Antimicrobial activity of Specific IgY against Streptococcus mutans (Streptococcus mutans에 대한 specific IgY의 항균력)

  • Antimicrobial effects of the specific IgY separated from eggs which were laid by hens vaccinated with Streptococcus mutans were investigated. The comparison tests of vaccination, addition levels of crude specific IgY, and innoculation concentration were applied by microscopic observation and turbidity test. Ten% addition of crude specific IgY obtained from vaccinated hens showed agglomerative clusters of S. mutans cells in supernatants and sediments, while crude IgY produced by non-vaccinated hens showed no cluster. IgY addition above 5% showed agglutinating clusters of most S. mutans cells and there was definite difference between IgY addition below 2.5% and above 5%. Concentration tests of crude IgY revealed that antimicrobial effects were differentiated by addition level and addition over 10% produced satisfactory results with turbidity test. The cluster size was dependent upon concentration of crude IgY addition. $10^5\;cfu/mL$ inoculation showed agglutinated cells and extent of agglomeration was proportional to cell numbers. Study of inoculation levels showed that 10% addition of crude IgY decreased turbidity effectively regardless of number of S. mutans cells. Plaque formation decreased to 75% with 15% addition of specific IgY concentration. These results implied that IgY separated from eggs laid by S. mutans-vaccinated hens might prevent dental caries caused by S. mutans.

Volatile Flavor Components in Cooked Black Rice (취반된 흑미의 휘발성 향기 성분)

  • Volatile flavor components (VFCs) in cooked black rices (Suwon-415 and Chindo) were studied. The major reactions during cooking, which result in aroma volatiles, are the Maillard reaction between amino acids and reducing sugars, and thermal degradation of lipid. Black rices washed with water were soaked in 1.5 folds water and heated at $110^{\circ}C$ in oil bath for 30min. VFCs in cooked black rices were extracted for three hours by SDE and were analyzed by GC and GC/MS. A total of 91, 82 volatiles were identified in Suwon-415 and Chindo black rice, respectively. Suwon-415 was composed of 26 alcohols, 10 aldehydes, 5 acids, 11 esters, 15 ketones, 9 hydrocarbons, 3 furans, 3 nitrogen containing compounds and 9 sulfur containing compounds. Chindo was composed of 28 alcohols, 9 aldehydes, 4 acids, 12 esters, 14 ketones, 5 hydrocarbons, 3 furans, 3 nitrogen containing compounds and 4 sulfur containing compounds.

Characteristics of bitter peptides from Doenjang (된장의 쓴맛 펩타이드 특성)

  • Conventional Doenjang, three kinds of Improved Doenjang(A; koji: soybean: salt=53: 100: 33, B; koji: soybean: salt=100: 100: 40, C; koji: soybean: salt= 200: 100: 40) prepared with different ratio of koji and salt were made to study the changes in the general contents, characteristics bitter peptides, correlations between bitterness and overall eating quality. 1. Total nitrogen contents increased a little, and amino nitrogen contents in all samples increase markedly. Especially, Amino nitrogen contents of conventrional Doenjang increased more than others. Reducing sugars of doenjang prepared with Asp. oryzae were higher than conventional Doenjang and increased throughout the aging period and Doenjang prepared with Asp. oryzae were more acidic. 2. To characterize bitter peptides in fermented Doenjang, peptides were extracted with 2: 1(v/v) chloroform-methanol and separated by Gel chromato-graphy with Sephadex and TLC. After Gel chromatography and TLC, each fraction examined presence of bitterness and evaluated intensity of bitterness. Amino acid composition of the fractions showing bitter tastes were as follows. Conv. peak 1-1 Trp-(Asp, Arg, Thr, Ser, Glu, Pro)-Phe Imp. A peak 1 Trp-(Glu, Val, Arg, lie)-Phe Imp. B peak 1 Trp-(lie, Pro, Asp, Lys, Val, Glu)-Trp. Imp. C peak 1-2 Trp-(Try, Thr, Glu, Pro, Gly)-Phe 3. Sensory evaluation revealed that correlation coefficient between bitterness and overall eating guality was not high.

