Ontogeny of Haustorial Xylem in Parasitic Angiosperm Cuscuta austrailis R. Brown (기생피자식물 실새삼(Cuscuta australis R. Brown)의 흡기내 목부의 개체발생)

  • 기주식물 (토끼풀, Trifolium repens L.)에 기생하는 실새삼(Cuscuta australis R. Brown)의 흡기내 목부의 개체발생 과정을 광학현미경으로 조사하였다. 흡기의 목부 분화를 암시하는 최초의 해부학적 특징이, 흡기 자체내에서가 아니라, 그 흡기가 형성되어 있는 실새삼 줄기에서 나타났다. 즉 실새삼 줄기의 중심주의 유관속 내부 및 유관속 사이에서 세포분열 활성이 관찰되었다. 이 분열로부터 유도된 세포들은 짙게 염색된 세포질과 뚜렷한 핵을 갖고 있었다. 흡기가 기주조직에 침입하여 생장함에 따라, 실새삼 줄기의 중심주에서 시작된 세포분열 활성은 유관속 사이를 거쳐서 흡기의 기부를 향하여 구정적으로 확장되었다. 이와 동시에, 실새삼 줄기의 목부에 인접해 있던, 짙은 세포질 밀도를 갖는 세포들이 목부요소로 분화하기 시작하였다. 결국, 이미 형성되었던 짙게 염색된 세포들을 따라서 흡기의 목부는 구정적으로 분화하였다. 실새삼 줄기의 중심주에서 일어나는 유조직 세포들의 분열활성 및 이로부터 흡기의 목부 분화를 구정적으로 유도할 수 있는 가능성에 대하여 논의하였다.

  • 대마에 대한 분석 결과, 인피 섬유의 경우 리그닌 함량은 7.6%로 다른 목질계 바이오매스나 일년생 초본류와 비교하여 매우 낮은 리그닌 함량을 나타냈으며 탄수화물 함량은 65.4%로 목질계 바이오매스와 유사하고, 초본류보다는 오히려 높았다. 목부는 리그닌 및 탄수화물 함량이 활엽수와 유사한 경향을 나타내었는데 특히 높은 자일란의 함량이 확인되었다. 또한 회분 함량이 인피섬유와 목부에서 각각 5.0%와 2.3%로 낮은 값을 나타내어 당화 및 발효 공정에 적용될 때 회분에 의한 공정 장애(스케일링 등)의 가능성을 낮추어 줄 것으로 사료되었다. 최종적으로 이러한 화학적 분석을 통해 대체에너지 생산을 위한 자원으로서 대마의 가능성을 확인할 수 있었다.

Contents of Mineral Elements and Cytokinins in Xylem Sap of Two Oriental Melon Cultivars Affected by Rootstocks (참외품종과 대목종류에 따른 목부액 내의 무기성분 및 시토키닌 함량)

  • Contents of mineral element and cytokinin in the xylem sap of 'Keumdongee' and 'Tongilhwang' oriental melons were compared with those in oriental melons grafted onto 8 rootstocks. The effect of grafting on the fruit quality of oriental melon was also investigated. Flesh firmness varied with rootstocks. Soluble solids contents in the placenta tissue of grafted 'Tongilhwang' were higher than that in the 'Keumdongee'. Electric conductivity of the xylem sap in own-rooted plants was higher in 'Keumdongee' than in 'Tongilhwang', but it increased in 'Tongilhwang' once they were grafted. The sap volume per plant was greater in 'Keumdongee' than in 'Tongilhwang'. The mineral concentrations varied considerably depending on the rootstock used. Xylem sap of grafted oriental melons contained a higher amount of mineral ions, especially $NO_3{^-}$ and $PO_4{^-}$, than did the sap in own-rooted plants. The increase in the mineral levels in sap due to grafting was most apparent in 'Tongilhwang'. Xylem sap from both 'Keumdongee' and 'Tongilhwang' contained trans-zeatin (t-Z), trans-zeatin riboside (t-ZR), and dihydrozeatin riboside (DHZR). Small amounts of isopentenyl adenine (IPA) and isopentenyl adenine riboside (IPAR) were also detected. Trans-zeatin riboside was the most abundant, followed by t-Z. Cytokinin concentration in 'Keumdongee' was not significantly influenced by rootstock type used, although the highest concentration of cytokinins in 'Keumdongee' was obtained with 'Chamtozwa' rootstock. However, the cytokinin concentration in 'Tongilhwang' increased with grafting irrespective of rootstock type used.

Effect of Cucurbitaceous Crops and Rootstocks on the Xylem Sap, Amount Mineral Element and Plant Hormone (박과채소의 대목에 따른 복부분비액, 무기성분 및 식물호르몬의 함량)

  • Experiments were conducted to investigate the bleeding xylem sap characteristics, mineral contents and endogenous plant hormon contents in cucumbeer and origental melen plant grafted onto different rootstocks and various cucubitaceous crops developed for rootstocks. The volume of xylem sap, expressed as milliliter per 100g plant flesh weight, was high in the Heukzong rootstock plant as compared to the other rootstock plant,. Acidity(pH) and electric conductivity(EC) values in breeding xylem sap were not much influenced by the kind of rootstocks. Incucumber plant grafted onto different rootstocks, the contents of xylem sap in total nitrogen, phosphorus and K were increased by grafting, whereas those of Ca and Mg were decreased. Rootstocks had no influence on the total nitrogen contents in xylen sap of ‘Sineuncheon’, but decreased K Contents. Conecntration of cytokinin-like substances in origental melon xylem sap was a little higher in Sintozwa and ungrafted.

