납(Pb)이 생쥐 신장세포에 미토콘드리아 미세구조 및 전자전달계에 미치는 영향 (Effects of Lead on the Ultrastructure ana the Electron Transport System of Mitochondria of Mouse Kidney)

  • To investigate the effects of lead on the electron transport system and ultrastructure of mouse kidney mitochondria, various lead acetate concentrations were treated in vitro and respiration rate, enzyme activities were measured. Ultrastructural changes at state IV respiration were also observed. To compare with in vivo experiments, mouse were injected intraperitoneally of 100 mg lead acetate per kg body weight and state IV respiration rate and enzyme activities were measured. Ultrastructure of renal proximal tubular cells were also observed. In in vitro treatement, decreased state IV respiration, decreased enzyme activities, ruptured membranes and inhibition of condensed to orthodox transformation were observed. In in vivo treatment, decreased state IV respiration and decreased enzyme activities were observed after 24 hrs of i.p. injection. Cytochrome c oxidase activity showed twice the inhibition compared to NADH-CoQ reductase activity at 24 hrs. Continuous decreased state IV respiration was observed after 48 and 72 hrs of injection, however, the enzyme activities were increased to control level. Lead-protein complex which probably inhibits the toxic effects of lead appeared. To conclude, dominant effect of lead on the electron transport system appeared at cytochrome c oxidase activity, and the increased enzyme activities may be a result of appearance of lead-protein complex.

HL-60세포에서 disulfiram의 항암작용과 미토콘드리아 안정성에 대한 연구 (Anti-tumor activity and mitochondrial stability of disulfiram in HL-60 cells)

  • Disulfiram (DSF) is a member of the dithiocarbamate family that can bind copper. Recent studies have shown that DSF has anti-cancer activities, but the mechanism has not been clarified. Therefore, it is important to study the action mechanism of DSF to maximize its anticancer effects. A human leukemia cell line, HL-60, was used in this study. HL-60 cells were treated with DSF and the cellular metabolic activity was measured. DSF increased the cell death of HL-60 cells in annexin V-fluorescein isothiocyanate/propidium iodide staining analysis. In addition, DSF decreased the mitochondrial membrane potential (MMP) of the HL-60 cells. The cytotoxicity of DSF on HL-60 cells was observed at 0.4 μM. Interestingly, the reduction of MMP by DSF was recovered by N-acetyl-L-cysteine, an inhibitor of reactive oxygen species (ROS) production. This suggests that the decrease in MMP by DSF is closely related to the production of ROS in HL-60 cells, which indicates the relationship between the apoptosis of HL-60 cells by DSF and the role of the mitochondria. This study provides clinicians and researchers with valuable information regarding the anti-cancer activity of DSF in terms of the action mechanism.

고려인삼의 미토콘드리아 DNA의 분자생물학적 특성연구 (The Characterization of Mitochondrial DNA of Korean Ginseng (Panax ginseng C.A. Meyer))

  • This study was focused on the characterization of mitochondrial DNA (mtDNA) for molecular 9enetical approach of energy Production related mechanism in Panax ginseng. The simple and efficient method of mtDNA isolation from ginseng has been developed by modification of recently advanced methods. This procedure can successfully apply to mtDNA isolation of several plants. mtDNA of etiolated shoot and one-year root were digested with restriction endonucleases, but that of 6-year root not. Any difference was not observed in the restriction endonuclease digestion patterns among the ginseng variants. Molecular size of ginseng mtDNA was estimated at least 159 kb by the restriction endonuclease fragment analysis. The 4.5 kb extra band at the lane of EcoRII treatment could be observed in restriction patterns digested with the methylation sensitive endonucleases, BstN I and EcoRII. For construction of mitochondrial genomic library of ginseng, mtDNA was partially digested with EcoRl, and packaged with EMBL4 phage vector. Genomic library was screened and purified for further research including restriction mapping of ginseng mtDNA, and cloning of the genes. The gene of ATP synthase A subunit was cloned from the purified EMBL4 library clone No. 16. Now, clone No. 16 is subcloned for structure gene sequence analysis.

생쥐 신경계의 미세구조에 미치는 Acrylamide의 영향 (The Effect of Acrylamide on the Ultrastructures of Nervous System of the Mouse)

  • Acrylamide의 신경독성에 대해 조사하기 위하여 초기운동실조증이 유도된 생쥐의 신경계에 대한 Acrylamide의 영향을 관찰한 결과, 중추 및 말초신경조직에 있어서의 손상이 보여 척수의 전반적 공포현상, 신경원 및 신경교의 세포손상 그리고 말초신경섬유의 퇴행현상들이 보였다. 또한 이들에 대한 미세구조적 관찰에 의하여 신경원에서의 비정상적 미토콘드리아 및 척수전근 섬유에서의 수초의 균혈화 현상등이 야기됨을 알았으며 좌골신경에서의 수초의 부분적 붕괴 및 축삭형질내의 퇴행현상들도 보였다. 위와 같은 실험결과는 Acrylamide로 인한 중독의 영향이 말초신경장애의 증상으로 나타나나 최초의 신경독성효과는 중추 및 말초신경계에 대해 거의 유사한 시기에 작용함을 시사한다.

  • 고고 유적지에서 출토된 뼈에서 추출한 DNA는 부식산과 풀빅산 등의 토양성분과 콜라겐 등 다양한 오염물질이 포함되어 있어 DNA의 추출 및 분석이 매우 어렵다. 본 연구에서는 phenol 추출법, silica 추출법 등 두 가지 대표적인 고대 DNA 추출법의 효율을 DNA 증폭 결과에 의하여 비교하였다. 또한 울트라급의 초음파를 시료 용해에 적용한 후 silica 추출법으로 DNA를 분리한 방법이 기존의 phenol 및 silica 추출법에 비하여 미토콘드리아 DNA와 아밀로제닌 유전자 증폭 결과가 더 우수한 것으로 나타났다.

