최근 약 10년 동안 마이크로 유체 시스템은 다양한 세포연구에 광범위하게 활용되며 생물학과 의학 분야에 새로운 전기를 가져왔다. 마이크로 유체 시스템 내의 유동현상을 설계하고 연구에 적용하는 일은 기계공학도가 중추적인 역할을 해야 하는 분야이다. 유동장 내에서 관찰되는 세포의 반응은 매우 흥미로울 뿐 아니라 학술적으로 막대한 파급력있는 결과를 가져오기도 한다. 이 글을 통해 전통적인 접근법으로는 성취하기 힘들었던 세포연구들이 마이크로 유체 시스템을 이용하여 어떻게 효과적으로 수행되는지 소개하고자 한다.
다중 암의 동시 진단 기술에 대한 관심이 전 세계적으로 증가하는 추세이며, 진단 난이도를 낮추기 위해 혈액과 같은 미량의 바이오 유체를 이용하여 질병을 진단하는 미세 유체 소자 기반의 액체 생검 기술이 연구되고 있다. 바이오 유체를 이용하여 형광 영상 등을 통해 분석물질의 농도를 측정하는 광학적 바이오 센싱에 있어 민감도를 향상시키기 위한 기술개발이 필요하다. 본 논문에서는 모세관력에 의한 자가구동 기반의 마이크로 채널의 기하학적 구조와 미세 유체 현상만으로 수동적 자기 혈장 분리 기술과 유체 혼합을 통한 분자 인식 활성화 기능을 구현하는 형광 다중 암 진단 센서 플랫폼 구조를 제안하고 설계하였다. 설계된 센서의 혈장 분리부의 성능에 영향을 미치는 파라미터를 확인하기 위해 채널의 수력학적 직경과 종횡비, 유체의 점도를 변수로 설정하여 딘 와류 형성 여부를 시뮬레이션을 통해 확인하였고 최적의 센서 플랫폼 구조를 제시하였다.
알지네이트 하이드로 젤은 해조류에서 추출되는 천연 고분자인 알지네이트가 칼슘 또는 마그네슘 양이온과 이온가교(Ioninc cross linking)를 형성할 때 알지네이트의 고분자 구조가 칼슘, 마그네슘 양이온을 감싸면서 형성되는 고분자이다. 알지네이트 하이드로 젤은 높은 생체적합성(Biocompatibility)으로 인해 세포 재생을 위한 조직공학 및 재생의학, 약물전달 등의 제약 관련 분야에 광범위하게 적용될 수 있는 물질로 많은 연구가 이루어지고 있다. 본 연구에서는 마이크로 플루이딕 칩을 이용하여 알지네이트 튜브를 제조하였다. 먼저 유동 포커싱 방식(flow focussing)을 유도할 수 있는 PDMS(Polydimethylsiloxane) 마이크로 플루이딕 칩을 제조하였다. 마이크로 플루이딕 칩은 CNC(Computer Numeric Control) milling machine을 이용한 template를 만들고 NOA mold를 이용하여 최종 PDMS 칩을 제작하였다. 튜브를 만들기 위한 마이크로 채널은 내부 채널 ($200{\times}200um$), 중간 채널 ($200{\times}200um$) 및 외부 채널 ($200{\times}200um$)로 구성되며 내부, 중간, 외부의 유체가 합류하는 수집채널은 폭 500 um, 깊이 200 um로 구성되었다. 운반체로는 5%의 acetic acid를 함유한 mineral oil를 이용하였으며 내부의 core flow는 $H_2O$로 하였다. 중간 유체인 2% 알지네이트 프리폴리머는 칼슘 이온의 존재 하에서 젤화 과정이 매우 빠르기 때문에 마이크로 채널 내부에서의 반응을 제어하고 막힘을 방지하기 위해 수용성 복합 칼슘-에틸렌 디아민 테트라 아세트산 (EDTA)을 사용하였다. 본 마이크로 플루이딕 칩에 각각의 유체를 이동시켰을 때, 운반체인 oil phase의 수소이온은 중간 유체인 알지네이트 프리폴리머와의 계면을 통해 확산되어 Ca-EDTA 복합체로부터 칼슘 양이온의 방출을 유발하게 된다. 방출된 칼슘 양이온은 알지네이트 고분자와의 이온 가교를 통해 알지네이트 하이드로 젤을 형성하여, 각 유체의 flow에 따라 알지네이트 튜브를 쉽고 빠르게 제조 가능하였다. 본 연구에서 제조된 알지네이트 튜브는 인체 내 장기간 약물 전달을 위한 나노섬유로 활용하거나 인공혈관을 구성하는 extracellular matrix로 활용될 잠재력을 가지고 있어 추후 활발한 연구개발이 진행될 예정이다.