Fungicidal Activity of Domestic Plant Extracts against Six Major Phytopathogenic Fungi (국내산 식물체 추출물의 여섯 가지 주요 식물병원권에 대한 살균활성)

  • Methanol extracts from 207 samples of 118 plant species in 44 families were tested for their fungicidal activities against six phytopathogenic fungi. Extracts of Thuja orientalis leaf, Cinnamomum loureirii leaf, Lindera erythrocarpa barks and leaf, Pinus koraiensis wood, Hovenia dulcis wood, Koelreuteria paniculata barks, Styrax japonica wood, Camelia japonica leaf and Cleyera japonica leaf showed very strong fungicidal activity against more than two phytopathogenic fungi at a concentration of 2000 ppm. As a naturally occurring fungicide, these plants could be useful as new fungicidal products against various plant diseases induced by plant pathogens.

Anti-Inflammatory Effect of Essential Oils Extracted from Wood of Four Coniferous Tree Species (침엽수 4종 목부 정유의 항염증 효과 평가)

  • The aim of this study was to evaluate the anti-inflammatory effects of essential oils extracted from the wood of Chamaecyparis obtusa, Pinus densiflora, Pinus koraiensis, and Larix kaempferi. Essential oils were extracted by hydrodistillation, and their chemical components were determined by GC/MS. Major chemical components of these essential oils were ${\alpha}$-cadinol (19.25%), ${\tau}$-muurolol (14.20%), and ${\alpha}$-pinene (13.74%) in C. obtusa; ${\alpha}$-pinene (47.16%), longifolene (14.31%), ${\beta}$-phellandrene (11.78%), and ${\beta}$-pinene (11.02%) in P. densiflora; ${\alpha}$-pinene (13.49%) and longifolene (10.79%) in P. koraiensis, and geranyl linalool (23.58%) and ${\alpha}$-pinene (18.57%) in L. kaempferi. To evaluate the anti-inflammatory effects of essential oils, lipopolysaccharide (LPS)-induced RBL-2H3 mast cells were treated with these essential oils; then, the changes in the mRNA expression level of the cytokines IL-4 and IL-13 were examined. Further, degranulation was evaluated by measuring ${\beta}$-hexosaminidase release. After LPS-induced RBL-2H3 mast cells were exposed to $10^{-7}%$ of all types of essential oils, the gene expression levels of IL-4 and IL-13 within the cells remarkably decreased. The relative mRNA expression level of IL-4 was 69.6% in P. densiflora, 63.2% in P. koraiensis, 55.1% in C. obtusa, and 45.8% in L. kaempferi compared with that in the group treated with LPS. The mRNA expression level of L-13 should a similar trend. The inhibitory rate of IL-13 mRNA expression of P. densiflora, P. koraiensis, C. obtusa, and L. kaempferi was 57.8%, 57.1%, 51.1%, and 34.5%, respectively. ${\beta}$-Hexosaminidase release significantly decreased following the treatment with the four types of essential oils. The rate of ${\beta}$-hexosaminidase release were 38.1% C. obtusa; 33.0% P. densiflora; 27.4% P. koraiensis; and 9.1% L. kaempferi. Among all types of essential oils, that extracted from P. densiflora wood showed the highest anti-inflammatory activity. These results show that the tested essential oils exert an anti-inflammatory effect through the inhibition of degranulation and expression of cytokines.

Comparative Anatomy of the Secondary Xylem in the Root and Stem of Some Korean Betulaceae (수종의 한국산 자작나무과 식물에 있어서 뿌리와 줄기의 이기목부의 비교해부)

  • A comparative anatomy between the secondary xylem in the root and stem of Korean Betulaceae, including 5 genera and 6 species, was carried out in this study. Anatomical characteristics of the secondary xylem in the root and stem are as follows: Diameter of vessel and fiber is wider in the root than the stem, while the number of vessel and fiber per unit area is fewer in the root than the stem. The length of vessel element is longer in the stem than the root, whereas length of the fiber is longer in the root than the stem. Number of bar in the perforation plate is more in the stem than the root, and the angle of perforation plate is broader in the root than the stem. Number of ray per unit area is more in the root than the stem.

Anatomical Characteristics of Kenaf Grown in Reclaimed Land - Volumetric Composition and Cell Dimension - (간척지에서 재배된 양마(kenaf)의 해부학적 특성(II) - 구성 비율 및 세포의 치수 -)

  • Anatomical characteristics of volumetric composition, fiber length, vessel diameter, and crystalline properties of cellulose in kenaf (Hibiscus cannabinus L.) planted in the reclaimed land of Buan-si, Korea were examined to understand the growth characteristics using a light microscopy and an X-ray diffraction method. The samples of kenaf were taken from six positions (3 cm, 35 cm, 70 cm, 105 cm, 280 cm, and 320 cm) of each stem over the growth period (July, August, September, and October) after seeding in the mid-May. In the kenaf stem, phloem constituted 10 to 15 %, xylem 66 to 82%, and pith 7 to 19%. The ray, bast fiber, and remainder comprised 50%, 20%, and 30% of the phloem, respectively. The volume of vessel, ray, and fiber in the xylem was approximately 10, 15, and 75%, respectively. The proportion of cell wall was 30.92% at the base of stem and 46.40% at the top of stem, respectively. The average length of bast fiber and xylem fiber was about 2.8 mm and 0.9 mm, respectively. Radial and tangential diameters of vessel increased with the increase of growth period, while they decreased with increasing the stem height. Relative crystallinity ranged from 70 to 79% in phloem and from 50 to 56% in xylem. Cellulose crystallite width was about 3 nm both in the phloem and xylem. Thus, the volumetric composition and cell dimensions in the phloem and xylem appeared to be varied with the growth period and the stem height.
