Selection of koji and yeast strain for improvement of Choungju quality (청주의 주질 개선을 위한 국 및 효모의 선정과 그 발효 특성)

  • To improve the quality of Choungju. a kind of rice wine, two different types of koji were prepared and compared : one from wheat bran with Aspergillus usamii mut. shirousami Y-79 and the other from rice with A. oryzae, and yeast strains from cereal wine mashes were newly isolated and applied for the brewing method. Levels of the related enzymes such as glucoamylase, ${\alpha}-amylase$ and acid protease in the wheat bran koji were higher than those in the rice koji, whereas vice versa in the case of acid carboxypeptidase. An amount of $2{\sim}3%$ wheat bran koji to the weight of total rice was adequate for saccharification of the mash and resulted in improved duality of the fermented mash, accompanied by decrease in koji ordor and amino acidity. When the solution of wheat bran koji and the isolated yeast strains were employed, the better Choungju taste was obtained in comparison with those fermented with Japanese sake yeasts, the strain K-7 and 9, due to the lower content of organic acids especially succinic acid. The amino acidity of the fermented mash was able to be controlled to some extent, when the rico types of koji and the isolated strains were employed, by changing the ratio of the two koji types. However, the application of the rice koji with the isolated strains was not desirable for the brewing process because organic acids were produced in excess and ethanol fermentation was retarded.

Enzyme Production by the Mutant of Aspergillus oryzae (국균변이주(麴菌變異株)에 의한 효소생산(酵素生産))

  • One hundred and fifty one mutant strains were obtained from the parent strain Aspergillus oryzae MF by ultra-violet ray irradiation. Among those mutants a strain, Asp. oryzae UM-36 which hyperprodued protease, was selected and its morphological characteristics and the production of enzymes protease, ${\alpha}$-amylase, and glucoamylase on wheat bran koji and on soy-sauce koji were studied. The results obtained were as follows 1. The selected mutant showed slower growth and weak sporulation on malt agar and on Czapek agar than the parent strain. 2. The conidiophores of the mutant were generally shorter than those of the parent when grown on malt agar. 3. Sectoring in colonies was not found when grown on malt agar and on Czapek agar. 4. The level of protease production by the mutant was increased approximately 1,4-fold higher on wheat bran koji and 2-fold higher on soysauce koji than by the parent. 5. The production of ${\alpha}$-amylase and glucoamylase by the mutant were also increased as compared with the parent on wheat bran koji and on soy sauce koji. 6. In the case of parent strain and mutant strain, the highest activity of protease appeared after three days in wheat bran medium at $30^{\circ}C$ incubation, but the highest activities of ${\alpha}$-amylase and glucoamylase appeared after two days.

  • Several mutants were isolated from the parent strain of Aspergillus niger CF: the first mutant strain CF-11 was obtained by UV irradiation, and the second mutant strain CF-21 and CF-22 were from NTG (N-methyl-N'-nitroso-N-nitroso-guanidine) treatment on the CF-11. These mutants were characterized, and their enzyme and acid production on wheat bran Koji and wheat flour Koji were studied. Asp. niger CF-22 mutant appeared to be tan type which conidial heads were discolored. It's glucoamylase activity was inreased approximately two times and its ${\alpha}-amylase$ about 50 percent as compared with that of the parent strain of Asp. niger CF, when grown on wheat bran Koji under the optimal conditions. Asp. niger CF-21 mutant showed slower growth rate and poor sporulation than the wild type, although its conidial heads were not discolored. Approximately 4-fold increment in its acid production was observed as compared with the weld type. The activities of glucoamylase and ${\alpha}-amylase$ of the Asp. niger CF-22 and CF-21 mutants were higher than those of the wild type, but their protease activity was rather lower. The maximum production of glucoamylase by the Asp. niger CF-22 mutant was obtained after 2 to 3 days incubation on wheat bran at 30 to $35^{\circ}C;$ ${\alpha}-amylase$2 days incubation at 30 to $35^{\circ}C$. The maximal levels of acid production by the mutant CF-21 was appeared after 2 days incubation on wheat bran Koji, and after 3 days on wheat flour Koji at $30^{\circ}C$. Little differences in the levels of acid production were observed between on wheat bran and flour Koji.

