Antitumor Activity of Seaweeds toward Sarcoma-180 (해조류의 Sarcoma-180에 대한 항암효과)

  • Protein-polysaccharides(PP) extracted from four species of gulfweed, fusiforme, sea-mustard and sea-tangle were examined for antitumor activity toward sarcoma-180 implanted in mice, ICR. Polysaccharides in protein-polysaccharides extracted from gulfweed, fusiforme, sea-mustard and sea-tangle were 61.48, 55.61, 34.06 and 30.28%, respectively. Monosaccharides of four seaweeds consisted of glucose, galactose, mannose, fructose and xylose, and major amino acids consisted of glutamic acid, aspartic acid, cystein, valine and glycine. Antitumor activity of the protein-polysaccharides extracted from sea-mustard showed the highest inhibition ratio of 69.76% when PP was injected to the mice at the dose of 100mg/Kg/day. PP of sea-tangle showed the maximum survival ratio of 25.22% when injected at the dose of 100mg/Kg/day.

Prenatally Diagnosed Uncommon Mosaic Autosomal Trisomy

  • Prenatal diagnosis of rare autosome mosaicism involvingchromosomes other than chromosome 13, 18, 21 or the sex chromosome is encountered prognostic dilemma during genetic counseling. We report four cases of level III uncommon mosaicism of trisomy 5, 16 and 20,diagnosed prenatally. In case 1 with mosaic trisomy 20, there was a higher mosaic ratio of trisomy 20 in the repeat amniocentesis (62.1%) than in the first (36.6%) with normal fetal ultrasound finding except for a relatively small aorta on a 3-vessel view of the fetal heart. Case 2 showed a low rate of mosaic trisomy 20 (5.25%) in cultured amniocytes but normal karyotype in the repeat amniocentesis, who delivered a normal healthy baby. Case 3 showed a 13.6% of trisomy 16 mosaicism in the 30 cells of cultured amniocytes. Sixty cells from a fetal blood sample at termination showed non-mosaic 46,XX normal karyotype, while skin fibroblasts had 22.5% trisomy 16 in 40 metaphases. The autopsy showed ventricular septal defect (VSD). Case 4 with low grade mosaicism (10.5%) of trisomy 5 resulted in elective termination, though the ultrasoumd showed growsly normal fetus. Although level III mosaicism is regarded as true mosaicism, it is difficult to predict the outcome of the fetus with rare mosaic autosome trisomy. Therefore mosaic autosome trisomy of fetus should be carefully interpreted with more various approaches including repeat sampling and targeted fetal ultrasound.

Volcanic stratigraphy and petrology of Cretaceous volcanic rocks in the eastern part of the Euiseong Basin (의성분지 동부에 분포하는 백악기 화산암류의 화산층서와 암석학적 연구)

  • In the eastern part of the Euiseong Basin acidic~intermediate volcanic rocks widely distribute on the Cretaceous sedimentary basement. Coeval granitic rocks and dyke rocks intruded into the volcanic rocks. Volcanic stratigraphy of study area are andesite lava, dacitic lapilli tuff, dacitic flow-banded lava, rhyolitic bedded tuff, rhyolitic massive tuff, dacitic massive lava, rhyolitlc welded tuff occur from the lower to the upper strata. $SiO_2$ content of the volcanic rocks range from 51 to 74 wt.%. With the increase of $SiO_2$, the contents of $TiO_2$, $Al_2$$O_3$, MgO, FeOT MnO, CaO, $P_2$$O_{5}$ decrease but those of $K_2$O increase. The contents of $Na_2$O show dispersive variation. This trend is quite sim-ilar to the major oxide variation in the volcanic rocks from the Yucheon sub-basin. The geochemical natures indicate that the volcanic rocks in the study area are discriminated to the island-arc type high K to medium K calc-alkaline rocks. The compositional variation of the volcanic rocks can be explained by the plagioclase fractionation of the volcanic magmas originated from similar source materials. The volcanic stratigraphy seems to have formed by at least two eruptive sequences of andesitic to rhyolitic and dacitic to rhyolitic magmas which underwent crystallization differentiation.

