Methane oxidation and the production potentials of ground soil (soil A) and garden soil (soil B, C, & D) in an urban school were evaluated, and the methanotrophic and methanogen communities in the soil samples were quantified using quantitative realtime PCR. The methanotrophic community in the raw soil A sample possessed a $6.1{\times}10^3$ gene copy number/g dry weight soil, whereas those in the raw soils B~D samples were $1.6-1.9{\times}10^5$ gene copy numbers/g dry weight soil. Serum bottles added with the soil samples were enriched with methane gas, and then evaluated for their methane oxidation potential. The soil A sample had a longer induction phase for methane oxidation than the other soils. However, soil A showed a similar methane oxidation potential with soils B~D after the induction phase. The methanotrophic community in the enriched soil A sample was increased by up to $2.3{\times}10^7$ gene copy numbers/g dry weight soil, which had no significantly difference compared with those in soils B~D ($1.2-2.8{\times}10^8$ gene copy numbers/g dry weight soil). Methane production showed a similar tendency to methane oxidation. The methanogens community in raw soil A ($1.7{\times}10^5$ gene copy number/g dry weight soil) was much less than those in raw soils B~D ($1.3-3.4{\times}10^7$ gene copy numbers/g dry weight soil). However, after methane gas was produced by adding starch to the soils, soil samples A~D showed $10^7$ gene copy numbers/g dry weight soil in methanogens communities. The results indicate that methanotrophic and methanogenic bacteria have coexisted in this urban school's soils. Moreover, under appropriate conditions for methane oxidation and production, methanotrophic bacteria and methanogens are increased and they have the potential for methane oxidation and production.

Two UASB reactors were operated to investigate the effect of high concentration of sulfate on anaerobic digestion of propionate using an upflow anaerobic sludge blanket (UASB) reactor. An organic loading rate of $1.2kg\;COD/m^3{\cdot}d$ and a hydraulic retention time of 1.6 d were maintained during this study. In the absence of sulfate, the UASB reactor achieved about 95% removal of chemical oxygen demand whereas in the presence of $2,000\;SO_4^{2-}mg/L$, the COD removal rate decreased to 83% due probably to the inhibition of dissolved sulfide inhibition. Interactions between the methane producing bacteria (MPB) and sulfate reducing bacteria (SRB) were measured to investigate the competition between MPB and SRB. The MPB consumed average 58% of the available electron donors at $COD/SO_4^{2-}$ ratio of 1. Propionate was consumed mainly by SRB, converting sulfate into sulfide and suppressing the methane production. The specific methanogenic activity (SMA) using acetate and propionate increased as microorganism acclimated to the substrate.
본 실험의 목적은 다양한 펄프제조 조건과 여러 형태의 리그노 셀루로우스 성분이 펄프폐수의 혐기성 생분해에 미치는 영향을 규명하는 것이다. 실험에 사용된 폐수는 일반적으로 펄프 제조시에 발생되는 폐수를 대상으로 하였으며, 펄프제조 조건은 TMP공정과 소다 펄프공정을 적용하였다. 혐기성 생분해 가능성 시험 및 독성실험은 $35\pm 2\circ$C의 중온상태에서 입상슬러지를 식종물질로 사용한 회분식 반응조를 이용하였다. TMP공정의 배출되는 폐수는 산으로의 전환율이 총 COD기준으로 68-87%로 매우 높은 혐기성 생분해 가능성을 보였다. 그리고 TMP공정폐수는 일반적으로 제지폐수 처리시 독성농도라고 알려진 농도에서도 메탄생성균에 독성을 주지 않았고, 또한 COD 10g/l의 농도에서도 처리에 저해가 일어나지 않았다. 반면에, 알카리성 상태에서 준비된 펄프폐수의 경우는 생분해성이 매우 낮아서 대략 50%정도의 산전환율을 보였으며, 메탄생성균에 상당한 저해를 주었다. 메턴생성균의 활성도에 50%저해를 주는 농도는 2.1~5.4 gCOD/l였다. 알카리성 펄프폐수의 독성에 대한 추가 실험결과 펄프내 wood resin이 산이나 중성 pH부근에서는 잘 용해가 되지 않고 알카리성 상태에서 쉽게 용해되어 메탄생성균에 저해를 나타내는 것으로 밝혀졌다. 따라서 펄프제조시 나무성분이 알카리성분과 접촉할 경우 후속하는 혐기성 처리공정의 메탄생성균에 심각한 저해를 줄 수 있다.

