The Effect of Soil Conditioners on the Rutin Biosynthesis and the Yield of Buckwheat(Fagopyrum esculentum moench) (메밀의 rutin 생합성과 수량에 대한 토양개량제의 효과)

  • This experiment was conducted to investigate the effects of soil conditioners, such as lime, borax, poultry waste sawdust manure (P.W.S.M) and mixed oil cakes (M.O.C), on the rutin biosynthesis and the yield of buckwheat. The content of phosphorus ($P_2O_5$) in buckwheat plants before flowering was higher at before flowering than that of flowering time at all plots. The content of nitrogen (N), potassium ($K_2O$), calcium (CaO) and magnesium (MgO) was lower at before flowering than those of flowering time at all plots. The contents of rutin in buckwheat plant at flowering time was higher than that at before flowering content of rutin in leaf was higher than that in stem. The contents of rutin in buckwheat plant was the highest at the plot treated with borax (B) compared to other plots. Correlation between rutin and amino acid was positive in buckwheat leaf, while it was negative in buckwheat stem. The yield of buckwheat was 21 percent higher at the plot treated with borax than the plot applied with the other three ingredients.

Physiological Functionality of Various Extracts from Danmemil and Legumes (단메밀과 콩 추출물들의 생리 기능성)

  • Physiological functionalities of various extracts from Danmemil and legumes were determined and its optimal extraction conditions were also investigated. Angiotensin I-converting enzyme (ACE) inhibitory activity and tyrosinase inhibitory activity of Danmemil were higher in water extracts (53%, 58%) than those of ethanol extracts. However, its electron-donating ability was the highest in ethanol extracts (72%). ACE inhibitory activity and electron-donating ability of Black bean No. 1 and Taekwangkong(one of bean) were higher in water extracts than those of ethanol extracts, whereas SOD-like activity was the highest in ethanol extracts. ACE inhibitor and tyrosinase inhibitor of Danmemil were maximally extracted when it were treated with 20 times of distilled water at 35$^{\circ}C$ for 24 h and 36 h, respectively. Its electron donating compound was maximally extracted by treatment of 50$^{\circ}C$ for 18 h. ACE inhibitor of Black bean No. 1 was extracted maximally when it was treated with distilled water (1 :20) at 20$^{\circ}C$ for 24 h, whereas the other functional compounds were maximally extracted at 20$^{\circ}C$ for 18 h.

Monitoring on the Tea with Steaming and Drying Process of Germinated Buckwheat (메밀순의 증숙 및 건조에 따른 침출차 특성 모니터링)

  • To make the germinated buckwheat tea, soluble solid contents, total flavonoid contents and organoleptic properties were investigated under various steaming time and drying temperature. The optimum condition of soluble solid contents were 6.93 min of steaming time and 73.59$^{\circ}C$ of drying temperature. Total flavonoid contents were maximum under the condition of 5.22 min of steaming time and 79.05$^{\circ}C$ of drying temperature. The optimum condition of overall palatability was 6.00 min of steaming time and 77.33$^{\circ}C$ of drying temperature. The optimum ranges of soluble solid contents, total flavonoid contents and overall palatability of the tea were 5.4∼7.0 min of steaming time and 75∼8$0^{\circ}C$ of drying temperature. The values expected in the optimum ranges were also similar to the experimental values.

Increase of the D-chiro-inositol and D-Pinitol Contents by Abiotic Stress in the Buckwheat Seedlings (쓴메밀 유식물에서 비 생물학적 스트레스에 의한 D-chiro-inositol과 D-pinitol의 함량 증가)

  • D-chiro-Inositol, which is the isomer of myo-inositol, is a well known drug for the treatment of type II diabetes. The methylated form of D-chiro-inositol, D-pinitol and D-chiro-inositol are synthesized when the plants are exposed to the abiotic stresses such as drought, salinity and low temperature as osmoprotectants. In soybean, myo-inositol is converted to ononitol by O-methyltransferase, and ononitol is converted to D-pinitol by ononitol epimerase and finally converted to D-chiro-inositol by demethylase. However there have been some reports that in buckwheat, myo-inositol can be converted to D-chiro-inositol directly. This study was conducted to determine the changes of soluble cyclitols in buckwheat seedlings after exposure to salt and drought stresses by GC-FID. The results indicated that myo-inositol may be the precursor of D-chiro-inositol biosynthesis.

