A Comparative Study on the Lipid Components of Barley and Malt -I. Composition of Neutral Lipids- (보리와 맥아(麥芽)의 지방질(脂肪質) 성분(成分)에 관한 비교(比較) 연구 -제(第)1보(報) : 중성지질(中性脂質)의 조성(組成)-)

  • Neutral lipids of the total lipid extracted from 4 representative varieties of barley grown in Korea and their corresponding malt were studied. Total lipids of barley and malt were solvent extracted with chloroform:methanol:water (1.0 : 1.0 : 0.9, v/v), The total lipids were fractionated into neutral and polar lipids by silicic acid column chromatography, and neutral lipids fraction was separated by thin layer chromatography and quantitated by TLC scanner. The fatty acid compositions was determined by gas liquid chromatography. The average content of total lipid in the 4 barleys and their malts were 3.3 and 2.9%, and average of neutral lipids content in the barley and malt lipids were 73.8 and 68.5%, respectively. Among the neutral lipids, triglycerides and free fatty acids were the major components, and triglycerides content decreased and free fatty acids content increased during malting. Sterol esters, free sterols, 1,3-and 1,2-diglycerides were the minor components of the neutral lipids, and contents of those components showed increasing tendency during malting. The major fatty acid composition of the total lipids were linoleic, palmitic and oleic acids, and in general, the malts had lower amounts of unsautrated fatty acids and high amounts of saturated fatty acids. Fatty acid composition of neutral lipids was of almost the same pattern as that of the total lipids.

  • Polar lipids of the total lipid extracted from 4 representative varieties of barley grown in Korea and their corresponding malt were studied. The average content of glycolipids and phospholipids in barley were 8.9 and 17.3% and their average content of malt were 12.3 and 19.2%, respectively. Among the glycoliplds of the barley, digalactosyl diglycerides and monogalactosyl diglycerides were the major components, and the malts showed somewhat lower amounts of those components. Steryl glycosides and cerebrosides were the minor components of the glycolipids, and malts of the barley showed somewhat increased amounts of those components. Phosphatidyl cholines, lysophosphatidyl cholines, diphosphatidyl glycerols, and phosphatidyl ethanolamines were the major components of the phospholipids for the barley and represented 85 to 90% of the total phospholipids. The malts had lower amounts of phosphatidyl cholines and the lyso analog, and higher amounts of phosphatidyl ethnolamines and diphosphatidyl glycerols. The fatty acid composition in the glycolipids and phospholipids were similar to the pattern in those of the neutral lipids. But glycolipids and phospholipids fractions contained a higher percent of linoleic and palmitic acid than other lipid fractions, respectively.

  • The proximate compositions, fatty acids, and amino acids of mung bean(Phaseouls aureus) were determind: 1. The proximate compositions of mung bean were 24.80% crude protein, 4.75% crude fiber, 4.75% crude ash, 46.03% carbohydrate and 0.82% fat. 2. Saponification number, iodine number and non-saponifiable content of the lipids extracted from mung bean were 154.99, 117.05 and 14.83% respectively 3. The protein of mung bean was composed of glutamic acid (15.92%) and aspartic acid (12.09%) as major amino acids and considerable amounts of leucine (8.19%), arginine(7.31%) and pheylalanine (6.41%). The essential amino acid content including lysine(8.3%), threonine (3.5%) and tyrosine (2.83%) was higher than those of rice and barley which are deficient in those amino acids. 4. The lipids were composed of 35.5% linoleic acid, 15.5% linolenic acid and palmitic acid, 37% stearic acid, 5% oleic acid as major components, and 0.4% myristic acid, 1.0% arachidonic acid and 1.2% behenic acid as minor components. The saturated and unsaturated fatty acid ratio of oil extracted with di-ethyl ether from mung bean was $42{\sim}43/57{\sim}58%$.

