Anaylsis of ${\gamma}$-aminobutyric Acid (GABA) Content in Germinated Pigmented Rice (발아 유색미의 GABA(${\gamma}$-aminobutyric acid) 함량 분석)

  • The level of ${\gamma}$-aminobutyric acid (GABA) in pigmented rice was quantitatively and qualitatively determined by high pressure liquid chromatography/fluorescence detection (HPLC/FLD). In this study, the recovery rate and limit of detection (LOD) of GABA were 122.4${\pm}$2.4% and 0.23 ${\mu}g/g$, respectively. The geminating pigmented rice samples, which were harvested in Paju, Korea, were soaked in water at $18^{\circ}C$ for 20 hr. After soaking, the samples were germinated at $30^{\circ}C$ for about 24 hr. GABA content was highest (293.0 ${\mu}g/g$) in the germinated red rice. Furthermore, GABA levels in the germinated rice increased significantly by up to 11.1 and 24.7-fold as compared to non-germinated rice and milled rice, respectively. The GABA concentrations of non-germinated rice, with the exception of red rice, were significantly higher than those of milled rice by 7.6-20.6-fold.

Fermentation Characteristics of Cheonggukjang made of Germinated Soybean under Light Condition (명조건에서 발아시킨 콩을 이용한 청국장의 발효 특성)

  • This study was conducted to investigate the fermentation characteristics of cheonggukjang prepared with soybean germinated under light condition. The moisture content and pH of cheonggukjang made with soybean germinated under light condition (CGL) were higher than that of cheonggukjang made with non-germinated soybean (CNG). The color of CGL was darker than that of CNG and cheonggukjang made with soybean germinated under dark condition (CGD). The number of live bacteria in the CGL ($10.7{\pm}0.3\;\log\;CFU/g$) and CGD ($10.7{\pm}0.4\;\log\;CFU/g$) was higher than that of CNG ($9.7{\pm}0.4\;\log\;CFU/g$). The levels of isoflavone in CGL, CGD and CNG were 2,038.0, 1,993.6 and 1,747.0 ug/g, respectively. There were no significant differences in the appearance, aroma, taste and overall acceptability of the cheonggukjang made with different germination conditions. Therefore, it was expected that the cheonggukjang prepared with soybean germinated under light condition could be practically used as a new fermented food.

Antimutagenic Activities of the Germinated Specialty Rices in E. coli and V79 Cultured Cell Assay Systems (E. coli와 V79 배양세포계에서 발아특수미의 항돌연변이 활성)

  • To evaluate the antimutagenic activity of the specialty rices, a giant embryonic rice and a pigmented rice, we measured the inhibitory effect on the chemically induced mutagenesis in E. coli and V79 cultured cell system, as well as on DNA strand scission induced by oxidative damages in vitro. When the inhibitory activity to mitomycin C-induced mutagenesis using SOS chromotest in E. coli cell was measured, the activities decreased in the following order: germinated pigmented rice (40.4%) > germinated giant embryonic rice (37.1%) > pigmented rice (35.5%) > germinated brown rice (15.7%) > giant embryonic rice (14.0%) > brown rice (0.8%). The activities for inhibiting mitomycin C-induced DNA strand scission decreased in the order of pigmented rice > giant embryonic rice > germinated pigmented rice > germinated brown rice > brown rice > germinated giant embryonic rice. We also determined antimutagenic activities of the specialty rices using the suppressing effect on 6-TG resistant colony formation by 4-NQO in V79 cells as a mutagenicity index. The order of antimutagenicity was germinated giant embryonic rice (53.2%) > pigmented rice (40.0%) > brown rice (21.2%) > germinated brown rice (14.4%) > giant embryonic rice (0.23%); in contrast, germinated pigmented rice showed promoting effect on 4-NQO-induced mutagenesis.

