Antioxidative and Antigenotoxic Activity of White and Yellow Chrysanthemum morifolium Ramat Extracts (백국과 황국 추출물의 항산화활성 및 항유전독성 효과)

  • Chrysanthemum morifolium (C. morifolium) is a perennial plant herb widely distributed in Korea and has been used in a traditional herbal remedy for various diseases. This study was conducted to determine antioxidant activities and antigenotoxic effect in water, acetone, ethanol and methanol extracts from white and yellow C. morifolium flowers (WC and YC). The antioxidants properties were evaluated on the basis of total phenolic content (TPC), DPPH radical-scavenging activity (RSA) and superoxide dismutase (SOD)-like activity. The highest TPC (5.09 g/100 g GAE) showed in YC methanol extract. The DPPH RSA activity of WC and YC water extracts increased as its concentration increase from 50 to 1000 mg/mL, respectively, and the lowest $IC_{50}$ of DPPH RAS showed in YC of $25^{\circ}C$. Also, WC solvent extracts showed significantly higher DPPH RSA than YC solvent extracts. The SOD-like activity of YC water extracts were higher than WC water extracts. And, YC acetone extract and WC methanol extract showed significantly higher SOD-like activity than WC acetone extract and YC methanol extract, respectively. The antigenotoxicity of WC and YC extracts were determined by measuring inhibitory effects of $H_2O_2$ induced DNA damage in human leukocytes using the comet assay, resulting that the ethanol extracts of WC and YC showed a significant antigenotoxic effect against oxidative stress. These results suggest that C. morifolium has significant antioxidant activity and protective effect against oxidative DNA damage.

  • The solvolysis of 1-naphthalene-and 2-naphthalenesulfonyl chlorides in acetone-water mixtures have been studied by means of conductometry. The solvent effect and mechanism have been discussed in terms of variation in solvent composition and activation parameters. The reaction was predominantly $S_N2$ type, but bond breaking increased with the increase of water content. The rate constant for 1-naphthyl compound was smaller than that of 2-naphthyl compound due to the ground state stabilization effect and peri-hydrogen effect, and the latter effect was similar to that in ethanol-water mixtures.

  • The Raman stretching frequency of nitrile group was affected by change of solvents and its in concentration in same solvent. In the case of acetonitrile, nitrile group stretching frequencies were observed in the region of 2247.3∼2254.9 cm-1 with various solvents. While in benzonitrile, they were found in the region of 2226.1∼2230.3$ cm^{-1}. With the addition of water in acetonitrile,νC≡N was shifted to high frequency from 2250.1 cm^{-1} in pure acetonitrile to 2257.7 cm^{-1} in 90% water forwhich had with higher volume % of water caused higher hydrogen-bonded equilibrium between methyl protons and water. The νC≡N frequency for nitrile group was shifted to high frequency by solvent inductive effect with the increasing mixed solvent (CHCl_3/CCl_4)$mole% ratio.

다양한 용매 속 NaCl 의 분자 동역학 시뮬레이션 : Umbrella sampling 을 이용한 Potential of Mean Force 계산

  • 용매화 된 이온들간의 상호작용은 화학을 비롯한 여러 분야에서 중요하게 다뤄지고 있다. 계산화학에 있어 다양한 용매에 용매화 된 이온들간의 상호작용은 분자동력학 시뮬레이션(MD)을 이용한 PMF 계산을 통해 많이 연구되어 왔는데, 본 그룹에서는 물을 표현할 수 있는 다양한 모델과 Salt solution 하에 Na+ 이온과 Cl- 이온 사이 상호작용을 PMF 계산을 통해 알아보았다. 첫 번째로 Polarizable water model인 Tip4p-fq를 사용한 PMF 계산 결과는 아직 진행 중에 있으며 결과적으로는 기본적으로 CHARMM water force field에서 제공해주는 Tip3p 모델 및 양자계산 방법 중 하나인 Ab initio를 이용한 결과와 비교, 분석 할 예정이다. 두 번째로 1M/2M/5M Salt solution의 서로 다른 농도를 사용하여 시뮬레이션 한 결과 1M과 2M에서는 물 속에서 시뮬레이션 한 결과와 큰 차이가 없었으나 농도가 짙은 5M 용매 내에선 $Na^+$이온과 $Cl^-$이온 사이 거리가 멀어질수록 주변 Salt에 영향을 받는 것을 확인 할 수 있었다. 결론적으로 본 연구를 진행함으로써 우리는 서로 다른 전하를 갖는 이온 사이 상호작용에 대한 이해를 더 높일 수 있었다.