Free Radical Scavenging Activities and Inhibitory Effect on Xanthine Oxidase by Acetone Extract from Buckwheat (수원 5호 메밀 채소 추출물의 유리라디칼 소거 및 Xanthine Oxidase 활성 저해)

  • To examine the characteristics of the antioxidative property of Buckwheat components, acetone extracts from a buckwheat, Suwon 5, was fractionated using five solvents. Hexane, ethylacetate, ether, butanol and water fractions were obtained. Butanol fraction showed the greatest electron donating ability and inhibitory effect on lipid peroxidation. It also showed the most excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. Spectrophotogram of butanol fraction was similar to that of rutin. Superoxide radical scavenging activity was related to the contents of rutin. Inhibitory effect of each fraction on xanthine oxidase was also measured. Butanol fraction had the strongest inhibitory effect on xanthine oxidase and $IC_{50}\;was\;3.1\;{\mu}g$. The inhibition type of butanol fraction on xanthine oxidase turned out to be a mixture of the uncompetitive and non-competitive modes.

Quality Characteristics of New Reddish Brown Color Rice Variety "Superhongmi" (슈퍼홍미 벼품종의 품질특성)

  • 'Superhongmi' is a new rice variety with reddish brown color developed from a crossing a high cyanidin 3-glucoside (C3G) content beeeds including Heugjinju and Suwon 425 and a big grain size breed such as Daeribbyeo. This variety heads on Sep. 5 and has 86.7 cm culm length. The fertility ratio of Superhongmi was about 81.7% and weight of 1,000-grain was about 26.8 g. The 1,000 grain weight of Superhongmi was heavier than that of Superjami. The total essential amino acid content of Superhongmi was lower than those of others. However, Superhongmi was high in tyrosine and arginine. Total polyphenol content of Superhongmi was 2 times higher than that of Heugjinju and flavonoid content was 8 times higher than that of Dongjinbyeo. In addition, hydroxy radical scavenging activity of Superhongmi was significantly higher than that of Dongjinbyeo. However, significant differences were not found in the newly breed colored rice. These results suggest that Superhongmi variety has very high value as a source of various functional food as well as stable food.

  • Chungkookjang, a traditional Korean fermented soybean food, was prepared by a commercial process. Cooked soybeans were fermented with Bacillus natto for 3 days and ripened with addition of 7% salt for 20 days. And the changes in the lipids during these chunqkookjang fermentation were studied. The total lipid content was decreased during chungkookjang preparation from 12.1% to 9.5%. Total lipid of cooked soybean consisted of 92.08% neutral lipids, 1.76% free fatty acids, 2.04% glycolipids and 4.12% phospholipids, respectively. During fermentation, as netural lipids were decreased, contents of free fatty acids and glycolipids were increased. The changes of phosphatidyl inositol and phosphatidyl choline in phospholipid fraction were observed and digalactosyl diglyceride in glycolipids fraction was significantly decreased during fermentation. Difercences were observed in the fatty acid compositions of glycolipids and phospholipids of cooked soybeans and chungkookjang. Oleic acid was the major fatty acyl moiety in neutral lipid and free fatty acid fractions, and palmitic acid was predominant in glycolipids and phospholipids. During fermentation, saturated fatty acyl moieties of glycolipids and phospholipids were increased.

Lipid Composition of Barley Flour Produced in Korea (한국산(韓國産) 보리가루의 지방질(脂肪質) 조성(組成))

  • Grains of naked barley (Baekdong cultivar) were polished, powdered and subjected to the successive extraction into free and bound liquid fractions. These were further fractionated into lipid classes and quantified by means of thin layer chromatography, column chromatography and gas-liquid chromatography. Contents of free and bound lipids in barley flour were 2.27% and 1.01%, which were decreased to 2.12% and 0.76%, respectively, after purification. Free and bound lipids were consisted of monoglycerides, diglycerides, triglycerides, free sterols, sterol esters, free fatty acids and polar lipids. Major constituents of free lipids were 56.2% triglycerides, 14.9% free fatty acids and 13.4% sterols while those of bound lipids were 73.8% polar lipids, 8.4% free fatty acids and 5.2% triglycerides. The content of non-polar lipids in free lipids was 93.6% as compared with 26.2% in bound lipids. However, phospholipids content in bound lipids was 55.5% as compared with 2.5% in free lipids, and glycolipids content in bound lipids was 19.4% as compared with 3.9% in free lipids. Major fatty acids in the free and bound lipid fractions were linoleic acid 52.1%, 54.8%, palmitic acid 24.8%, 30.0% and oleic acid 15.6%, 8.8%, respectively, showing similar patterns in both fractions. The amount of unsaturated fatty acids in free lipids was 72.8% as compared with 68.0% in bound lipids. In comparing the fatty acid composition of non-polar lipids, glycolipids and phospholipids, no difference was observed between free and bound lipid fractions while a slight difference was found among the lipid constituents.

