The Asian ladybird, Harmonia axyridis, might be distinguished in biological control agents for the several aphid species in the greenhouses as well as in the fields. Therefore, feeding behavior and predacious capability characteristics were observed. When ladybirds are searching for aphids as their preys, they use the maxillary palpus as contact reception. Otherwise, most of aphids escape from walking or chewing ladybird to other place (mainly move to near-by leaf, or fall down to the under-leaf or ground) instead of plant sucking. Consumed time of ladybirds for searching and feeding prey were significantly different and dependent on their development stage. The ladybird adults ate about 120 green peach aphids and 257 cotton aphids per day. The 3rd instar of ladybirds ate 2,420 and 2,750 eggs of greenhouse whitefly on the cucumber and tomato leaf, respectively, as well. However, ladybird instars taken greenhouse whitefly eggs could not continuously develope to adult, on the other hand, ladybirds taken aphids were normal condition.
Acid phosphatase(AP) of he aphid, Megoura crassicauda and the major component of the lady beetle's artificial diet, fresh chicken liver, was adapted as a model protein to study the digestion of diet proteins in the midgut of Harmonia axyridis. The lady beetle did not secrete its own AP into the lumen of the midgut. The aphid and the live chicken liver had AP which was still alive in enzymatic activity from the extract of the lumen of the midgut of the lady beetle. The digestive ability of the lady beetle on proteins turned out to be different depending on food sources. In the lumen of the midgut of the lady beetle, though most of AP of live chicken liver lost its activity withtin 12 hours, that of M. cassicauda kept strong enzymatic activity up to 24 hours.
The aphidophagous lady beetle, Harmonia axyridis, is induced to undergo reproductive diapause by low temperature treatment. The diapause of the adult beetle can be maintained without supplying the diet up to 5 months at $4^{\circ}C$. The diapause can be induced any time including during the summer, allowing the beetle to be supplied to the field for the control of aphids through the year. Fecundity and longevity after the reproductive diapause is though to be satisfactory enough for the control of aphids in the field, Methoprene, an analog of reproductive hormone of JH, increases the early fecundity of the beetle.
Application of the Asian ladybird (Harmonia axyridis) to control several species of aphids in the plastic green houses in mind, control effects of aphid populations regulated by the Asian ladybird were observed. The green peach aphid, the turnip aphid, and the cotton aphid were present on mustard plants, Angelia utlis, ornamental kales, and egg plants at greenhouses in spring, summer, and winter. Adults and larvae of the Asian ladybird used in experiments were collected from aggregated sites at Taejon in the autumn and reared on the cotton aphid in the laboratory. In winter, more number of adults and larvae of ladybirds than in other seasons were needed to control aphid population in successively double plastic greenhouses with supplied subterranean water for keeping warmth. In spring and summer, it was possible to keep the aphid populations low when necessary by manipulating ladybird populations according to the density of aphids. On the other hand, the innate increasing rate of aphid, the aphid population density at the time of applying ladybird, the predacious ability of ladybird at specific developmental stages, and needed periods should be taken into account to control aphids. In addition, the environmental factors, for example, optimum temperature and humidity should be considered to be biologically effective when ladybirds are released to greenhouses.
This experiment was conducted to select the selective aphicides to Myzus persicae adults and its predator, Harmonia axyridis adults. The effects of the selective aphicides on the longevity, fecundity and hatchability of predaror were examined. All experiments were tested at the recommended concentration of those aphicides. Mortalities of adult H. axyridis to pirimicarb (162.5 ppm), fenvalerate (50 ppm) and endosulfan (577.5ppm) were shown 0, 20 and 22.5%, respectively. Endosulfan and fenvalerate showed the mortality above 48% to eggs and larvae H. axyridis, but pirimicarb was not toxic to those stages. Pirimicarb did not affect to the longevity, fecundity and hatchability of H. axyridis, but endosulfan and fenvalerate severely affected to the predators. From these results, pirimicarb can be used in biological controls for M. persicae.
Enterobacteria were isolated in the gut of the predacious multicolored Asian ladybird beetle, Harmonia axyridis, and their effects to the development of H. axyridis were examined. Populations of H. axyridis in this experiment were collected from Kimjae at Cheonbuk province (JK population), Geumsan at Chungnam province (CK population) and laboratory population at Laboratory of Insect Physiology in Chungnam National University, Daejeon. Thirty-four enterobacteria isolates were purified and isolated from the digestive tract of H. axyridis, and a total of 4 strains were classified into group by analysis of 16S rRNA gene sequences. About 70% of total isolates were phylogenetic groups of Bacillus genus and Staphylococcus genus, and they were commonly separated from the digestive tract of H. axyridis. After investigating their susceptibility against antibiotics with 18 representative enterobacteria isolates, ofloxacin and penicillin were selected for examination in this study of their ability to inhibit the growth of all of isolates. In order to remove the enterobacteria from the aphids, ofloxacin and penicillin were given to the green peach aphid, Myzus persicae, and the turnip aphid, Lipaphis erysimi. These aphids were provided to H. axyridis as prey. The weight of pupa, developmental periods of each larval instar, the number of eggs and their hatching ratio of H. axyridis with treatment aphids were lower compared with non-treatment aphids. Staphylococcus saprophyticus is a representative enterobacteria and commonly isolated from the digestive tract of H. axyridis. In the absence of S. saprophyticus, the developmental periods of each larval instar increased; however, the weights of pupa, the number of eggs, and their hatching ratio decreased.
