncture. Through observation of scanning electron microscope, apical meristem in plumule showed the loose arrangement of cells, and abnormal leaf primordium formation and growth arrest of the primordium or no leaf primordium formation. Therefore, it is suggested that the germination arrest of carrot somatic embryos on liquid medium culture is due to the structural abnormality of the apical meristem in plumule.
