Effective Screening Condition for Low-Temperature Germinability of Rice (벼 저온발아성의 효율적 검정 조건)

  • This study was carried out to determine the desirable screeing condition for low-temperature germinability of rice seeds. The germination test was performed with fifty rice varieties from three countries, mainly including the recent Korean rice cultivars. The analyzed result of germination test under three temperature conditions(13$^{\circ}C$, 16$^{\circ}C$, $25^{\circ}C$) pointed out that 11 days after seeding at 13$^{\circ}C$ was the most efficient testing condition for low-temperature germinability in terms of mean and variation in germination rate. Highly significant correlations were observed between germination rates of 11 days after seeding at 13$^{\circ}C$ and germination coefficients, average days of germination at 13 for 20 days. Low-temperature germinability were highly positively associated with days to heading or amylose content, but negatively correlated with grain width.

  • 산채류의 종자 및 종묘는 시중 거래가격이 높게 형성되어 재배농가의 경영비 증가로 이어지고 있다. 특히 산채 꾸러미에 분류되는 작물 중 개미취(Aster tataricus), 섬쑥부쟁이(Aster glehnii ), 우산나물(Syneilesis palmata)은 주로 종묘의 형태로 공급되어 수급이 안정치 못하다. 이에 본시험은 개미취, 섬쑥부쟁이, 우산나물의 우량종자 생산을 위한 종자 결실률 향상 재배조건과 채종적기를 구명하고자 하였다. 시험작목을 전북 남원시 허브산채시험장에서 2016년 4월에 종묘를 노지에 정식하였다. 적정 채종 재배조건을 구명하기 위해 2018년 노지, 하우스, 55% 차광막을 설치한 처리구별 각 작물의 개화시기, 개화율, 생육특성, 결실률 등을 조사하였다. 또한 채종적기를 설정하기 위해서 각 작물별 개화 후 50일, 70일, 90일, 110일, 130일 간격으로 채종하여 결실률과 종자 발아율을 조사하였다. 개화가 시작된 일자는 개미취의 경우 하우스재배, 노지재배, 차광재배 순으로 빨랐으며 섬쑥부쟁이는 노지재배에서 개화시기가 가장 늦었다. 모든 작물의 개화가 종료되는 시점은 처리 간 차이가 없었다. 개화율은 개미취와 섬쑥부쟁이는 노지재배, 하우스재배, 차광재배 순으로 많았으며 우산나물은 노지재배시 개화율이 9.3%로 매우 낮았다. 개화기 생육특성을 조사한 결과 개미취는 초장과 엽폭이 차광재배, 하우스재배, 노지재배 순으로 커지는 경향이었으며, 섬쑥부쟁이는 하우스 재배에서 타 재배방법에 비해 우수하였다. 우산나물은 차광재배에서 생육이 가장 좋았다. 결실률은 개미취와 섬쑥부쟁이는 재배방법에 상관없이 결실률이 60%이상이었다. 반면 우산나물은 평균 31%로 타작물에 비해 상대적으로 결실률이 낮았으며 차광 재배일 때 43%로 가장 결실률이 좋았다. 개미취는 개화 50일 이후부터 종자 결실률이 65%이상으로 높았으며, 섬쑥부쟁이의 결실률은 개화 후 50일이 가장 좋았으며, 그 이후로 점차 결실률이 떨어지는 경향이었다. 우산나물은 개화 후 90~110일경에 가장 결실률이 높은 경향이었다. 개미취는 개화 후 70일일 때 가장 발아율이 낮았으며, 90일일 때 가장 발아율이 좋았다. 섬쑥부쟁이의 발아율은 개화 50일 이후부터 최종 채종시기까지 80%이상으로 우수하였다. 우산나물은 채종시기에 따라 발아율 정도가 매우 상이하여, 채종시기가 늦어질수록 발아율이 높아졌다. 따라서 개미취는 하우스 재배를 통해 개화 후 90일 때 채종하는 것이 가장 발아율이 좋았으며, 섬쑥부쟁이는 차광재배일 경우 개화 50일 이후에는 발아율이 90% 이상이므로 이때부터 종자비산 전까지가 채종적기로 판단된다. 우산나물은 타작물에 비해 다소 결실률이 떨어지나 차광재배 시 개화 110일 후 우량종자를 채종할 수 있을 것으로 여겨진다.

  • This experiment was conducted to evaluate the effect of early seed harvest on germination of tobacco(Nicotiana tabacum L.) seeds. Seeds of seven burley tobacco were harvested every two days from 8 to 30 days after pollination and tested for germination. The results are; 1. Seeds harvested 12days after pollination germinated but germination rates were low and varied across cultivars. These seeds will provide viable seeds adequate for breeding program. 2. Germination rates of seeds harvested 24-28 days after pollination were high and showed none significant differences among cultivars. 4. Over-ripen seeds showed low germination rates cused by after-harvest or temporary dormancy.

