The Change of Lipid Metabolism and Immune Function Caused by Antioxidant Material in the Hypercholesterolemic Elderly Women in Korea (고콜레스테롤혈증 여자 노인에서 항산화 물질 복용에 따른 혈중 지질 농도와 면역능의 변화)

  • This study was performed to examine the change of lipid metabolism and immune function caused by antioxidant material in hypercholesterolemic elderly women (serum total cholesterol $\geq$200 mg/dI). The subjects were 51 elderly women aged over 60 yrs. They were divided into antioxidant nutrients complex group (n = 25) and spirulina group (n= 26). Antioxidant nutrients complex (1 capsule/day) and spirulina (7.5 mg/day) were used for intervention for 8weeks. All the subjects were fully informed the purpose of study and gave written consents to participate in this study. Dietary intakes, anthropometric indices and blood assessment for lipid, immune function and antioxidant status were measured before and after supplementation. Either antioxidant nutrients complex or spirulina supplementation for 8weeks resulted in improved antioxidant status evidence by increased TAS (total antioxidant status) and decreased TB-ARS (thiobarbituric acid reactive substance) . This intervention led to decreased serum levels of total cholesterol, LDL-cholesterol, oxLDL, apolipoprotein B, IL-6 and IL-6 production by peripheral blood lymphocyte. In conclusion, the lipid profiles, immune function and antioxidant capacity were improved after either antioxidant nutrient complex or spirulina supplementation for hypercholesterolemic women. Therefore, improving antioxidant status using supplemen-tation could provide means of controlling cardiovascular disease in Korean elderly people.

Effects of Samultang on Immune Function during the late stage of Pregnancy in BALB/c mice (사물탕이 임신 말기 생쥐의 면역능에 미치는 영향)

  • The purpose of this research was to investigate effects of Samultang water extract (SMT) on cytokine production from immune cells during the late stage of pregnancy in BALB/c mice. SMT(500 mg/kg) was administered p.o. once a day for 7 days, and then thymocytes and peritoneal macrophages were separated. At the late stage of pregnant mice, the proliferation of thymocytes and the production of ${\gamma}-interferon$ in thymocytes were decreased as compared with normal group, but the production of interleukin-2 and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$, nitric oxide and phagocytic activity in peritoneal macrophage was increased as compared with normal group. At the late stage of pregnant mice administered with SMT, the production of interleukin-2 in thymocytes was decreased as compared with a pregnant group, but the proliferation of thymocytes, the production of ${\gamma}-interferon$ and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$ and nitric oxide in peritoneal macrophages were decreased as compared with a pregnant group, but phagocytic activity were increased. These results suggest that SMT has the regulative action on immune function of thymocytes and peritoneal macrophages at the late stage of pregnant mice.

Comparison of Nutritional Status and Immunocompetence of Elderly Women in Urban and Rural Area (거주환경이 다른 두 노인집단의 영양상태 및 면역능 비교)

  • The purpose of this study was to compare the nutritional status and the immunocompetence of elderly women residing in urban and rural areas. Dietary food records and anthropometric measurements were used to evaluate the nutritional status of subjects. The immune function of subjects was assessed by total and differential white blood cell(WBC) counts. Total B and T Lymphocytes, and T cell subsets were quantified by flow-cytometer. Immunoglobulin G, A, and M concentrations were also measured as an index of humoral immunity. Elderly women in rural area showed a relatively lower dietary intake of total energy, protein, and iron than did urban elderly women. Total WBC, neutrophil counts, eosinophil counts, and the percentage of neutrophils among total leukocytes were significantly higher in urban elderly women than in rural women. Although the numbers of lymphocytes were not significantly different, the percentage of Lymphocytes among total leukocytes as greater in rural elderly women than in urban. Both groups did not show any significant differences in numbers of T cell subsets and NK cells. Immunoglobulin G, A, and M levels were not significantly different between the two groups, but the numbers of subjects placed under the deficient range of immunoglobulins were greater in rural than in urban elderly women. from the present study, it could be suggested that poor nutritional intake may selectively affect the number of immune cells, thereby influencing the immunocompetence of elderly women. Antioxidative activity and Angiotensin Converting Enzyme Inhibitory activity of Fermented Medical Plants (DeulBit) and Its Modulatory Effects of Nitric Oxide Production (약용 식물 발효액(들빛)의 항산화, Angiotensin Converting Enzyme 저해 및 Nitric Oxide 생성 조절 효과)