  • The chemical composition of Undaria pinnatifida Laver (U.P. Laver) were determind. 1. Proximate compositions of U.P. Laver were 22.1% crude protein, 1.3% crude fat, 10.2% crude ash, 66.4% N.F.E, 3 0% crude fiber and 22.8% alginic acid. 2. U.P. Laver protein was chiefly composed of glutamic acid (3.24%) and aspartic acid (2.22%) and considerable quantities of leucine (1.98%), alanine (1.56%), valine (1.32%), lysine (1.22%) and phenylalanine (1.15%). 3. The major fatty acid contents of the lipid extracted from U.P. Laver were linolenic acid (45.2%), palmitic acid (26.14%) and linoleic acid (11.27%) and the minor fatty acid were myristic acid (8.41%), oleic acid (8.11%), and stearic acid (0.87%), respectively. The saturated and unsaturated fatty acid ratio of the oil extracted from U.P. Laver was 35.42/64.58 4. The vitamin contents of U. P. Laver were $28.1{\mu}g/g$ niacin, $25.0{\mu}g/g$ vit. E, $18.8{\mu}g/g$ vit. C. The minerals were composed 1.52% calcium, 0.32% phosphorus, 0.88% magnesium, 0.49% potassium and the others were small quantities.

Fractionation of Rice Bran Lipid and Storage Effects on Bran Lipid Composition (미강(米糠)의 지질성분(脂質成分) 및 저장(貯藏)중 지질특성(脂質特性) 변화(變化)에 관한 연구(硏究))

  • Lipids from rice bran (Indica type Milyang #23), both fresh and stored at $30^{\circ}C$ and 80% relative humidity for 5 weeks, were separated and analyzed for the determination and the storage effect on the bran lipid composition. Total lipids of fresh rice bran consisted of 89.9% neutral lipids, 8.0% glycolipids, 2.1% phospholipids and no significant changes of these fractions were noted during storage. Triglycerides(43.1%), diglycerides(13.8%) and hydrocarbon-esterified sterol(13.5%) among six fractions were considered as main components in neutral lipids. After storage triglycerides content significantly decreased as the free fatty acid increased in the neutral lipid fraction. Major components of the glycolipid fraction were acylsterolglycoside(43.1%) and sterolglycoside(30.3%). Phosphatidyl choline(39.8%), phosphatidyl serine(20.9%) and phosphatidyl ethanolamine(19.8%) were predominent in the phospholipid fraction. No significant changes of the composition were shown in fraction of the glycolipid or the phospholipid during the storage period. Major fatty acids of the total lipid fraction were oleic(44.3%), linoleic(32.5%) and palmitic acids(18.4%). The fatty acid compositions of the neutral lipid, the glycolipid and the phospholipid fractions were similar to the total lipid fraction. Small changes in fatty acid composition in each fraction were noted during the storage period. The acid value increased but iodine value decreased during the storage period. The values of peroxide and TBA increased gradually in the first three weeks, and then slowly decreased in the fourth and the fifth week of the storage.

Analysis of Fatty Acid Compositions and Biodiesel Properties of Seeds of Woody Oil Plants in Korea (국내 목본 유지식물 종자의 지방산 조성 및 바이오디젤 특성 분석)

  • In order to evaluate their potential as sources of biodiesel, oil content and fatty acid composition of seeds and fatty acid methyl ester (FAME) properties from seven woody oil plants in Korea were analysed. The oil content of seed of all woody plant species ranged from 15.1 (Ligustrum lucidum) to 70.3% (Camellia japonica) by dry weight. Fatty acid composition consisted mainly of oleic acid, linoleic acid, linolenic acid, palmitic acid and stearic acid, with oleic acid being the most abundant. The content of unsaturated fatty acids of all species was higher than saturated fatty acids. Oxidation stability of seed oils of all woody plants ranged from 2.25 to 8.62 hours/$110^{\circ}C$. Fatty acid methyl ester of Styrax japonica has been found to have the highest iodine value, indicating that unsaturated fatty acid content is higher than other seed oils. Cold filter plug point(CFPP) was varied over a wide range from $0^{\circ}C$ to $-13^{\circ}C$. The cold fluidity of FAME of Chionanthus retusa were excellent.
