Patterns of Utilizing Sole Carbon Source by Soil Microbes in a Forest Soil (토양 세균 군집의 유일탄소원 이용에 의한 지문분석)

  • This study was carried out utilzing ability of sole carbon sources in soil microbial communities used by Biolog GN microplate. Cluster analysis showed that soil microbial cornmuties were categorized into three groups as forest, non-forest soil and naked soil of microbial group. Soil microbial commutites in a forest soil of Qirercus mongoIica was divided into another group microbial communites in Qirercus dendata vegetation soil and Pinus dnzsqlora vegetation soil by Multidimensional scaling(MDS). Generally, sole carbon utilzing abilties were higher in order of polymer, amino acids and carboxylic acids, but it was lower in amides substrates carbon group. From the result: it was supposed that metabolic diversity of microbial communities was corresponded to vegetation succession.

PAHs로 오염된 광양만 퇴적토의 미생물 군집분석

  • 연안에 가까운 광양만 퇴적토를 대상으로 하여 PAHs 오염도와 말단제한절편 다형성 분석방법(T-RFLP method)을 이용하여 미생물 군집구조를 조사하였다. 조사된 10개 정점의 PAHs오염도는 80$\pm$20~5,690$\pm$580 (평균 1,499$\pm$1,716) ng/g dry wt.였으며 폐수종말처리장이 위치하는 하천 입구에 위치한 정점 (Wl)과 광양제철소 우수배출구 입구에 위치한 정점에서 상대적으로 높은 농도를 보였다. T-RFLP방법으로 조사한 미생물 군집은 10개 정점이 4개의 그룹으로 구분되었으며 정점 Wl에서 다른 정점과 다른 독특한 군집구조를 형성하였다. PAHs 농후배양시료에서의 미생물군집구조 변화와 비교해 볼 때 조사대상지역 퇴적토의 미생물 군집구조는 PAHs의 오염에 부분적으로는 영향을 받지만 입도, 유기물 등과 같은 환경요인이 군집구조를 결정하는 주된 요인인 것으로 생각된다.

Evaluation of Fluoride Removal Effect with Growth of Attached Microbial Community in Middle and Small Stream (중·소하천에서 부착미생물군집의 성장에 따른 불소 제거 효과 평가)

  • This study was conducted to understand the growth of attached microbial community in small and medium streams of Gyeonggi-do and the removal efficiency of fluoride with changes in growth. The sites monitoring were carried out for 12 weeks at the P1 and P2(P1 is the point where the discharge water flows. P2 is the downstream point of about 2 km flow), and investigated the water quality and the biomass analysis of the attached microbial community. The growth of the attached microbial community increased up to the 7th investigation, and it was observed that detachment phenomenon occurred. Influencing factors about growth of the attached microbial community were flow rate, velocity, and organic material(T-N & T-P). Meanwhile, fluoride content of attached microbial community also tended to increase until the 7th investigation, and decrease from the 8th. It is assumed that fluoride content has also be reduced with the detachment phenomenon of the attached microbial community. It is expected that this will contribute to the evaluation and management of the use of attached microbial community as a means of stream management. The application of techniques using the attached microbial community should include basic investigation of factors that may affect the growth of the attached microbial community and replacement of the attachment plate according to the time of removal.

Effects of Polycyclic Aromatic Hydrocarbons Contamination on the Community Structure of Microorganisms in Gwangyang bay Sediments (다환방향족 탄화수소가 광양만 퇴적토의 미생물 군집구조에 미치는 영향)

  • Effect of polycyclic aromatic hydrocarbons (PAHs) on the community structure of indigenous microorganisms in Gwangyang Bay sediments was investigated in Mar. & Aug.,2000. Microbial community structure was analyzed using 5'-terminal restriction fragment length polymorphism (T-RFLP) method. Microbial community structure based on T-RFLP method revealed that community differentiated by sampling period except station 1 located near the stream discharge site from Yeosu Industrial Complex. Even, microbial diversity was higher at stations showed relatively high concentrations of PAHs. The microbial community structure was severely changed during the enrichment culture with 1,000 ppm of PAHs mixture. It was also different between cultivated at 8$^{\circ}C$ and 30$^{\circ}C$. The results implied that temperature, poyosity, organic content and etc were more responsible than PAHs on the microbial community structure.

T-RFLP Analysis of Microbial Community Structure in Leachate from Landfill Sites (폐기물매립장 침출수내 미생물군집 구조 해석을 위한 T-RFLP의 활용)

  • Microorganisms are key-role player for stabilization of landfill sites. In order to evaluate the availability of T-RFLP(Terminal Restriction Fragment Length Polymorphism) for monitoring microbial community variations during stabilization of landfill sites, the phylogenic diversity of microbial community in the leachate from 4 different full-scale landfills was characterized by T-RFLP based on bacterial 16S rDNA. Main population of microbial community analyzed by T-RFLP was significantly similar with that of microbial community analyzed by clone library analysis. The results of T-RFLP analysis for main population of microbial community in the leachate from landfills with different landfill structures, waste types and landfill ages showed apparently different microbial diversity and structures. Therefore, long-term monitoring of microbial community in leachate from landfill sites by using T-RFLP is expected to be available for evaluation of landfill stability.