Nitrogen Conversion Factors and in vitro Protein Digestibility of some Seaweeds (수종해조의 단백계수와 in vitro Digestibility)

  • In an attempt to evaluate the nutritional quality of seaweed protein, the effects of heat treatment on the in vitro digestibility and trypsin inhibitor content in seaweed were determined. In this study, the nitrogen-to-protein conversion factors were also calculated on the basis of quantitative amino acid data. The results are as follows : 1. The in vitro protein digestbilty of red seaweeds (P. teoera anc P. suborbiculata) were ranged from 78.5 to 82.2, and green seawerd (E. linza) and brown seaweeds showed value under 80 in vitro digestibility. In general, trypsin inhibitor contents in brown seaweed were higher (0.33-0.54 mg/g) than those of red seaweeds (0.26-0.39 mg/g). And it is noted that the lowest trypsin inhibitor content was shown in green seaweed (E. linza) in spite of lowest in spite digestibility (78.5). 2. The in vitro protein digestibility of sun dried laver (P. tenera) was increased with cooling time (microwave heating), but it was not significant. Hot plate cooking raised the in vitro digestibility from 81. 1 to 84.5. The influence pot cooking time on trypsin inhibitor content was inversely proportional to in vitro digestibility. 3. Computed nitrogen factor, based on amino acid content (Factor method) and Kjeldahl nitrogen content (Kjeldahl mettled), were 5.83 (H. fusiforme)- 6.52 (P. tencra) as Factor method and 5.40 (U. pinnatifida)-6.29 (P. tenera) as Kjeldahl method. Individual value for each nitrogen conversion factor differed by species, especially in brown seaweeds. The best estimate of the protein content of seaweed can be calculated, from multiplying the summed amino acid content by conversion factor (Factor method).

Anticarcinogenic Effects of Sargassum fulvellum Fractions on Several Human Cancer Cell Lines in vitro (모자반 분획물의 in vitro에서의 항발암효과)

  • Despite many therapeutic advances in the understanding of the processes in carcinogenesis, overall mortality statistics are unlikely to change until there is reorientation of the concepts for the use of natural products as new anticarcinogenic agents. In this study, we investigated the anticarcinogenic activity, antioxidant and DPPH scavenging activity of Sargassum fulvellum (SF). SF was extracted with methanol, which was further fractionated into five different types: hexane (SFMH), ethylether (SFMEE), ethyl acetate (SFMEA), butanol (SFMB) and aqueous (SFMA) partition layers. We determined the cytotoxic effect of these layers on human cancer cells by MTT assay. Among various partition layers of SF, at starting concentration of 100 $\mu\textrm{g}$/mL, SFMEE showed very high cytotoxicity which were 92, 90 and 84% and kept high throughout 5 concentration levels sparsed by 100 $\mu\textrm{g}$/mL against all three human cancer cell lines: HepG2, HT-29 and HeLa. SFMEA showed a low cytotoxicity at the beginning concentration level, but as the concentration became denser, growth inhibition effect of cancer cell lines started to increase and at 500 $\mu\textrm{g}$/mL, it hit the highest, which were 91, 96 and 98% against the same three cell lines as above. We observed QR induced effect in all fraction layers of SF. SFMEE showed similar tendensy of QR induced effect as did against cytotoxicity. The QR induced effect of SFMEE on HepG2 cells at 25 $\mu\textrm{g}$/mL concentration indicated 3 times higher than the control value of 1.0 and SFMH tended to be concentration-dependent on HepG2 cells. At 100 $\mu\textrm{g}$/mL, the QR induced effects resulted a ratio, which was 2.5 times higher than the control value. In search for antioxidation effects of SF extract and partition layer, the reducing activity on the 1, 1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging potential was sequentially screened. The SFM has similar antioxidant activity as to BHT and vitamin C groups.