A Study on the Seed Characteristics and Germination Percent of Several Native Herb Species (자생 초본류 몇 종의 종자특성과 발아율에 대한 연구)

  • Seed characteristics-weight, seed length and width, pure seed percentage are measured on 15 native herb species, and seeds of six native herb species were tested on the seedbed in the forest with autumn seeding. Seeds of 13 species including Aster scaber were very small; seed length is less than 7mm and seed width is less than 3mm pure seed percentages of 10 native herb speciesin-cluding Ligularia fischeriwere were much lower than 5%. Germination percent of five native herb species were significantly different among the growing sites. The mean germination percent was highest 47.00% for Angelica gigas, 38.25% for Ligularia fischeri, 30.88% for PimPinella brachycarpa, 15.75% for Aralia continetalis, 10.00% for Cirsium setidens, and the lowest 9.32% for Aster scaber.

Effects of Plant Activators on Germination of K3PO4-primed Seeds of Pepper (Capsicum annuum L.) (생리활성물질 처리가 priming 고추종자의 발아에 미치는 영향)

  • Effects of plant activators on germination of primed pepper seeds were investigated. The percent germination and T50 of primed seeds with 0.1mM ASM were 98% and 0.96 day at the 7 days after seeding, respectively. However, the germination of primed seeds with 0.5mM ASM were suppressed as 17% at the 7 days after seeding. The percent germination of primed seeds with 0.01mM INA were 90% at the 2 days after seeding. However, those of primed seeds by 0.1mM INA were increased as 90% at the 5 days after seeding. The percent germination of primed seeds was not change by treatment of the BABA and JA.

Effect of Light Quality during Priming and Germination Temperature on Pepper Seed Germinability (파종 전 Priming과 광질 처리, 발아온도에 따른 고추종자의 발아율)

  • Pepper (Capsicum annum L.) used as a spice or fruit vegetable has been transplanted for cultivation to accompany the production of its seedlings in general. The experiment was done to measure the effect of its cultivars (Daemyng; Wanggochu), priming (chemicals; concentration; period), light quality (red; far-red; blue; dark) during priming, and germination temperature (25 or 15$^{\circ}C$ constant; 25/15$^{\circ}C$ alternating) on the rate of germination done under incandescent lamps until 9 days after sowing. Seed germination was better when primed with Ca(NO$_3$)$_2$ than with KNO$_3$. Priming for 6 to 12 days using the former chemical enhanced the final germination rate and shortened the elapsed days to 50% germination, $T_{50}$, compared to no-priming. Under $25^{\circ}C$ constant germination temperature, primed seeds at darkness or far-red light showed the highest rate until 5 days after sowing or the lowest one since 7 days after sowing, respectively. The germination response at 25/15$^{\circ}C$ alternating temperature did not followed the lowest rate of 15$^{\circ}C$ constant germination temperature but inclined to that of $25^{\circ}C$ constant recorded the greatest one. Under 3 germination temperature red light treated during priming elevated the rate since 7 days after sowing compared to the other light treatments, and the germination rate of each cultivar was affected by no-priming or priming, light quality during priming. which imply their interaction to the seed germination.

Effects of Priming Duration and Temperature on the Germination of Forage Seed (Priming 처리시간 및 온도가 목초종자의 발아특성에 미치는 영향)

  • Seed priming is an useful technique for increasing germination and early establishment of seedlings. In this experiment, the conditions for priming of pasture seeds (tall fescue, orchardgrass, alfalfa and white clover) have been optimized to ensure an early germination and more uniform growth of seedlings. The experiment was conducted in a split plot design with three replications. The main plots consisted of four different priming duration such as 2, 4, 6 and 8 days for grass, and 1, 2, 3 and 4 days for legume. The subplots were consisted of three priming temperature such as 10, 15 and $20^{\circ}C$. Effects of priming duration and temperature were different in all four pasture seeds examined. The germination of primed grass (tall fescue and orchardgrass) was the highest on 6 days in priming duration, whereas that of legume (alfalfa and white clover) was the highest on 2 days. In priming temperature, the germination of primed orchradgrass increased as decreasing temperature, however that of legume increased as increasing temperature. These results suggest that seed priming induced earlier and higher germination for all four pastures tested. However, the degree of priming effectiveness on pasture species depending on the priming duration and its temperature.