  • 난청의 약 50%는 유전적 요소가 원인이기 때문에 난청의 유전적 배경을 이해하는 것은 중요하다. 현재까지 150개 이상의 원인유전자들이 밝혀져 있다. 이번 종설에서는 유전성 난청의 분류, 유전성 난청 원인규명의 난점, 유전성 난청과 관련된 내이의 구조와 기능, 증후군성난청, 비증후군성난청, 미토콘드리아 유전성 난청, 그리고 다인자성 난청에 관해 논하고자 한다. 그리고 유전성 난청을 가진 환자의 치료적 접근과 유전적 상담을 간략하게 설명하고, 마지막으로 유전성 난청에 대한 앞으로의 연구 방향을 제시하고자 한다.

인삼의 Triol 및 Diol계 사포닌이 쥐의 신피질 미토콘드리아 의 Glutamine 이동에 미치는 영향 (Effect of Triol and Diol Fractions of Ginseng Saponin on Glutamine Transport into Rat Renal Cortical Mitochondria)

  • Attempts were made if diol and triol fractions of ginseng saponin affect on glutamine transport into rat renal cortical mitochondria, swelling, phosphate dependent glutaminase activity, and consumption of oxygen. The following results were obtained. When mitochondrial preparation from rat renal cortex was incubated in medium containing 14C-glutamine and either triol or diol fractions, radioactivity was shown to increase at both 10-6% and 10-5% triol fractions of ginseng saponin, but reduce in case of diol fraction. The remarkable acceleration of the rate of swelling of renal cortical mitochondria was observed in the presence of 10-1% trios and diol fractions but no accerelation at lower concentrations. The activity of phosphate dependent glutaminase from renal cortical mitochondria was slightly activated at 10-2% of triol fraction. However, there was no effect in case of diol fraction. Oxygen consumption by mitochondria from renal cortex was remarkably increased at concentrations of 10-5% and 10-6% triol fractions, but reduced in the case of diol fractions. On the basis of these observations it was concluded that triol fraction of ginseng saponin might increase the transport of glutamine into mitochondria by accelerating the respiratory chain and supplying additional energy to mitochondria, and physiological role of triol fraction was entirely different from that of diol fraction of ginseng saponin.

  • The topology of the enzyme has been investigated by biochemical studies including chemical labeling and cross linking. Thirteen subunits(polypeptides) of the cytochrome-c-oxidase have localistic characteristics of existing in the matrix side or cytoplasmic side in the mitochondria. In order to observe the distribution of the enzyme subunit on the mitochondria membrane, immunogold-labeling methods were employed. Antibody was obtained from the serum of immunized rabbit with enzyme subunit antigen which was obtained from cytochrome-c-oxidase of the beef heart muscle mitochondria. Beef heart muscle tissue as a tissue antigen was stained with immunized rabbit IgG and protein A gold complex. Electron microscopy has identified the existance of cytochrome-c-oxidase subunit $Mt_I,\;Mt_{II}\;and\;Mt_{III}$ on the membrane of cristae and outer chamber of mitochondria and the subunit $C_{IV}$ on the membrane of cristae and matrix of mitochondria. Particularly, the subunit $C_{IV}$ was also observed to exist in the sarcoplasm of muscle tissue.

오스뮴침착법에 의한 초파리 단안시각계의 미세구조 (Ultrastructural Study of Drosophila Ocellar Visual System by Osmium Impregnation)

  • 성충초파리의 단안을 일반적인 방법(conventional)과 오스뮴침착법(osmium impregnation)을 이용한 전자현미경적 수준에서 미세구조를 비교하였다. 오스뮴침착법에서는 세포내의 특정 소기관들이 강하게 염색되었는데, 그 중에서도 간상분체의 바로 아래에 있는 SRC가 강하게 염색되고, 복안에서와 같이 그물구조를 보이고 있었다. SSC, ER, 핵막, 색소과립 미토콘드리아 등이 강하게 오스뮴에 의해서 염색된 것이 관찰되었다. 이들 소기관들은 공통적으로 칼슘이온의 저장과 깊은 관계가 있는 것으로 알려져 있다. 결론적으로 오스뮴침착법에 의해 얻어진 강한 명암대비 및 막구조의 강조효과는 이들 칼슘저장고에 존재하는 칼슘이온과 오스뮴의 강한 결합에 의한 것으로 사료된다. 그러므로 저자들은 세포소기관들의 형태를 비교하는 데에는 오스뮴침착법이 유용한 방법이 될 것이다.

동해유입천에 서식하는 잔가시고기의 미토콘드리아 DNA를 이용한 계통분류학적 연구 (Molecular Phylogeny of Pungitius kaibarae from Mitochondrial DNA Sequences in Selected Streams Flowing into the East Sea)

  • The nucleotide sequence variations of mitochondrial DNA were investigated to understand genetic differentiation for five different Pungitius kaibarae populations at five study sites from selected four streams flowing into the East Sea. The complete sequences of mtDNA control regions of them were determined the constant sites 342 bp and variable sites 183 bp including parsimony infromative sites 122 bp. Based on the phylogenetic tree, five populations were monophyletic unit (97% MPand 100% NJ) and separated two groups (Myongpa-Songhuen stream group and others group). The population of Baebong stream (82nd bp, G-A) consistently formed them of the Jasan stream into a monophyletic unit even though it is closer then Myongpa stream (99th bp, T-C) from the Baebong stream. Further studies on the molecular phylogeny for the primary freshwater fish are needed to establish for the fish biodiversity conservation in mountainous and upland streams.