For treating tannery wastewater containing high sulfate and heavy metals, test was performed to assess their performance, competition between SRB (sulfate reducing bacteria) and MPB (methane producing bacteria), and the activity of MPB according to change of chromium concentrations. COD removal efficiency was above 70% at VLR (volumetric loading rate) of 2.0 gCOD/I.day and HRT (hydraulic retention time) of 18hrs at $35^{\circ}C$. In the competition between SRB and MPB, about 15% of the removed COD was utilized by SRB in the begining, but it became 43% at the end. It indicated that MPB was strongly suppressed by the occurrence of significant sulfate reduction since a large electron flow was uptaken by SRB. For the entire experiment, removal efficiencies of chromium concentration were more than 90%. Despite high removal efficiencies of chromium concentration, performance of reactor did not change significantly during the experimental periods. Expecially, chromium (III) is tannery wastewater is less toxic than chromium (VI).
The effects of EDTA(Ethylene diamine tetraacetic acid), Cu, temperature, and gas(methane and oxygen) composition on methanol production from methane with Methylosinus trichosporium were investigated. In this experiment EDTA was found to be a potential methanol dehydrogenase inhibitor since it causes methanol accumulation and 6mM was found to be optimum concentration of EDTA for methanol production. When Cu was added in culture media, the produced methanol concentration level was increased. Hence it is believed that Cu enhanced the particulate methane monooxygenase formation and consequently the addition of Cu could increase the methanol production from methane. In this experiment the optimum concentration of Cu was found to be 1mM for methanol production. When temperature was shifted down from $30^{\circ}C to 25^{\circ}C$, the methanol production level was enhanced by 50%. When the ratio of methane to oxygen in gas phase was increased to 2.3 from 1, produced methanol concentration was also enhanced by 100%.
The Study was conducted to develope the low temperature tolerant methane-producing bacteria(LTTB) and to increase the efficiency of anaerobic fermentation for the treatment of agricultural and livestock wastes at low temperature. The samples were collected from muddy soil, water logged sediment, organic layer and anaerobic sludge at three latitudes, $34.8{\sim}37.4\;^{\circ}N(Korea)$, $41.4\;^{\circ}N(USA)$ and $54.5{\sim}56.9\;^{\circ}N(Canada)$. They were used for determination of the methanogenesis rates for isolation and identification of the LTTB. The methanogenesis rate of smaples at low temperature were higher in the cellulose medium than methanol medium. The methanogenesis rate in the samples of subarctic region were $15{\sim}19$ moles/ml during 30 days at low temperature($8\;^{\circ}C$), whereas not detected in the samples of temperate region. The methanogenesis rate in the enrichment culture of subarctic samples were inhibited by the $40\;{\mu}g/ml$ of streptomycin + vancomycin or ampicillin + oleandomycin which were not effect to the methanogens. An inhabitation of high temperature tolerant methane producing bacteria was identified in the samples of temperate region, whereas that of the LTTB growing at $8{\sim}13^{\circ}C$ was identified in the subarctic region.

The effects of copper on the anaerobic degradation of propionate were studied using anaerobic batch reactors. The apparent inhibitory effects of copper on the anaerobic degradation of propionate could be observed from behaviors of intermediates, ultimate methane yield(UMY) and specific methanogenic activity(SMA) There was little inhibition at the concentration of $2.5mg\;Cu^{2+}/L$. Beyond this concentration, the inhibitory effects increased with increasing dose of coppers. The 50% inhibition of UMY and SMA occurred at copper dosage of 33.8 and $24.1mg\;Cu^{2+}/gVSS$, respectively. The inhibitory effect based on the UMY was gradually reduced with the operation time dueprobably to the acclimation of microorganisms and/or binding of the added copper by ligands(and possibly ion exchange sites)contained on the cell membrane and extracellular polymer matrix whereas it based on the SMA might exclude the this phenomena. Therefore, the methodology for interpretation of inhibition data based on the SMA was more accurated than the UMY. There was no inhibitory effect in batch reactors supplemented with sulfate due to an antagonistic action of the sulfate reducing bacteria. Propionate degradation was initially retarded for copper inhibited samples but it gradually degraded afterward. Based on the mass removal considering take into account the propionate to acetate conversion, propionate degradation may appeal more affected than acetate. This result revealed that the hydrogenotrophic methanogens were the most affected by copper.
The study investigated the biochemical methane potential (BMP) assay of cellulose supplementing with mixed methanogens and cellulolytic bacteria to improve anaerobic digestion for methane production. For the BMP assay, 7 different microbial supplementation groups were consisted of the cultures of mixed methanogens (M), Fibrobacter succinogenes (FS), Ruminococcus flavefaciensn (RF), R. albus (RA), RA+FS and M+RA+FS including control. The cultures were added in the batch reactors with the increasing dose levels of 1% (0.5 mL), 3% (1.5 mL) and 5% (2.5 mL). Incubation for the BMP assay was carried out for 40 days at $38^{\circ}C$ and anaerobic digestate obtained from an anaerobic digester with pig slurry as inoculum was used. In results, 5% FS increased total biogas and methane production up to 10.4~22.7% and 17.4~27.5%, respectively, compared to other groups (p<0.05). Total solid (TS) digestion efficiency showed a similar trend to the total biogas and methane productions. Generally the TS digestion efficiency of the FS group was higher than that of other groups showing at the highest value of 64.2% in the 5% FS group. Volatile solid (VS) digestion efficiencies of 68.4 and 71.0% in the 5% FS and the 5% RF were higher than other groups. After incubation, pH values in all treatment groups were over 6.4 indicating that methanogensis was not inhibited during the incubation. In conclusion, the results indicated that the hydrolysis stage for methane production in anaerobic batch reactors was the late-limiting stage compared with the methanogenesis stage, and especially, as the supplementation levels of F. succinogenes supplementation increased, the methane production was increased in the BMP assay compared with other microbial culture addition.
Molecular methods were employed to investigate microorganisms in a thermophilic continuous stirred tank reactor(CSTR) used for continuous $H_2$ production. The reactor was inoculated with heat-treated anaerobic sludge and fed with a glucose-based medium. Denaturing gradient gel electrophoresis showed dynamic changes of bacterial populations in the reactor during 43 days of operation. Gas composition was constant from approximately 14 days but population shift still occurred. Populations affiliated with Fervidobactrium gondwanens and Thermoanaerobacterium thermosaccharolyticum were dominant on 21 and 41 days, respectively. Keeping pH of the medium at 5.0 could suppress methanogenic activity that was detected during initial operation period. $CH_4$ and mcrA detected in the samples obtained from the reactor or inoculum suggested the heat treatment condition employed in this study is not enough to remove methanogens in the inoculum. PCR using primer sets specific to 4 main orders of methanogens suggested that major $H_2$-consuming methanogens in the CSTR belong to the order Methanobacteriales.