Whitening Effects of Angelica dahurica Radix Ethanol Extract (백지 에탄올추출물의 미백효능 연구)

  • To investigate whitening Effects of Angelica dahurica Radix Ethanol Extract (ADEE), we used melan-a cell line, brown guinea pig, and HMB-45. We treated with ADEE of 6.25, 12.5, 25, and 50 ${\mu}g/m{\ell}$ concentration in order to evaluate the effect of ADEE on cell viability and on morphological observation of melan-a cells. Also we were induced the artificial tanning spots by 1,500 mJ/$cm^2$ of ultraviolet B radiation on the backs of brown guinea pigs (approximately 450~500g) and then the test agent of $30{\mu}{\ell}$ was applied on the spots twice a day, five days a week, for five weeks respectively. The visible whitening effect was evaluated once a week. At the end of the experiment, the animals were sacrificed under anesthetization. The artificial tanning spots were obtained by biopsy punch and stained with HMB-45 to observe the gp100 proteins which were melanosomes. Our results show that cell viability was not reduce at ADEE concentrations between 6.25 and 50 ${\mu}g/m{\ell}$, melanin synthesis and melanocyte dendricity were decreased in ADEE treated melan-a cells increasing ADEE concentration. In the gross observation, ADEE treated groups had lower pigmentation than the vehicle control groups. And in the histological observation, ADEE treated groups had lower melanocytes than the vehicle control groups. Also in the quantitative analysis of the gp100 proteins using image analysis software, ADEE treated groups had a significantly lower value (p<0.001) than the vehicle control group and this resultsagreed with the results of observation under microscope. From these results, weconcluded that ADEE had positive whitening effect.

A Basic Study on BIM Library Business Model based on Building Material Information System (건축 자재 정보 시스템기반 BIM 라이브러리 비즈니스 모델에 관한 기초 연구)

  • In recent years, the use of BIM in domestic construction has been increasing, but it is used mainly for internal and external visualization rather than for BIM design including drawings. For this reason, several studies have highlighted the lack of libraries and economic problems for BIM design. In other words, for the most economically small domestic design offices, activating the BIM design is a negative factor because it is difficult to invest the necessary manpower and cost to produce and use the required BIM library. The BIM design can be more applicable if the library can be supplied economically, easily, and quickly. In this study, BIM library business model and consideration factors that can provide BIM library service economically, easily, and quickly by considering the existing building material selection task and BIM library are presented. The proposed business model will increase the number of suppliers of BIM libraries, which are lacking in Korea, and will enable an effective BIM design at low cost and effort. If a service that reflects the business model proposed in this study is made, an integrated database that can consistently share information in the design, construction, and maintenance stages will be constructed. This can monitor the changes in material and equipment information during the life cycle of the building. The database can be used to monitor any changes in material and equipment information throughout the life cycle of a building, so that it can be used as historical data for effective design and maintenance as well as for material and equipment upgrades.

EFFECT OF INCREASING APPLICATION TIME OF SINGLE BOTTLE ADHESIVES TO MICROTENSILE BOND STRENGTH OF DRIED DENTIN (수종의 단일병 접착제의 적용 시간 연장이 건조 상아질의 미세인장 결합강도에 미치는 영향)

  • The purpose of this study was to evaluate the effect of increasing application time of single bottle adhesives (SBA) to microtensile bond strength (MTBS) of dried dentin. To expose the superficial dentin surfaces, human molars were sectioned perpendicular to the long axis of tooth. $32\%$ phosphoric acid gels were applied for 15s and rinsed. The teeth were randomly assigned to 3 groups ; S group (Single Bond), O group (One-Step), P group (Prime & Bond NT). Each group was divided to 3 subgroups (W: dentin wipe with wet gauge and light cured immediately, D, dentin dried for 30s and light cured immediately, 30: dentin dried for 30s and light cured after applying SBA for 30s). Composite resin was built up on the dentin surface and sectioned to obtain 20 specimens with $1mm^2$ cross sectional area and the MTBS was measured. For Single Bond, the mean MTBS of S-W and S-30 group were higher than that of S-D group statistically (P<0.05). For One-Step, the mean MTBS of O-D group was statistically lower than that of O-W group (P<0.05). For Prime & Bond NT, the mean MTBS of P-30 group was statistically lower than that of P-D group (P<0.05).