  • This study was aimed to investigate the contents of flavonoids and the biological activity of fermented beverage of medical plants, DeulBit (DB). 50 g of Cassia semen (Cassia tora L.), 50 g of Omija (Schisandra chinensis Baillon.), 50 g of Gugija (Lycium chinense Mill), 50g of Menthae herba, 75 g of Chrysanthemum indicum Linne, 25 g of Dioscorea batatas, 5 g of Lindera obtusiloba Blume, 150 g of Polygonatum odoratum, 25 g of Glycyrrhiza uralensis, 25 g of Acanthopanacis cortex, 100 g of green tea (Camellia sinensis), and 100 g of Laminaria japonica was fermented with sucrose ($50.0{\sim}60.0^{\circ}Brix$.) and 0.5% of deep sea water in 10 L of distilled water for six months at room temperature. Total flavonoids contents of DB was calculated to $3.4{\pm}0.5\;{\mu}g/g$ and antioxidative activity of DB was measured by using DPPH radical scavenging and SOD-like activity. DPPH radical scavenging and SOD-like activity of DB was 96% and 29% at 100% of DB, respectively. In addition, DB indicated about 88% and 66% of the xanthine oxidase and angiotensin converting enzyme inhibitory activities at 1% and 10% of DB, respectively and showed fibrinolytic activity. Nitric oxide (NO) synthesis was increased to 15 times by addition of DB. In addition, NO productions of the macrophages RAW264.7 cells stimulated with lipopolysaccharide (LPS) were reduced to 40.4% by addition of DB. These results suggested that DB is significant role for antioxidative and fibrinolytic activity, and have the strong xanthine oxidase and angiotensin converting enzyme inhibitory activities.

Physiological Activities of Extracts of Cedrela sinensis leaves (참죽나무 잎 추출물의 생리활성)

  • The purpose of this study was to confirm the content of total polyphenol, antioxidative and immune activities of the extracts of Cedrela sinensis leaf. The content of total polyphenol of water extracts ranged from 46.5-59.6 mg/100 g, which was higher than other extracts using organic solvents such as EtOAc, $CH_2Cl_2$ and $C_6H_{14}$. The antioxidant activity of the water and organic solvents extracts showed 6-33% in terms of 2,2-diphenyl-picryl-hydrazyl (DPPH) scavenging activity. To analyze the immuno-stimulation activity of C. sinensis leaf extract, we investigated the effect of the extracts on NO synthesis which is important in host defense against bacterial infection. Hot water extracts significantly increased NO generation by RAW 264.7, macrophage cell line, while organic solvent extract has no significant effect on NO production. To further analyzed the anti-inflammatory effect of the extracts, we investigated the effects of the extracts on lipopolysaccharide(LPS)-induced NO generation. Organic solvent extracts of C. sinensis leaves showed strong inhibitory effect on NO production in LPS-stimulated RAW 264.7 cells. These results suggest that C. sinensis leaf extract may represent a useful immune stimulating agent and anti-inflammatory agent.

Immune Cell Stimulating Activity of Wheat Arabinoxylan (밀 arabinoxylan의 면역세포 활성화 작용)

  • Effects of wheat arabinoxylan on mouse spleen lymphocytes and peritoneal macrophages were examined in vitro. Among three wheat arabinoxylans (A1: low MW, A2: medium MW, A3: high MW), A3$(50{\sim}100\;{\mu}g/mL)$ increased the viability of spleen lymphocytes up to $114{\sim}125%$ of the control. A1 and A3 $(20\;{\mu}g/mL)$ increased the viability of lipopolysaccharide-treated lymphocytes synergistically. Viability of murine peritoneal macrophages treated with wheat arabinoxylans $(10{\sim}100{\mu}g/mL)$ was increased up to $135{\sim}175%$ of the control. The cytotoxic activity of macrophages against murine lymphocytic leukemic cell increased in the presence of wheat arabinoxylan. Phagocytic index of macrophages treated with wheat arabinozylans $(20\;{\mu}g/mL)$ significantly increased $197{\sim}232%$ compared with the control, and lysosomal phosphatase and myeloperoxidase activities also increased significantly (p<0.05). Treatment of wheat arabinoxylans tended to decrease nitrite production, but significantly stimulated $H_2O_2\;and\;O_2$ productions of macrophages (p<0.05). These results indicate that the immunostimulating effect of wheat arabinoxylan may be closely related with lysosomal enzyme activity and reactive oxygen intermediate production of macrophages.

