Effects of Cortisol on the Steroidogenesis and the Apoptosis of Human Granulosa-Lutein Cells (Cortisol이 사람 과립-황체화 세포의 스테로이드 생성과 세포자연사에 미치는 영향)

  • Cortisol is present in high concentration in the ovary and its receptor is expressed in the ovarian cells. Moreover, cortisol is known to have a role in steroid synthesis and cell metabolism in human granulosa and lutein cells. However, little is known of the role of cortisol presenting in high concentration in the follicles after LH surge on the granulosa-lutein cells. Therefore, the this study we evaluated the apoptosis and the production of progesterone $(P_4)$ and estradiol $(E_2)$ in the granulosa-lutein cells that are obtained during oocyte-retrieval after treatment with 5, 50, and $500{\mu}g/m\ell$ cortisol and 1 IU/$m\ell$ FSH. Results of DNA fragment analysis and TUNEL assay demonstrated that DNA fragmentation and the rate of apoptotic cells were increased in a dose-dependent manner showing a significant increase in 50 and $500{\mu}g/m\ell$ cortisol treated cells. We found, however, that FSH did not suppress the apoptosis of the cells induced by cortisol. In the results of chemiluminescence assay for $P_4$ and $E_2$, $P_4$ production was decreased by cortisol treatment, whereas $E_2$ was not changed. We also demonstrated that FSH did not inhibit the suppressive effect of GnRH on $P_4$ production as the result of apoptosis. The present study suggests that cortisol of high concentration could cause the apoptosis of human granulosa-lutein cells by suppressing the production of $P_4$. However, we need more studies to elucidate the mechanism by which cortisol induces apoptosis in human granulosa-lutein cells in view of the fact that our results are inconsistent with previous reported data.

Effects of Dietary Ginger and Scutellaria Dry Powder on Antioxidant Capacity and IgG Concentration in Colostrum and Plasma of Sows (모돈에 대한 생강 및 황금 분말 급여가 초유와 혈청의 항산화력과 IgG 농도에 미치는 영향)

  • The objective of this study was to investigate the effects of dietary ginger and Scutellaria on antioxidant capacity and IgG concentration in plasma and colostrum of sows. Twenty-four sows (Landrace $\times$ Yorkshire) in parity 2 were assigned to 3 treatment groups (8 sows per treatment) from 30 day before farrowing up to weaning on day 21 after farrowing. The treatment diets included 1) CON (Basal diet), 2) GDP (0.3% ginger dry powder was used as the replacement of wheat in basal diet) and 3) SDP (0.3% Scutellaria dry powder was used as the replacement of wheat in basal diet). No differences were found in antioxidant capacity and IgG concentration in plasma of sows among all the three treatments. Antioxidant capacity in colostrum was not affected by replacement of ginger and Scutellaria dry powder. IgG and protein concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. However, fat and lactose concentration were not affected by treatments. The total amino acid concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. Plasma antioxidant capacity was not affected by treatments in piglet on 0 day and 21 day, but tended to increase by supplementation of ginger. On 0 day, plasma IgG in piglet was not detected by replacement of ginger and Scutellaria dry powder. However, plasma IgG concentration in piglets on 21 day was significantly higher (P<0.05) in GDP than that in CON and SDP. These results indicate that colostrum IgG concentration in sows and plasma IgG concentration in piglets were improved by feeding ginger dry powder 30 days before farrowing in comparison to basal diet.

Fluconazole prophylaxis in high-risk, very low birth weight infants (고위험 극소저체중 출생아에서 fluconazole 예방요법)

  • Purpose : Fluconazole prophylaxis for very low birth weight (VLBW) infants has been shown to reduce invasive fungal infection and its mortality. This study aims to evaluate the effect of fluconazole prophylaxis in VLBW infants on the incidence and mortality of fungal infection. Methods : VLBW infants with endotracheal intubation and central vascular access admitted to the Neonatal Intensive Care Unit (NICU) at Chonnam University Hospital were enrolled. Twenty eight infants of 7-month baseline period from January to July 2005 ('non-fluconazole group') were compared with 29 infants of a 7-month fluconazole period from January to July 2006 ('fluconazole group'). Results : Gestational age, birth weight, sex, mode of delivery, frequency of twin pregnancy, chorioamnionitis, antenatal steroid and rupture of membranes were similar between the fluconazole and non-fluconazole groups. The rate of extremely low birth weight (ELBW) infants, frequency and duration of prophylactic antibiotics, total parenteral nutrition and umbilical catheters, duration of intralipid, mechanical ventilation and nasal continuous positive airway pressure (CPAP) were also not significant. However, frequency of percutaneous central venous catheterization (PCVC) and intralipid administration, and duration of PCVC were significant between the two groups. The overall incidence and mortality of fungal infection were not different between the two groups. Although the Malassezia infection was increased in the fluconazole group (P<0.05), candida was significantly decreased compared to the non-fluconazole group (P<0.05). Conclusion : Fluconazole prophylaxis in high risk VLBW infants decreased the candida infection significantly. Antifungal prophylaxis is recommended in terms of cost effectiveness, but further study is needed to clarify the reason for the increase of Malassezia infection.