Characteristics of Germinated Colored Rice as a Potential Raw Material for Sikhe (잠재적 식혜원료로서 발아유색미의 특성)

  • This work was to study the possibility of using germinated colored rice as a raw material for sikhe. Colored rice was soaked in water at $15^{\circ}C$ for 2 days and then germinated at 15, 20, 25 and $30^{\circ}C$ for upto 10 days. The higher the germination temperature, the higher the germination speed, the rate of increase in ${\alpha}-amylase$ activity, and the rate of increase in extract and its sugar content. The viscosity of extract rapidly decreased first and then slightly increased during germination. The germination speed of colored rice was lower than that of brown rice at $25^{\circ}C$. The germinated colored rice can be used for sikhe not as a saccharifying agent but as a substitute for white rice for the nutritional purpose.

Screening of Physiological Functionality of Germinated Giant Embryonic Rices (발아 거대배아미의 생리활성 효과 검정)

  • Changes in physiological functionality of giant embryonic rice by germination process were investigated using 70% ethanolic extract of the rices. Physiological functionality was evaluated by determining the reducing power, phenolic compound content, GABA content, and antimutagenicity. The results showed that the order of reducing power of the non-germinated rices was Nampung giant embryonic rice>normal rice> Whachung giant embryonic rice, however, the activity was high as the order of Whachung giant embryonic rice> Nampung giant embryonic rice> normal rice by germination process. About 3-fold activity increase was observed for Whachung giant embryonic rice, while, the activity of Nampung giant embryonic rice and normal rice decreased by the same treatment. The phenolic compound content of three rice cultivars were found to be almost same levels. Germination of rice increased the content of phenolic compounds by 2.6-fo1d without any considerable changes for both Nampung giant embryonic rice and normal rice. The GABA contents was highest in Whachung giant embryonic rice, followed by Nampung giant embryonic rice, normal rice in either germination or non-germination condition. The germination increased the GABA contents by more than 2.4-fold for all rice cultivars tested. We also found an increase in the antimutagenic activity by germination process for all cultivars, where the activity was the greatest for Whachung cultivar.

Takju Brewing Using the Uncooked Germed Brown Rice at Second Stage Mash (2단담금에서 무증자 발아현미를 이용한 막걸리 제조)

  • In general cooked rice would be used as a major raw material for making Takju. In this case quality, taste and storage time of Takju were not fully satisfied. Fermentation conditions for Takiu mash were examined by using the germed brown rice in this study. In case of the germed brown rice on 2nd stage mash, alcohol was slowly generated in comparison with the cooked rice. Reducing sugar was slowly produced and the amount of reducing sugar was low. The sugar content was created at a uniform rate. The pH was shown to be higher in mash of the germed brown rice than that of the cooked rice. Acidity change showed a similar inclination to pH change. The degree of yeast growth on the mash of the germed brown rice was revealed to be slightly lower than that of the cooked rice. Temperature of mash was kept to be constant after 3 days from fermentation. Fusel oil produced from the mash of the germed brown rice was less gernerated in comparison with the cooked rice. Amount of amino acid in case of the germed brown rice was indicated to be higher. Takju made with the germed brown rice was shown to be 1.3 times in overall taste, 1.5 times in refreshing as compared with Takju made with the cooked rice. However there are no differences between them in flavor and color of Takju. In overall acceptance Takju made with the germed brown rice was shown to be 1.3 times as compared with Takju made with the cooked rice. In conclusion the germed brown rice was expected to be able to be better in Takju quality.

Antioxidative Activity of Germinated Specialty Rices (발아 특수미의 항산화 활성)