A numerical study of the mixing of two fluids(pure water and a solution of glycerol in water) in a microchannel was carried out. By varying the glycerol content of the glycerol/water solution, the variation in mixing behavior with changes in the difference of the properties of the two fluids(e.g., viscosity, density, diffusivity) was investigated. The mixing phenomena were tested for three micromixers: a square mixer, a three-dimensional serpentine mixer, and a staggered herringbone mixer. The governing equations of continuity, momentum and solute mass fraction were solved numerically. To evaluate mixing performance, a criterion index of mixing of mixing uniformity was proposed. In the systems considered, the Reynolds numbers based on averaged properties were 1 and 10. For low Reynolds number (Re = 1), the mixing performance varied inversely with mass fraction of glycerol due to the dominance of molecular diffusion. The mixing performance by diffusion deteriorated due to a significant reduction in the residence time of the fluid inside the mixers.
레이저 광 산란을 이용한 검출 시스템 및 레이저를 이용한 광학적 검출에 있어서 높은 발광 강도를 통해 궁극적으로 높은 효율의 광 산란 신호를 광검출기에서 얻기 위해서는 발광 레이저빔을 미세유체 칩의 채널 중앙에 집광하는 것이 매우 중요하다. 본 논문을 통해 레이저 광 산란을 이용한 세포 검출을 위해 PDMS 마이크로렌즈가 집적화된 PDMS 미세유체 칩을 소개하고자 한다. 기존에 제작된 PDMS 미세유체 칩 위에 간편히 정렬하여 올려놓아 사용함으로써 검출 효율을 증가시킬 수 있는 PDMS 마이크로렌즈를 제작하였다. PDMS 마이크로렌즈는 포토레지스트 리플로우와 PDMS 복제 몰딩에 의해 제작되었다. 이 제작 방법은 간단하며 높은 치수 정확성 및 좋은 마이크로렌즈의 성능을 제공한다. PDMS 미세유체 칩 위에 집적화된 PDMS 마이크로렌즈가 적혈구를 이용한 레이저 광 산란을 통한 세포 검출 실험에서 레이저 강도를 증가시켜 신호대잡음비 및 감도를 증가시킴을 검증하였다.
An analysis has been conducted of the flow characteristics and pumping performance of a thermopneumatic micropump with electrically conducting fluid. In the present study, considered is a thermopneumatic micropump for electrically conducting fluids with electromagnetic resistance alternately exerted at the inlet and outlet by alternately applied magnetic fields. A model of Prescribed Deformation is used for the motion of the membrane. Here, the pumping performance of the micropump and flow characteristics of the electrically conducting fluid are investigated in the range of Hartmann number less than 30. The current numerical study shows that the net flow rate through the micropump is almost proportional to the strength of the applied magnetic field.
In the microfluidic devices the most important thing is mixing efficiency ol various fluids. In this study a newly designed miler is proposed to enhance the mixing effect with the purpose to apply it to microchannel mixing in a short future. This design is composed of a channel with cross baffles periodically arranged on the both bottom and top surfaces ol the channel. To obtain the yow patterns, the numerical computation was performed by using a commercial code, ANSYS CFX 10.0. To evaluate the mixing performance, we computed Lyapunov exponent and obtained Poincare sections. it was shown that our design provides the excellent mixing effect.
Nowadays, the development of microfluidic chip [i.e. biochip, micro-total analysis system ($\mu$-TAS) and LOC (lab-on-a-chip)] becomes more active, and the microchannels to deliver fluid by pressure or electroosmotic forces tend to be more complex like electronic circuits or networks. For a simple network of channels, we may calculate the pressure and the flow rate easily by using suitable formula. However, for complex network it is not handy to obtain such information with that simple way. For this reason, Graphic User Interface (GUI) program which can rapidly give required information should be necessary for microchip designers. In this paper, we present a GUI program developed in our laboratory and the simple theoretical formula used in the program. We applied our program to simple case and could get results compared well with other numerical results. Further, we applied our program to several complex cases and obtained reasonable results.