This study was conducted to select the acaricides selective to H. axyridis and examine the effects on phytophagous mites and natural enemies after application of selected acaricides in the apple orchard. All the acaricides tested were more toxic to T. urticae and P. ulmi than to H. axyridis, and also were more toxic to egg than to the larva and adult of H. myridis. Azocyclotin and fenpropathrin were the highest and lowest selective acaricides to H. axyridk, respectively. The density of phytophagous mites was high affected by frequency of acaricide application. Significant difference in the changes of the density of natural enemies was not be observed according to frequency of acaricide application and kinds of acaricide. Acaricide treatment showed high effect on the population densities of A. womersleyi and Oriur spp., while less effect on Chrysopa spp.

Bacterial density distributions of gut microbes in the digestive organs of Harmonia axyridis collected from three different sources (JK, CK, and CJ) were $6.0{\times}10^4$ CFU/gut under aerobic culture condition and $8.0{\times}10^6$ CFU/gut under anaerobic culture condition. Seven colony types were observed under aerobic condition and three types of similarity were detected under anaerobic condition. In total, 116 strains, including 34 strains under aerobic condition, were isolated from the digestive organs of H. axyridis. Based on the analysis of the 16S rRNA gene sequence, aerobic gut microbes were assigned to the Proteobacteria, Actinobacteria, Firmicutes, and Deinococcus-Thermus. A large number of isolates belonged to the genus Bacillus and Staphylococcus of the Firmicutes commonly found in H. axyridis from different sites. Anaerobic gut microbes were found to be similar according to colony morphological, phylogenetic analysis using ARDRA. Eighty-two anaerobic gut microbes were clustered into 17 different ARDRA types according to HaeIII. Representative anaerobic gut microbes in each ARDRA group were divided into five species of ${\gamma}$-Proteobacteria based on 16S rRNA gene sequence analysis; Hafnia alvei, Enterobacter ludwigii, Enterobacter kobei, Pseudomonas oryzihabitans and Pseudomonas koreensis. Phylogenetic analysis indicated that about 70% of the isolates belonged to ${\gamma}$-Proteobacteria, suggesting predominance of gut microbes.

Elytra of Harmonia axyridis exhibit varied color patterns. In the present study, we deciphered the genetic basis for intraspecific diversity of elytra color patterns in H. axyridis, using amplified fragment length polymorphism (AFLP). Twenty-eight AFLP reactions were performed to generate a total of 2,741 bands. Of these, 20 bands were polymorphic for each color pattern. The polymorphic bands showed differences of genetic character among different color patterns of H. axyridis. Among them, ten candidate AFLP markers were color-linked. S1, S2, and S20 markers were detected in Succinea 1 and 2 variants of H. axyridis, whereas S3 and S5 were specifically detected in the Conspicua variant. S15, S18, and S19 were specific to the Succinea 2 variant. Polymerase chain reaction (PCR) products of these ten AFLP markers were sequenced. BLAST analysis of these sequences against the GenBank database revealed their homology to DNA fragments of unknown function. Based on the color-linked AFLP markers, sequence characterized amplified region (SCAR) markers were designed for PCR amplification of genomic DNA. Of the ten AFLP markers, five were successfully converted into SCAR markers, which could discriminate elytra color polymorphism in H. axyridis.
Esterase activity was observed after pesticides treatment in eggs of H. axyridis to select low toxicity pesticide. Egg esterases of H. axyridis were examined using an esterase substrate(${\alpha}$-naphthyl acetate). Three esterase isozymes were detected and the activities were inhibited by organophosphorus insecticide (Chlorpyrifos and Phenthoate), organochlorine insecticide(Methidation), triazole fungicide(Hexaconazole and Triflumizole), and pyrimidine fungicide(Nuarlmol). Fecundity and hatchability in adults and eggs of H. axyridis were examined on selected pesticides. Fecundity and hatchability were significantly reduced from H. axyridis adults and eggs treated with the pesticides and the fungicides showed strong inhibition of esterase isozymes activities. However, we also observed the pesticides and the fungicides showed low or non-inhibition of esterase isozymes activities affected on fecundity and hatchability in adults and eggs.