Decomposition of 2,4,6-Trinitrotoluene (TNT) by Gamma Ray Irradiation (감마선 조사에 의한 2,4,6-Trinitrotoluene (TNT)의 분해)

  • The purpose of this study was to evaluate the potential of a gamma irradiation to decompose 2,4,6-trinitrotoluene(TNT) in an aqueous solution. The decomposition reaction of TNT by gamma irradiation was a pseudo first-order kinetic over the applied initial concentrations($25{\sim}100mg/L$). The dose constant was strongly dependent on the initial TNT concentration. The removal of TNT was more efficient at pH below 3 and at pH above 11 than at neutral pH(pH 5-9). The required irradiation dose to remove over 99% of TNT was 40, 80 and 10 kGy, individually at pH 2, 7 and 13. The dose constant was increased by 1.6 fold and over 15.6 fold at pH 2 and 13, respectively, when compared with that at pH 7 When irradiation dose of 200 kGy was applied, the removal efficiencies of TOC were 91, 46 and 53% at pH 2, 7 and 13, respectively. Ammonia and nitrate were detected as the main nitrogen byproducts of TNT and glyoxalic acid and oxalic acid were detected as organic byproducts. The results showed that a gamma irradiation was an attractive method for the decomposition of TNT in an aqueous solution. However, regarding the application of high energy radiation for the TNT decomposition and mineralization, an application of an acidic pH below 3 to the solution before irradiation should be considered.

  • These experiments were carried out to study influences of the kind of yeasts and of brewing condition on fermentation of Takjoo mash. The results obtained were as follows: 1. Kind of yeasts and the number of yeasts in mash. When the first stage mash was fermented at $20^{\circ}C$ for $1.5{\sim}2.5$ days and at $25^{\circ}C$, $30^{\circ}C$ for $1{\sim}2days$, in the second stage mash that was fermented at high temperature, the number of yeasts was less as compared with the case of fermentation at low temperature, but the living yeasts number of Takjoo yeast strain Dm-1 was more than those of sake yeast, strain No. 7. 2. Kind of yeasts and composition of ripened mash. 1) In the secondstage mash that was fermented at high temperature($30{\sim}35^{\circ}C$), alcohol percentage of ripened mash using the selected Takjoo yeasts (strains: Dm-1, Y-1) was remarkably higher than the case of another yeasts (strains: No.7, No.6, No. 396, No. 1). 2) Acidity of mash had a little differences between strain Dm-1 and strain No. 7. 3) In the second stage mash that was fermented at high temperature ($30{\sim}35^{\circ}C$), the amount of Formol-N using strain Dm-1 was remarkably less than strain No.7. 3. Brewing condition and alcohol percentage of mash. 1) The fit amount of wheat bran Kuk addition per material was 3 percentage and it was adequate to use the mixture of wheat flour Kuk 20 percentage and wheat bran Kuk 1-2 percentage. 2) Though brewing concentration of the first stage mash was duiluted by twice of general brewing concentration, the yeast reproduction was normal. 3) In addition of wheat flour $80{\sim}140g$ per 180ml water, alcohol percentage of the mash increased almost propotionally according to the increase of the amount of wheat flour. 4) It was recognized that three stage brewing was superior in method to two stage brewing at present.