Clinical Investigation about the Result of Surgically Treated Myasthenia Gravis (중증 근무력증의 수술적 치료결과에 대한 임상적 고찰)

  • Myasthenia gravis is a rare autoimmune disease involving acetylcholine receptor and its autoantibody on neuromuscular junction. The methods of treatment are medical treatment and surgical thymectomy. In this paper we analyzed the result of thymectomy and the factors affecting the postoperative symptom improvement. Material and method : This study obtained medical records of 37 patients who received the thymectomy for myasthenia gravis from March 1986 to December 1998. Result Out of 37 cases, 21 cases(57%) showed improvement, of which 8 cases (50%) in the group of thymoma(n=16), and 13 cases (62%) in the group of thymic hyperplasia(n=21) showed the improvement of symptoms. Postoperative complications were respiratory insufficiency due to aggravation of symptoms after operation, including tracheal intubation for ventilator support in 9 cases, pneumonia in 3 cases, pneumothorax in 2 cases and left vocal cord palsy in 1 case. There was one postoperative mortality. The relation between postoperative improvement and sex(P=0.3222), age(P=0.7642), thymic pathologic variants,(P=0.4335) and classification of thymoma(P=0.20) showed no statistically significant correlation. However, the lower grade of preoperative symptoms can predict the lower grade of postoperative symptoms significantly(P=0.0032). Follow up study to 36 postoperative survivors was performed in October 2002 based on the out-patient records and call with patients. Out of 36 cases, 33 cases(91.7%) could be investigated and 3 cases could not. Mean follow up period was 83.2 months. Out of 33 cases, 25 cases(75.8%) showed symptomatic improvement, of which 8 cases(53.3%) in the group of thymoma(n=15) and 17 cases(94.4%) in the group of thymic hyperplasia(n=18) showed the improvement of$\boxUl$ symptoms. Conclusion : In myasthenia gravis, thymectomy showed the good improvement, and more important factor affecting the improvement of symptoms was the grdae of preoperative symptoms. Also midterm and long term follow up results showed good symptomatic improvement.

The Effects of RGDS Tetrapeptide on the Calcification of the Bovine Pericardium Transplanted Subcutaneously in Rats (흰쥐에서 RGDS tetrapeptide가 소 심낭 이식절편의 피하이식 후 석회화에 미치는 영향)

  • All kinds of tissue valves must be pretreated for the inactivation of immunologic properties and the strengthening of tissue before implantation. However, the tissue valves are gradually denatured with the calcification process and they eventually lose their functions. Recent reports have shown the existence of specific calcium binding non collagenous proteins in the calcified area of implanted biomaterials. This experiment was intended to confirm the effect of pretreatment with RGDS(Arg-Gly-Asp-Ser) tetrapeptide on the calcification of subcutaneously implanted bovine pericardium in rats. RGDS tetrapeptide has the same amino acid sequence of attachment site of specific calcium binding non collagenous proteins. Material and Method: All bovine pericardial pieces were fixed with 0.6% glutaraldehyde. The pretreatments were done using 5 different methods, groupI, with normal saline for 60 minutes, groupII, with 0.5% GRSD(Gly-Arg-Scr-Asp) tetrapeptide solution for 60 minutes, group III : with 0.5% RGDS(Arg-Gly-Asp-Ser) tctrapeptide for 30 minutes, group IV ; with 0.5% RGDS for 60 minutes, and group V : with 0.5% RGDS for 120 minutes. The pretreated bovine pericardial pieces were implanted subcutaneously at the abdominal sites of rats. 30 days after the implantation, the implanted bovine pericardial tissue were examined radiologically, biochemically, and histologically to measure the severity of calcification. Result: On the radiological examination, group I ; 68.42$\pm$3.06, group II , 64.25$\pm$5.58 showed significant difference with group III: 48.00$\pm$3.57, group IV; 43.67$\pm$2.31, and group V ; 2.58$\pm$2.47(p<0.05). There was no difference between group I and II(p=0.105). On the biochemical examination, the amount of calcium in group I was , 33.09$\pm$6.59 mg, in group II ; 28.12$\pm$5.50mg, in group III ; 25.42$\pm$7.67mg, in group Ⅵ ; 20.51$\pm$5.11mg, and in group V : 15.43$\pm$4.25mg.