Transcriptomic Profile Analysis of Jeju Buckwheat using RNA-Seq Data (NA-Seq를 이용한 제주산 메밀의 발아초기 전사체 프로파일 분석)

  • In this study, transcriptome analysis was conducted to collect various information from Fagopyrum esculentum and Fagopyrum tataricum during the early germination stage. Total RNA was extracted from the seeds and at 12, 24, and 36 hrs after germination of Jeju native Fagopyrum esculentum and Fagopyrum tataricum and sequenced using the Illumina Hiseq 2000 platform. Raw data analysis was conducted using the Dynamic Trim and Lengths ORT programs in the SolexaQA package, and assembly and annotation were performed. Based on RNA-seq raw data, we obtained 16.5 Gb and 16.2 Gb of transcriptome data corresponding to about 84.2% and 81.5% of raw data, respectively. De novo assembly and annotation revealed 43,494 representative transcripts corresponding to 47.5Mb. Among them, 23,165 sequences were shown to have similar sequences with annotation DB. Moreover, Gene Ontology (GO) analysis of buckwheat representative transcripts confirmed that the gene is involved in metabolic processes (49.49%) of biological processes, as well as cell function (46.12%) in metabolic process, and catalytic activity (80.43%) in molecular function In the case of gibberellin receptor GID1C, which is related to germination of seeds, the expression levels increased with time after germination in both F. esculentum and F. tataricum. The expression levels of gibberellin 20-oxidase 1 were increased within 12 hrs of gemination in F. esculentum but continuously until 36 hrs in F. tataricum. This buckwheat transcriptome profile analysis of the early germination stage will help to identify the mechanism causing functional and morphological differences between species.

Construction of Data System on Seed Morphological Traits and Functional Component in Tartary Buckwheat Germplasms (쓴메밀 유전자원의 종자특성과 유용성분 변이에 관한 자원 정보 구축)

  • This study analyzed the phenotypes and chemotypes of 74 tartary buckwheat (Fagopyrum tataricum) germplasms using principal component analysis and cluster analysis. The average seed size of tartary buckwheat germplasm was 5.2 × 3.4 mm, which is smaller than the seed size of common buckwheat. The dark browned colored ovate or elliptic shape was mostly observed in collected germplasm. The average content of rutin was 1,393 mg per 100 g dry weight (DW) in tartary buckwheat seed. Similarly, the flavonoid and polyphenol contents ranged from 253 to 2,669 and 209 to 1,823 mg, respectively, per 100 g DW in the collected germplasm. The three components (PC1, 2, and 3) of principal component analysis revealed 68.55% of the total variance of the collected accessions. Cluster analysis using descriptors showed that 74 accessions were clustered into five groups. The study showed that the most interesting resources for functional breeding programs are: Five resources (HLB1004, HLB1005, HLB1007, HLB1009, and HLB1013) due to the rich rutin, polyphenol, and flavonoid.

Growth of Intestinal Bacteria and Intestinal Inflammation of Sprout Extract from Common Buckwheat and Tartary Buckwheat (일반메밀과 쓴메밀의 새싹 추출물의 장내 유익균 증식 및 염증조절 효능 평가)

  • We aimed to assess the potential growth-promoting effects of buckwheat sprout on intestinal bacteria and their anti-inflammation effects in a cellular model of intestinal inflammation. The growth of Bifidobacterium longum ssp. infantis BT1 was enhanced with the addition of the sprout extract of tartary buckwheat. Further, in the inflammatory model cells cultured with Raw 264.7 cells were treated with buckwheat sprout including each 10 probiotics before the addition of lipopolysaccharide (LPS) to induce inflammation in Raw 264.7 cells. Buckwheat sprout in both Bifidobacterium longum ssp. infantis BT1 and Lacticaseibacillus paracasei LPC5 significantly reduced the production of NO and PGE2. The above results indicate that buckwheat sprout extract which contains with various physiologically active substances such as rutin, quercetin, and choline is effective in suppressing NO and PGE2 production, which are inflammation-related indicators. The present study suggests that buckwheat sprout could induce positive effects on the intestinal beneficial bacteria and in anti-inflammation.

Production and Characteristics of Cello- and Xylo-oligosaccharides by Enzymatic Hydrolysis of Buckwheat Hulls (메밀껍질의 효소분해에 의한 기능성 올리고당의 생산 및 특성)

  • This study was conducted to produce oligosaccharides from buckwheat hull by using commercial enzymes. Yields of oligosaccharides obtained by enzymatic hydrolysis of the cellulose and hemicellulose fractions were 132.37 and 393.04 g/kg, respectively. Xylose, glucose, fructose, xylobiose, xylotriose, cellobiose, and cellotriose were detected in the hydrolysate produced from buckwheat hull. Antioxidant activity of oligosaccharide from cellulose fraction (OSC) reduced with increasing hydrolysis time; however, the antioxidant activity of oligosaccharide from hemicellulose fraction (OSF) increased as the hydrolysis time was prolonged. OSF and OSC showed higher increase in viable counts compared to the control. As a result, oligosaccharides produced from buckwheat hull by enzymatic hydrolysis showed antioxidant activity and prebiotic effects. It is suggested that utilization of oligosaccharides produced from buckwheat hull as functional food materials may be improved when hydrolysis time and conditions are controlled for this purpose.
