Ribonucleotide Reductase Activity of Schizosaccbarornyces pombe Is Inhibited by Escherichia coli RecA Antibody (Schizosaccbarornyces pombe에서 Escherichia coli RecA 항체에 의한 Ribonucleotide Reductase 효소활성 저해)

  • We have previoosly demonstrated that the RecA-like protein of Schizosaccharomyces pombe (S. pombe) is immunologically related to Escherichia coil (E. coil) RecA protein and that the cellular level of the protein is significantly increased by inhibitors of nucleotide pool-forming enzymes such as hydroxyurea (HU) and methotrexate (MTX) (lee and Park, 1994; lee et al., 1994). In this study, we report that the ribonudeotide redudase activity of S. pombe is inhibited by E. coil RecA antibody, as determined by thin layer chromatography using [5-$^3$H]CDP as a substrate. The relative activity of ribonucleotide reductase was dramatically inhibited by 100 mM of flu (26.4% reduction) in in vitro assay, compared to that of non-treated control. The ribonucleotide reductase activity was also inhibited by immunoprecipitation with E. coil RecA antibody (43.3% reduction). These results indicate that the strudure of S. pombe ribonucleotide reductase is in part similar to that of E. coil RecA protein.

Biosynthesis of Organic Germanium Using Cordyceps militaris (동충하초를 이용한 유기게르마늄의 생산)

  • In the present study, an attempt has been made to produce a high quality medicinal mushroom Cordycops militaris supplemented with organic Ge. Cordycops militaris was cultivated in SDAY liquid medium containing yeast extract 10 g, peptone 10 g, glucose 40 g per liter and chrysalis media supplemented with inorganic Ge at 100, 500, 1000, and 5000 ppm concentrations. The greatest organic Ge production of 442.4 ppm/g and 284 ppm/g were observed in SDAY liquid and Chrysalis media cultures supplemented with 100 ppm inorganic Ge respectively. Similarly, 4,509.7 ppm/g and 1,058 ppm/g of organic Ge were obtained from liquid and chrysalis media cultures supplemented with 5,000 ppm and 1,000 ppm inorganic Ge, respectively. In addition, higher concentration of organic Ge was obtained in mycelia than fruiting bodies. These results indicate that the concentration of organic Ge increase with decreasing inorganic Ge concentration in the medium. This is the first report on production of high valuable Cordyceps militaris contained with organic Ge.

  • In this paper, it is predicted that the tendency of planning and genre films to produce high - quality films will continue, but it is necessary to have high value and meaningful films and solid storytelling. From the blockbuster warships to arts and low-budget films, the movie genre is centered around fiction and storytelling. However, in the case of Princess Deokhye, it is difficult to recognize any categories of entertainment or artistic due to the fact that it is highly visually and has a high historical status and low awareness. And the distortions of the facts have caused a lot of controversy. However, it seems that Dongju and Park are usually composed of individual - oriented narratives and can be freed from the artistic approach and historical facts in the material. THE DISTRIBUTIONS AND TRENDS IN MALOCCLUSION PATIENTS - A 10 year study of 2155 patients from YDSH (연세대학교 영동세브란스병원 교정과에 내원한 부정교합 환자의 분포 및 경향에 관한 연구)

  • Distributions and trends were examined in 2155 malocclusion patients who had been examined and diagnosed at Deparment of Orthodontics in Youngdong Severance Hospital over a 10 year-period from 1984 to 1993. The results were as follows ; 1. The number of patients per you had nearly quadrupled during the 10 year-period, with females($58.3\%$) outnumbering males ($41.7\%$). 2. Age distribution had shown 7-12 year-old group being the largest($53.3\%$), but percentages of 7-12 year-old and 13-18 year-old groups had been decreasing while that of above-19 patients had gradually incresed. 3. Distributions in the types of malocclusion according to the Angle's Classification had shown $27.8\%$ for Class I, $22.6\%$ for Class II division 1, $7.9\%$ for Class II division 2, and $41.6\%$ for Class III. Percentages of Class I patients had decreased while percentages of Class II and III patients had increased. 4. Non-extraction cases ($75.4\%$) had outnumbered extraction cases ($24.6\%$), with a general trend toward fewer extraction cases over the 10 year period. 5. Patients who had orthognathic surgery had been increasing, with Class I, II, and III cases comprising $8.8\%,\;16.9\%\;and\;74.3\%$, respectively. There had been three times as newly 2-jaw operations as 1-jaw operations. 6. Nearly $4\%$ of the subjects had facial asymmetries, with Class III cases being the most frequent. 7. Patients with TMD symptoms comprised $4.6\%$, with an increasing trend. The symptoms had occurred more frequently in older patients and in females. 8. Geograpic distributions showed majority (3/4) of patients from the nearby areas, namely Gangnam-gu, Seocho-gu and Songpa-gu.

Electrofusion of Yeast Cells and Their Genetic Analysis Using RAPD-PCR (효모세포의 전기융합 및 융합세포의 RAPD-PCR을 이용한 유전적 분석)

  • In the present study, an attempt has been made to produce hybrid yeast strains of different useful and dominant characteristics. The hybrid yeast strains were produced by electrofusion and their genetic analysis were performed by RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction). The protoplast of Saccharomyces cerevisiae KCTC 7904 and Zygosaccharomyces rouxii KCTC 7966 were obtained above 92% when treated with lyticase at $30^{\circ}C$ for $60{\sim}90$ min after the pretreatment of $1{\sim}2%$ 2-mercaptoethanol at $30^{\circ}C$ for $15{\sim}20$ min. The fusant was produced from paired protoplast stage under the electric pulse at high frequency conditions (1.5 MHz/50 pV, 615 $V/256\;{\mu}sec$) within glass-platinum made electrofusion chamber. Changes in RAPD patterns in mother cells and hybrid cells proved that the fusant contains two types of yeast gene originated from its parent. Furthermore, fermentation characters exhibits by the fusant cell confirmed its genetic changes. These results suggest that genetically stable hybrid yeast strains of economic importance can be produced by electrofusion technique and these electrofused yeast cells have an enormous impact in biotechnology and biomedicine.