Comparison of Direct-labeling Method of Antibody with $^{99m}Tc$ and $^{188}Re$ (농양이식백서에서 $^{99m}Tc,\;^{188}Re$ 직접표지항체의 비교)

  • Purpose: We investigated the direct labeling method of antibody with $^{99m}Tc$ and $^{188}Re$ and examined the stability and function of these labeled compounds in in vitro and in vivo. Materials and Methods: Disulfide bond of nonspecific human IgG was reduced to -SH group by 2-mercaptoethanol. Stannous ion was used to reduce $^{99m}Tc$ and $^{188}Re$. The stability of $^{99m}Tc$-IgG and $^{188}Re$-IgG was estimated upto 24 hrs. Biodistribution was evaluated in abscess bearing rats at 4 and 24 hr post-injection of $^{99m}Tc$ or $^{188}Re$ labeled IgG. Results: The number of -SH group per reduced IgG molecule was 2.34. The labeling yield of $^{99m}Tc$-IgG and $^{188}Re$-IgG were 90% and 95%, respectively The stability of $^{99m}Tc$-IgG at 1, 4, 6 and 24 hr was 91%, 83%, 78%, 7% and that of $^{188}Re$-IgG at 1, 4, 16 and 24 hr was 94%, 80%, 47%, 42%, respectively. At 4 hr post-injection of $^{99m}Tc$-IgG, high uptake was found on kidney, blood, stomach and abscess ($9.42{\pm}0.68,\;1.43{\pm}0.24,\;0.86{\pm}0.18,\;0.72{\pm}0.10$ %ID/g, respectively). The uptakes at 24 hr were kidney, abscess,.itomach, and blood in descending order. In case of $^{188}Re$-lgG, high uptake at 4 hr post injection appeared on kidney, blood, abscess and stomach ($3.92{\pm}0.62,\;1.32{\pm}0.08,\;0.88{\pm}0.01,\;0.26{\pm}0.06$, respectively). The uptakes at 24 hr were kidney, abscess, blood and stomach in descending order. The abscess to blood uptake ratio of $^{99m}Tc$-IgG was 0.5 at 4 hr and 2.02 at 24 hr and that of $^{188}Re$-IgG was 0.67 and 1.29. Conclusion: $^{99m}Tc$-IgG and $^{188}Re$-IgG canbe labeled efficiently with direct labeling method. However, $^{99m}Tc$-IgG and $^{188}Re$-IgG, labeled with direct method, was unstable. Further study is needed to enhance the stability of the antibody labeling.

Effect of Horse Grazing Intensity on Changes of Sasa quelpaertenis Nakai Vegetation and Physiological Characteristics in Horses (말 방목 강도가 제주조릿대(Sasa quelpaertenis Nakai) 식생과 말의 생리적 변화에 미치는 영향)

  • This study was to investigate the effect of the horse grazing intensity on the vegetation of the S. quelpaertensis, and the physiological changes of the horse. This experiment was conducted at the community of the S. quelpaertensis (altitude of 550m) with the grazing intensity of 1.0AU(500kg), 1.5AU(750kg) and 2.0AU(1,000kg) at 30m × 30m for one week, and then the growth characteristics of the S. quelpaertensis, the changes of the vegetation, the damages on trees and the changes in the physiological characteristics of the horses was investigated and compared before and after the horse grazing. As the results, the 2.0AU grazing land showed that the culm number had the inverse correlation (r=-0.902, P<0.01) to the other lands and the species diversity index of lower-level vegetation showed the correlation (r=0.773, P<0.01) as increasing the grazing intensity.

Anti-inflammatory Effect of Oxya chinensis sinuosa Ethanol Extract in LPS-induced RAW 264.7 Cells (LPS로 유도된 RAW 264.7세포에 대한 벼메뚜기(Oxya chinensis sinuosa) 에탄올 추출물의 항염증 효과)

  • Although the grasshopper Oxya chinensis sinuosa has long been used as food in Korea, there is little data on its functional effects. In this study, we investigated the anti-inflammatory effect of O. c. sinuosa ethanol extract (OCE) in RAW 264.7 mouse macrophage cells treated with lipopolysaccharide (LPS) for induction of inflammation. First, we determined that there is no cytotoxicity at $2,000{\mu}g/ml$ or less of OCE in RAW 264.7 cells. To evaluate the anti-inflammatory effects of OCE, we investigated expression levels of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-6, and pro-inflammatory enzymes such as inducible nitric oxide synthase (iNOS) and cyclo-oxygenase-2 (COX-2) in LPS-induced RAW 264.7 cells. In addition, we examined whether OCE could inhibit translocation of NF-${\kappa}B$ p65 into the nucleus in LPS induced RAW 264.7 cells. As a result, we found that the mRNA and protein levels of TNF-${\alpha}$ and IL-6 decreased in LPS-induced RAW 264.7 cells after treatment with OCE in a dose-dependent manner. In addition, we confirmed a $2,000{\mu}g/ml$ concentration of OCE inhibited translocation of NF-${\kappa}B$ p65 by immunnostaining and Western blot analysis, and a decrease in the protein expression levels of iNOS and COX-2. Accordingly, we suppose that OCE has an anti-inflammatory effect through down-regulation of TNF-${\alpha}$, IL-6, iNOS, and COX-2 related to ${\kappa}B$ p65 inflammatory signaling pathways.