Comparison of Early Germinating Vigor, Germination Speed and Germination Rate of Varieties in Poa pratensis L., Lolium perenne L. and Festuca arundinacea Schreb. Grown Under Different Growing Conditions (생육환경에 따른 Poa pratensis L., Lolium perenne L. 및 Festuca arundinacea Schreb.의 초종 및 품종별 발아세, 발아속도 및 발아율 비교)

  • Research was Initiated to investigate germination characteristics of cool-season grasses (CSG). Several turfgrasses were tested in different experiments. Experiments I and III were conducted under a room temperature condition of 16$^{\circ}C$ to 23 $^{\circ}C$ and under a constant light condition at 25 $^{\circ}C$, respectively. An alternative environment condition that is a requirement for a CSG germination test by International Seed Testing Association (ISTA) was applied in the Experiment II, consisting of 8-hr light at 25 $^{\circ}C$ and 16-hr dark at 15 $^{\circ}C$. In each experiment, data such as early germinating vigor, germination speed and germination rate were evaluated. Six turfgrass entries were comprised of two varieties each from Kentucky bluegrass (KB, Poa pratensis L.), perennial ryegrass (PR, Lolium perenne L.), and tall fescue (TF, Festuca arundinacea Schreb.), respectively. Significant differences were observed in early germinating vigor, germination speed and germination rate. Early germinating vigor as measured by days to 70% seed germination was variable according to environment conditions, turfgrasses and varieties. It was less than 6 days in PR and 6 to 9 days in TF. However, KB resulted in 11 to 13 days under an alternative condition and 11 to 28 days under a room temperature condition. The germination speed was fastest in PR of 7 to 10 days and slowest in KB of 14 to 21 days. However, intermediate speed of 10 to 14 days was associated with TF. There were considerable variations in germination rate among turfgrasses according to different conditions. Generally, PR and TF germinated well, regardless of environment conditions. However, a great difference was observed among KB varieties, when compared with others. Under a room temperature condition, total germination rate was 71.0% in Midnight and 77.7% in Award. And it increased under an alternative condition, which was 81.7% and 91.7% in Award and Midnight, respectively. However, the poorest rate was found under a constant temperature condition, resulting in 18.0% in Award and 15.3% in Midnight. These results suggest that an intensive germination test required by ISTA be needed prior to the decision of seeding rate, including early germinating vigor and germination speed as well as total germination rate. KB is very sensitive to environment conditions and thus its variety selection should be based on a careful expertise.

Cryopreseryation and Germination of Native Aquilegia Species Seeds by Predehydration Treatment (건조 전처리에 의한 자생 Aquilegia속 식물 종자의 초저온 저장과 발아)

  • Predehydration effects for cryopreservation in -196$^{\circ}C$ liquid nitrogen were studied in Korean native Aquilegia buergeriana var. oxysepala Kitamura and A. flabellata var. pumila Kudo seeds. Aquilegia species seeds were adjusted to moisture contents between 3.2 and 9.7% by air dry treatments. Seeds were placed in paper envelopes after submerged in liquid nitrogen and rewarming in 38 $^{\circ}C$ water. Seeds moisture contents by duration of drying were identified as controlling factors in the survival of Aquilegia species seeds for cryopreservation. Aquilegia species seeds having approximate 5% moisture content were able to withstand cooling to -l96$^{\circ}C$. Undehydrated seeds of Aquilegia buergeriana var. oxysepala Kitamura after being cryopreservated in liquid nitrogen have a 10.9% of moisture content and show 52.5% in germination. But, Aquilegia buergeriana var. oxysepala Kitamura seeds dehydrated by drying for 60 min. to have 6.0% of moisture content before cryopreservation show 84.7% in germination test. Properly dehydrated seeds after being stored in liquid nitrogen showed over 60% in germination rate and also shows an uniform sprouting time,11~13 days in average. Any morphologically deformity in germinating beds has not been observed. Results from this study suggest that Aquilegia species seeds can cryopreservation in liquid nitrogen if the seed moisture content is controled by a proper amount of dehydration.

Study on the Germinability of Sesame Seeds at Lower Temperature (참깨 품종들의 저온발아성에 과한 연구)

  • The objective of this study was to test germinability of sesame (Sesamum indicum L.) varieties at low temperatures. Seeds of III sesame varieties representing different maturity groups and countries were tested at four different temperatures between $10^{\circ}C$ and $16^{\circ}C$. Average germination percentages of III varieties were 92%, 57% and 3.3% at $16^{\circ}C, 14{\circ}C$ and $12^{\circ}C$.respectively. The highest daily germination percentage was observed after 4 days of incubation at $16^{\circ}C$, while it was after 8 days at $14^{\circ}C$. A regression analysis indicated that $15^{\circ}C$ as the minimum temperature for satisfactory germination in field conditions. In general, Korean local collections had better germinability than introduced varieties from 6 countries at different latitudes. 'San dong', a Korean local cultivar. showed the highest germination percentage (24%) at $12^{\circ}C$ when the average germination percentage of all varieties was as low as $3.3^{\circ}C$. Early maturity varieties germinated better at $12^{\circ}C$ and $14^{\circ}C$ than medium or late maturity groups, but no difference was observed at $16^{\circ}C$. Although larger seeds showed better germinability within a variety, there seems to be no relationship between seed size and germinability in sesame.in sesame.