  • Calcium hydroxide is used as a root canal medicament with its several pharmacological effects. However, it has been known that the usage of calcium hydroxide in the root canal system before canal filling with gutta-percha and zinc oxide eugenol-based cement induced change in the properties of root canal cement which might adversely affect sealing ability of the canal filling. The purpose of this study was to identify the reactivity of calcium hydroxide-eugenol compound made from chemical interaction of between calcium hydroxide and zinc oxide eugenol. Chemical properties of calcium hydroxide, eugenol, zinc oxide eugenol, calcium hydroxide-eugenol and calcium hydroxide-zinc oxide eugenol compound were analyzed using XRD. FT-IR Spectrophotometer and FT-NMR Spectrometer. The results were as follows: 1. The compound made from interaction between calcium hydroxide and zinc oxide eugenol was as follows : 2. Calcium hydroxide was shown to make chemical bond (ionic bond) with eugenol. 3. Since bonding between $Ca^{2+}$ and eugenol is simple ionic nature, under water existence, calcium hydroxide-eugenol compound may be ionized easily and its physical property be deteriorated.

EFFECT OF FIBER DIRECTION ON THE POLYMERIZATION SHRINKAGE OF FIBER-REINFORCED COMPOSITES (섬유 보강 복합레진의 섬유 방향이 중합수축에 미치는 영향)

  • The aim of this study was to evaluate the effect of fiber direction on the polymerization shrinkage of fiber-reinforced composite. The disc-shaped flowable composite specimens (d = 10 mm, h = 2 mm, Aeliteflo A2, Bisco, Inc., IL, USA) with or without glass fiber bundle (X-80821P Glass Fiber, Bisco, Inc., IL, USA) inside were prepared, and the longitudinal and transversal polymerization shrinkage of the specimens on radial plane were measured with strain gages (Linear S-series 350${\Omega}$, CAS, Seoul, Korea). In order to measure the free polymerization shrinkage of the flowable composite itself, the disc-shaped specimens (d = 7 mm, h = 1 mm) without fiber were prepared, and the axial shrinkage was measured with an LVDT (linear variable differential transformer) displacement sensor. The cross-section of the polymerized specimens was observed with a scanning electron microscope to examine the arrangement of the fiber bundle in composite. The mean polymerization shrinkage value of each specimen group was analyzed with ANOVA and Scheffe post-hoc test (${\alpha}$=0.05). The radial polymerization shrinkage of fiber-reinforced composite was decreased in the longitudinal direction of fiber, but increased in the transversal direction of fiber (p<0.05). We can conclude that the polymerization shrinkage of fiber-reinforced composite splint or restoratives is dependent on the direction of fiber.

Study of endodontic working length of Korean posterior teeth (한국인의 구치부 근관작업장에 관한 연구)

  • Objectives: The aim of this study was to investigate average working lengths of Korean posterior teeth and evaluate validity of endodontic file length. Materials and Methods: The endodontic working length of the posterior teeth of 670 Korean patients were measured than each mean value and standard deviation were investigated than the frequency deviation and standard deviation per each length were calculated. Results: Among the canals of premolar, 66.5% of canal length was marked under 20 mm by endodontic working length and 95.4% could be measured under 22 mm and Among the canals of molars, 95.5% of canal length was marked under 20 mm endodontic working length. Conclusions: With the result of measurement of endodontic working length of premolars of Korean, it suggested that 23 mm endodontic file is more proper than the 21 mm and 25 mm file on the market.

Construction of Component Repository for Supporting the CBD Process (CBD 프로세스 지원을 위한 컴포넌트 저장소의 구축)

  • CBD(Component Based Development) has become the best strategical method for the business application. Because CBD is a new development paradigm which makes it possible to assemble the software components for application, it copes with the rapid challenge of business process and meets the increasing requirements for productivity. Since the business process is rapidly changing, CBD technology is the promising way to solve the productivity. Especially, the repository is the most important part for the development, distribution and reuse of components. In component repository, we can store and manage the related work-products produced at each step of component development as well as component itself. In this paper, we suggested a practical approach for repository construction to support and realize the CBD process and developed the CRMS(Component Repository Management System) as implementation product of the proposed techniques. CRMS can manage a variety of component products based on component architecture, and help software developers to search a candidate component for their project and to understand a variety of information for the component. In the paper, a practical approach for component repository was suggested, and a supporting environment was constructed to make CBD to be working efficiently. We expect this work wall be valuable research for component repository and the entire supporting Component Based Development Process.