Effect of Dietary Selenium on the Colon Carcinogenesis in Male ICR Mice

  • Selenium is an essential micronutrient for normal body function and functions as an essential constituent of selenoproteins. This study was carried out to investigate effect of selenium on the formation of colonic aberrant crypt foci (ACF) and tumor formation in a mouse model. Five-week old ICR mice were acclimated for one week and fed different selenium diet (0.02, 0.1, and 0.5 ppm) for 12 weeks. Animals received three intraperitoneal injections of azoxymethane (10 mg/kg B.W. in saline for 3 weeks), followed by 2% dextran sodium sulfate in the drinking water for a week. There were four experimental groups, including a normal control group and three different selenium levels groups. After sacrifice, the total numbers of aberrant crypt (AC) and ACF were measured in the colonic mucosa after methylene blue staining. The number of tumors was noted for tumor incidence. Liver selenium concentration was measured using ICP-AES method. Gutathione peroxidase (GPx) activity was determined using a GPx assay kit in the liver and colon. TUNEL assay and proliferating cell nuclear antigen (PCNA) staining were performed to examine the cell apoptosis and cell proliferation, respectively. Immunohistochemistry of $\beta$-catenin was also performed on the mucous membrane tissue of colon. The activity of GPx in the liver and colon was decreased in the selenium-deficient diet group while it was increased in the selenium-overloaded diet group. Apoptotic positive cells were increased in the selenium-overloaded diet group but decreased in the selenium-deficient diet group. PCNA staining area was decreased in the selenium-overloaded diet group. In addition, the $\beta$-catenin protein level in the selenium-deficient diet group was increased but decreased in the selenium-overloaded diet group. These results indicate that dietary selenium might exert a modulating effect on colon cancer by inhibiting the development of ACF and colon tumor formation in this mouse model.

Influences of Feeding Seleniferous Whole Crop Barley on Growth Performance, Blood and Carcass Characteristics, and Tissue Selenium Deposition in Finishing Barrows (셀레늄함유 청보리 급여가 거세비육돈의 생산성, 혈액 및 도체특성, 조직 내 셀레늄 축적에 미치는 영향)

  • The present study has been conducted to investigate the effects of feeding seleniferous whole crop barley (WCB) to finishing pigs on their growth performance, blood and carcass characteristics as well as on tissue selenium deposition. A total of 40 cross-bred barrows ((Landrace${\times}$Yorkshire)${\times}$Duroc) were allotted to five replicates of four treatments. Each replicate was arranged to 2 pigs per pen; the experimental period lasted for 6 weeks. The finishing pigs were fed diets containing 0.1 (non-seleniferous WCB as a control), 0.2, 0.4 and 0.6 ppm of selenium (Se) by supplementing the diets with seleniferous WCB. The isonitrogenous and isocaloric diets containing 5% non-seleniferous or seleniferous WCB were formulated. Feeding seleniferous WCB did not affect (p<0.05) the feed intake and BW gain. Total blood lipid concentration was significantly (p<0.05) decreased with increasing Se levels. Total blood cholesterol concentration for the control was significantly (p<0.05) higher than that for 0.4 and 0.6 ppm of Se treatments. Increasing the Se levels in WCB significantly (p<0.05) decreased blood triglyceride concentration; however, the levels increased immunoglobulin G and selenium concentrations. Feeding seleniferous WCB did not affect the carcass rate, backfat thickness and meat quality as well as yield grades. The Se concentration in the kidney, liver and loin were significantly (p<0.05) increased with increasing levels of seleniferous WCB. The results indicated that feeding seleniferous WCB may improve the blood characteristics related to lipid metabolism and thus, could produce selenium-fortified pork. Moreover, it is shown that the dietary optimal selenium level to depose selenium in porcine tissues by utilizing seleniferous WCB would be 0.4 mg of Se/kg of ration. Moreover, when 100 g of pork produced from pigs raised under such condition is served to consumers, it meets the minimum recommended daily requirements (40 ${\mu}g$) of dietary selenium proposed by the World Health Organization (1996).