  • Effects of modifier and soaking on extraction of triterpenoid saponin (glycyrrhizin) from liquorice were examined using supercritical $CO_2(SC-CO_2)$ at 50 MPa, $60^{\circ}C$, and flow rate of 3 mL/min, and glycyrrhizin content was analyzed by HPLC. Additon of undiluted methanol, ethanol or isopropanol as modifier to $SC-CO_2$ had little influence on extraction yield of glycyrrhizin. Soaking process using water increased the extraction yield as the sample to solvent ratio was increased. Addition of 70% methanol, ethanol or isopropanol to $SC-CO_2$ significantly increased the extraction yields, with 70% methanol resulting in the highest yield. When water at 90% (w/w) of sample weight was used for soaking, the extraction yield and rate increased, 70% ethanol-modified $SC-CO_2$ was almost equal to that obtained using 70% methanol.

  • The enzymatic hydrolysis of Castor oil for the mass production of ricinoleic acid was studied to find out the optimum conditions such as solvents and the weight ratio of substrate to enzyme. Three different lipases were tested for the hydrolysis of castor oil: lipase from Porcine Pancrease(lipsase PP), lipase from Candida cylindracea(lipase CC), lipase from Candida Rugosa(lipase CR). The poor mass transfer in water caused a low degree of hydrolysis of castor oil. To overcome this problem, organic solvents were used. Among organic solvents tested, hydrophobic solvents gave better results of hydrolysis than hydrophilic solvents. Organic solvents also lowered or changed the effect of pH. Isopropyl ether made complete hydrolysis of castor oil. The ratio of water to isopropyl ether and the ratio of weight ratio of lipase to castor oil were important for the hydrolysis of castor oil. At 30$^{\circ}C$ castor oil was completely hydrolyzed by 4 wt% of lipase in the mixture of isopropyl ether and water(1:1 in volume).

  • The electrochemical reduction of iodine in non-aqueous media have been studied by polarography, chronopotentiometry, cyclic voltammetry and controlled potential coulometry at dropping mercury electrode and platinum, gold and amalgamated platium electrodes. In amphiprotic solvents such as methanol, ethanol, isopropanol and pyridine, iodine were reduced to iodide ions via one step reduction involving 1 electron and in aprotic solvents such as acetonitrile, dimethylformamide and dimethylsulfoxide via two step reduction involving all 3 electrons. The reductions of iodine give well defined polarograms at dropping mercury electrode and irreversible chronopotentiograms at platinum electrode, but less defined irreversible chronopotentiograms at gold and amalgamated platinum electrodes, those are all diffusion controlled. The diffusion coefficients of iodine in various solvents were estimated from the chronopotentiometric data and Sand equation.

  • To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.

  • The rates of solvolysis for 2-thenoyl chloride have been measured in aqueous acetone and aqueous ethanol at various temperatures ranging from 20 to $40^{circ}C$. The activation parameters and the Grundwald-Winstein's slope are determined by the analysis of solvolysis rates. The results indicated that the reaction rates of solvolysis are considerably slower than those of the reaction for benzoyl chloride due to the electron donating effect of thiophene nucleus. The results also showed that the reaction proceeds with the $S_N1$ mechanism in water-rich solvents whereas the $S_N2$ character increases with the decrease of water content, and overall reaction is subject to entropy control.

공용매 첨가가 비대칭 기체 분리막의 구조 및 투과도에 미치는 영향

  • 비대칭 분리막의 제조에 있어서는 용매와 비용매의 교환에 의한 상분리 현상을 이용하기 때문에 용매와 비용매의 성질이 막 제조 공정에 중요한 변수가 된다. 고분자 용액에 공용매를 첨가할 경우 용액의 점도, 용매와 비용매의 상호 작용 정도 등의 용액의 성질을 변화시킬 수 있다. 따라서 이들의 변화에 따른 분리막의 구조 변화를 관찰하여 그 상관 관계를 규명하면 분리막의 미세 구조 조절 및 제어가 가능할 것이다. 본 실험에서는 폴리이미드(PI)를 고분자로 사용하였고, 용매로는 NMP, 공용매로는 $\gamma$-butyrolactone($\gamma$-BL)을 사용하여 고분자의 농도가 15 wt%인 용액을 제조하였다. 제조한 용액을 유리판에 균일한 두께로 casting 한 후 물을 비용매로 사용하여 immersion precipitation 방법으로 막을 제조하였다. 제조한 막은 24시간 동안 물 속에 방치하여 용매를 충분히 제거한 후 상온에서 24시간 이상 건조시켰다. 투과도 측정은 soap bubble flow 방법으로 시행하였으며 단면 구조는 주사전자 현미경을 사용하여 관찰하였다.