  • Functionality changes by germination of giant embryonic rice and pigmented rice were evaluated with focusing on antioxidative activities of 70% ethanolic extracts. Overall, reducing power of giant embryonic rice and pigmented rice was higher than that of normal brown rice, and the germination of rices tend to enhance their reducing powers. In vitro and ex vivo experiments employing linoleic acid peroxidation and rabbit erythrocyte membrane peroxidation systems, respectively, revealed inhibitory effect on lipid peroxidation was highest in pigmented rice, followed by giant embryonic rice, and normal brown rice from high to low order. Superoxide radical-scavenging activity decreased in order of pigmented rice > giant embryonic rice > normal brown rice, and germination also enhanced their superoxide scavenging ability compared to non-germinated controls. Hydroxyl radical-scavenging ability was highest in pigmented rice, followed by giant embryonic rice, and normal brown rice. Despite marked enhancement in hydroxyl radical-scavenging ability of normal brown rice by germination, order of scavenging ability was not altered among germinated rices. Same trend as with in vitro ROS scavenging was observed for ex vivo scavenging potency on ROSs generated by TPA stimulation in HL-60 cells. Germination-associated differential increase in ROS scavenging ability of pigmented rice and giant embryonic rice, characterized by no induction of cytotoxicity, was observed.

Effect of germination and temperature on the antioxidant activity of coffee (발아 및 발아온도가 커피의 산화방지 활성에 미치는 영향)

  • The present study aimed to investigate the effect of germination and temperature on the antioxidant activity of coffee. Green beans were germinated at 20 and $40^{\circ}C$. Germinated green beans were dried and roasted. Ground coffee was brewed at $90^{\circ}C$ for 5 min. Coffee samples were analyzed for antioxidant compounds and for its antioxidant activity. The total polyphenol content (TPC) and total flavonoid content (TFC) in coffee brewed with coffee beans germinated at $20^{\circ}C$ (CG20) were significantly higher than those in coffee brewed with non-germinated coffee beans (CNG). The same tendency was observed on 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging assays. TPC and TFC of coffee brewed with germinated coffee beans decreased with an increase in germination temperature from 20 to $40^{\circ}C$. In conclusion, germination of coffee beans contributes to an increase in its antioxidant activity. However, setting the appropriate temperature for germination is an important factor in determining the antioxidant activity of coffee.

Characteristics of grain powder as a viscosity agent (증점제로서의 곡류 가루 특성 연구)

  • For increasing both the viscosity and intake of calories and nutrients, four cereal powders: Goami (GR), waxy barley (WBL), germinated brown rice (GBR), and oat (OT) were added to rice porridge, and the properties of the porridge were compared with that containing a commercial thickener. Among the samples, the porridge with WBL showed the highest viscosity, whereas that with GR showed the lowest. With regards to color properties, the porridge with GR showed the highest whiteness, while those with OT or GBR had the highest yellowness, affecting the color of the rice porridge. The viscosity level of porridges with cereal powders did not reach that of the porridge with the commercial thickener at $90-95^{\circ}C$; however, at $55-60^{\circ}C$, the viscosity of the porridge with cereals was enough to replace the commercial thickener. With regards to organoleptic tests, the overall acceptability was mainly attributed to color, and there were no significant differences in other properties. From these results, it seems that the cereal powders could be suitable substitutes for commercial thickeners.

Antiproliferation Effects of Germinated-Korean Rough Rice Extract on Human Cancer Cells (한국산 발아 벼 추출물의 여러 가지 암세포주에 대한 증식 억제 효과 비교)

  • This study was conducted to investigate the effects of 70% ethanol extracts of various germinated-rough rice cultivars: ('Ilpum', 'Goami2', 'Keunnun', 'Sulgaeng', 'Baegjinju', and 'Heugkwang') on proliferation of human cancer cell lines (MKN-45, HCT116 and NCI-H460). Antiproliferation effects of rough rice on different cancer cell lines were higher in after germination than before germination. The viability of HCT-116 colon cancer cells was lowest at 18.89% in after germination of 'Heugkwang' at 1.0 mg/mL. The cell viability of MKN-45 lung cancer cells and MKN-45 stomach cancer cells were in the range of 5~10% in after germination of 'Ilpum', 'Goami2', 'Baegjinju', and 'Sulgaeng', and 'Heugkwang' at 1.0 mg/mL. These results suggest that germinated rough rice might have a potential preventive effect on human cancer cells.