청주의 제조와 판매현황

  • 오늘날의 청주(淸酒)는 SAKE라는 이름으로 일본(日本)을 대표(代表)하는 술로 여겨지고 있으나 사실 일본식(日本式) 청주(淸酒)의 제법(製法)도 결국(結局)은 우리나라에 그 근원을 두고 있다. 오늘날의 청주(淸酒)는 우리의 술 빚는 법이 일본(日本)으로 전래된 후, 일본(日本)에서 잘 다듬어져 다시 우리나라에 상륙(上陸)한 것으로 現在의 청주(淸酒)의 근본도 그 뿌리는 백제를 비롯한 우리나라에 있음은 주지할 만한 사실이다. 그러나 우리 고유의 술빚는 법이 체계적으로 정리되지 못하고 사라져 버린데에는 우리의 전통사상의 일부인 과학, 기술 노동을 천시하는 풍조와 술빚는 일을 특히 여성의 전유물로 여겨 양조법의 정립이 이루어지지 못한 것도 큰 이유라 하겠다. 자가용(自家用)으로만 사용되던 주조(酒造)에서 탈피하여 공업적(工業的)인 규모의 청주생산은 1883년 1월 후꾸다라는 일본인이 부산에 세운 공장을 시초로, 1917년 현 백화양조의 전신이라 할 수 있는 조선주조주식회사 설립으로 본격화되었다. 우리의 전통청주(약주)는 곡류와 누룩을 이용하여 빚는데, 누룩은 밀기울을 잘 다져서 장기간에 걸쳐 제조하며, 누룩에는 당화효소를 분비하는 곰팡이류와 알콜발효를 할 수 있는 효모류가 함께 부착되어 있다. 따라서 호화시킨 곡류와 누룩을 함께 넣어서 술을 빚게되면 당화와 발효가 동시에 일어나게 된다. 일본식 청주도 Koji라 불리는 국(麴)을 제조하는 것을 달리할 뿐 우리 전통청주와 제법의 맥락은 같다. Koji는 찐쌀에 곰팡이류를 인위적으로 접종하여 배양한 것으로, 결국 당과용 효소를 얻는다는 것은 누룩과 같은 원리이나 곰팡이균을 분리하여 접종하는 것이 다르다. 한국의 청주판매 현황을 보면 80년대 초반 급격한 감소추세를 보였으나, 업계가 적극적인 판촉활동을 벌임과 동시에 새로운 TYPE의 냉청주(冷淸酒)를 개발하여 여름철이 비수기였던 시장 상황을 완전히 깨뜨려 사계절의 주류로 만드므로서 뚜렷한 회복, 증가추세로 바꾸었다. 앞으로 좀더 잘 다듬어지고 현대 이미지에 맞는 청주가 지속적으로 등장하여야 하며, 결국 이러한 민족고유의 맛이 가미된 청주 TYPE의 주류가 우리나라를 대표하는 국주로까지 발전할 수 있으리라 확신한다.

Microbial Studies on the Korean Traditional Soju -Part 1. Characteristics of Fungal Amylases Produced by the Isolate from the Native Youngkwang Koji- (우리나라 재래 소주에 관한 미생물학적 연구 -제1보. 영광 토종국에서 분리한 곰팡이의 Amylase 활성-)

  • The most active strain for the amylase activity from the native Youngkwang koji, was isolated and identified as Aspergjllus awamori. The optimal conditions for the production of ${\alpha}-amylase$ and glucoamylase were investigated and the properties of these enzymes were also examined. Maximum yields of ${\alpha}-amylase$ and glucoamylase were obtained at $30^{\circ}C$, pH 5.0 for days. The production of these two enzymes were increased with the addition of maltose, urea, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$, and $CaCl_2{\cdot}2H_2O$. The activities of these enzymes were maximized at $50^{\circ}C$, $pH\;5{\sim}6$. The heat stabilites of ${\alpha}-amylase$ and glucoamylase were maintained at $50^{\circ}C$ for 20min and 40min, respectively. Also, the addtion of $MgSO_4{\cdot}7H_2O$, $K_2HPO_4$, and $CaCl_2{\cdot}2H_2O$ salt increased the heat stabilities of these enzymes.

Studies on the Utilization of Sweet Potatoes for Takju Brewing (탁주양조원료(濁酒釀造原料)로서 고구마의 이용(利用)에 관(關)한 연구(硏究))