Optimal Conditions for the Production of Immunostimulating Polysaccharides from the Suspension Culture of Angelica gigas Cells. (면역증강성 다당 생산을 위한 참당귀 세포배양의 최적조건)

  • An Immunostimulating polysaccharide was produced from the suspension culture of Angelica gigas H4, plant cells. In order to enhance the polysaccharide production by the A. gigas cell culture, medium composition and physical conditions were optimized. Schenk and Hildebrandt (SH) medium was selected as an optimal basal medium for the growth of A. gigas. The maximum cell and polysaccharide concentration obtained in SH medium were 15.8 g DCW/l and 0.85 g polysaccharide/l, respectively, at $25^{\circ}C$ under dark condition. For the enhanced polysaccharide production, a polysaccharide production medium (PPM) was established by modifying Gamborg B5 medium with optimized carbon sources, growth regulators, organic and inorganic elements. Optimal initial pH and temperature were 6.0-6.6 and $20^{\circ}C$, respectively, and the dark condition was better than the light condition. The maximum polysaccharide concentration of 1.5 g/l could be obtained through the optimization of the medium composition and physical conditions.

Enhancing Effect of Pleurotus ostreatus Extracts on Mouse Spleen and Cytokine Cells Activation (느타리버섯 물 추출물 투여에 의한 마우스 비장세포 및 사이토카인 활성효과)

  • Pleurotus ostreatus have been used as a traditional remedy and food source. Its anti-inflammatory, anti-oxidation effects have been previously reported. The production of cytokines (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$) secreted by LPS- and non LPS-stimulated macrophages, were detected by ELISA assay using a cytokine kit. Mouse splenocytes proliferation increased with Pleurotus ostreatus water extracts supplement at 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after a 48hr pre-treatment with the mitogen (ConA or LPS). The cytokine production (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$), measured by a cytokine ELISA kit, increased on water extracts supplementation. This in vitro study suggested that supplementation with Pleurotus ostreatus water extracts may enhance the immune function by regulating the splenocyte proliferation and enhancing cytokine production.

Immunomodulating activities of Sarcodon aspratus (능이버섯의 면역 조절능)

  • This study was carried out to evaluate the immunomodulatory capacity of edible mushrooms, including Sarcodon aspratus, Letinus edodes and Grifola frondosa in mice. BALB/c mice were administered 50, 500, and 1000 mg/kg body weight of various mushrooms five times a week over 4 weeks through oral administration. The control mice were administered distilled water. No significant changes in body weight were observed. IL-4 and $IFN{\gamma}$ production was evaluated with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hr. The mice group administered Sarcodon aspratus, Grifola frondosa tend to higher ratio of $IFN{\gamma}$ versus IL-4 than the other groups. In addition, the ratio of plasma IgG2a versus IgG1 was also elevated in mice treated with Sarcodon aspratus. These results indicated that Sarcodon aspratus can enhance type-1 helper T cell-mediated cellular immunity. And also, S. aspratus seems to be one of the most useful mushrooms for immunomedicine.

The Purification and Immunogenicity of Pneumococcal Surface Protein (PspA) from Invasive Streptococcus pneumoniae KNIH1156 Isolated in Korea (국내 임상 분리주 Streptococcus pneumoniae KNIH1156으로부터 PspA 단백 항원의 정제 및 면역원성 확인)

  • Pneumococcal surfacce protein A (PspA) is an important virulence factor and an antigenically variable surface protein of the pneumococci. To purify the PspA from S. pneumoniae KNIH1156 , a clinical isolate (type 19F), we have taken advantage of the fact that PspA is released from the surface of pneumococci into the medium by growing in a CDM-ET medium and PspA is capable of binding human lactoferrin, the iron carrier protein. PspA of S. pneumoniae KNIH1156 was purified from culture supernatant by human lactoferrin (hLf) affinity chromatography. The purified PspA was confirmed with anti-PspA antiserum and also had the binding capacity to hLf specifically. To determine whether the purified PspA could elicit protection in mice against pneumococcal inflection, we immunized the mice with purified PspA and subsequently challenged with S. pneumoniae KNIH1156. Immunization with purified PspA protected mice from 500 times the $LD^{50}$ of S. pneumoniae KNIH1156. Therefore, it has been shown that purified PspA fromS. pneumoniae KNIH1156 (type 19F) is a protective immunogen.