  • Since the development of the next generation sequencing (NGS) technology, 16S rRNA gene sequencing has become a major tool for microbial community analysis. Recently, human microbiome project (HMP) has been completed to identify microbes associated with human health and diseases. HMP achieved characterization of several diseases caused by bacteria, especially the ones in human gut. While human intestinal bacteria have been well characterized, little have been studied about other animal intestinal bacteria. In this study, we surveyed diversity of livestock animal fecal microbiota and discuss importance of studying fecal microbiota. Here, we report the initiation of the fecal microbiome project in South Korea.

Dynamics of Attached Microbial Community on the River with Gravel Riverbed (자갈하상 하천에서 부착미생물군집의 거동)

  • This study was carried out in Osan river to quantitatively investigate behavior of attacked microbial community (AMC) for enhancing self-purification process of river. We gained the results such as follows throughout long-term monitoring at in-situ river. The biomass of AMC had higher in the riffle than the almost stagnant pool and they were more developed in the riffle with high current velocity (HCV). Although the fast flowing current affects negatively to growth of the AMC during the early phase when the community gets attached to the benthic substrate, it was observed that it affected positively to their growth during the intermediate and later phase after the community is adapted to the substrate. When turbulence due to external pressure (storm or discharge of dam and reservoir) occurs, the degree of separation depends upon the flowing strength and the type of the external pressure. Since the community is not all separated, recovery is rather fast. Therefore, this study found that the degree of reduction of the pollutant by self-purification of the stream is depended upon the riverbed shape and the AMC contributes to self-purification positively or negatively in river. Therefore, the riverbed shape must be constructed in accordance with the characteristics of water quality in stream. Furthermore, the technique of installing the water channel structure appropriate for each section must be developed to maximize self-purification ability.

Application of Methodology for Microbial Community Analysis to Gas-Phase Biofilters (폐가스 처리용 바이오필터에 미생물 군집 분석 기법의 적용)

  • There are four key factors for gas-phase biofilters; biocatalysts(microorganisms), packing materials, design/operating techniques, and diagnosis/management techniques. Biofilter performance is significantly affected by microbial community structures as well as loading conditions. The microbial studies on biofilters are mostly performed on basis of culture-dependent methods. Recently, advanced methods have been proposed to characterize the microbial community structure in environmental samples. In this study, the physiological, biochemical and molecular methods for profiling microbial communities are reviewed, and their applicability to biofilters is discussed. Community-level physiological profile is based on the utilization capability of carbon substrate by heterotrophic community in environmental samples. Phospholipid fatty acid analysis method is based on the variability of fatty acids present in cell membranes of different microorganisms. Molecular methods using DNA directly extracted from environmental samples can be divided into "partial community DNA analysis" and "whole community DNA analysis" approaches. The former approaches consist in the analysis of PCR-amplified sequence, the genes of ribosomal operon are the most commonly used sequences. These methods include PCR fragment cloning and genetic fingerprinting such as denaturing gradient gel electrophoresis, terminal-restriction fragment length polymorphism, ribosomal intergenic spacer analysis, and random amplified polymorphic DNA. The whole community DNA analysis methods are total genomic cross-DNA hybridization, thermal denaturation and reassociation of whole extracted DNA and extracted whole DNA fractionation using density gradient.

Variation of Nitrogen Removal Efficiency and Microbial Communities Depending on Operating Conditions of a CANON Process (CANON 공정에서 운전조건에 따른 질소 제거효율 및 미생물군집 변화)

  • Nitrogen removal is one of the most important issues about wastewater treatment because nitrogen is a primary pollutant caused various problems such as eutrophication. We developed a CANON microbial community by using AOB and ANAMMOX bacteria as seeding sources. When 100 mg-N/L of influent ammonium was supplied, the DO above 0.4 mg/L showed a very low TN removal efficiency while the DO of 0.3 mg/L showed TN removal efficiency as high as 71.3%. When the influent ammonium concentration was reduced to 50 mg/L, TN removal efficiency drastically deceased. However, TN removal efficiency was recovered to above 70% after 14 day operation when the influent nitrogen concentration was changed again from 50 mg-N/L to 100 mg-N/L. According to the operating temperature from $37{\pm}1^{\circ}C$ to $20{\pm}1^{\circ}C$, TN removal efficiency also rapidly decreased but gradually increased again up to $70.0{\pm}2.6$%. The analysis of PCR-DGGE showed no substantial difference in microbial community structures under different operational conditions. This suggests that if CANON sludge is once successfully developed from a mixture of AOB and ANAMMOX bacteria, the microbial community can be stably maintained regardless of the changes in operational conditions.

Climate Effects on Greenhouse Gas Emissions and Microbial Communities in Wetlands (기후변화가 습지 내 온실기체 발생과 미생물 군집구조에 미치는 영향)

  • Global climate changes including elevated $CO_2$, drought, and global warming may influence greenhouse gas emissions in wetlands. A variety of microbial communities including denitrifiers and methanogens play a key role in determining such processes. In this paper we summarize current knowledge on the effects of climate changes on $CO_2,\;CH_4$, and $N_2O$ production and microbial communities mediating those processes in wetlands. Elevated atmospheric $CO_2$ and warming generally increase gas emissions, but effects of droughts differ with gas type and drying level. The responses of microbial community to climate changes in terms of composition, diversity and abundance are still in question due to lack of studies in wetlands. Based on the present review, it is suggested that future studies on microbial processes should consider microbial community and relationships between microbial function and structure with diverse environmental factors including climate changes. Such knowledge would be crucial to better understand and predict accurately any shifts in ecological functions of wetlands.