  • In order to utilize sweet potatoes for the material of Takju, brewing experiments with raw sweet potatoes, sweet potato chips powder and its koji were conducted; and various tests were carried out on effect of the treatments of acid, alkali, polyphenol oxidase inhibitor, oxidizing and reducing agents upon the prevention against coloring of sweet potato chips by steaming, and on peeling effect of sweet potatoes by the alkali and heat treatments. The results obtained were as follows. 1) In the case of brewing with raw sweet potatose, each plot showed low acid and ethanol content, and its finished Takju had an undersirable color and odor. The plots which were mashed after peeling showed higher ethanol contents than the plots mashed without peeling. 2) In the case of brewing with sweet potato chips powder, each plot contained considerably more amount of ethanol than the plots brewed with raw sweet potatoes, white it contained less amount of acid. The ethanol contents of the plots using wheat bran koji were $10.5{\sim}11.4$ per cent 4 days after mashing, and were higher than those of the plots using malts powder. Their finished Takju was inferior in quality because of the lack of acid and being darkened gradually in process of time. 3) The kojies which were made of sweet potato chips powder with Neurospora sitophila or Aspergillus oryzae had good appearance, but the Takju mashes brewed with these contained remarkably less amount of ethanol. 4) Effect of the treatments of acid, alkali, polyphenol oxidase inhibitor and organic solvents such as ether and ethanol upon the prevention against coloring of sweet potato chips was not recognized. Alum and burnt alum were effective a little on the decolorization, and among the oxidizing and reducing agents tested, potassium permanganate was most effective. 5) Darkening of sweet potato chips powder in course of heating after mixing with water was not affected by pectin and amino acids, but by tannin. 6) Sweet potatoes were not peeled easily by friction after soaking in the boiling solution of 3 per cent alkali for 6 minutes and peeled in boiling water for 12 minutes. From the viewpoint of the results above mentioned, it seems to be necessary to study further on the isolation of microorganisms which are able to decompose the coloring substances and yeasts which are adequate for the fermentation of sweet potatoes in order to utilize sweet potatoes for Takju brewing, because brewing with raw sweet potatoes, sweet potato chips powder and its koji was unsuccessful, and effect of the various treatments on the decolorization of sweet potatoes was not recognized.

  • 94 Cellulase producing strains were isoated from soils, composts, rotten woods and straws, and gastric contents and feces of herbivorous animals in various places. Among them, the strain MC-9, MC-10, MC-53 and MC-61 were found to be highly active in the degradation of carboxy methyl cellulose. Their cultural conditions adequate for the cellulase formation and effects of inorganic salts and various organic substances added to the wheat bran media were investigated. The results obtained are as follows; 1. Optimum conditions for the cellulase formation were MC-9: pH 5.5, temp. $35^{\circ}C$, incubation time 5 days, MC-10: pH 5.5-6.0, temp. $30^{\circ}C$, incubation time 5 days, MC-53: pH 3.5, temp. $30^{\circ}C$, incubation time 5 days, MC-61: pH 3.5-4.0, temp. 30-$35^{\circ}C$, incubation time 5 days. 2. Their cellulase activity in their optimum conditions were MC-9: CMC-LP(liquefying power). 87.7%, CMC-SP(saccharifying power) 3.20 glucose mg./gm. of the cultures/min., MC-10: CMC-LP 82.9%, CMC-SP 2.48 glucose mg./gm. of the cultures/min., MC-53: CMC-LP 72.4%, CMC-SP 1.76 glucose mg./gm. of the cultures/min., MC-61: CMC-LP 87.1%, CMC-SP 2.08 glucose mg./gm. of the cultures/min. 3. Additions of inorganic salts to the wheat bran media were not significant for the cellulase formation, but additions of soybean film and orange-peel pomace promoted the CMC-liquefying power 3 to 5 percent in wheat bran cultures of the strains.

Producton of Yeast Spores from Rice Wine Cake. (주박으로부터 효모포자의 생산)

  • Rice wine cake (RWC) is the solid waste obtained after rice wine fermentation. For the mass production of the spores of yeast Saccharomyces from RWC, the optimum pretreatment condition of RWC, the optimum composition of culture medium, and the optimum culture condition were examined. For sporulation, yeast cells were grown in the pre sporulation medium (PSM), transferred into sporulation medium (SM) containing 1 % potassium acetate, and incubated in a rotary shaking incubator at $25^{\circ}C$ for 4 days. The supernatant of the mixture of RWC and water was used as the presporulation medium (PSM). The optimum temperature and time for the pre-incubation of the mixture of RWC and water (1:2) to obtain maximum sporulation yield were $V^{\circ}C$ and 24 hr, respectively, and optimum culture time in PSM was 48 hr. Using these optimum conditions, the asci number obtained was 0.72$ 1.06${\times}$10^{8}$$m\ell$. The addition of wheat coat koji into SM increased the final number of asci to beTEX>$10^{8}$ $m\ell$. Spores were formed in the SM with the initial pH of 7-11, but no spores were formed in the SM with the initial pH of 5. To save the time and effort to pretreat the RWC, 2% and 0.5% RWC without any pretreatment were directly added into PSM containing 1 % brown sugar and SM, respectively, and the maximum asci number of $1.27${\times}$10^{8}$ /$m\ell$ was obtained.