Trace Components and Functional Saccharides in Marine Algae -2. Dietary Fiber Contents and Distribution of the Algal Polysaccharides- (식용해조류중의 미량요소와 특수기능성 당질 -2. 산지와 채취시기별 식이성섬유질 함량의 변화와 해조다당류의 분포-)

  • This report described on the soluble, insoluble, and total dietary fiber level depending on the harvesting seasons and habitats in 9 species of marine algae. Total dietary fiber contents were comprised $25.4-38.1\%$ (dry basis) in green laver and $35.4-43.8\%$in sea staghorn of green algae, $34.2-48.8\%$ in sea mustard, $37.5-47.8\%$ in seaweed fusiforme, $42.9-71.3\%$ in gulf weed, and $37.1-45.1\%$ in sea tangle of brown algae, and $31.3-40.5\%$ in laver, $51.5-60.4\%$ in seaweed dilatata, and $57.1-65.8\%$ in seaweed furcata of red algae. Relatively high levels of both soluble and insoluble dietary fibers were found in seaweed furcata and gulf weed. The ratio of soluble dietary fiber to total dietary fiber was the highest in green laver $(43.7-64.8\%)$, sea mustard $(17.5-31.3\%)$, and seaweed furcata $(44.7-63.2\%)$ in their respective groups. The highest level of algal polysaccharides was confirmed to be an alkali-soluble alginic acid $(9.0-15.1\%)$ in whole brown algae, porphyran$(5.8\%)$ in laver, agar $(20.0\%)$ in seaweed furcata, and carrageenan $(23.8\%)$ in seaweed dilatata of red algae.

Production of Total Reducing Sugar and Levulinic Acid from Brown Macro-algae Sargassum fulvellum (거대 갈조류 모자반으로부터 환원당과 레불린산의 생산)

  • Recently, many biofuels and chemicals converted from renewable resources have been introduced into chemical industries. Sargassum fulvellum is a brown macro-algae, which is found on the seashores of Korea and Japan. In this work, the production of total reducing sugar and levulinic acid from S. fulvellum, using dilute-acid catalyzed hydrothermal hydrolysis and statistical methodology, was investigated. As a result, 15.28 g/l total reducing sugar was obtained under the optimized conditions of $160.1^{\circ}C$, 1.0% sulfuric acid, in 20.2 min. Furthermore, 2.65 g/l levulinic acid was obtained at $189.5^{\circ}C$, 2.93% sulfuric acid, in 48.8 min.

STUDIES ON THE EXTRACTION OF SEAWEED PROTEINS 4. Precipitation Conditions and Nutritional Evaluation of Isolated Seaweed proteins (해조단백질 추출에 관한 연구 4. 추출단백질의 심전조건 및 영양적 평가)

  • For the effective utilization of diverse and abundant resource of seaweeds in Korea as a food protein supplment, extraction conditions of water, salt, and alkali soluble proteins were investigated in previous work(Ryu and Lee, 1977: Lee et al., 1977: Lee et al., 1978). The present study as a part of the serial work was thus aimed to find the conditions of isolation and purification of extracted proteins, and to evaluate the nutritional quality of the isolated seaweed proteins in terms of amino acid composition, chemical score, protein score, modified essential amino acid index(MEAAI), and in vitro digestibility presented as pepsin-pancreatin digest residue index (PPDRI). As for the isolation of extracted proteins, TCA treatment was more effective for the proteins from rhodophyceae and Chlorophyceae while the precipitation at isoelectric point was more desirable for Phaeophyceae proteins. In amino acid composition, water soluble protein fraction was superior to the other fractions in Porphyra suborbiculata whereas both water and alkali soluble fractions seemed to bo more benefitial for Enteromorpha linza and Ulva pertusa; the extraction with alcohol-alkali mixed solvent for Undaria pinnatifida and Sargassum fulvellum. Glutamic acid and aspartic acid content was particularly high in all protein fractions examined. The total amino acid content of Porphyra suborbiculata and Enteromorpha linza was almost equivalent to that of dried whole egg although the essential amino acid content was lower. A comparative analysis was made on the inedexes between raw seaweed powder and isolated protein. Chemical score of Porphyra suborbiculata and Ulva pertusa was approximately 35 and 56 in cafes of raw powder and isolated protein respectively while only 10 to 16 for raw powder of Undaria pinnatifida and Sargassum fulvellum and 30 to 35 for their isolated proteins. Protein score of all isolated proteins was in the range of 63 to 73 which indicates that isolated protein would be mere valuable than the fern of raw seaweed powder. Digestibility by means of PPDRI was found to be extremely low in case of raw powder but it could be doubled in case of isolated protein yielding 67 to 70 for Porphyra suborbiculata and Ulva pertusa.
